Biodiversitas 24: 1117-1124. The Next Generation Sequences (NGS) is one of the established metabarcoding methods for analyzing microbial communities. Investigation of the cyanobacterial community in eutrophic freshwater habitats is important, especially for those known as toxic-released cyanobacteria. This study aimed to analyze the structure community of cyanobacteria in Agung and Sunter Barat Lakes in North Jakarta, DKI Jakarta Province using the NGS method. The physico-chemical parameters were also measured. Sampling plots were selected by purposive sampling method. The result of the study successfully analyzed the bacterial community structure and portrayed the dominancy of the cyanobacterial population on the two lakes. Several cyanobacteria genera previously reported in 2008 research were found, including Arthrospira and Planktothrix. The diversity index in Agung Lake (4.7) was higher than in Sunter Barat Lake (3.7). Two dominant populations were found, which were Raphidiopsis (in Agung Lake) and Planktothrix (in Sunter Barat Lake). Both genera have been acknowledged as toxin-releasing cyanobacteria, which could be harmful to the water-biotic community as well as human health. The presence of potentially toxic cyanobacteria in the recreation area of Agung Lake should be taken into consideration for biomonitoring management.

Title The effect of different honey concentrations on the ultrastructure profile of spermatozoa in Dewa Mahseer (Neolissochilus soro). Biodiversitas 24: 1025-1031. Dewa Mahseer (Neolissochilus soro) is one of Indonesia's endemic freshwater fish at risk of population decline due to water pollution and asynchronous maturation of gonads. One of the efforts to conserve this species is preserving their spermatozoa using honey as a protector at certain temperatures. Therefore, this study aims to evaluate the effect of multiflora honey concentrations on ultrastructure spermatozoa profile of Dewa Mahseer. The temperature was maintained at 4°C and stored for 48 hours, followed by the cement dilution (1:10) in a stock solution containing fish ringer and honey. Transmission electron microscopy (TEM) examined the spermatozoa profiles at a magnification of ×1,500. Furthermore, ANOVA one-way analysis of variance was conducted to test the effect of head length, width, and area of spermatozoa on honey. The results showed a statistically significant difference in head width (F=13.929, p=0.001). Also, there is a difference in area between honey spermatozoa, fresh spermatozoa, and without honey at a cold of 4°C. The fresh spermatozoa stored at cold temperatures without honey observed considerable membrane damage. The novel aspect is an investigation of the effect of honey as a natural protector in the storage of Neolissochilus spermatozoa at 4°C for 48 hours. This study can be applied as a guide for the 48 hours of spermatozoa in a cold temperature in fish farming.

Short Communication: Leopard cat (Prionailurus bengalensis) distribution in the Cisokan Hydropower Plant, West Java, Indonesia. Biodiversitas 23: 6247-6253. Globally, leopard cats (Prionailurus bengalensis) can be found in various land cover types. Information about leopard cats is still low in Indonesia, especially in Java. Java Island has high human activities that impact leopard cats, such as the UCPS hydropower development area, a non-conservation area. The UCPS hydropower plant is in the upper catchment area of ??the Cisokan River as a tributary of the Citarum River. Although leopard cats can adapt to various land cover types, monitoring their existence is necessary. This study aims to reveal the distribution of leopard cats in the UCPS hydropower development area, Cianjur Regency and West Bandung Regency, West Java, Indonesia. Sign survey and literature were applied in this study. Fifty-eight leopard cat findings were found in the UCPS hydropower area. Leopard cats were found in 16 of the 47 grids from 2012-2022. After 5-10 years, leopard cats still use the same area in certain areas to meet their needs.In eight of 16 grids, 50% of the grids used by leopard cats were habitats used repeatedly. They were most commonly found in shrubs (39%), riparian (20%), and pine plantations (15%). Leopard cats were located at 445-895 masl and are most commonly found at 400-800 masl. Leopard cats are found in the UCPS development area and Perhutani Land. After 5-10 years, leopard cats still use the UCPS development area, so the habitat still fulfills their daily needs. Further monitoring is needed to determine the ecological status of the leopard cat population during the construction and post-construction process.

Abstract. Widodo CJ, Taufik M, Khaeruni A, Mallaranggeng R. 2023. Determination of Begomovirus on chili plants (Capsicum sp.) in Buton and Muna Islands, Southeast Sulawesi, Indonesia. Biodiversitas 24: 741-751. Begomovirus disease is one of the main factors inhibiting chili cultivation which could harm plants and result in crop failure. This study aimed to determine the symptoms of begomovirus disease in chili plants in Buton and Muna, Southeast Sulawesi. The methods used are disease incidence survey, insect vector observation, molecular identification using the polymerase chain reaction (PCR) technique, and Sanger sequencing (Applied Biosystem 3500). The survey was conducted in Southeast Sulawesi province, covering Bau-Bau City, Buton Regency, Central Buton, North Buton, Muna, and West Muna. The results showed that the incidence and severity of Begomovirus disease were observed highest in Buton islands, 73% and 49.5% (Buton City), followed by North Buton, Central Buton, and Buton, with the incidence and severity of Begomovirus, respectively, of 71.74% and 59.5%; 69.32% and 51.5%; and 68% and 61.5%. While in Muna Regency, 63.16% and 38.5%, West Muna, 73.79% and 53.50%. On the other hand, the highest percentage of the whitefly population was in Bau-Bau City, 24%, while the lowest was in West Muna, 15.5% supporting this observation. Furthermore, PCR analysis successfully amplified symptomatic isolates from Buton and Muna islands at a band length of 580 bp. On the other hand, Sanger sequencing analysis revealed the existence of the Indonesian Pepper Yellow Leaf Curl Virus (PepYLCIV) with more than 96% genetic homology to the Gene Bank data of MN094866 PepYLCIV Capsicum annuum Indonesia and MN738463 PepYLCIV Chili pepper Indonesian. This information is the first report of the presence of Begomovirus on the two islands.

Rhizoctonia solani J.G.Kühn and Xanthomonas oryzae are two main pathogens in rice plants that cause sheath blight and bacterial leaf blight disease. Controlling these two diseases using plant extracts is an alternative, environmentally friendly method. Plant extracts are known to have the ability to inhibit microbial growth. The purpose of this study was to determine the ability of Kaempferia galanga L. (aromatic ginger) extract to inhibit the growth of R. solani and X. oryzae in-vitro and to determine the antimicrobial compounds contained in the extract. First, K. galanga was extracted using a maceration technique with 96% ethanol. Next, followed by an antifungal test on R. solani with a diffusion well technique and an antibacterial test with a disc agar technique against X. oryzae using two types of solvents (water and 96% ethanol) to obtain a concentration gradient (2, 4, 6, 8, and 10%). Then, the content of K. galanga compounds was examined using GC-MS. The results showed that K. galanga could inhibit the growth of pathogenic fungi and bacteria, as indicated by the formation of an inhibition zone. Chemical compounds such as 2-propenoic acid, 3-(4-methoxyphenyl)-, and ethyl ester are the dominant compound in K. galanga. In addition, several compounds with antimicrobial activity were found in the K. galanga extract, including germacrene-D, 1,8-cineol, borneol, caryophyllene, jasmone, and heptadecane.

Abstract. Zahara Y, Suhartono S, Nirliana N. 2022. Abundance and antimicrobial susceptibility of Shigella spp. isolated from aquatic environments in Banda Aceh, Indonesia. Biodiversitas 23: 6420-6427. Accumulating pathogenic bacteria in aquatic systems increases waterborne infections, including shigellosis. The infection is causatively associated with Shigella spp. within the water system. This study aimed to isolate and determine the abundance of Shigella spp. collected from three rivers in Banda Aceh, Indonesia, namely Krueng Lamnyong, Krueng Aceh, and Krueng Neng water, and their antibiotic susceptibility. The bacterial enumeration was conducted using the MPN method followed by bacterial isolation using XLD and SSA selective media, whereas antibiotic susceptibility was evaluated using the standards of CLSI. Results showed that there was no significant difference (p=0.059) in the bacterial abundance among rivers i.e., 4.67 MPN/mL in Krueng Lamnyong, 27.33 MPN/mL in Krueng Aceh, and 18.33 MPN/mL in Krueng Neng. In total, 19 isolates of Shigella spp. were isolated, of which four isolates were from Krueng Lamnyong, nine isolates from Krueng Aceh, and six isolates from Krueng Neng. Antibiotic susceptibility test showed that the isolates had resistance to tetracycline, penicillin, and nalidixic acid. More than half of the isolates were resistant to more than five antibiotic classes. The study indicates that rivers are the main reservoir for the transmission of pathogenic bacteria in aquatic systems and some efforts are needed to reduce potential health risks and ensure water quality in aquatic systems.

The ecology of suweg (Amorphophallus paeoniifolius) in the Citanduy and Cimanuk Watershed, West Java, Indonesia. Biodiversitas 23: 6520-6529. Suweg (Amorphophallus paeoniifolius (Dennst.) Nicolson) is one of the plants distributed in Indonesia, including in the Citanduy and Cimanuk Watershed. Amorphophallus grows in various land cover types. This species exhibits wide agroecological adaptation to dry and moist lands. Citanduy Watershed passes through the Leuwikeris Dam, and the Cimanuk watershed passes through the Kertajati International Airport. These projects can affect the condition of the surrounding environment, including biodiversity. This study was conducted to observe the distribution, population, and habitat characteristics of the suweg in the Citanduy and Cimanuk watersheds. Exploratory surveys and intensive studies were applied in this study. An exploratory survey was conducted randomly through direct observation. An intensive study was carried out to determine the ecological description of suweg in several areas, namely Kuta Subvillage, Ciangir Subvillage, and Pasiripis Village, with existing projects, still have strong local cultures, and the discovery of suweg growing. Suweg is growing in several sites, including the Citanduy and Cimanuk Watersheds, with certain elevations and environmental conditions. The Summed Dominant Ratio (SDR) of suweg in Kuta Subvillage, Ciangir Subvillage, and Pasiripis Village in the home garden and garden, respectively, are 8.50 and 13.77; 10.07 and 19.25; and 0 and 22.92, which indicate the population or density of suweg is low. The abiotic and biotic environmental parameters observed at each site varied.

Abstract. Hamzah N, Husna, Ruslin, Arba M. 2022. The application of medicinal plants in the local community of Gantara Forest, Southeast Sulawesi, Indonesia. Biodiversitas 23: 6557-6563. The local community around Forest Management Unit (FMU) Gantara (Gantara Forest) has traditionally utilized medicinal plants, especially people in Liwu Metingki Village of Muna District, Southeast Sulawesi Province, Indonesia, yet the least information on its application has been recorded. This study investigated the traditional application of medicinal plants by the local community nearby Gantara Forest, specifically Liwu Metingki Village. Exploratory research was implemented using the multi-method approach of field observation and a semi-structured interview with 17 traditional practitioners living around Gantara Forest. The study successfully collected and identified 34 potential medicinal plants in 20 families to treat 21 diseases. Most plants have a habitus of the tree (39%), which 67% of the local community apply as traditional herbs. Boiling has become the most popular practice in using medicinal plants of the local community (45%) to obtain benefits from the plants against internal diseases (37%). This finding demonstrates the vital role of sustainable management of Gantara Forest for human health. With this finding, the community is strongly expected to understand the significant role of medicinal plants, so they can attempt to protect the plants by, for example, planting them in their yards.

Biodiversitas 23: 6254-6261. Characterization is an important activity for the study of genotype diversity and similarity. The study was aiming at obtaining information about the diversity and similarity of melon groups and to determine morphological characters as differentiators between melon groups. The genotypes tested were IPB240, IPB283, IPBM21, IPBM23, IPBME5, IPBMETA9, UME20, UME38, UME39, UME90, UME91, UME98, UME99, UME100, UME101. Characterization was based on UPOV and IPGRI guidelines for qualitative and quantitative characters. Two qualitative characters showed no variation in all tested genotypes, namely sex expression (EK) and secondary skin color outside the groove (WKSDA). Qualitative characters that have high diversity were groove color (WA), maximum width between grooves (LMAA), groove color intensity (IWA), and flower stalk thickness (KTBN). The UME98 genotype had the most different color appearance from the other genotypes on the heatmap. The color and depth of fruit grooves are distinguishing characteristics of all genotypes. There were genotype differences in the inodorus and reticulatus types. Unlike the IPBMETA9 genotype, UME100, UME101, and UME38 formed the same clusters with the characteristics of the makuwa group. Melon group makuwa had different characteristics with the inodorus and reticulatus groups. The distinguishing characters for the makuwa type were groove depth, ratio length to diameter, the diameter of hermaphrodite flower stalks, fruit diameter, and fruit skin thickness. Melon groups in the inodorus and reticulatus groups had high similarities, making it difficult to find specific characters in all melon genotypes observed.

The local knowledge and potential of suweg (Amorphophallus paeoniifolius) in the Citanduy and Cimanuk watersheds of West Java, Indonesia. Biodiversitas 24: 370-378. The Indonesian archipelago is rich in natural resources and ethnic diversity. Each ethnic group interacts with the surrounding environment in ways that often differ. Interaction with the environment creates local knowledge that is complex in terms of language, practical understanding, beliefs, and social interaction. One useful natural resource that grows in many areas of Indonesia, especially in watershed ecosystems, is Suweg (the elephant foot yam) which has the scientific name Amorphophallus paeoniifolius (Dennst.) Nicolson. Local communities, such as in the Citanduy and Cimanuk watersheds of West Java, use suweg for various purposes in their lives. The study reported here aimed to document the local knowledge and development potential for suweg in the Citanduy and Cimanuk watersheds, West Java, Indonesia. The research used mixed observation methods, predominantly qualitative. The data collection technique was purposive, based on interviews (structured, semi-structured, and in-depth interviews) with several informants or community groups who were considered competent to know suweg. Data were analyzed through emic and ethical approaches. Data analysis was conducted by cross-checking, summarizing, and synthesizing. The results of the research showed that suweg grows in a range of environmental conditions on managed agroecosystem lands. The corms of suweg are peeled, cooked and eaten in a variety of side dishes. The leaves of suweg have been used as food for fish in pond-system aquaculture. The results of this study suggest that suweg has the potential for expanded development, based on sociological and ecological evidence.
