Abstract

In this work, the Graphene oxide (GO) nanosheets were synthesized by a electrochemical exfoliation method. The morphological, structure and optical properties were investigated by using Field emission scanning electron microscope (FE-SEM), X-ray diffraction (XRD), Raman spectroscopy, FTIR and UV-VIS spectroscopy. FE-SEM images of GO nanosheets confirmed the presence of layered structure. The crystalline size and number of layers of GO nanosheets were estimated by XRD analysis.  The UV-VIS spectrum of GO nanosheets shows maximum and shoulder absorption peaks. The oxidation of graphene was confirmed via FTIR analysis by the appearance of functional groups of oxygen. Raman analysis showed the formation of peaks D, G and 2D. The antibiofilm activity of different concentrations of GO nansheets against Pseudomonas aeruginosa (P. aeruginosa) as a model of more antibiotics resistant Gram-negative bacteria was investigated. These results confirm that the GO nanosheets material damages the biofilm of P. aeruginosa. Therefore, It can be considered as an anti-bacterial material. Keywords: Graphene Oxide, Nanosheet, Electrochemical Exfoliation, Antibiofilm, Pseudomonas aeruginosa.

Abstract

Indonesia is a country with a high prevalence of hepatitis B virus infection and can develop into chronic hepatitis in the community. Indonesia is a multiethnic country with a vast territory, so it is estimated that the prevalence of each hepatitis B virus subtype varies greatly in different regions. Sequences in the S region gene encoding amino acids can show hepatitis b virus subtypes through exclusive variations in subtype determinants of HBsAg. This hepatitis B virus subtype is epidemiologically important, because it can show differences in geography and ethnicity in its spread. This study aims to analyze the hepatitis B virus subtype in HBsAg positive subjects in Mengwi, Badung, and Bali-Indonesia. The subjects of the study were 75 patients who came to the Mengwi I Badung Primary Health Center. All serum samples were examined for HBsAg by the ELISA method. In HBsAg positive serum samples, hepatitis B virus subtypes are determined. The DNA is extracted from serum samples that are HBsAg positive. Amplification of portions of DNA S gene from serum HBsAg positive was performed by polymerase chain reaction first-round and second-round. The nucleotide sequence of the hepatitis B virus is converted into amino acid sequences and multiple alignments are performed. The hepatitis B virus subtype was determined by amino acid substitution analysis at positions 122, 127, 134, 159, 160 and 177 in the S gene assisted by using a computer program. The subjects with HBsAg positive were obtained from 11 out of 75 samples (14.7%). The results of hepatitis B virus subtype analysis found that almost all samples, 10 of 11 samples (90.9%) had adw2 subtypes, while one sample (9.1%) had adrq + subtype. Keywords: HBsAg, Subtype, HBV, Mengwi.

Abstract

Hepatitis C virus (HCV) infection is a major health problem worldwide where130-150 million people are chronically infected with HCV, which progress to cirrhosis or hepatocellular carcinoma. A total of 120 subjects were recruited to assess the correlation between gene polymorphisms (IL-10-1082 and -819) with the susceptibility to HCV infection in Iraqi patients by using allele specific PCR. The serum level of IL-10 was assessed by ELISA. Gene polymorphism results of IL-10-1082A/G (rs1800896) showed significant increase in AA distribution (P=0.0076) and allele A frequency (P=0.049) among the control group while AG distribution were significantly higher in patients (P=0.0082). Although, G allele frequency had but an association (P=0.049), but the homozygous GG mutant genotype had no significant differences between the two groups. While the genotype analysis of the second SNPs of IL-10-819C/T (rs1800872) showed that neither CC nor CT genotypes distribution were associated with HCV infection (0.1627 and 0.2559, respectively) but the TT distribution and allele frequency had a suggestive association in patients (P=0.0483 and 0.0257, respectively). Results of IL-10 serum levels were higher by two-fold in patients than in healthy controls. It appeared that the IL-10-1082 AA genotype had a protective effect against HCV infection but the TT genotype of -819 mutations had a suggestive association with susceptibility to infection with HCV in Iraqi patients. Such correlation may indicate a protective role for IL-10 and could be used as a predictive risk susceptibility biomarker for progressive infection with HCV.
