Abstract

ABSTRACTHybridization program was started in PKBT IPB Bogor in 2003, entangles of 12 parental cultivars, consisting of five type Smooth Cayenne cultivars and seven type Queen cultivars. The cross yielded 195 genotypes with various different character combinations. The result of cluster analysis based on morphological characters showed that there were 33 groups of hybrid at the degree of genetic similarity of 50%. The result of principal component analysis indicated that, between yield component characters and result most importantly, were fruit weight, fruit diameter and fruit length which were main supporting character of variance in hybrid result of the crosses. Descriptive results of fruit yield and quality characters showed three to five classes with the highest number of individuals around the mean value for each character. Fruit weight, crown weight, fruit length, fruit diameter, flesh thickness, core diameter, total soluble solid (TSS), total acid, vitamin C, pH, plant height, and peduncle length, were characters controlled by nuclear genes.Key words : hybridization, variability, Smooth Cayenne, Queen, genotype

Abstract

ABSTRACTThe objective of this research was to determine the effect of coating materials and BAP concentration on inhibition of ripening process of mangosteen. The experiment used factorial completely randomized design with two factors, and three replication s. Coating materials as the first factor consisted of control (without coating) , bee wax 6%, and chitosan 2%. The second factor was concentration of Benzil Amino Purine (BAP), with 0, 5, 10, 15, and 20 ppm. Non destructive observations were weight loss, diameter decrease, peel and calyx color development. While, destructive observations were fruit hardness, total soluble solid (TSS), total titratable acidity (TTA) and opened ability. The result showed that bee wax was effective to inhibit weight loss. Interaction of bee wax and BAP 20 ppm inhibited peel and calyx color changes during storage.Key word: coating material, cytokinin, benzil amino purine, calyx, storage

Abstract

ABSTRACTMolecular and biochemical characteristics of Fusarium oxysporum f.sp. cubense (Foc) were detected. Six of Indonesian Foc isolates were artificially inoculated on “Ambon Kuning” banana. DNA of one-week culture isolates was extracted by three methods prior to PCR assay using Foc TR4 (tropical race 4) specific primer. Activity of extracellular enzyme was determined with reduction sugar, agar diffusion and SDS-PAGE assays. Statistical analysis revealed that all isolates insignificantly caused Fusarium wilt symptoms on tested banana with disease severity index ranging from 3 to 3.6. Maximum DNA concentration was obtained by CTAB method (766.25 µg mL-1), followed by SDS and alkaline l ysis methods, i.e. 553.75 and 211.25 µg mL-1, respectively. PCR analysis showed that Bnt2 and Kjg1 isolates positively reacted to TR4 of Foc primer (DNA size of 1400 bp approximately). Reduction sugar and agar diffusion assays demonstrated that Kjg1 isolate significantly produced more extracellular enzyme, with 6.53 × 10-2mg mL-1 in conce ntration and 20 mm in halo diameter. Meanwhile, SDS-PAGE assay viewed diverse bands of tested fungi (20.6 to 80 kDa), representing four extracellular enzymes. Positive PCR results highlighted the presence of Foc TR4 infecting banana in Indonesia. Various activities of extracellular enzymes did not influence the pathogenicity of Foc.Key words: pathogenicity, DNA concentration, extracellular enzyme

Abstract

ABSTRACTThe objective of this research was to study the differences effects of interstock on scion growth, flowering and fruiting of ‘Nambangan’ pummelo (Citrus grandis (L.) Osbeck). The research was conducted from January to December 2005 in a green house of Cikabayan Research Station IPB, Dramaga. Th e research was arranged in randomized completely block design with singe factor i. e. kind of intertock consisted of Flying Dragon, Troyer, Citrumelo, and Rangpur Lime. All interstocks were grafted on Javanche Citroen rootstock. The result showed that the growth, flowering and fruiting of ‘Nambangan’ pummelo was significantly affected by kind of interstock. Citrumelo interstock tended to induce more vegetatif with low generatif growth of ‘Nambangan’ scion. Troyer, Rangpur Lime and Flying Dragon resulted in moderate growth of ‘Nambangan’ scion, however only Flying Dragon induced ‘Nambangan’ scion to produce more flower and fruit as compared with other interstocks. There was no significant different in carbohydrate content in leaves and C/N ratio among scion with four interstock studied.Key words: interstock, Nambangan, pummel o, flowering

Abstract

ABSTRACTThis research was about in vitro propagation of Pitcher Plant Nepenthes mirabilis. The aims of this research were to determine: 1) the influence of kind and concentration of in vitro medium and also plant growth regulator on germination of Nepenthes mirabilis, 2)the effect of BAP and NAA on shoot multiplication of N. mirabilis. This research consisted of three experiments and all experiments used Completely Randomized Design. On the first experiment MS (Murashige and Skoog) and KC (Knudson C) were used, i.e. full (1), three-fourth (3/4), one-half (1/2) and one-fourth (1/4) Salt Concentration. On the second experiment 10 composition media were used, i.e. ½ MS, ½ MS+TDZ 0.01 ppm, ½ MS+IAA 0.1 ppm, ½ MS+GA3 10 ppm, ½ MS+150 ml coconut water, 1/4KC, 1/4 KC+TDZ 0.01 ppm, ¼KC+IAA 0.1 ppm, 1/4KC+GA3 10 ppm and 1/4KC+ 150 ml coconut water. On the third experiment BAP with 0, 0.5, 1, 2 ppm and NAA 0, 0.1, 0.2, 0.5 ppm were used as factor for shoot multiplication. The result showed that KC medium was the best medium for germination percentage of N. mirabilis (64%). MS or KC medium with ½ or ¼ salt concentration was the suitable for germinating time of N. mirabilis (average 39 days after showing). Adding of TDZ, IAA, and GA3 significantly increased germination percentage (70 – 90%) and decreased germinating time (27 – 38 days after showing). Culture growth on medium with BAP 0 - 1 ppm was the best medium for shoot and leave induction. The best medium for root growth of N. mirabilis was MS or KC medium without NAA.Key words: Nepenthes mirabilis, MS, KC, TDZ, IAA, GA3, BAP, NAA, in vitro propagation, medium
