Abstract

Pandanus conoideus is exclusively grown in papua island and its neighbor areas and its fruit (buah merah) has been utilized as food supplement by the native papua people for a long period. recently it was found that its extract oil (SMB) contains contents of carotenoids ,especially a novel micronutrient beta-cryptoxanthin .we consider that SBM is a potent chemopreventive supplement against some types of cancer and provide health benefits for chronic diseases .in the present studies, we evaluated acute ad subacute toxicities of SBM using Sprague Dawley rats.Oral acute toxicity of SBM using female rats was not observed and LD 50 was more than 2 ml/kg in subacute toxicity study ,SMB was orally administered at 0.1,0.3 and 1.0 ml/kg to male and female rats for consecutive 4 weeks .no findings associated with SBM observed we consecutive 4 weeks .no findings associated with were observed we conclude that SBM is a safe food supplement.

Abstract

The mammary gland contains adult stem cells that are capable of self-renewal and are likely target for neoplastic transformation leading to breast cancer. In this study, we developed a cell culture derived from the mammary glands of cynomolgus monkeys (Macaca fascicularis)(MfMC) and furthermore identified the expression of markers for stemness and estrogenreceptor-associated activities. We found that the primary culture can be successfully subcultured to at least 3 passages, primarily epithelial-like in morphology, the cultured cells remained heterogenous in phenotype as they expressed epithelial cell markers CD24, CK18, and marker for fibroblast S1004A. Importantly, the cell population also consistently expressed the markers of mammary stem cells (ITGB1 or CD29 and ITGA6 or CD49f), mesenchymal stem cells (CD73 and CD105) and pluripotency (NANOG, OCT4, SOX2). In addition to this, the cells were also positive for Estrogen Receptor (ER), and ER-activated marker Trefoil Factor 1, suggesting an estrogen responsiveness of the culture model. These results indicate that our cell culture model is a reliable model for acquiring a population of cells with mammary stem cell properties and that these cultures may also serve as a reservoir from which more purified populations of stem cell populations can be isolated in the future.

Abstract

Cynomolgus macaques (Macaca fascicularis) are an established animal model of asthma, which exhibit different responses to allergen exposure that are clinically relevant. The chemokine ligand gene (CCL7) encodes Monocyte Chemotactic Protein-3, which has an important role in asthma pathogenesis. While CCL7 polymorphism in humans is associated with asthma phenotype, very little is known about CCL7 in nonhuman primate models of respiratory disease. The objective of this study was to isolate and characterize CCL7 gene in cynomolgus macaques of Indonesian origin. In this study, we used sequencing and bioinformatics technique for gene isolation, characterization, and protein 3D structure prediction. We isolated a 2253 base-pair (bp) sequence of CCL7 in cynomolgus macaques, which exhibited 95% similarity in coding sequence to human CCL7. The amino acid sequence was more closely clustered with human CCL7 than with that of rodents. Importantly, the predictive protein structure of CCL7 was similar to that in humans. These similarities in CCL7 suggests the potential of cynomolgus macaque as a translational model to study asthma, particularly in the context of genetics and role of chemokines such as CCL7.

Abstract

Coleus amboinicus (Lour) (torbangun) has been used traditionally as a breast milk stimulant, flavoring agent in many cuisines, and reported to possess many pharmacological values. The aim of this study was to explore the utility of the Pressurized Liquid Extraction (PLE) technique to identify the phytochemical contents of torbangun. In this study, total phenolic content and flavonoid in torbangun were determined using spectrophotometric method. The GC-MS analysis was used to identify the chemical constituents of torbangun, which was sequentially extracted with a wide range of solvent or solvent mixture from nonpolar-moderately polar and finally polar solvent. Total phenolic and flavonoid contents in the methanol extract of torbangun were 42.17±2.96 mg GAE/g and 11.20±0.58 mg QE/g, respectively. The identified chemical constituents in torbangun were sugar, hydrocarbon, ketone, terpenes, phenolics, fatty acid, fatty alcohol, steroids, alkaloids, and others, whereas the dominant constituents were phenolic compound, alkane, and sugar. In conclusion, this study demonstrated the effectiveness and rapid extraction of the PLE technique. Many valuable compounds in torbangun were extracted using gradual composition of solvent and were able to identify certain compounds in different polarities of solvents that are important in functional food preparation, pharmaceutical, and metabolomics research. Abstract

Rice bran is a by-product of rice-milling process. The rice bran is a coarse powder substance that is rich in bioactive components, including ferulic acid, ?-oryzanol, ? -sitosterol, tokotrienol/tocopherol, trisin, r-coumaric, sinapic, syiringic and phytic acid. The phenolic compounds of rice bran have potential of preventing cancer. The compounds could be increased by SSF (Solid State Fermentation). Enzymes produced by microbes during the process of fermentation are amylase, xylanase, protease, lacase, esterase, and ? -glucosidase. Enzymes which help increase the phenolic compounds in the rice bran include: lacase, esterase and ? - glucosidase. Factors affecting the amount of increases of phenolic compounds in SSF are the types of microbes being used and duration of fermentation.

Abstract

Sel punca memiliki peran penting dalam perkembangan jaringan normal maupun onkogenesis, termasuk pada kejadian kanker payudara. Keterkaitan sel punca dan pembentukan kanker payudara perlu diteliti agar dapat ditemukan metode pencegahan dan terapi baru yang efektif. Teknik kultur mamosfir telah terbukti memperkaya populasi sel punca pada sel kelenjar susu manusia dan satwa primata. Penelitian ini bertujuan untuk mengembangkan model onkogenesis kelenjar susu secara in-vitro dengan induksi sinar gamma pada kultur mamosfir yang berasal dari kelenjar susu monyet ekor panjang (Macaca fascicularis). Analisis morfologi sfir dan uji imunofluoresensi untuk γH2AX dilakukan setelah pajanan iradiasi 4 Gy selama 62 detik pada kultur mamosfir. Penanda molekuler sel punca dewasa dan sel punca kanker dievaluasi menggunakan qRT-PCR. Hasil penelitian menunjukkan bahwa iradiasi sinar gamma menimbulkan kerusakan DNA (double strand break) di dalam sel-sel penyusun mamosfir. Pajanan radiasi juga mengubah morfologi sfir. Ekspresi penanda untuk sel punca dewasa kelenjar susu (CD49f atau Integrin Alpha 6; ITGA6) dan sel punca kanker (CD133 atau Prominin-1; PROM1 dan CD44) lebih tinggi pasca iradiasi dibandingkan dengan kontrol. Temuan ini menunjukkan bahwa kultur mamosfir iradiasi yang berasal dari kelenjar susu monyet berpotensi dikembangkan sebagai model in-vitro untuk studi onkogenesis, terutama yang berkaitan dengan deregulasi sel punca dan risiko kanker payudara.

Abstract

Nonhuman primates (NHPs) play as indispensable animal model in biomedical research for studying a variety of human health issues, diseases and disorders, therapies, and preventive strategies. Since the immunological and physiological responses of NHPs, at some extent, to experimental viral infections are similar to humans, it is possible that studies of dengue infection in NHPs may aid understanding of dengue infection in humans. In this study,we used pigtailed macaques (Macaca nemestrina) as the experimental animal to study dengue-3 (DEN-3) virus infection.We evaluated DEN-3 viral distribution and replication sites after a primary infection in all collected tissues. Sequential localization in tissue of DEN-3 virus was studied in pigtailed macaques euthanized three days post viral inoculation (10 pfu mL ). Pigtailed macaque that was inoculated subcutaneously or intravenously; showed the highest viremia (62.94 pfu mL and 58.62 pfu mL ) detected by one step reverse transcription real time PCR. The virus inoculated in pigtailed macaques by subcutaneous injection was rapidly disseminated from the inoculation site to the lymph nodes, adrenal glands, kidneys, heart, thyroid, liver, prostate gland, and seminal vesicles. Meanwhile, dissemination of dengue virus in pigtailed macaques inoculated intravenously was detected in lymph nodes, thymus, salivary glands, liver, and prostate gland. This study suggested that the above mentioned-tissue specimens are involved or affected by DEN-3 virus replication and the route of infection seemed to have influenced the virus dissemination.

Abstract

BACKGROUND: C-C motif Ligand 7 (CCL7) has been reported to be associated with asthma severity in humans. Cynomolgus macaques (Macaca fascicularis; Mf) are often used as animal model of asthma but little is known about Mf genetic profile such as polymorphism. Our aim was to identify CCL7 polymorphism in Mf as a potential surrogate marker for identification of allergen responsiveness in the Mf model of asthma.METHODS: Real-time PCR was performed on archive of bronchoalveolar fluid samples previously collected from Mf that were exposed to allergen. Expression of CCL7 mRNA was evaluated, and sequencing technique was used to identify polymorphism in this gene.RESULTS: The results showed that CCL7 expression did not differ between Mf, despite a trend of lower expression in Mf that exhibited high response to allergen. By direct DNA sequencing of CCL7, 10 sequence variants were identified; three in promoter region (-460 G/A, -459 A/G, -456 -/A ), two in exon 1 (9 A/G, 65 G/C), four in intron 1 (135 T/C, 254 T/C, 420 T/C, 453 A/G), and one in intron 2 (1205 T/A).CONCLUSION: There was an association between Mf sensitivity to allergen with CCL7 promoter polymorphism at (-456 -/A). These results suggest that CCL7 may be a potential genetic marker to identify Mf sensitivity to allergen, which could be a useful tool to efficiently select for Mf model of asthma.KEYWORDS: asthma, CCL7, allergy, Ascaris suum, nonhuman primate
