Abstract

Dinding sel jamur Trichoderma tersusun atas senyawa kitin yang sangat kokoh dan resisten terhadap aktivitas enzim. Teknik dan metode spesifik diperlukan untuk mendegradasi dinding sel jamur. Penelitian ini bertujuan untuk menganalisis metode isolasi DNA terbaik dalam melisiskan dinding sel jamur dengan membandingkan beberapa metode isolasi. Metode isolasi yang digunakan adalah DNeasy Plant Mini Kit, kombinasi DNeasy Plant Mini Kit dan pemanasan, serta kombinasi DNeasy Plant Mini Kit dan cara fisik (menggerus sampel dengan menggunakan mortar dan pestle dalam nitrogen cair). Penelitian ini dilakukan pada bulan Maret sampai September 2018 yang bertempat di Laboratorium Mikrobiologi, Laboratorium Bioteknologi dan Genetika, serta Laboratorium Penelitian Terpadu, FMIPA, UNP. Data dianalisis secara kualitatif dengan elektroforesis gel agarose dan secara kuantitatif dengan menghitung nilai kemurnian dan konsentrasi DNA menggunakan Nanodrop. Hasil penelitian menunjukkan bahwa metode kombinasi DNeasy Plant Mini Kit dengan cara fisik memberikan hasil isolasi DNA dengan konsentrasi yang tertinggi yaitu 2,4 μg/ml dengan nilai kemurnian 1,857. Pada metode DNeasy Plant Mini Kit dan metode kombinasi DNeasy Plant Mini Kit dengan pemanasan, konsentrasi DNA yang diperoleh berturut-turut 1,211 µg/ml dan 0,933 µg/ml dengan nilai kemurnian 1,728 dan 1,708. Hasil uji elektroforesis menunjukkan adanya pita DNA berupa garis tipis pada sampel yang diisolasi dengan kombinasi metode kit dan cara fisik, sedangkan pada kedua sampel lainnya tidak ditemukan adanya pita DNA. Dapat disimpulkan metode isolasi DNA yang dapat dijadikan rujukan untuk mendegradasi dinding sel jamur Trichoderma adalah metode kombinasi kit dengan nitrogen cair dengan catatan dibutuhkan penambahan kadar isolat yang digunakan.Kata Kunci: Isolasi DNA, Dinding sel, Trichoderma

Abstract

Writing this paper aims to introduce the king's wooden tree (Koompassia excelsa) to the general public, as the background of this writing because North Sumatra especially in the Tangkahan Langkat forest has one tree with a height of almost 88 meters with very large roots and very hard wood, but not much people know it, this can happen because of lack of exposure and lack of publicity both directly and with the mass media. The method used in this study is the Exploration Method and directly record the sample during the trip. The area observed in the data collection process is 40 hectares divided into 2 regions, namely Region A (15 Hectares) and Region B (25 Hectares). The purpose of this study is to explain that the Koompasia excelsa species is an original species called Timber The king and saw its abundance in the Gunung Leuser Tangkahan Langkat Forest which is adjacent to the Natural Protection Forest of Bukit Barisan National Park in North Sumatra. Based on the results of research in the Gunung Leuser National Park Forest area, North Sumatra it can be concluded that the tualang tree can be found in lowland habitats with altitudes reaching 350 m above sea level and slope rates varying from 40-60. The dominance of species and associations is one of the factors that influence the adventure habitat, but that does not mean that other species do not influence the adventure habitat, such as altitude, soil type, and species of animals that have not been observed and analyzed further. In addition, it is necessary o carry out an inventory of adventurers to estimate the population in North Sumtatra, so that if there is a decline in population, conservation status can be improved.

Abstract

Hyperglicemia in DM Type will induce macrofag to produce cytokin proinflamation TNF-α. High levels of TNF-α in DM Type 2 can lead to more severe insulin resistance and become involved in endothelial dysfunction which lead to complications of the disease. This research aims to know differences between TNF-α level in DM type 2 controlled and uncontrolled. This research used obervational analizing method and cross sectional study comparative approach. Blood samples are grouped to 35 patient DM type 2 controlled and uncontrolled in the medicinal treatment in Padang State University polyclinic. The level of serum TNF-α will be examined in Biomedical Laboratory FKUA using ELISA method. TNF-α level in the diabetes type 2 controlled on average 7,55 ± 0,43 pg/ml whereas uncontrolled on average 149,28 ± 26,82 pg/ml. The Mann-Whitney test showed a p value < 0,001. There are significant differences in the levels of TNF-α in patients with DM type 2 controlled and uncontrolled. Key Words : Diabetes Mellitus type 2 controlled and uncontrolled, TNF-α

Abstract

Tanda dan simbol pencegahan bahaya adalah alat komunikasi keselamatan yang penting, mereka membantu menunjukkan berbagai bahaya yang ada di laboratorium. Pada saat yang sama, simbol pencegahan bahaya memperingatkan praktikan agar selalu waspada terhadap bahaya tersebut dengan memberikan informasi dan instruksi keselamatan yang dibutuhkan. Simbol pencegahan bahaya bisa mengkomunikasikan pesan dengan cepat dan mengatasi hambatan bahasa. Pada tahun 2015, perubahan besar terjadi pada label bahan kimia dan SDS secara global. Ada bahasa baru dari simbol pencegahan bahaya yang harus dipahami oleh para pengguna bahan kimia. Beberapa produsen bahan kimia telah mengeluarkan jenis simbol baru untuk pencegahan bahaya. Hal ini sebagai implikasi dari telah diadopsinya regulasi GHS (Globally Harmonized System) di beberapa negara Uni Eropa dan Amerika Serikat. Banyak orang yang bekerja dan mengajar dengan bahan kimia masih belum mengetahui secara rinci tentang apa yang telah berubah dan apa artinya bagi mereka.

Abstract

Jabon (Anthocephalus cadamba [Roxb] Miq.) is one type of tree that has a high prospect for industrial plantations and reforestation in Indonesia, due to its very fast growth and relatively easy silvicultural treatment. The growth of a plant is strongly influenced by photosynthesis. The process of photosynthesis can take place because of the presence of pigments called chlorophyll. The formation of chlorophyll is influenced by environmental factors. Therefore, studies have been carried out on the chlorophyll content of Jabon leaves in different locations. This research was conducted from March to May 2019. The samples came from the Sungai Nyalo, Pesisir Selatan, and Lubuk Alung, Padang Pariaman. While the chlorophyll content of Jabon leaves was tested at the Laboratory of Plant Physiology, Department of Biology, FMIPA UNP. This research is a descriptive study that was analyzed by the T test at a significant level of 5%. The results showed that there were differences in the chlorophyll content of Jabon leaves in the Sungai Nyalo, Pesisir Selatan with value 44,26 (mg/g) and Lubuk Alung, Padang Pariaman with value 19,45 (mg/g).

Abstract

 Abstract -- Endophytic bacteria are microorganisms that exist in the system of plant tissues such as leaves, twigs, roots and they can form colonies without causing damage to the plant. The endophytic bacteria may produce secondary metabolites in accordance. One potential medicinal plant has endophytic bacteria are plant greeting (Syzygium polyanthum Wight). Part of the plant Salam particularly frequently used is part of the leaf, which serves as an antibacterial and antifungal. Because the leaves contain compounds bay leaf tannins, flavonoids and essential oils. The porpuse of this research was to find of to obtain isolates of endophytic bacteria and determine the antimicrobial activity of bay leaf (Syzygium polyanthum Wight).This research is descriptive that has done on January - February 2015 of Microbiology Laboratory at Biology Departement FMIPA, UNP. The parameters observed in the determination of isolates is a form colonies, colonies edge, elevation colonies, colony color, and Gram staining of  leaves (Syzygium polyanthum Wight). The antimicrobial activity was observed with the formation zone of inhibition around the paper disc. Obtained were 11 isolates of endophytic bacteria from leaves, 5 isolates of endophytic bacteria have the ability to produce antimicrobial.Isolates had the highest antimicrobial activity that isolates DSA 1-1 with a 6.42 mm diameter inhibition zone. Keywords: antimicrobial, endophytic bacteria, bay leaf.

Abstract

Abstract. The objective of this research is to know the effect of giving of n-hexan extract of A. Paniculata leaf on the reproduction function on female mice and when hypodermic of how many dose of extract n-hexan most having an effect on the reproduction function on female mice. Extract gift conducted by inoculation. Mice or test animal obtained from Hall of Animal Research (BPH) of Regency Maros. Old age the mencit 10-12 week of weighing of mean body 21g, while test substance used by leaf of Andrographis paniculata obtained in regency of Bone of subdistrict of Sibulue of countryside of Pattiro Bajo. Parameter perceived that is s the number of implantation, number of life fetus, gestation loss, dead foetus, embryo resorbsi, corpus luteum, and death pascaimplantasi. This Research represent the research of experiment and desain research used that is Complete Random Device ( RAL) consisted of by three treatment group five times is restating so that obtained by fifteen combination. Result of research indicate that the gift of n-hexan extract of A. Paniculata leaf at dose level tested degrade the number of implantation, number of life fetus, improving number of gestation loss, and number of died fetus , also generate the effect fitotoksik of body weight of mice mains. From inferential research result that extract of n-hexan A. Paniculata leaf can effect on the reproduction function on female mice with the hypodermic time most having an in with day of pregnancy to-0 until pregnancy day to-4.

Abstract

This research was aims to determine the response of jabon seed germination  toward duration of combustion and light. This research was conducted from January to June 2018 at the Biology Laboratory and FMIPA UNP greenhouse. The design used was RAL with 2 factors. The first factor is the duration of combustion and the second factor is the light treatment. Based on the design obtained 10 treatment combinations and each treatment was repeated 3 times so that there are 30 units of experiment. Parameter observed is the number of seeds  germination. The results showed that light treatment, combustion duration treatment, and interaction between combustion duration and light give good effect on the number of seeds  germination. The seeds with the combustion treatment for 15 (A2) minutes had the highest number of sprouts that was 342.83 sprouts per 0.2 gram of jabon seeds. For light factor, many seeds germinate on the shelter treatment (B2) of 349.59 seeds per 0.2 gram of seed. While the combination of treatments showed that the best number of seeds germinated was in the combustion treatment 12.5 minutes with germination in the sheltered place (A1B2) that produced 564.67 sprouts per 0.2 gram of seed.

Abstract

Protease is an enzyme that can hydrolyze proteins into simpler compounds such as small peptides and amino acids. The enzymes produced by microorganisms can be isolated by separating cells by centrifugation. This study aims to determine the biological characteristics of the PrTK 02 bacterial isolate and the characteristics of the crude extract of the protease enzyme produced by the PrTK 02 bacterial isolate. The study was conducted by inoculating 1 ose PrTK 02 bacteria on the Nutrient Broth (NB) media. Pre-culture was carried out at 37 °C for 48 hours with an agitation speed of 120 rpm. Protease enzyme extraction was carried out by centrifugation of bacterial growth media at a speed of 3000 rpm for 15 minutes at 4 °C. Supernatant as a sample was tested for protease activity and protein content. The results of this study were obtained from the protease enzyme from bacterial isolate PrTK 02 marked by a clear zone in the medium of high school and the highest activity at the time of production 24 hours with the specific activity of the enzyme 1.2154 unit / mg protein. Biochemical and microscopic identification shows that the isolate of PrTk 02 bacteria is a class of Bacillus sp. PrTk 01 which is a Gram-positive bacteria that is found in tempeh.

Abstract

White rice is considered as a food have hyperglycemic, it can increase the risk of Diabetes Mellitus. Changes in blood hematocrit value is very risky for people with Diabetic. High hematocrit value increases the risk of hypertension in patients with Diabetic. Meanwhile, a low hematocrit values can lead to anemia in patients. Thus it made to reduce the risk of diabetic disease has to limit consumption of white rice. In fact, white rice glycemic response varies widely, influenced by processing, varieties, and chemical composition.West Sumatra white rice have high amylose deemed able to increase blood glucose slowly. The purpose of this research was to determine the effect of white rice varieties West Sumatra on blood hematocrit value mice (Mus musculus L. Swiss Webster) males. This research used Completely Randomized Design with 7 treatments and 4 replications. The treatment were 0,5 mL sugar solution as a control, and 0,5 mL of  rice flour Bakwan, Ciredek, Cantiak Manih, Randah Putiah, Mundam and Anak Daro. Blood hematocrit value mice (Mus musculus L. Swiss Webster) males measured on day 7th and 15th. Data analyzed by ANOVA and DNMRT at significance level of 5%. The results showed white rice varieties West Sumatra affect blood hematocrit values mice (Mus musculus L. Swiss Webster) males. White rice varieties resulted the highest hematocrit values found in male mice is rice Ciredek of Solok. Meanwhile, the normal hematocrit values found in male mice were given rice Mundam of Pariaman. Rice Mundam can be consumed by people with Diabetes because it provides stable results for hematocrit value. Keywords: Rice White, Blood Glucose, Mus musculus, Hematocrit
