Abstract

Penelitian ini bertujuan mengetahui motilitas dan kemampuan memfertilisasi sperma kucing yang berasal dari epididimis yang disimpan pada suhu 4° C. Epididimis disimpan dalam media phosphate buffer saline (PBS) pada suhu 4 C selama 1, 3, dan 6 hari. Viabilitas spermatozoa diamati dengan pewarnaan hoechst-propidium iodine. Fungsi biologis spermatozoa dievaluasi melalui teknik kultur in vitro dengan fertilisasi mikro dan perkembangan embrio di dalam media kultur CR1aa. Hasil penelitian menunjukkan persentase spermatozoa hidup pada hari ke-1, 3, dan 6 penyimpanan masing-masing adalah 81,0; 71,7; dan 70,7% (duktus deferens), 84,0; 81,2; dan 63,2% (kauda epididimis), 84,0%; 75,0; dan 74,7% (korpus epididimis). Persentase pronukleus (PN) yang didapat dengan teknik intra cytoplasmic sperm injection (ICSI) menggunakan spermatozoa epididimis pada hari ke-1, 3, dan 6 hari penyimpanan masing-masing adalah 8,0; 10,0; dan 5,9%. Preservasi epididimis pada suhu 4 C dalam PBS dapat digunakan untuk fertilisasi dan menghasilkan embrio kucing secara in vitro.

Abstract

The aim of this research was to find out the normal estrogen and progesterone profile in local goat estrous cycle.  Five female Aceh local goats which the criteria clinically in good condition, have been giving birth before, 1.5-3.0 years old, and showed minimal twice regularly estrous cycle. All samples were synchronized with 0.5 ml cloprostenol (Estrone, Bioveta) using randomly infection pattern. The data obtained were analyzed descriptively. Blood serum was collected for examination of estrogen and progesterone content on 0, 7, 14, and 21 day of estrous cycle (day 0 of estrous cycle). Hormone analysis was conducted using ELISA. The results showed that estrogen and progesterone profile on day 0, 7, 14, and 21 were 62.21; 28.61; 29.84; 79.89 pg/ml and 0.0; 8.6; 14.7; 0.0 ng/ml respectively.Keywords: estrogen, progesterone, estrous cycle, Aceh local goats

Abstract

Penelitian ini bertujuan menentukan frekuensi pemberian vaksin DNA koi herpesvirus (KHV) yang menghasilkan kelangsungan hidup tinggi pada ikan mas yang diinfeksi KHV. Frekuensi pemberian vaksin yaitu vaksinasi satu kali seminggu (K1), vaksinasi dua kali seminggu (K2), vaksinasi tiga kali seminggu (K3), ikan tidak divaksin dan diuji tantang KHV (K4), dan ikan diinjeksi dengan larutan phosphate buffered saline (K5). Pemberian pakan dilakukan 2 kali sehari secara satiasi.Mortalitas pada K1; K2; K3; dan K4 masing-masing adalah 33,33±5,77; 20,00±10,00; 6,67± 5,77; dan 43,33±5,77%, sedangkan kelangsungan hidup relatif (RPS) K1; K2; dan K3 masing-masing adalah 23,07±13,32; 53,84±23,07; 84,60±13,32%. Disimpulkan bahwa pemberian pakan mengandung vaksin DNA anti-KHV dengan frekuensi tiga kali dalam satu minggu efektif meningkatkan kelangsungan hidup ikan mas yang diinfeksi KHV.

Abstract

 Mesenchymal stem cells (MSC) derived from white adipose tissue are considered to be an appropriate therapeutic candidate for various human diseases due to their natural ability to maintain homeostasis. They have also been considered for use as a therapeutic alternative due to their sufficient availability and ease of harvesting. MSC play an important role in the process of adipogenesis and energy metabolism that is directly related to the pathogenesis of various degenerative diseases and metabolic syndromes. The purpose of this study was to obtain MSC cultures from white adipose tissue biopsied from three adult males Macaca fascicularis and to evaluate these cultures qualitatively and quantitatively regarding their characteristics and ability to differentiate. Polymerase chain reaction (PCR) was used to confirm the expression of mesenchymal stem cell-specific gene markers, and a specific growth medium was used to differentiate these cells into adipocytes, chondrocytes, and osteocytes. The results obtained showed that the MSC culture from M. fascicularis WAT had a fibroblast-like morphology with a spindle shape. The MSC markers of CD73, CD90 and CD105, were also expressed. The MSC culture was also able to differentiate into adipocytes, chondrocytes, and osteocytes. Based on these results, white adipose tissue from Macaca fascicularis was found to have potential as a source of MSC.

Abstract

This study was conducted to determine the effect of water salinity changes on methionine content in tiger grouper juvenile (Epinephelus fuscoguttatus). A total of 2,560 tiger grouper juveniles were used and divided into five groups consist of 1 control group (without exposed to salinity changes) and 4 treatment groups. The salinity was changed every 2, 3, 4, and 6 hours in treatment A, B, C, and D, respectively. Salinity levels were changed during 24 hours by lowering salinity level from 32 psu to 22 psu. Twenty five of fish were collected from each treatment for methionine content analysis. Data were analysed using one way analysis of variance (ANOVA). The results showed that there was a decrease in methionine content in all treatments. A significant decrease (P0.05) of methionine content in treatment A, B, and C were observed after 20 hours (1.15%), 18 hours (1.27%), and 16 hours (1.24%), respectively. While at 0 hours (control), the methionine content was 2.02%. Methionine level in treatment D was not significantly different (P0.05) compared to control group. As conclusion, rearing the tiger grouper juvenile with salinity fluctuations every 6 hours did not lead to methionine deficiency.

Abstract

Penelitian ini bertujuan menentukan hubungan clonal methicillin resistance Staphylococcus aureus (MRSA) antara isolat sapi dan manusia. Staphylococcus aureus yang digunakan dalam penelitian ini berasal dari infeksi kulit manusia di Yogyakarta (10 isolat) dan dari susu sapi perah (11 isolat) yang berasal dari Yogyakarta, Solo, dan Boyolali. Identifikasi bakteri dan uji resistensi S. aureus terhadap methicillin telah dilakukan pada penelitian sebelumnya. Hubungan genetika S. aureus antar isolat sapi dan manusia ditentukan menggunakan teknik single enzyme amplified fragment length polymorphism (AFLP). Hasil penelitian diketahui bahwa berdasar analisis AFLP S. aureus memperlihatkan 15 pola genetika (A sampai O) dan dapat dikelompokkan ke dalam 7 klas (I sampai VII). Staphylococcus aureus isolat asal sapi dari daerah yang berdekatan (Boyolali dan Solo), dapat dikelompokkan dalam 1 klas (kecuali 1 isolat sapi dari Yogyakarta), isolat sapi dan manusia dari Yogyakarta dapat dikelompokkan dalam beberapa klas. Masing-masing klas terdapat isolat S. aureus yang telah resisten terhadap MRSA. Hubungan genotipe S. aureus dalam penelitian ini dapat digunakan untuk mengetahui distribusi clonal antara isolat sapi dan manusia dan dapat digunakan sebagai kontrol adanya infeksi MRSA di Indonesia.

Abstract

ABSTRACT The purpose of this research was to examine the effect of various sugars addition in Tris extender on quality of frozen thawed epididymal spermatozoa of ram. Collected spermatozoa was divided into five tubes and centrifuged at 3,000 rpm for 30 minutes. Supernatants were removed and pellets were diluted with Tris extender (control), Tris extender + 0.4% dextrose (dextrose), Tris extender + 0.4% maltose (maltose), Tris extender + 0.4% lactose (lactose), and Tris extender + 0.4% sucrose (sucrose), respectively. There were no significantly difference among treatment on mean percentages of motile (MS), live (LS) and intact plasma membrane (IPM) after equilibration. Mean percentages of post-thawed MS, LS, and IPM for dextrose (43.0, 53.8, and 53.4%), maltose (44.0, 54.8, and 52.8%), lactose (44.0, 52.8, and 52.8%), and sucrose (44.0, 55.0, and 53.4%) were significantly (P0.05) higher than control (37.0, 44.0, and 40.8%). In conclusion, addition of 0.4% dextrose or 0.4% maltose or 0.4% lactose or 0.4% sucrose in Tris extender increased quality of frozen thawed cauda epididymal spermatozoa of ram.

Abstract

Penelitian ini bertujuan mempelajari deformasi sendi tarsus yang mengalami tarsitis kronis. Penelitian ini dilakukan dengan mengamati  perubahan ukuran sendi tarsus pada preparat kaki belakang kuda delman dan deformasi skelet sendi tarsus pada kuda delman. Dari hasil penelitian ditemukan adanya pembesaran di bagian medial tarsus kuda dan pembesaran pada lingkar tarsus kuda 3-7 cm dari bentuk normal dengan rata-rata lingkar tarsus kuda delman 23,18 cm dan rata-rata lingkar tarsus pada kuda yang mengalami tarsitis 27,83 cm. Kelainan berupa perlekatan tulang (ankylosis) dan pertumbuhan tulang baru (exostosis) pada permukaan tulang-tulang sendi tarsus kuda. Pada gambaran radiografiditemukan adanya penyempitan ruang sendi, kerusakan permukaan tulang dan pertumbuhan tulang baru yang abnormal. Deformasi yang terjadipada sendi tarsus preparat kaki belakang kuda delman diduga disebabkan oleh aktivitas yang berat semasa hidupnya.

Abstract

This study aims to determine the effects of adding propionic acid in the poultry diets on the internal organs of broiler chickens. The livestock used were 180 broilers with Cobb strain. This study used a completely randomized design (CRD) consisting of 3 treatments and 6 replications.The treatments in this study consisted of: P0= commercial feed without propionic acid addition; P1= commercial feed with addition of 0.5%propionic acid; and P2= commercial feed with 0.75% propionic acid. The parameters observed included live weight, spleen percentage, gizzardpercentage, liver percentage, and small intestine length. The results showed that the addition of propionic acid in the broiler feed had a significant effect (P0.05) on the live weight and had no significant effect (P0.05) on the percentage of spleen, percentage of gizzard, percentage of liver, and small intestine length. The conclusion of this study was that the addition of propionic acid in poultry feed at a dose of 0.75% did not negatively affect the internal organs and the length of the small intestine of broiler chickens.

Abstract

The research has been done to know prevalence of toxoplasmosis in livestock, to search the source of infection that have potency to cause toxoplasmosis in human and to make a map of toxoplasmosis infection area in Banda Aceh. The method used in this research was serologic test, Card Aglutination Test (CATT). Data of fertile women in this study are collected through questioner. The result of this research showed that the toxoplasmosis prevalency number of society in Banda Aceh was 3.15%, while at livestock were varied respectively: goats 40%, chickens 25%, cattles 23%, duck 20%, cats 16%, buffalos 15%, and sheeps 10%. The potencial sources of to coures toxoplasmosis in human are: goat, chicken, cattle, duck, cat, buffalo, and sheep. The area found toxoplasmosis in humans and animal were: Baiturrahman, Kuta Raja, Ulee Kareng, Kuta Alam, and Syiah Kuala.
