Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak akar pasak bumi (Eurycoma longifolia Jack) terhadap peningkatan konsentrasi testosteron domba Waringin. Dalam penelitian ini digunakan dua ekor domba waringin jantan berumur ±4 tahun. Penelitian ini merupakan penelitian eksperimen dengan menggunakan rancangan pola bujur sangkar latn 2 x 2 sehingga hewan percobaan diberi perlakuan 20 mL akuades per oral sebagai kontrol (P1) dan 20 mL larutan yang mengandung 45 mg/kg bobot badan ekstrak pasak bumi yang diberikan per oral (P2). Perlakuan diberikan selama enam hari pada pukul 09.00 WIB. Pada bulan ke-1 domba pertama mendapat perlakuan P1 sedangkan domba kedua mendapat perlakuan P2. Sebaliknya, pada bulan ke-2 domba pertama mendapat perlakuan P2 sedangkan domba kedua mendapat perlakuan P1. Koleksi darah untuk pemeriksaan konsentrasi hormon testosteron dilakukan lima jam setelah pemberian ekstrak akar pasak bumi (Eurycoma longifolia Jack). Analisis konsentrasi testosteron dilakukan menggunakan metode enzyme linked immunosorbent assay (ELISA) yang diukur pada hari 1, 3 dan 6. Data yang diperoleh dianalisis dengan uji sidik ragam pola bujur sangkar latin. Hasil analisis statistika menunjukkan bahwa volume semen, motilitas spermatozoa, viabilitas spermatozoa dan abnormalitas spermatozoa pada domba yang diberikan ekstrak akar pasak bumi (E. longifolia Jack) menunjukkan perbedaan yang signifikan (P<0,05) dibandingkan dengan kontrol namun konsentrasi testosteron tidak menunjukkan perbedaan yang signifikan (P>0,05). Disimpulkan bahwa pemberian ekstrak akar pasak bumi meningkatkan kualitas spermatozoa namun tidak menyebabkan peningkatan konsentrasi hormon testosteron pada domba waringin.

Abstract

ABSTRAKEscherichia coli dikenal sebagai bakteri indikator sanitasi, keberadaanya dalam pangan menunjukkan bahwa pangan tercemar oleh lingkungan. Penelitian ini bertujuan mengetahui angka prevalensi bakteri E. coli pada meja dan peralatan pedagang daging ayam broiler. Sampel yang digunakan yaitu 20 swab meja, pisau, dan talenan pedagang daging ayam broiler di dua Pasar Tradisional Kota Banda Aceh. Penelitian ini menggunakan metode Total Plate Count (TPC) dengan larutan Buffer Peptone Water (BPW) dan mengidentifikasi E. coli menggunakan media selektif diferensial, yaitu dengan Eosin Methylene Blue Agar (EMBA). Hasil penelitian menunjukkan bahwa 2 meja, 2 pisau, dan 3 talenan di Pasar Tradisional Seutui positif tercemar E. coli dengan angka prevalensi cemaran E. coli pada meja (40%), pisau (40%), dan talenan (60%), sedangkan di Pasar Tradisional Peunayong terdapat 2 meja dan 2 pisau positif tercemar E. coli dengan angka prevalensi cemaran  E. coli pada meja (13%) dan pisau (13%). Hasil kesimpulan yang didapat yaitu rata-rata angka prevalensi cemaran E. coli secara keseluruhan pada peralatan pedagang dari dua Pasar Tradisional di Kota Banda Aceh yaitu 18,33%.Kata kunci : Escherichia coli, prevalensi, peralatan pedagang, pasar tradisional  ABSTRACTEscherichia coli is known as an indicator of sanitation bacteria, its presence in food indicates that food is polluted by the environment. This study aims to determine the prevalence of  E. coli bacteria on the table and equipment of broiler chicken meat traders. The samples used were 20 table swabs, knives, and cutting boards of broiler traders in two Traditional Markets in Banda Aceh City. This study uses the Total Plate Count (TPC) method with Buffered Peptone Water (BPW) solution and identifies E. coli using differential selective media, namely Eosin Methylene Blue Agar (EMBA). The results showed that 2 tables, 2 knives, and 3 cutting boards in the Seutui Traditional Market were positively contaminated with E. coli with the prevalence of E. coli contamination on the table (40%), knives (40%), and cutting boards (60%), while in the Peunayong Tradisional Market there are 2 tables and 2 positive knives contaminated with E. coli with the prevalence rate of E. coli contamination on the table (13%) and knives (13%). The conclusion obtained is the average prevalence rate of E. coli contamination as a whole on the equipment of traders from two Traditional Markets in the City of Banda Aceh is 18.33%. Key words : Escherichia coli, prevalence, equipment traders, traditional markets

Abstract

ABSTRAK Penelitian ini bertujuan mengetahui gambaran histopatologi hati tikus putih yang diinjeksi formalin dengan dosis bertingkat. Penelitian ini menggunakan hati 24 ekor tikus putih strain Wistar jantan umur 4-5 bulan dengan berat 180-200 gram. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) pola searah dengan 4 kelompok perlakuan. Kelompok kontrol (KO) adalah tikus yang hanya diberi pakan standar tanpa perlakuan apapun. Kelompok perlakuan 1 (K1), 2 (K2), dan 3 (K3) diberi pakan standar dan diinjeksi formalin secara intraperitoneal dengan dosis 1; 2,5; dan 5 mg/kg bb per hari. Penelitian ini dilakukan secara berkelanjutan selama 14 hari. Pada hari ke-15 semua tikus dibunuh dengan menggunakan kloroform, kemudian sampel organ hati diambil untuk dibuat sediaan histopatologi. Data yang diperoleh dianalisis menggunakan ANAVA dan apabila terdapat perbedaan yang nyata dilanjutkan dengan uji Duncan. Hasil analisis statistik menunjukkan rata-rata (±SD) jumlah sel hati yang mengalami degenerasi parenkim pada K0, K1, K2, dan K3 berturut-turut adalah (2,40 ± 0,34), (3,40 ± 0,34), (4,73 ± 0,410), dan (5,53 ± 0,50). Jumlah sel hati yang mengalami degenerasi hidropis pada K0, K1, K2, dan K3 berturut-turut adalah (0,26 ± 0,23), (3,13 ± 0,30), (4,33 ± 1,00), dan (5,40 ± 0,87). Jumlah sel hati yang mengalami nekrosis pada K0, K1, K2, dan K3 berturut-turut adalah (0,66 ± 0,23), (2,06 ± 0,30), (2,53 ± 0,46), dan (3,60 ± 1,00). Formalin berpengaruh nyata (P0,05) terhadap jumlah sel hati yang mengalami degenerasi parenkim, degenerasi hidropis, dan nekrosis. Pemberian formalin dengan dosis bertingkat selama 14 hari pada tikus putih menyebabkan perubahan histopatologi sel hati berupa degenerasi parenkim, degenerasi hidropis, dan nekrosis. Semakin tinggi dosis yang diberikan, semakin banyak kerusakan sel hati yang ditemukan.  ABSTRACK This study aimed to determine the histopathology of rat liver induced by gradual dosage of formaldehide. This study used 24 liver of Wistar rats aged 4-5 months, weighing of 180-200 grams. The research design used was a completely randomized design (CRD) with 4 unidirectional pattern group treatments. Eachgroup treatment consisted of six rats. The control group (KO) rat given only the standard feed without any treatment. Treatment group 1 (K1), 2 (K2), and 3 (K3) were fed with standard feed and injected intraperitoneally with formaldehide doses of 1, 2.5, and 5 mg/kg bw. The treatment was carried out for 14 consecutive days. On day 15 all rats were eutanized using chloroform. Then the liver samples taken for histopathological preparations. Data were analyzed using ANAVA and then followed by Duncan test. Statistical analysis showed that the average (± SD) number of parenchyma degeneration of liver cell on K0, K1, K2, and K3 were (2.40±0,34), (3.40±0.34), (4.73±0.410) and (5.53±0.50) respectively. The number of hydropis degeneration of liver cell on K0, K1, K2, and K3 were (0.26±0.23), (3.13±0.30), (4.33±1.00), and (5.40±0.87) respectively. The number of liver cell necrosis in K0, K1, K2, and K3 were (0.66±0.23), (2.06±0.30), (2.53±0.46), and (3.60±1.00) respectively. Formalin effect significantly (P0.05) on the number of parenchyma degeneration, hydropis degeneration, and necrosis of liver cell. The higher the dosage given, the more damage the liver cells were found.

Abstract

Differential display technique was used to identify differentially expressed gene ofswordtail fish (Xiphophorus spp.) that induced by 1 µg/ml copper for 24 hours. From eightprimers that have been used for differential display, seven of them showed the differentialgenes which were not present in the control. The reamplification of these fragments rangingfrom below 100 to 200 bp. But only two fragments were successfully cloned and sequenced,those are H-AP10 and H-AP11. Sequence analysis showed that both of the sequences revealedhigh similarity with Homo sapiens stress-associated endoplasmic reticulum protein, a signalgenerated that induces apoptosis.Keywords: differential display, swordtail fish, copper pollution, mRNA expression, biomarker

Abstract

The objective of this study was to evaluate the effect of storage time of epididymis at 5 oC on spermatozoa quality of Aceh local goat. This research was conducted at Reproduction Laboratory, FKH Unsyiah from May to June 2009. Nine epididymis from local goat at the age of 1.5-2.0 years were used in this study. The epididymis were collected from abattoir in Banda Aceh. The experimental design used in this study was completely randomized design with 3 treatments and 3 replications. Spermatozoa quality which consist of sperm concentration, motility, amount of life sperm, and sperm abnormality were examined after the collection of epididymis on day 0 (H-0), day 1 (H-1), and day 3 (H-3) post storage at 5 oC. Data were analyzed using one-way analyses of variance (ANOVA). The result showed that no significant difference (P0.05) seen in the spermatozoa quality after different storage time. The average of spermatozoa concentration on H-0, H-1, and H-3 were 318x107/ml, 282x107/ml, and 241x107/ml respectively. On the average, the percentage of spermatozoa motility on H-0, H-1, and H-3 were 82,27±2,75; 80,25±2,83; and 78,07±0,92%, respectively. Life spermatozoa observed on H-0, H-1, and H-3 were 82,29±2,71; 80,63±1,87; and 80,09±3,31%, respectively. Observation on spermatozoa abnormality showed that the average of spermatozoa abnormality on H-0, H-1, and H-3 were 7,23±0,27; 8,21±0,55; and 10,75±3,14%, respectively. It could be concluded that the spermatozoa quality were not affected by the storage time at temperature of 5 oC.

Abstract

Penelitian ini dilakukan untuk mengetahui efek suplementasi tepung daun jaloh (Salix tetrasperma Roxb) (TDJ) dalam pakan terhadap nilai diferensial leukosit dan ketahanan hidup setelah uji tantang dengan Aeromonas hydrophila pada ikan nila (Oreochromis niloticus) yang dipelihara pada akuarium diberi peningkatan suhu lingkungan. Penelitian ini menggunakan rancangan acak lengkap faktorial 2x4. Faktor pertama adalah penambahan jumlah tepung daun jaloh dalam pakan yaitu: 0% (P1), 5% (P2), 10% (P3), dan 15% (P4) dari berat pakan dan faktor kedua adalah suhu air dalam akuarium yaitu 29±1 C (S1) dan 35±1 C (S2), sehingga didapat 8 kombinasi perlakuan yaitu: P1S1, P2S1, P3S1, P4S1, P1S2, P2S2, P3S2, dan P4S2 dengan ulangan 10 ekor ikan per perlakuan. Sebanyak 80 ekor ikan nila dengan bobot badan 40-50 g secara acak dibagi ke dalam 8 perlakuan. Perlakuan dilakukan selama 30 hari. Pada hari ke 31 dilakukan pengambilan sampel darah dan uji tantang. Hasil penelitian ini menunjukkan bahwa suplementasi pakan dengan TDJ pada konsentrasi 5-15% tidak berpengaruh terhadap nilai diferensial leukosit ikan nila yang dipelihara pada suhu lingkungan yang berbeda. Suplementasi TDJ dalam pakan sebanyak 5-10% dapat mengurangi kematian ikan akibat infeksi Aeromonas hydrophila terutama pada suhu 35±1 C. Dari hasil penelitian ini dapat disimpulkan bahwa ikan nila yang dipelihara pada akuarium dengan suhu air 35±1 C dan diberi pakan yang disuplementasi TDJ 5-10% dapat meningkatkan daya tahan tubuh ikan nila.

Abstract

The aim of this research is to find out the histopathological changes of erythrocyte and the quantity of immature erythrocyte of tegal duckling treated with lead (Pb). Twenty five young female ducks with the age of 7 days and 60.55.07 g body weight alloted into 4 treatment groups and 1 control group (Ko) with 5 replication each. The treatment group fed with K1 (5.7), K2 (11.4), K3 (17.1), and K4 (22.8) mg Pb/kg body weight for 10 days, respectively. Young ducks showed clinical symptoms such as the color of feces were brown to black, green color of diarrhea, anemic and decreasing body weight. Histopathologically, erythrocyte showed that immature and abnormal in shape (like teardrops, dumb bell, and bottles). The result of variant analysis followed by linier regression shows that the higher the dosage of Pb in diet the higher the quantity of immature erythrocyte andabnormal erythrocyte shape found (P0.01).

Abstract

This study was conducted to determine the effect of water salinity changes on methionine content in tiger grouper juvenile (Epinephelus fuscoguttatus). A total of 2,560 tiger grouper juveniles were used and divided into five groups consist of 1 control group (without exposed to salinity changes) and 4 treatment groups. The salinity was changed every 2, 3, 4, and 6 hours in treatment A, B, C, and D, respectively. Salinity levels were changed during 24 hours by lowering salinity level from 32 psu to 22 psu. Twenty five of fish were collected from each treatment for methionine content analysis. Data were analysed using one way analysis of variance (ANOVA). The results showed that there was a decrease in methionine content in all treatments. A significant decrease (P0.05) of methionine content in treatment A, B, and C were observed after 20 hours (1.15%), 18 hours (1.27%), and 16 hours (1.24%), respectively. While at 0 hours (control), the methionine content was 2.02%. Methionine level in treatment D was not significantly different (P0.05) compared to control group. As conclusion, rearing the tiger grouper juvenile with salinity fluctuations every 6 hours did not lead to methionine deficiency.

Abstract

Telah dilakukan penelitian untuk mengisolasi dan mengidentifikasi protein 28 kDa (GRA2) dari stadium takizoit dan bradizoit Toxoplasma gondii strain lokal. Cairan asites mencit galur Balb/c digunakan sebagai protein sampel melalui elektroforesis (SDS-PAGE), serum kambing untuk uji antigenitas protein 28 kDa (GRA2) dan serum darah kucing untuk melihat tingkat sensitivitas dan spesifisitas protein 28 kDa (GRA2). Data yang diperoleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa protein 28 KDa (GRA2) telah dapat diisolasi dari stadium takizoit dan bradizoit Toxoplasma sp. Nilai OD405 antigenesitas ES adalah sebesar 0,578 sedangkan pada antigen GRA2 (28 kDa) diperoleh nilai OD405 adalah sebesar 0,178. Tingkat sensitivitas protein 28 kDa (GRA2) terhadap antibodi dalam serum darah kucing terinfeksi adalah sebesar 61,53% dan nilai spesifisitasnya adalah 33,33%. Tingkat sensitivitas protein ES terhadap antibodi dalam serum darah kucing terinfeksi adalah 76,92% dan tingkat spesifisitasnya adalah 33,33%.

Abstract

The objective of this research is to know Aceh cattle follicle dynamic under environmental heat stress condition. This research was conducted on December 2012 until August 2013. Animals used in this research were 20 Aceh cattle aged 5-8 years old, weighing of 150-250 kg, and had at least 2 regular cycles. All cattle used were clinically in good body condition score. The cattle were divided into two groups of 10, used in two different period of times (December-January and July-August) to examine the effect of environmental heat to their follicle dynamic. Each group is separated into two different keeping management; one group was kept in pens while the others in the pasture, 5 cattle in each management group. Research timing was based on information acquired from Indrapuri Agency for Meteorology, Climatology, and Geophysics (BMKG) which predicted that extreme weather would last from July to August while December to January would be relatively normal in Aceh region. All cattle’s estrous cycle were synchronized by 5 mg/mL of PGF2α intramuscularly (Lutalyse™, Pharmacia Upjohn Company, Pfizer Inc.). Ultrasonography examination was performed to monitor ovary follicle’s growth and dynamic during one cycle. Days during ovulation marked by standing heat was regarded as Day 0 of estrous cycle. Follicular dynamic examination during estrous cycle on December-January and also on July-August, both penned or pastured cattle showed the follicle waves that was 3 follicle growth waves. The size of follicle growth on first wave (1st DF) on cattle kept in pen on July-August and December-January were relatively similar. The size of follicle growth on first wave for cattle kept in pasture on July-August and December-January were relatively similar. To conclude, Aceh cattle’s follicle dynamic is not change during environment heat stress condition and is not affected by different cattle-keeping management.
