Abstract

An experiment was carried out to analyze the application of laparoscopic technique for oocyte collection, in vitro embryo production and embryo transfer in sheep. The first experiment was conducted to observe effect of gonadotropin stimulation on follicle development and laparoscopic technique for oocytes aspiration. In the second experiment, effect of culture system on the embryo development in vitro was assessed and in the third experiment, the application of laparoscopic for embryo transfer has been conducted. The result showed that single dose of gonadotrophin was sufficient to support follicle development significantly and it could help follicle visualization. It also showed that laparoscopic ovum-pick up could be conducted weekly without any restriction The second series experiment showed CR1aa culture system was better than TCM 199 (29.90°/o vs 8.00%) and the changing of media was required to ensure better metabolism process for embryos. The third experiment revealed that embryo transfer could be conducted with an aid from laparoscope. In conclusion, single dose PMSG stimulation is sufficient to support follicle development for /aparoscopic ovum-pick up, the culture media changing affects the successful rate of in vitro embryo production (8% vs 25.66%) and the laparoscopy technique can be used safely for embryo transfer on sheep.Keyword: laparoscopic, oocyte, embryo transfer, sheep

Abstract

The objective of this research is to study the scaling up of bionsecticide production from Bacillus thuringiensisvar. israelensisusing onggok (a cassava by-product) as a carbon source. The insecticide produced was used to eradicate Aedes aegypti larvae. The product was a crystal protein produced during bacterial sporulation. Scaling up from laboratory to pilot plant scale was done using two methods, i.e. constant agitation power per unit volume (Pg/V) and constant oxygen transfer coefficient (kLa). The results showed that yield of product per substrate (Yp/s) of Pg/V based product with the value of 3.52 ± 0.02 spora per gram substrates was higher than Yp/s of  kLa based product with the value of 2.96 spora per gram substrate.  Logarithmic value of viable spore count (log of VSC) was also higher, i.e. 7.23 ± 0.30 for Pg/V based product as compared to 7.17 ± 0.20 for kLa based product. Substrate efficiency was also higher in Pg/V based (92.47%) than kLa based (64.87%). LC50 of Pg/V based product was lower (0.49 μg/ml) meaning that it was more toxic than kLa based product (0.62 μg/ml). Amino acid content of Pg/V based product was also higher than kLa based product. Constant Pg/V method was suggested as a based on the scaling up of bioinsecticide production of B. thuringiensis israe/ensison industrial scale. Abstract

The research was conducted to examine the seedling of mangium (Acacia mangium and sengon (Paraserianthes falcataria) on compacted soil. Soil was compacted with a proctor test at the different level of bulk density (0.9;1.0; 1.1; 1.2; 1.3 g/cm3 ) with 10 replications. The compacted soil was put into the plastic pot. After 6 month growth in pot the seedling growth response was measured.The result showed that the root penetration and seedling height linearly decrease with the increase of bulk density. The root of seedling of mangium (Acacia mangium and sengon (Paraserianthes fa/cataria) could not penetrate deeply at the hardest soil (1.3 g/cm3 ). Roots needs space to grow. The space was occupied by soil particles and soil pores. Root may influence chemical and physical properties of soil. In related to the compacted soil the root penetration is the important variable which is affected by the soil hardness. Under the condition of decrease penetration, seedling can not grew well in lack of nutrient to be absorbed by the root. On the other hand the bulk density 1.3 g/cm3 showed difference for seedling height response. However, the growth response was significantly reduced at the level of 1.3 g/cm3. The bulk density 1,3 g/cm3 was the critical limit of soil hardness for the seedling of Mangium and Sengon growth.

Abstract

Accurate and up-to-date information of marine resources is needed for optimum resource utilization. If the required information cannot be obtained with traditional field surveys, remote sensing as a tool for providing fast, efficient, and accurate information is used to detect the changes. The study aims to use remote sensing data for assessing distribustion and condition of marine resources in Nias water's before and after tsunami. Marine resources pararameter derived remote sensing data are sea surface temperature (SST), suspended solid matter, chlorphyll-a concentration, coral reef and mangrove distributions. Mapping and differences in coral reef and mangrove distribution were evaluated using two scans of Landsat image in 1989 and 2005. Sea surface temperature was derived from NOAA-AVHRR data, and chlorophyll-a concentration was derived from SeaWiFS data. The study showed that there were no significant changes of coral reef and mangrove distribution, SST, chlorophyll-a concentration after and before tsunami. Spectral analysis of SST and chlorophyll-a showed that the variability of the parameters was affected by monsoon system and global climate change, namely El Nino Southern Oscillation and Indian Ocean Dipole.Keywords: remote sensing, satellite, Nias, tsunami, chorophyll-a, coral reef, mangrove

Abstract

In the last five years, several earthquakes struck several places in Indonesia and thousands of people died caused of their masonry house. Therefore, the idea of bamboo utilization in the form of structural sandwich panel components for anti-seismic pre-fabrication house is one of appropriate way out. This research describes the performance and behavior of engineered structure wooden-bamboo sandwich panel in full sized test. The wall frame being use were meranti wood, this frame attached by plywood as face and back, then 5 em bamboo cutting as core; the fastener used in this study were isocyanate resin and nails. This study carried out by experimental data of full scale structural tests on shear wall (ISO 22452). These result showed that the seismic reliability of the wall frame system made of bamboo sandwich panel was appropriate and will be useful for prefabrication houses as anti-seismic and environment friendly residential building.

Abstract

Rainfall zoning analysis with fuzzy clustering method has been performed at the centre of paddy area in the northern coast of Banten Province and West Java Province. Rainfall data recorded in the 1980-2006 period from 62 rainfall stations in the northern coast of Banten Province and from 75 rainfall stations at Karawang and Subang in the northern coast of West Java Province have been used in this analysis. For the first analysis a calculation of arithmetic mean values representing EI-Nino, La-Nina and Normal condition has been performed. Next, a fuzzy clustering analysis is applied to these mean values. The clustering analysis consists of two steps. First, a symmetric and reflective compatibility relation matrix describing a distance function between rainfall stations is calculated. Second, a fuzzy equivalency relationship i.e. a transitive approach of fuzzy compatibility matrices is determined. The results of analysis indicate a difference in the equivalency level among the stations under the EI-Nino, La-Nina and Normal conditions in the northern coast of Banten Province and West Java Province. Based on the 75°/o equivalency level, in the northern coast of Banten area can be grouped into four rainfall zones under EI-Nino condition, two zones under La-Nina condition and three zones under Normal condition. On the other hand, in the northern coast of West Java area can be grouped into three zones under EINino condition, two zones under La-Nina condition, and four zones under Normal condition.Keywords: Arithmetic means values, EI-Nino, La-Nina, Fuzzy clustering, Rainfall zoning

Abstract

Paederus fuscipes is an important predators in soybean fields, including toward Helicoverpa armigera. Predation evaluation using a cage are very common, giving significant suppressed the exposed pest population. However, predation evaluation in open space is rarely conducted. This study was to investigate the dispersal capability of P. fuscipes, and to evaluate the effectiveness of the beetle release in the predation on H. Armigera eggs and larvae. Observations were made in a soybean field in Mekarwangi Village, Cianjur, during soybean planting season in 2005 and 2006. In the dispersal study, all beetles were collected from the field and tagged on the elitra. As many as 1187 tagged beetle were released. The dispersal was observed in the area within a radius of 40 m from the release point. In the predation study, eggs and larvae of H.armigera from laboratory rearing were used as the prey. Soybean plots with exposed prey were treated by releasing the beetle with different densities, i. e., 100-400 beetles, and a control. The released beetles were from laboratory rearing and beetles were collected from the field. The study indicated that the tagged beetles that were released to the soybean field actively move but their dispersal were relatively slow. Until 5 days after release, most beetles were remain in the area surrounding release point, within a radius of 5 m. Release of predators to the soybean field, suppressed H. armigera egg and larva populations for both vegetative and generative stages, but were not significantly different between treatment and control plots, except for the egg population on vegetative stage. It indicates that population of the natural predator in soybean field were very complex and potential to be used for controling pest populations. Abstract

Snake fruit is an exotic fruit from Indonesia, and nowadays has increasing in demand. There is opportunity to market minimally processed snake fruit, but this product becomes very highly perishable because of increasing in respiration rate. The objective of this research was to determine edible coating formulation in combination with modified atmosphere packaging, in order to maintain quality and to prolong shelflife of minimally processed snakefruit. The research was started by preparation of salak fruit, edible coating, measurement of respiration rate, determination of optimum atmosphere composition and critical parameter, and storage in modified atmosphere packaging. First, best atmosphere composition for minimally prcessed snakefruit was determined, then storage of minimally processed snakefruit was conducted after selection of the appropriate plastic film. Analysis was conducted on quality and organoleptic characteristics. Optimum gas composition storage for minimally processed snake fruit is 4±1%0[2] and 14±2% C0[2]. Minimally processed snake fruit with edible coatings made from pectin and chitozan combined with modified atmosphere packaging can extend shelflife and maintained quality until eight days, while without edible coating the minimally-processed snake fruit in with modified atmosphere packaging can extend until 6 days only.

Abstract

Efficacy and quality of medicinal plant extracts depend on chemical composition therein. Therefore, to ensure its efficacy orquality, the chemical composition of extracts have to be analyzed based on its chemical marker or its chemical pattern. This study tried to develop a method for extracts quality assay based on infrared spectrum in fingerprint region and extracts inhibition activity to xanthine oxidase. Five extracts from kunyit (Curcuma domestica), temulawak ( C xanthorriza), jahe (Zingiber officina/e), temukunci (Boesenbergia pandurata), and cabe jawa (Piper retrofractum) were tested to inhibit xanthine oxidase activity. The bioactive data of extracts and its infrared spectrum was analyzed using PCA and PLS-DA (partial least squarediscriminant analysis). Almost all extracts that were tested showed inhibition activity to xanthine oxidase activity, except the extract of jahe. The result showed that PCA can group all of extract into each of their region. Meanwhile, PLS-DA has shown to be a good prediction model for classifying extracts based on fingerprint region of its mR Spectrum and its biological activity. Abstract

Testicular cell transplantation technology can be used in fish seed production engineering. In this study, optimization of transplantation using donor cells from young gouramy and triploid tilapia (3N) as recipient. Triploid tilapia is produced using heat shock method. The testes of male gouramy (body weight of 400- 850 g) was dissociated using 0.5% trypsin. Dissociated testicular cells was injected into the peritoneal cavity of tilapia larvae. Analysis of donor cell colonization was carried out using PCR method with DNA template that had been extracted from the gonad of 2-month-old tilapia. PCR was performed using specific primers for the growth hormone gene and (3-actin as an internal control of DNA loading. The results of nucleoli preparation showed that the success of triploidyzation was 88.5%. The gonad size of diploid (2N) and 3N recipient were relatively similar, while in not transplanted 3N tilapia was rudimentary. PCR results showed that the transplanted 3N tilapia has a DNA band of the same size with gouramy, while in control was not. This indicated that donor cells have been colonized in the gonads of recipient. The donor cell colonization in recipient 3N (78%) was higher than that of 2N (50%). Further research is required to determine the ability of donor cells differentiate into sperm and eggs in recipient gonad.
