Abstract

Barramundi (Lates calcarifer, Bloch, 1790) also known as  barramundi has high commercially important. Its utilization is not limited to meat, but also the swim bladder it produces. Knowledge of the genetic aspects of barramundi (Lates calcarifer, Bloch, 1790) is very important in designing genetic improvement and conservation activities for this fish. The purpose of this study was to identify the species of barramundi collected from Pesisir Pantai Payum, Merauke Regency using the Cytochrome Oxidase I gene. DNA extraction used the Blood & Tissue Kit (Qiagen Kit), while the primers used for molecular identificationthe primers were FISH-F1 (5’- TCA ACC AAC CAC AAA GAC ATT GGC AC -3’) and FISH-R1 (5’- TAG ACT TCT GGG TGG CCA AAG AAT CA -3’). The results of this study indicated that the samples collected were barramundi species (Lates calcarifer, Bloch, 1790) with a similarity of 99.61-100% with 9 point mutations. Genetic distance and the level of similarity of DNA sequences greatly determine the formation of a phylogenetic tree. Molecular approaches can be used in the management and conservation of economically important fish species, especially barramundi (Lates calcarifer, Bloch, 1790).

Abstract

Pumpkin leaf plant (Cucurbita moschata D.) is commonly used to treat gastric pain, jaundice, hipercholesterol, and contains antibacterial compounds. Nature products derived from plants are often used to treat facial skin disease by inhibiting acne-causing bacteria. Staphylococcus epidermidis has important role in acne formation. This research aims to determine the antibacterial activity, Minimum Inhibitory Concentration (MIC), and Minimum Bactericidal Concentration (MBC) of 70% ethanol extract and n-hexane against Staphylococcus epidermidis. Pumpkin leaf extracts were obtained by maceration technique then diluted to obtain the extracts with concentrations of 10%, 25%, and 50%. The antibacterial activity test of pumpkin leaves was carried out in vitro by well diffusion method on Mueller Hinton Agar (MHA) media, while MIC and MBC was determined by dilution method using different concentration of extract. The results of this research showed that there was inhibition zone formed at concentration 10%, 25%, and 50% of 70% ethanol extract of yellow pumpkin leaves. According to the result, the largest inhibition zone was obtained at concentration of 50% (20.02 ± 0,28 mm). The MIC value was found at concentration of 12.5% and MBC value was determined at 50% concentration of ethanol extract. Meanwhile, n-hexane extract of yellow pumpkin leaves has no antibacterial activity against S. epidermidis. It can be concluded that ethanol extract of yellow pumpkin leaves has potent antibiotic material for S. epidermidis.

Abstract

Information about plants used as traditional medicines is passed orally from generation to generation. Semata Village is one of the villages that still use plants in the process of traditional medicine. As the age grows, it is feared that public knowledge of plants as the basis for traditional medicine will decrease. Written documentation is necessary to be done by conducting ethnobotanical studies of medicinal plants. This study aimed to determine the types of plants used as the source of medicine by the community in Semata Village. The research was conducted using the descriptive method through interviews, observation, and identification of plants as the data collection techniques. The information was obtained from 43 informants consisting of shamans, massage therapists, and the community of Semata Village. The community of Semata Village used 60 species from 37 plant families that are believed to treat 44 types of disease symptoms. The dominant part of plants used is the leaves, while the plants that are often used come from the Zingiberaceae family. Kunyik Itam is the typical plant of Semata Village that is starting difficult to obtain. Rhizome of Kunyik Itam  is used as medicine for asthma, cancer, and itching.
