Abstract

The Gracilaria genus is a potential source of natural and environmentally-friendly alternatives in improving the survival and growth of shrimp. This study aims to identification immunostimulant molecules extract G. verrucosa and evaluate the utilization of G verrucosa extract as an immunostimulant in improving survival and growth of L. vannamei. Seaweed extraction used ethyl acetate then formulated in the diets. The immunostimulant molecule in the G. verrucosa was analysis. The shrimp were fed a test diet containing extract G. verrucosa at a dose of 2 g kg-1 or extract G. verrucosa-free control diets for 42 days. Shrimps were fed diets containing extract with a specific duration. The observation on the survival and growth of L. vannamei was performed after maintenance at the Laboratory for six weeks. Following, diets containing extract was tested in the field (pond shrimp farm) at the same dose of extract for 58 days. Shrimp was feed diets containing extract once a week, once in the early culture, and diet control, then the survival and growth shrimp were analysis. Concentrations of sulfates and carbohydrates in G. verrucosa ethyl acetate-extract were 24.21% and 13.41%, and crude protein 3.64%. GC-MS pyrolysis results show that G. verrucosa polysaccharide is similar to immunostimulant molecules. The survival shrimp gave diets containing G. verrucosa extract formulation was higher than that of shrimps fed controls diet. Abstract

Lais fish of Ompok hypophthalmus is one of high economic fish in Kampar river. It should be protected fromdecreasing of it population estimated due to decreasing of habitat quality and increasing of exploitation. Theobjectives of the research are to study reproduction biology of lais fish as the basic data for conservation. Thisresearch was conducted from January 2007 to January 2008. The results of O. hypophthalmus reproductionaspect show that the smallest female of maturity is 22,9 cm and male is 22,6 cm; the spawning season onSeptember to November; O. hypophthalmus is more appropriate spawning location to oxbow lake that closerelation with tributary; the spawning pattern indicated total spawner fish; the fecundity ranges from 3111 to 11164eggs and the egg diameter ranges 0,41-1,13 mm.

Abstract

This study aims to develop a strategy for the sustainability of shrimp aquaculture using an ecosystem approach or EAA in Pinrang Regency, South Sulawesi. A series of analyzes were carried out, namely the environmental carrying capacity analysis of aquaculture using pond environmental feasibility standards, analysis of critical factors for the sustainability of aquaculture using multidimensional scaling analysis, analysis of the sustainability status of aquaculture using pairwise comparison analysis and analysis of shrimp aquaculture management strategies based on EAA. using hierarchy process analysis. The results showed that the following strategies were needed: a) accelerating spatial planning and implementing programs in accordance with the directions for spatial use and control; b) institutional strengthening of capital cultivators in order to complement and improve facilities and infrastructure in accordance with the SOP; and c) increasing the level of education and providing a social and economic security system for members of the shrimp farming community

Abstract

Molecular marker is useful in the development of testicular cells transplantation for detecting donor-derived germ cells in the recipient gonad. In this study, a giant gourami (Osphronemus goramy) vasa-like gene (GgVLG) was cloned and characterized for use as a molecular marker for germ cells in this species. Nucleotide sequence analysis revealed that GgVLG comprises 2,340 bps with an open reading frame of 1,962 bps encoding 653 amino acids. The deduced amino acid sequence contained 17 arginine-glycine or arginine-glycine-glycine motifs and eight conserved motifs belonging to the DEAD-box protein family. The GgVLG sequence showed high similarity to Drosophila vasa, common carp vasa homolog and tilapia vasa homolog for 66.2, 85.9, and 90.7%, respectively. In adult tissues, the GgVLG transcripts were specifically detected in ovary and testis. In situ hybridization analysis showed that GgVLG mRNA was detected in oocytes of the ovary and spermatogonia of the testis. There was no signal detected in the spermatocytes, spermatids and other gonadal somatic cells. Thus, consensus sequences, specific localization of GgVLG mRNA in the germ cells, amino acid sequence similarity and phylogenic analysis all suggest that GgVLG is the giant gourami vasa-like gene. Further, GgVLG can be used as a molecular marker for giant gourami germ cells.

Abstract

Testicular cell transplantation technology can be used in fish seed production engineering. In this study, optimization of transplantation using donor cells from young gouramy and triploid tilapia (3N) as recipient. Triploid tilapia is produced using heat shock method. The testes of male gouramy (body weight of 400- 850 g) was dissociated using 0.5% trypsin. Dissociated testicular cells was injected into the peritoneal cavity of tilapia larvae. Analysis of donor cell colonization was carried out using PCR method with DNA template that had been extracted from the gonad of 2-month-old tilapia. PCR was performed using specific primers for the growth hormone gene and (3-actin as an internal control of DNA loading. The results of nucleoli preparation showed that the success of triploidyzation was 88.5%. The gonad size of diploid (2N) and 3N recipient were relatively similar, while in not transplanted 3N tilapia was rudimentary. PCR results showed that the transplanted 3N tilapia has a DNA band of the same size with gouramy, while in control was not. This indicated that donor cells have been colonized in the gonads of recipient. The donor cell colonization in recipient 3N (78%) was higher than that of 2N (50%). Further research is required to determine the ability of donor cells differentiate into sperm and eggs in recipient gonad.

Abstract

The utility of cytochrome b gene as a molecular marker to obtain genetic diversity and phylogenetic relationship among Kryptopterus spp. consist of Kryptopterus limpok and Kryptopterus apogon from Riau Province has been studied. The universal primers of (F) L14841 and (R) H15149 were used to amplify the cytochrome b gene. The results of multiple allignment were 159 nt (coding 53 amino acids). Kryptopterus limpok from Kampar and Indragiri River form a phylogeny cluster at 80% bootstrap value. Kryptopterus apogon from Kampar and Indragiri River form a phylogeny cluster at 99% bootstrap value.Keywords : cytochrome b, Kryptopterus limpok, Kryptoperus apogon.

Abstract

Tujuan penelitian ini adalah untuk menjelaskan pengaruh pemberian ekstrak hipofisis ikan mas terhadap pematangan gonad ikan sidat tropis betina (Anguilla bicolor bicolor). Ikan sidat dengan bobot 600 gram telah digunakan sebagai materi biologis. Ekstrak hipofisis ikan mas dengan tingkat 0, ¼ dan ½ dosis dan tiga tipe frekuensi penyuntikan yakni setiap 1, 2 dan 3 minggu dijadikan sebagai perlakuan. Parameter yang diamati adalah Gonado somatic index (GSI), Hepatosomatic index (HSI), diameter folikel, kadar lemak dalam folikel, serta kadar glikokonjugat dalam folikel dan hati. Hasil percobaan menunjukkan bahwa penyuntikan ekstrak hipofisis ikan mas dengan ½ dosis yang dilakukan setiap minggu selama 10 minggu memberikan pengaruh yang terbaik.Kata kunci: gonad, sidat, Anguilla bicolor bicolor.

Abstract

Penelitian ini bertujuan untuk mengetahui dosis optimal Inhibitor Aromatase (IA) pada perkembangan oosit ikan mas koki. Dosis yang digunakan adalah k = kontrol (disuntik NaCl fisiologis), Pl = 2.5 mg/kg berat tubuh (b.t), P2 = 7.5 mg/kg b.t., dan P3 = 12.5 mg/kg b.t. Untuk mengetahui pengaruh IA terhadap perkembangan gonad diukur kandungan hormon estradiol-l7β dalam plasma darah dan level protein gonad mulai dari awal perlakukan kemudian setiap tujuh hari sekali, yaitu hari ke-7, ke-14, dan ke-21. Hasil penelitian menunjukkan bahwa, pada hari ke-7 hasil semua perlakuan P1, P2, P3 menunjukkan penurunan hormon estradiol- 17β diikuti penurunan level protein gonad, hasil analisis pengamatan histologi menunjukkan terjadi atresi pada sel gonad. Pada hari ke-14 kadar hormon estradiol-17β perlakuan menunjukkan terjadi peningkatan diikuti peningkatan level protein gonad. Pada hari ke-21 kadar hormon estradiol-17β perlakuan sudah sama dengan kadar hormon kontrol namun level protein gonad masih dibawah level protein gonad kontrol.Kata kunci: estradiol 17ß, perkembangan oosit, level protein gonad.

Abstract

The aim of the present study was to investigate the growth performance of Nile tilapia immersed in 17α-methyltestosterone (MT) and recombinant giant grouper growth hormone (rElGH), and fed a rElGH-enriched diet. A total of 200 tilapia larvae aged 10 days and 14 days post hatching was immersed in 1 liter of 30 g/L saline water for 3 minutes, then in freshwater containing either MT, MT+rElGH, or rElGH for 4 hours. The MT dose was 2.0 mg/L and rElGH was 2.5 mg/L. As control was treatment without MT and rElGH. Fish were maintained in 250-L aquariums for 6 weeks and then moved to net cages for 8 weeks of rearing. The rElGH-enriched diet was fed to the 6-week-old fish for a month, and then fed a non-enriched diet. Each treatment was triplicates. The result showed that the daily growth rate (DGR), biomass gain (BM), survival (SR) and feed (artemia nauplii) consumption during maintenance in the aquariums in the rElGH and MT+rElGH treatments were higher (P<0.05) than in the control. The higher DGR and BM at net cage rearing and lower feed conversion ratio (P<0.05) were also found in MT and MT+rElGH treatments. None of the treatments showed any difference in survival (P>0.05). Profits estimation in the MT and MT+rElGH treatments were 63.38% and 57.91% higher than the control. The present study concluded that higher Nile tilapia farming performance could be obtanied by MT immersion in the larval phase and feeding juvenile on the rElGH-enriched diet.

Abstract

Synodontis eupterus male represents only 5-10 % of farmer total synodontis production, which limits male availability for reproduction purpose. A method widely used in overcoming the low male production was masculinization through synthetic hormones, which was restricted due to its adverse impact on the environment. Natural ingredients from plant were demonstrated in various studies to substitute the 17α-methyltestosteron hormone on masculinization. This research aimed at evaluating the effects of Javanese long pepper extract (JLP) through immersion on synodontis fish larvae. The research was conducted using factorial design with two faktor (4x2) consisted of eight treatments: 17α-methyltestosteron 2 mg L-1 (MT) immersion for 5 and 10 hours, JLP dose 0.125 mg L-1 (P1) immersion for 5 and 10 hours and dose 0.25 mg L-1 (P2) immersion for 5 and 10 hours compared to the control treatment without immersion (P0). A hundred of synodontis larvae of 10 days old after hatching each replication were used in the immersion treatments. The results showed that JLP treatments produced 25-40 % of male synodontis age four months, 1-2 % females and above 60 % intersex. While at age five months, the percentage of intersex fish decreased to 20-40 %, the female fish increased to  60-62 %, and male fish ranged 20-35 % in JLP compared to the control P0 (15 %). The dose of JLP 0.25 mg L-1 increased mortality of about 14-54 %.
