Abstract

Abstract. Al-Darraji ZDG, Mohammed-Ameen MKM. 2023. Phenotypic and genotypic diversity of yeast isolated from cancer patients and their environment and their antifungal sensitivity pattern. Biodiversitas 24: 4166-4174. This study aimed to investigate yeast diversity and their antifungal susceptibility patterns in 853 samples collected from cancer patients, their apparently healthy companions, their beds, their tables, and indoor hospital air. The samples were cultured and identified using classical phenotypic characteristics and a molecular method utilizing amplification of ITS conservative regions of rRNA. The sum of phenotyping and genotyping identification revealed that 102 yeast isolates included 6 genera and 14 species. Candida sp. was the most dominant genera (84.31%), followed by Naganishia sp. (10.78%). Candida albicans (24.50%) and C. krusei (16.66%) were the most prevalent yeast species. Fluconazole, caspofungin, and voriconazole exhibited potent antifungal activities against the most yeast species with low minimum inhibitory concentration (MIC) values compared with high MIC (64 µg/mL) values against Meyerozyma guilliermondi, Naganishia difflunes, and C. tropicalis. To conclude, the antifungal activity was isolate- and species-specific and the higher antifungal concentrations led to a more-rapid expression of activity. The high isolation rate of yeasts from beds and tables, advocated the use of various monitoring systems to ensure thorough cleaning and consistent disinfection of surfaces around patients and health care providers. The study findings showed that the molecular method was superior to the phenotyping technique in identifying yeast isolates. While similar research has been performed in different locations, the current project represents a unique milestone for this particular institution, and offers an opportunity to advance research in this area.

Abstract. Mascari?as HJC, Otadoy JB. 2023. Economic valuation of seagrass ecosystem in Maribojoc Bay, Bohol, Philippines. Biodiversitas 24: 4427-4435. The seagrass ecosystem is one of the most productive and precious ecosystems. In the Philippines, however, more data on the economic valuation of all seagrass ecosystem services must be collected. The study estimated the total economic value of the seagrass ecosystem, including use and non-use values. Resource user groups include municipal ?shers, gleaners, and tourism business operators. The sample size was determined based on the total number of resource users in Maribojoc Bay, Bohol, Philippines and using a confidence coefficient of 95% (P = 0.05). The total economic value of the seagrass ecosystem in Maribojoc Bay is PhP 52.8 million (US$ 1.1 million). Direct use values, or market benefits, are PhP 35.4 million (US$ 708 thousand), accounting for 67% of the total economic value. Indirect and non-use values, are PhP 17.3 million (US$ 346 thousand), accounting for 33% of the total economic value. On a per hectare basis, the economic value of the seagrass ecosystem in Maribojoc Bay is PhP 46,722 (US$ 934). The results of this valuation can be an input in decision-making to manage and conserve the seagrass ecosystem as well as maintain the economic security of Maribojoc Bay. The findings of this study are valuable for developing a comprehensive information management and decision-support system for the management and protection of the seagrass ecosystem. However, more research is required to evaluate the range of ecosystem services provided by seagrass ecosystems across the country, focusing on qualitative value attributions linked to the social-ecological dynamics of seagrass systems.

Characterization of type I L-asparaginase encoding gene from the thermohalophilic bacterium Bacillus subtilis CAT3.4 from Wawolesea Hot Spring, Southeast Sulawesi, Indonesia. Biodiversitas 24: 5167-5178. This study aims to determine the molecular characteristics of the gene encoding type 1 L-asparaginase from CAT3.4 thermohalophilic bacterial isolates from the hot springs of Wawolesea, North Konawe, Southeast Sulawesi. This enzyme can be used as a cancer therapy agent and prevents the formation of acrylamide in food products. It is a kind of exploratory research. The characterization was done by amplifying the ansA gene sequence encoding L-asparaginase from CAT3.4 isolate by polymerase chain reactions (PCR) technique using primers AsnBac1-F1 (5'-ACGCGATATTTCTTTTGGCCGG-3') and AsnBac1-R1 (5'-CAGTGAAGAGGTGCATGGTATG-3'). The amplified PCR product was used as a template for sequencing by the Sanger method. The amino acid coding regions (CDS) obtained were bioinformatically characterized using the NCBI website for BLASTn analysis, restriction site and hydrophobicity profiles were analyzed using the BioEdit program, phylogenetic tree was analyzed using the MEGA X program, and type 1 L-asparaginase amino acid sequence was analyzed using the Expasy translate program. The characterization results showed that the target gene has a high similarity to the ansA gene sequence of 20 Bacillus subtilis strains, i.e., 99-100%, is closely related to the ansA gene of Bacillus subtilis strain SRCM103629 and Bacillus subtilis strain GOT9, can be identified using restriction enzymes MluI and BstI to differentiate the species of organisms they produce, has CDS encoding 329 amino acids with a dominant composition of polar amino acids (54.1%) and has a hydrophobicity profile of amino acids dominated by hydrophilic regions. All these characteristics have confirmed that the characterized gene is the ansA gene encoding type 1 L-asparaginase from Bacillus subtilis. The finding of the ansA gene from Bacillus subtilis in a thermohalophilic region in Southeast Sulawesi, Indonesia is a novelty of the research.

Diet analyses of the pelagic thresher shark, Alopias pelagicus (Lamniformes: Alopiidae), from the Lombok Strait waters, Indonesia. Biodiversitas 24: 3708-3714. The pelagic thresher shark, Alopias pelagicus, is an endangered shark species estimated to be declining in the Pacific and Indian Oceans because of overexploitation, including from target fisheries and bycatch. Despite its importance in providing better information for species management, the ecological aspects of this species are still rarely studied. This study aims to provide information on the feeding ecology of the pelagic thresher sharks from the Lombok Strait. Stomach samples from 178 specimens were sampled from 2020-2021, and 149 stomachs that contained food contents were analyzed for species diet characterization. Prey Species Accumulation Curve slope values are <0.1 and the percentage of prey coverage for all sharks combined is 92.1%, suggesting reliable inventory in the number of prey represented. The resulting diet indexes suggest that A. pelagicus in Lombok Strait waters is a specialist predator (Ba=0.3) with few prey variations (H=2.21) and no competition between sexes and maturity stages of the species (C=0.64 and 0.81, respectively). The top three prey species found for A. pelagicus in the study area are the Auxis thazard (85.8 %IRI), Sthenoteuthis oualaniensis (8.5 %IRI), and Dasyscopelus spinosus (1.11 %IRI). A better understanding of the overlap of A. pelagicus and its diet, which includes economic fish commodities, will inform authorities to develop better fisheries and conservation management for the species in Indonesia.

Biodiversitas 24: 4175-4184. In agricultural practices, ensuring an adequate supply of potassium to plants is crucial for optimal growth and productivity. However, the exchangeable K is tightly bound to soil minerals such as mica, feldspar, and clay minerals, making it unavailable for plant uptake. K-solubilizing bacteria K can dissolve potassium from the mineral layer and be available to plants. The Previous study has found 11 bacterial isolates capable of solubilizing K in Aleksandrov solid media. This study aimed to select the best K-solubilizing bacteria for solubilizing K in soil and to perform molecular identification of these bacteria. A novel finding from this study is that specific KSBs enhance the levels of exchangeable K in the soil through various mechanisms, as evidenced by increased exchangeable Ca, Mg, and soil pH. Additionally, the research identified two newly discovered bacterial species capable of potassium solubilization: Paraburkholderia phymatum and Burkholderia paludis. Furthermore, the study suggests the existence of an unknown mechanism for K solubilization, indicated by the observed increase in soil pH during the process.

Effect of different extraction solvents on the yield and enzyme inhibition (a-amylase, a-glucosidase, and lipase) activity of some vegetables. Biodiversitas 24: 3320-3331. The present study investigated the in vitro inhibition of a-amylase, a-glucosidase, and lipase, and antioxidant activities (DPPH, metal chelating, and FRAP) of Allium cepa L., Apium graveolens L., Coriandrum sativum L., and Petroselinum crispum (Mill.) Fuss using solvent 80% ethanol, absolute ethanol, acetone, methanol, n-hexane, chloroform, and hot water. The total phenolic content (TPC) and the FTIR and GC-MS characterization spectra were compared with the activities. The TPC of 80% ethanol extract in A. graveolens L. was the highest among the extracts, with 23.78 mg GAE/g extract. The 80% ethanol extract exhibited the most potent antioxidant activity on DPPH radical scavenging (A. graveolens, 125.57 mg AEAC/g extract), metal chelating (C. sativum, 92.85 mg EECC/g extract), and FRAP activities (C. sativum, 46.98 mg AEAC/g extract). The 80% ethanol extract showed the highest anti-a-amylase (P. crispum, 30.61 mmol ACE/g extract) and anti-a-glucosidase (A. cepa, 595.28 mmol ACE/g extract) activities. In comparison, the absolute ethanol extract of C. sativum showed the highest anti-lipase activity (42.10% inhibition). According to the FTIR spectra, the 80% ethanol extract of the four green leafy vegetables is predicted to have several active compounds. GC-MS identified the compounds responsible for the activity. Studies on the recovery potential of active compounds as oral agents to treat diabetes mellitus with certain solvents are very useful in producing oral agents and functional foods to prevent diabetes mellitus.

Population genetic structure of the bentfin devil ray (Mobula thurstoni) in the South Indonesia Sea with limited sample based on ND5 gene. Biodiversitas 24: 3743-3749. Bentfin devil ray (Mobula thurstoni) is one of five Mobula rays with a worldwide distribution. This species has high economic value and play an essential ecological role. However, due to their biological characteristics, such as low reproductive rates and late maturity, these species are highly vulnerable to overfishing and slow to recover from depletion. Genetic information, such as this species's population structure and genetic diversity is limited. This study aimed to determine the genetic variation and population structure of M. thurstoni in the South Indonesia Sea based on mitochondrial DNA from ND5 gene. Twenty-two samples of M. thurstoni were successfully collected from three locations Palabuhanratu (n=5), Muncar (n=13), and Savu Sea (n= 4). The haplotype diversity (Hd) ranged from 0.782 to 1.00 (mean=0.839), and Nucleotide diversity (?) ranged from 0.004 to 0.018 (mean=0.007). Then, no significant population differences (P-value > 0.05) were observed between locations, indicating that M. thurstoni population in the South Indonesian Sea represents a single genetic population. Additionally, the haplotype distribution also indicated the presence of two shared haplotypes between locations. Consequently, co-management strategies for sustainable fishing and conservation must be implemented across these three locations to ensure the sustainability of this species in the nature habitat.

Abstract. Bahtiar B, Kasim M, Hati YIP, Ishak E. 2024. Population dynamics of mud clam Geloina expansa (Mousson, 1849) in degraded mangrove area at Kendari Bay, Indonesia. Biodiversitas 25: 616-623. Mud clams, Geloina expansa (Mousson, 1849), locally known as kalandue, are bivalves living in mangrove forests. These mud clams are currently experiencing ecological pressure, and are susceptible to the degradation of mangrove areas and the exploitation of the clams. This research was conducted in the mangrove forests of Kendari Bay for a year, from November 2016 to October 2017. Geloina expansa samples were collected throughout the mangrove forest area, and the water temperature was measured simultaneously with G. expansa sampling. The width of G. expansa shell was measured using a caliper. Population structure, growth, mortality, and exploitation rate data were analyzed using the Bhattacharya method, the von Bertalanffy inverse function, the width converted catch curve, and Pauly's empirical formula, all accommodated in the FiSAT II program version 3.0. The results showed that the G. expansa population was spread out from two and three-size groups, i.e., juvenile, adult, and old-size groups, dominated by the older group. Geloina expansa growth model follows the Lt = 10.08-(10.08-0.025) e-0.6t, which generally consists of 1.5-2.5 years of maturity. The total mortality (Z) of G. expansa was 2.26, in which the highest contributing factor was natural mortality (M) as opposed to fishing mortality (F), with values of M = 1.72 and F = 0.56. Thus, the exploitation rate (E) was within the low category, with a value of 0.24.

Abstract. Amelia T, Liliasari, Kusnadi, Aditiawati P. 2023. Isolation and characterization of potential indigenous bacteria from the former bauxite mining area for heavy metal reduction. Biodiversitas 24: 5096-5104. This study aimed to isolate, identify, and characterize some indigenous bacterial strains from the former bauxite mining area of Tanjungpinang, Bintan Island, Indonesia. Furthermore, this study focused on evaluating the potential of these bacteria for reducing the heavy metals lead (Pb) and chromium (Cr). The measurements of heavy metal concentrations at four sampling points, collection of soil samples as a bacteria source, and laboratory assessments for bio-removal capabilities were all conducted. Screening experiments were performed to identify bacterial strains resistant to heavy metal, using basic growth media such as Nutrient Agar (NA) and Nutrient Broth (NB) enriched with 100 ppm Pb and Cr metals. The reduction of heavy metal was analyzed with atomic absorption spectrophotometry (AAS), while the bacteria species were determined using MALDI TOF-MS. A modified version of the Kirby Bauer method was employed for toxicity reduction testing. Two bacterial strains were identified as Pb and Cr reducers and demonstrated resistance to both metals. Based on a 99.9% similarity value, the isolates were identified as Bacillus altitudinis (Isolate A) and Klebsiella pneumoniae (Isolate B), reducing Pb by approximately 72.7% and 34.5%, and Cr by 87.4% and 86.2%, respectively. The results indicated reduced toxicity in the metal-enriched media, with bacterial growth following three hours of incubation but no toxicity after 21 hours.

Biological aspect approach in sustainable management of coral catshark Atelomycterus marmoratus (Anonymous [Bennett], 1830) in Bali Strait, Indonesia. Biodiversitas 24: 5873-5882. Coral catshark (Atelomycterus marmoratus) is a shark species that thrives in many coral reefs. to evaluate the biological aspects of A. marmoratus and their type of growth curve for a time series to support their sustainability. Sampling was performed from August 23rd, 2019, to March 31st, 2020. The morphological analysis was performed by recording or photographing to reduce possible errors. Then, the length-weight relationship, sex ratio, length at first captured and matured, and spawning ratio were evaluated. The present study demonstrated that the coral catshark captured in the cohort that produces two cohorts only happens in February. The sex ratio is 41.6% male and 48.4% female, which is considered healthy, whereas the remaining percentage is so severely damaged that its gender cannot be determined. The FISAT-II software analysis results in a von Bertalanffy growth formula of Lt = 76.21 (1-e-2.01(t+0.0596)) with t max = 1.49/ year and L max = 77.39 cm. They grow steadily or tend to slow down after they reach the age of 1.5 years, with an asymptotic length of 77.39 cm. The Lc value (67.5 cm) is more significant than their Lm value (53 cm), indicating that their sustainability is highly possible to maintain because only fish that have passed the spawning stage are captivated. The spawning potential rate (SPR) is 35%, with M = 0.66, E = 1.85, and F = 0.78. These results indicate that the exploitation rate of coral catshark is moderate because 20% < SPR < 40%.  In conclusion, the A. marmoratus length-weight relationship is categorized as negative allometric, where the length grows faster than the weight.
