Biophysical assessment of the plant biodiversity of Northern Negros Natural Park, Negros Island, Philippines. Biodiversitas 24: 583-602. Data sets on plant bioresources in Northern Negros Natural Park are critical in planning for conservation management and strategies. This study aimed to (i) conduct a survey of plant species in NNNP and determine their conservation status, (ii) assess the species diversity pattern and vegetation structures, (iii) establish altitudinal zones, and (iv) analyze the environmental factors affecting species distribution. Standard vegetation analysis and secondary data were utilized in the data collection process. Results showed 242 species, 72 families of 78 tree species, 90 shrubs and small trees, and 37 epiphytes. About 112 species are threatened, 35 of which are Philippine endemics. Two altitudinal zones were identified using the cluster analysis: Zone I (643 to 1,256 m asl.), dominated by Crypteronia paniculata, Acer laurinum, Weinmannia hutchinsonii, and Syzygiumsp.; and Zone II (1421 to 1470 m asl.) dominated by Dacrydium cf. beccarii. Zone I was further subdivided into three clusters: Zone IA (643 to 985 m asl.), Zone IB (908 to 913 m asl.), and Zone IC (822 to 1,182 m asl.). Canonical correspondence analysis illustrated that environmental factors (N, Altitude, and human disturbance) influenced the zonation of vegetation or species abundance. These results are important in understanding the landscape and crafting strategic interventions and management plans for conservation.

Enterobacter hormaechei KIMS8 and Enterobacter cloacae KIMS10 isolated from Kapuas River, Kalimantan, Indonesia as indigenous multi-resistant bacteria to copper and dyes. Biodiversitas 23: 6661-6668. Study on the characterization of multiple copper and dyes resistance in bacteria has so far been quite limited. It is, therefore, of interest to isolate and characterize such bacteria. The study was aimed to isolate bacteria from the Kapuas River in Indonesia and measuring copper resistance as well as dye-resistance and decolorization abilities. Copper resistance was determined by measuring the minimum inhibitory concentration (MIC) of copper, while dye-resistance was measured by observing changes in colony color and clear zone formation and decolorization ability was determined by spectrophotometry. It was found that 2 out of the 15 isolated strains showed the highest copper resistance with the MIC of 7 mM. The two bacterial strains KIMS8 and KIMS10, were grown in solid media supplemented with 300 ppm of methylene blue or reactive black dye. Bacterial decolorization assays showed that KIMS8 was able to decolorize up to 90% methylene dye and up to 11.3% of reactive black dye whereas KIMS10 was able to decolorize 94.9% of methylene blue dye and 12.1% of reactive black dye. Molecular characterization by 16S rRNA gene sequencing of these two strains showed that KIMS8 and KIMS10 were identified as Enterobacter hormaechei and E. cloacae, respectively.

Abstract. Husaini IPA, Maulany RI, Nasri N, Ngakan PO. 2022. Diversity and use of traditional medicinal plant species in Bantimurung-Bulusaraung National Park, Indonesia. Biodiversitas 23: 5539-5550. The people of Minasatene (Pangkep District) in Bantimurung-Bulusaraung National Park, South Sulawesi (Indonesia) have used traditional medicinal plants for many centuries. However, this local knowledge has not been recorded. This study aims to examine the diversity of plant species used to treat various ailments in the area. A set of semi-structured questionnaires was distributed to 60 respondents, consisting of traditional healers, elderly people, herb sellers and local people of Minasatene. This was used to obtain a demographic profile, vernacular names of plants and their uses, plant parts used, modes of preparation and methods of application. The diversity and use of the medicinal plant species in a traditional context were descriptively analyzed. The data were further extrapolated using Use Value (UV) to analyze the relative importance of plant species based on the number of uses; Fidelity Level (FL) to determine the specific uses of each plant species and preference over other species, and Informant Consensus Factor (ICF) to investigate an agreement between each group of diseases and related remedies. The study reveals that there were 74 species of medicinal plants belonging to 65 genera and 44 families being used as traditional medicines, which have been known to treat 54 ailments. Among the plant species, soursop (Annona muricata) had the highest UV (0,95), while the fig tree species, Ficus septica (100%) had the highest FL with the most frequent citation. From the data gained, it was found that skeletomuscular disorders had the highest ICF value (0,92).

Maleo birds (Macrocephalon maleo) have a habitat in the forest near the beach or in the middle of the forest near the hot springs. However, in Pekaloa Village, East Luwu District, Maleo birds are found in Lake Towuti, South Sulawesi, Indonesia. This, of course, shows that the habitat of Maleo birds on the shore of the lake is new information, especially on the island of Sulawesi. This research was conducted in Pekaloa Village, Towuti Sub-district, East Luwu District, South Sulawesi. This study was conducted to collect data on the characteristics of Maleo Senkawor (Macrocephalon maleo) nests. Secondary data collection began from scientific journals and works, as well as discussions with the surrounding community and the TWA manager of Lake Towuti, while primary data was obtained from measurements and direct observation in the field, as well as laboratory analysis. In this study, five nest plots with 13 nest holes were used, and the vegetation analysis used example plots measuring 175 m2. The characteristics of the nest pit have an average depth of 0.6 m, a surface diameter of 0.41 m, a temperature of 33.6oC, and a pH of 6.2. Vegetation analysis found eight species around the Maleo Senkawor bird's nest. Vegetation analysis used the density formula: highest frequency density 19.5 and highest frequency 0.57, lowest density 0.02 and lowest frequency 0.14. This study forms the basis of new information in the conservation of Maleo birds with the characteristics of the habitat conditions for nesting Maleo birds, especially in the coastal area of the lake.

Abstract. Amzeri A, Badami K, Santoso SB, Sukma KPW. 2022. Morphological and molecular characterization of maize lines tolerance to drought stress. Biodiversitas 23: 5844-5853. The early step in assembling varieties with early maturity, high productivity, and resistance to drought stress is to carry out morphological and molecular characterization among lines resistant to drought stress. The research aimed to characterize the morphology and molecular characteristics of maize lines resistant to drought stress to develop breeding and cultivation programs. The research used a randomized complete block design with twenty genotypes (maize lines) as treatment and three replicates. A total of 57 morphological characters and seven RAPD primers were used to assess the relationship based on morphological characters and RAPD markers. The results showed that the similarity coefficient based on the morphological characters was 0.69-0.97, while the similarity coefficient based on the RAPD marker was 0.64-0.95. The phenotypic diversity coefficient (PDC) was greater than the Genotypic Diversity Coefficient (GDC) in all observed quantitative characters. The broad sense heritability values of the tested maize lines ranged from 0.83 to 0.99. Production per hectare was significantly positively correlated with eight tested quantitative characters. G1 and G15 were the genotypes that can be used as parents to assemble hybrid maize varieties, while G10, G11, G16, and G17 could be used as maize for cultivation programs.

Physicochemical properties in NOR tomato line MA 131-6-3 after treated with ethephon and calcium carbide induced ripening. Biodiversitas 24: 3029-3037. Tomato fruit Postharvest Losses (PHL) from rotting, Physiological Weight Loss (PWL), inadequate nutrition, and other factors can lower fruit quality. Tomato is included as climacteric fruit which shows a significant increase in respiration and ethylene. The use of tomato with an extended shelf life due to genetic engineering, such as a Non-Ripening (NOR) tomato, can lower PHL. This experiment was carried out to evaluate physicochemical characteristics of NOR tomato fruit (lineMA 131-6-3 as result of selection F6)during postharvest after being treated by Ethephon (ET) (375, 750, 1,125, and 1,500 ppm), calcium carbide (CaC2) (5, 10, 15, and 20 gkg?1), and control. The experimental design was arranged in Randomized Completely Block Design with three replications.The ambient temperature was 28.30±1.75°C and relative humidity of 60.69±2.33%. The results showed the fruit ripening and physicochemical altered with CaC2 treatment, then followed by yellow but did not alter to the red color for all treatments. The value of reduction sugar, respiration, ethylene, PWL, and pH increased, meanwhile the fruit firmness decreased caused by CaC2. Furthermore, ET could increase rotting during storage.

Abundance, distribution mapping and DNA barcoding of Procontarinia robusta (Diptera: Cecidomyiidae), a mango gall midge in Bali, Indonesia. Biodiversitas 23: 6428-6436. Gall midge (Procontarinia robusta) is an important pest of mango plants in various countries in the world, including Indonesia. This pest causes very serious damage to mango leaves which until now has not been reported. This study aims to map the distribution and abundance of the pest population and identify the pest species using the DNA barcode method on mango tree plantations in Bali, Indonesia. The survey method was used to collect data from various districts and cities in Bali Province, Indonesia. Mitochondrial COI primers were used to identify DNA barcodes. The results showed that the highest population abundance of P. robusta was found in Denpasar City. These pests have spread evenly throughout the Province of Bali, from the lowlands to the middle and highlands. Through a molecular approach, the insect pest that causes mango leaf gall in Bali Province is identified as P. robusta as the first report that can be used by researchers, related agencies, and farmers to be alert and ready with strategies and control tactics in the future. Further research is needed related to monitoring using sex pheromones or plant volatiles and the search for natural enemies for monitoring purposes and initiation towards biological control.

Pathogenicity of entomopathogenic fungi isolated from Periplaneta americana against Dolichoderus sp.. Biodiversitas 23: 5461-5468. Dolichoderus sp. was found to cause a problem for plants, buildings, and households. Many efforts have been made to regulate such animals, but they have not always been successful. Biological control has gained significance in studies as an alternative to chemical control, in which fungi and nematodes are among the successful microorganisms in managing insects. This study aimed to understand the toxicity impact of cockroach-entomopathogen against Dolichoderus sp. Isolated entomopathogens were then administrated to Dolichoderus sp. colonies under laboratory conditions to assess their mortality efficiency. The fungal suspension was sprayed on the ant colonies with concentrations of 107, 108, and 109 conidia/mL. Aspergillus carbonarius KIF3, and Penicillium sp. KIF4 were identified as entomopathogenic fungi. In this investigation, 15 individuals of Dolichoderus sp. were employed in each treatment, with three replications. Pure distilled water with 1% molasses and 0.1% Tween 80 was used as a positive control, whereas sterile distilled water was used as a negative control. The result showed that the entomopathogenic fungus Aspergillus carbonarius KIF3 significantly affected mortality in black ants when administered 109 conidia/mL conidial suspension. It was also observed that the presence of the Dolichoderus sp. population in nature is often associated with mealybugs. There is an opportunity to use an ecological control approach in black ant populations by utilizing entomopathogenic fungi.

Abstract. Erniati, Erlangga, Andika Y, Muliani. 2023. Seaweed diversity and community structure on the west coast of Aceh, Indonesia. Biodiversitas 24: 2189-2200. One of the commodities that continues to be developed by the Indonesian government in fisheries is seaweed. The potential of seaweed as a food ingredient in Aceh is very abundant. However, diversity and community structure of seaweeds on the west coast of Aceh have not been studied much. Therefore, this study has been carried out to assess the diversity and community structure of seaweeds on the west coast of Aceh based on transect method. Sampling has been carried out in all districts/cities (6 districts and 1 city) of west coast of Aceh. Altogether, a total of 50 species have been identified; of which, maximum number of species belongs to Phaeophyta (21 species) followed by Chlorophyta (16 species) and Rhodophyta (13 species). The estimated value of diversity (H'), uniformity (E), and dominance (D) index is 1.02, 0.50 and 0.51 respectively. Based on the univariate diversity indices analysis, it has been found that seaweed on the west coast of Aceh is in the moderate category. Furthermore, the water quality parameters revealed the existence of conducive environmental conditions for the growth and life of seaweed. So, it is proposed that use of appropriate seaweed culture techniques can be carried out on the west coast of Aceh to able to make Aceh a seaweed center in Indonesia.

Abstract. Nairfana I, Nikmatullah A, Sarjan M, Tandeang A. 2022. Variability of secondary metabolites from leaves of Ziziphus mauritiana obtained from different locations in Sumbawa, Indonesia. Biodiversitas 23: 4948-4958. Bidara (Ziziphus mauritiana L.) is an evergreen shrub that grows in many parts of the world, and is an important plant in arid and semi-arid regions for the environment, food, and health. The plant has been used traditionally as medicine, and more recently, growing scientific evidence demonstrated the active compounds of bidara leaves. However, little is known about the plant's secondary metabolite profiles in different environments. This research aimed to characterize the secondary metabolites profile of bidara leaf obtained from the different habitats of semi-arid Sumbawa Island, Indonesia, and to evaluate their relationship based on cluster analysis of the secondary metabolite diversity. The leaf samples were collected from bidara plants obtained in 3 districts on Sumbawa Island: West Sumbawa (SB), Sumbawa (KS), and Dompu (DP) Districts, each from different ecosystems: lowland coastal/savanna, lowland near residential areas, and mid-highland areas of residential or forest-edge. The leaf samples were extracted by maceration with multilevel extraction using solvents with decreased polarity (h-hexane, ethyl acetate, ethanol 70%, methanol, and distilled water, respectively), followed by phytochemical screening for the presence of alkaloid, flavonoid, saponin, and phenolic. The results showed variation in the phytochemical components of bidara leaves obtained from a different environment, with secondary metabolites of DP leaves being more diverse than KS and other SB. All samples contained polar alkaloids and flavonoids, while the DP1 samples also contained non-polar flavonoids. The SB1 and DP samples also contained saponin. Interestingly, phenolic was only detected in the polar extracts of DP1 leaves. Cluster analysis based on the secondary metabolites profile distinguished the SB1 and DP1 bidara from others. Further study is required to understand if the variation is resulted from bidara adaptation to a different environment or also due to genetic variation of the bidara in Sumbawa Island.
