Abstract

The Effect of Giberelin Acid (GA3) Application to Seed Yield of Hybrid Rice. Seed yield of rice hybrid on three line system has weakness in panicle exertion and outcrossing that cause low yield.  GA3 has been proved to increase the seed set through increasing panicle exertion and improved outcrossing. The purpose of this study was to determine the effect of GA3 application to increase seed yield of hybrid rice (F1). The study was conducted in Singamerta Experimental Farm, Banten Assesment Institute for Agricultural Technology from May to October 2013. The experiment was arranged in a split plot designed with four replications. The main plot were parental lines (cytoplasmic male sterility/CMS and restorer/R) of hybrid i.e. HIPA 8 (A1 and BP51-1), HIPA 6 (A2 and B8094), HIPA Jatim 3 (A6 and PK88) and HIPA 14 SBU (A7 and BH33d-Mr-57-1-2-2). Sub plot were the  frequency of GA3 treatments i.e. control (W0), two (W1), and three (W3) times application of GA3. GA3 dosage was 200 ppm, the best dose from the previous studies. The results showed that the GA3 applications increased plant height, stigma exertion, panicle exertion, duration of floret opening and panicle length. Two times application (W1) of  GA3  gave the best results as indicated by the highest seed yield (1429 kg/ha) that was better than three times application of GA3 that gave seed yield 1215 kg/ha and control with yield of 703 kg/ha.Keywords : GA3, hybrid rice, seed yieldABSTRAKHasil benih padi hibrida sistem tiga galur memiliki kelemahan, yaitu rendahnya eksersi malai dan tingkat penyerbukan silang,sehingga hasilnya rendah. Percobaan terdahulu menunjukkan bahwa aplikasi GA3 dapat meningkatkan hasil melalui peningkatan eksersi malai dan penyerbukan silang. Penelitian ini bertujuan untuk mengetahui pengaruh frekuensi aplikasi GA3 terhadap peningkatan hasil benih padi hibrida (F1). Penelitian dilakukan di Kebun Percobaan Singamerta, Balai Pengkajian Teknologi Pertanian (BPTP) Banten pada Mei sampai Oktober 2013. Penelitian menggunakan rancangan petak terbagi dengan empat ulangan. Petak utama yaitu tetua padi hibrida (galur mandul jantan/GMJ dan restorer) yaitu HIPA 8 (A1 dan  BP51-1), HIPA 6 (A2 dan B8094), HIPA Jatim 3 (A6 dan PK88) dan HIPA 14 SBU (A7 dan BH33d-Mr-57-1-2-2). Anak petak ialah tiga taraf frekuensi penyemprotan GA3 terdiri atas : kontrol (W0), dua kali aplikasi (W1) dan tiga kali aplikasi (W2). Dosis GA3 yang digunakan adalah 200 ppm yang merupakan konsentrasi terbaik dari hasil penelitian terdahulu. Hasil penelitian menunjukkan bahwa aplikasi GA3 mampu meningkatkan tinggi tanaman, eksersi malai, eksersi stigma dan durasi bunga membuka dibandingkan kontrol. Hasil yang terbaik dicapai pada perlakuan GA3 dengan dua kali aplikasi (W1). Hasil benih  pada perlakuan W1 mencapai 1429 kg/ha, lebih tinggi dibandingkan dengan aplikasi tiga kali GA3 (1215 kg/ha) dan kontrol (703 kg/ha).Kata kunci : GA3, padi hibrida, hasil benih

Abstract

Buah jeruk JC harus dipetik pada saat masak fisiologis untuk mendapatkan benih bermutu tinggi. Pada umumnya benih masak fisiologis saat berat kering benih optimum dan vigor optimum. Sementara itu benih JC bersifat poliembrioni, diduga dapat memengaruhi mutu benih. Penelitian ini bertujuan mendapatkan informasi tentang karakter poliembrioni, menentukan tingkat kemasakan fisiologi benih dan mengetahui pengaruh tingkat kemasakan benih terhadap multiple seedling dan off type pada benih JC. Penelitian dilaksanakan di Laboratorium Terpadu, Nurseri, dan Kebun Percobaan Balai Penelitian Tanaman Jeruk dan Buah Subtropika, pada Bulan Juli-Oktober 2012. Analisis deskriptif digunakan untuk mengetahui karakter poliembrioni benih JC. Percobaan di nurseri menggunakan rancangan acak kelompok lengkap satu faktor tingkat kemasakan buah dengan tiga ulangan. Pengamatan terhadap beberapa variabel dilakukan untuk mengetahui mutu fisik, fisiologis, genetis, dan jumlah semaian. Hasil penelitian menunjukkan bahwa benih JC mempunyai 1–6 embrio yang dapat tumbuh menjadi 1–4 semaian. Berat kering benih JC tidak dipengaruhi secara nyata oleh tingkat kemasakan buah karena adanya keragaman tingkat kemasakan embrio di dalam benih. Tingkat kemasakan fisiologis benih JC dapat ditandai dengan indeks vigor, kecepatan tumbuh, dan daya berkecambah optimum yang diperoleh pada saat buah berwarna kuning lebih dari 90% merata dengan karakter warna kulit benih krem kecoklatan dan embrio  dominan krem. Tingkat kemasakan buah tidak berpengaruh nyata terhadap persentase multiple seedling dan persentase semaianoff type. Penelitian lebih lanjut diperlukan untuk mendapatkan multiple seedling yang banyak dan semaian off type yang sedikit.

Abstract

DNA finger printing of hybrid rice variety and their parental lines are important molecular traits as a tool in plant variety protection, DUS test, and seed purity assessment. Molecular markers including microsatellite (SSR) markers have been used in varietal assessment. This study was aimed to identify specific SSR markers that can be used to distinguish parent lines of hybrid rice and to assess seed purity. The experiment was conducted in 2011 at Biology Molecular Laboratory of ICABIOGRAD, Bogor. Sixteen SSR markers were used to identify parental lines of five hybrid varieties of rice. Two hybrid varieties (F1), Hipa 6 and Hipa 7, were used in the seed purity assessment. Seven markers were polymorphic, and six of them (RM206, RM263, RM276, RM346, RM335, RM570) were capable of distinguishing parental lines of the five hybrid varieties. RM 346 marker could be used for testing seed purity of Hipa 6, while RM 206 marker for Hipa 7. Specific SSR marker was capable of distinguishing a false hybrid rice that might not be recognized by the grow out test in the field.

Abstract

Maize productivity could be improved by using good quality of seed of high yielding hybrid varieties. The objective of this research was to study the effect of rhizobacteria and P fertilizer on physiological quality of maize hybrid seed and maize productivity. The experiment was conducted at Lembar Village, West Lombok District, West Nusa Tenggara Province and at Seed Science and Technology Laboratory IPB from April to December 2012. The experiment was arranged in a split plot design with three replications. The main plot was P fertilizer (untreated, 50 kg, 100 kg, 150 kg, and 200 kg SP36/ha), and the subplot was rhizobacteria treatments (untreated, rhizobacteria B42, and rhizobacteria ATS4). The results showed that application of 100 kg SP36/ha increased maize yield compared to that of unfertilized. Rizhobacteria ATS4 increased plant height and productivity of maize. Application of ATS4 rhizobacteria followed by 100 kg SP36/ha fertilizer increased the percentage of good quality seeds. Application of rhizobacteria could reduce the use of SP36 fertilizer up 50% of the recommended rate. Rizobakteri ATS4 and fertilizer P 100 kg SP36/ha increased the physiological quality of harvested seeds after four month storage at 21-25oC temperature and 53-62% relatif humidity.

Abstract

Swamp land ecology is a potential land to increase the crop production, especially rice. The research was aimed to increase the production and seed quality of IPB 3S rice variety in swamp land ecology. The research was arranged in Factorial Randomized Complete Block Design with three replications. The first factor was dose of Zinc, consist of 0 kg ha-1, 15 kg ha-1, 30 kg ha-1, and 45 kg ha-1. The second factor was seed treatment by probiotic microbial, consist of control, Bacillus sp, Bacillus sp (+), Chromobacterium sp, Chromobacterium sp (+). Zinc fertilization couldn’t increase the yield characters and seed quality of IPB 3S rice variety. Microbia fertilization could increase the highest yield characters (plant height, tillers, productive tillers, panicle length, weiht per plant, and seed weight per grove), and Bacillus microbial increased the seed quality (Vigor index). Interaction of zinc fertilization (15 kg ha-1) and Chromobacterium increased the yield characters (flag leaf length, panicle length, and seed weight per grove), and the interaction of zink fertilization (30 kg ha-1) and Bacillus increased seed quality (vigour index).

Abstract

The low production of hybrid rice seed is influenced by the syncronized of flowering time between CMS and restorer. One of several ways to syncronize both two CMS and restorer was use gibberellins. Gibberellins as growth regulator worked by triggered plant growth and flowering. The objective of this research was to obtain the optimal concentration of GA3 to support the  increase hibryd rice seed production. The experiment was arranged in a split plot design based on randomized complete block design. The first factor was three hybrid rice varieties (V) i.e. V1 Hipa-8 (A1 and PK91), V2 Hipa-14 (A7 and PK92) and V3 Jatim-3 (A6 and PK88) belong to the Center for Rice Research (BB Padi) as the main plot and the second factor was four level concentrations of gibberellins (GA3) i.e. G0 (without GA3), G1 (GA3 150 ppm) G2 (GA3 200 ppm) and G3 (GA3 250 ppm) as a subplot. The treatment was  performed by spraying GA3 with spray volume of one liter per a plot on the female and male parent. The results of experiment showed that concentrations of GA3 treatment significantly increased plant height, angle of floret opening and decreased number of empty spikelets. The treatment interaction of GA3 concentration and variety significantly effected on percentage of increasing plant height and productivity. The highest productivity was rice hibryd of  A7 (Hipa-14) of 1 000 kg/ha with application of 200 ppm GA3.

Abstract

The aim of this research was to know the growth various accesions of physic nut in coastal area. This research was conducted from March until October 2011 at Wonokerto Kulon coastal area, Pekalongan. The evaluated accesion were derived from 14 selected parent plants originated from Central Java, South Sulawesi, West Java and West Nusa Tenggara. The accesions were selected based on provenance trials in the previous research. The seeds were germinated in the seed nursery. The seedlings were planted in to the field (8 WAP). This research was arranged in randomized complete block design. The result of this research showed that Bogor 80-11-5 and IP-2P 110-1-4 are the best accesion. Bontomaramu 1-1, Medan and Pinrang. Sukabumi, Bengkulu 3, Gunung Tambora, Lombok 59-1-2, Aceh Besar, Bima, Bone, Dompu and Luwu Utara are not good to be planted in coastal area.Keywords: accesion, physic nut, coastal area

Abstract

Indonesia has extensive acid soil but its utility is still limited because of its low nutrient content. Jatropha is a plant which can grow on various soil types, includes on marginal one, and has used as raw material for biodiesel. Therefore, the growth of jatropha on acid soil need to be studied as an effort for developing jatropha and utility of acid soil in Indonesia. To initiate the effort, research to grow various jatropha genotypes on acid soil was done. The objectives of this research was to obtain a potential tolerant jatropha genotype for acid soil. The research was conducted at UPTD Development of Dryland Technology at Singabraja Village, Tenjo Subdistrict, Bogor, West Java, from November 2010 – August 2011. This research using Randomized Complete Block Design (RCBD) with jatropha genotype as factor. The result showed that there were differences in growth among the jatropha genotypes. There are five jatropha genotypes with better growth in acid soil (pH 5.0) compare to others, i.e. Medan I-5-1, Dompu, IP-2P-3-4-1, Sulawesi, and Bima M.Keywords: physic nut, genotypes, Jatropha curcas, acid soil

Abstract

Kondisi lingkungan simpan (suhu dan RH) dan jenis kemasan merupakan faktor penting yang mempengaruhi daya simpan benih. Tujuan penelitian ini adalah untuk mengetahui pengaruh pengusangan cepat secara fisik terhadap viabilitas dan vigor benih koro pedang serta mendapatkan ruang simpan dan jenis kemasan yang tepat untuk mempertahankan viabilitas dan vigor benih koro pedang selama penyimpanan. Penelitian terdiri atas 2 percobaan. Percobaan I menggunakan Rancangan Kelompok Lengkap Teracak dengan lama penderaan fisik sebagai faktornya, yang terdiri atas 0, 30, 60, 90, 120, 150, dan 180 menit. Percobaan II menggunakan rancangan percobaan tersarang dengan 2 faktor. Faktor pertama berupa ruang simpan yang terdiri atas ruang suhu kamar dan ruang ber-AC. Faktor kedua jenis kemasan yang terdiri atas 3 taraf yaitu karung plastik, jerigen plastik, dan plastik polypropylene. Hasil percobaan I menunjukkan bahwa penderaan fisik dengan suhu dan RH tinggi efektif menurunkan viabilitas dan vigor benih serta meningkatkan kadar air benih koro pedang. Hasil Percobaan II menunjukkan bahwa penyimpanan benih koro pedang pada ruang suhu kamar menghasilkan kadar air, viabilitas, dan vigor benih lebih tinggi dibandingkan penyimpanan pada ruang ber-AC. Abstract

Koro pedang saat ini dikembangkan sebagai komoditas substitusi kedelai karena kandungan proteinnya yang cukup tinggi. Tujuan penelitian ini adalah untuk menguji viabilitas benih koro pedang dengan pengujian tetrazolium dan pengujian respirasi benih. Penelitian ini menggunakan tiga lot benih koro pedang yang berbeda yaitu lot benih A (dipanen bulan Maret 2014 di Bojong Bogor, disimpan dalam ruang kamar), lot benih B (dipanen bulan November 2013 di Ciherang Bogor, disimpan dalam ruang ber-AC), dan lot benih C (dipanen bulan September 2012 di Leuwikopo Bogor, disimpan dalam ruang ber-AC). Penelitian ini terdiri dari dua percobaan. Percobaan 1 ialah pengujian tetrazolium. Percobaan 2 ialah penguijian respirasi benih. Masing-masing percobaan menggunakan rancangan acak lengkap dengan pengusangan cepat secara fisik sebagai faktornya, yang terdiri atas 0, 8, 12, 16, dan 20 jam. Hasil percobaan 1 menunjukkan bahwa pola topografi pewarnaan tetrazolium benih koro pedang belum bisa digunakan untuk mengevaluasi mutu fisiologis benih koro pedang. Hasil percobaan 2 menunjukkan bahwa metode pengujian respirasi benih dengan cosmotector belum bisa digunakan untuk mengevaluasi mutu fisiologis benih koro pedang.
