Abstract

Abstract. Rosidi, Suswoyo I, Mugiyono S, Ismoyowati, Tugiyanti E. 2024. Genetic variability of IGF1 and IGF2 and correlation to body weight in Kedu chicken of Indonesia. Biodiversitas 25: 4846-4852. This study aims to examine differences in genetic variability using Insulin Growth Factors (IGF) to estimate the body weight of selected Kedu chickens. A total sample 37 chickens consist of 10 White Kedu, 10 Cemani Kedu, 10 Red Kedu, 5 Black Tongue Kedu, 1 Blorok Kedu, and 1 Lurik Walik Kedu. Next, their blood is taken from the chicken samples and analyzed in the laboratory. The identification of the IGF-1 and IGF-2 genes polymorphism was done by PCR and sequencing methods. The results showed two Single Nucleotide Polymorphism (SNP) of IGF1: c.166 T>A and c.169 A>G and IGF2: c.248 G>A and c.540 C>T. The frequency of genotypes was informed on GG, GA and AA with values of 0.71, 0.1 and 0.19, respectively. The base pair of 540 showed the existence of genotypes CC, CT and TT with values of 0.28, 0.62, and 0.1 respectively. The value of heterozygosity obtained at each base pair was c. 248 and the heterozygosity value was 0.3648, while the heterozygosity value of c. 540 was 0.4838. The correlation analysis showed that at c. 248 had a value of 0.57, which was relatively greater than that at c. 540, namely 0.15. The c. 248 genotypes of AA had greater body weight than the rest. In conclusion, AA genotypes in the IGF-1 and IGF-2 genes had a relatively high body weight compared to that of other genotypes, but IGF-1 had a relatively low association (correlation), so incompatible as a marker for future selection. An IGF marker can be used to estimate the body weight of the upcoming offspring of Kedu chickens.

Antibacterial activities and molecular identification of endophytic fungi isolated from mangrove Avicennia marina. Biodiversitas 24: 6923-6933. This research explored the antibacterial potential of endophytic fungi from the Avicennia marina's (Forssk.) Vierh leaves, stem bark, and root, located in Blanakan, Subang District, West Java. The screening process for antibacterial activity potential was conducted using the Kirby Bauer disk diffusion method. Subsequently, the molecular screening results of the most promising fungi species were identified using Internal Transcribed Spacer (ITS) markers. The results revealed the isolation of 30 fungal isolates. Among these, seven endophytic fungi exhibited significant antibacterial activity against Staphylococcus aureus ATCC 29213 and Vibrio harveyi ATCC 5339, with inhibition zones ranging from 7.88±1.52 to 23.60±0.77 mm. Through molecular identification, 7 endophytic fungi were identified, including Penicillium chrysogenum, Cladosporium anthropophilum, Trichosporon asahii, Cladosporium sphaerospermum, Fusarium verticillioides, Meyerozyma carpophila, and Penicillium steckii. Notably, C. anthropophilum exhibited the highest inhibition zones against S. aureus and V. harveyi measuring 23.60±0.77 and 21.80±0.26 mm, and showed (Minimum Inhibitory Concentration) MIC values of 15.625±0.98 and 31.25±0.39 ?g/mL, respectively. In this study, endophytic fungi isolated from different parts of A. marina exhibited promising antibacterial activity, with Cladosporium anthropophilum from the stem bark showing the highest potency against S. aureus and V. harveyi, suggesting their potential as a source of antibacterial agents. To the best of our knowledge, this is the inaugural study uncovering the isolation and antibacterial potential of endophytic fungi from the genera Cladosporium, Trichosporon, Fusarium, and Meyerozyma. These fungi were extracted from the bark, leaves, and roots of A. marina, situated within the unique mangrove ecosystem of Blanakan Sub-district, Subang District, West Java, Indonesia.

Detection of Plasmodium in small ruminants in Yogyakarta, Indonesia, using a nested PCR assay. Biodiversitas 24: 6722-6726. Limited investigation of natural malaria infection in small ruminants in Yogyakarta needs to be explored. Plasmodium sp., as the causative agent of malaria, has been proven to infect ungulates, including small ruminants. This study aimed to investigate the presence of natural malaria infection in goats and sheep in Yogyakarta. A total of 303 blood samples were collected from goats and sheep in this study from four selected areas by a cross-sectional study. A microscopic examination detected the Plasmodium stage in a thin blood smear. Nested Polymerase Chain Reaction (PCR) performed molecular detection, targeting cytochrome b (cytb) as a gene target. It was found that no Plasmodium stage was detected in thin blood smears. However, three out of 70 blood samples from Girimulyo were Plasmodium-positive based on nested PCR assay. The results highlight that molecular assay by nested PCR is more sensitive than microscopic examination. Molecular analysis revealed natural Plasmodium infections in Ettawa cross-bread goats, with a proportion of 0.0429 (95% CI: 0.0089-0.1202). There was no statistical difference between sex and natural malaria infection in goats. This finding suggests that molecular analysis is necessary to detect natural malaria infection with extremely low parasitemia levels. Therefore, further study with larger sample sizes and broader geographical representation is needed to fully understand these malaria infections in goats and sheep.

Biodiversitas 25: 998-1006. Fipronil is a widely used active ingredient for controlling Nilaparvata lugens Stal. (Hemiptera: Delphacidae), but the effect exerted on fitness ratio and composition of endosymbiont microbes remain unknown. Therefore, this study aimed to investigate effects of insecticide by assessing fitness ratio and bacterial composition of endosymbionts in N. lugens before and after applying sublethal fipronil concentrations. A pair of LC15-equivalent fipronil-treated imago were introduced into treatment pots planted with rice 30 days after transplanting. Additionally, N. lugens endosymbionts were identified using full-length primers 27F and 1492R targeting the 16S rRNA gene. The results showed that fipronil application increased the total number of nymphs and fitness ratio of N. lugens in both IR64 and ciherang rice varieties. Similar observations were obtained with erythromycin application, although the differences were not statistically significant compared to the control. Fipronil application tended to cause an insignificant increase in the percentage of hatched eggs, while samples lacking this treatment contained abundant quantities of an endosymbiont, Acinetobacter soli. After applying insecticide, a significant alteration occurred in the composition of endosymbionts, with a substantial increase in Delftia acidovorans and D. lacustris, which both contributed to the enhanced fitness and tolerance of N. lugens to fipronil. These results showed the intricate interactions between bacteria and insects, as well as the mechanisms underlying insecticide resistance, thereby providing valuable insights for the development of new pest management strategies.

Mangrove gastropod distribution based on dominant vegetation classes and their relationship with physicochemical characteristics on fringe mangroves of Lembongan Island, Bali, Indonesia. Biodiversitas 25: 156-166. Mangrove ecosystems are important in supporting diverse marine life, offering refuge, reproductive grounds, and sustenance to various species. However, mangroves possess distinct zoning patterns determined by dominant species due to environmental variations that significantly impact the biodiversity of biota, particularly gastropods. A study on the Indonesian island of Nusa Lembongan revealed distinct communities of coexisting gastropod species in each mangrove vegetation zone. We discovered 29 distinct species of mangrove gastropods, with the Sonneratia mangrove zone having the most and the Lumnitzera zone having the least. Certain species, including Amphidromus perversus, Cerithium adustum, and Echinolittorina sundaica, were detected in only one of the mangrove zones. In each mangrove zone, the composition of gastropod species changed depending on the substrate type, e.g., soil, mangrove trunks, or mangrove leaves. The Shannon-Wiener diversity index values range from low to high diversity, and the community structure index values in mangrove zones vary. The evenness index varied by region, with Avicennia, Bruguiera, and Sonneratia having the highest index value. The findings indicate that mangrove vegetation zones are essential for maintaining diverse gastropod populations, with distinct composition and abundance patterns influenced by the distance of each mangrove zone from the ocean. This data may influence conservation efforts to preserve mangrove habitats and the diverse array of organisms that depend on them.

Abstract. Daminova N, Nosirov S, Akbarov F, Tojibaev K, Temirov E. 2024. Distribution and conservation of the narrow sub-endemic shrub, Lonicera paradoxa in Pamir-Alay, Central Asia. Biodiversitas 25: 439-448. Lonicera paradoxa Pojark. (Lonicera ser. Alpigenae Pojark. ex A.Byalt) is a shrub with a geographically limited range. This species is included in the national red book lists of Uzbekistan, Kyrgyzstan, and Tajikistan and is considered one of the rare species in danger of extinction in nature. The species grows in a significant elevation from 1600 to 3200 m on the Northern slopes of the Alay Range, Central Asia. In this study, we investigated the geographic distribution of L. paradoxa based on scientific literature, herbarium data collections, and field surveys. We then modeled the current and potential future geographic distribution using Maxent. We also conducted a propagation study for its conservation under laboratory and greenhouse conditions. Our fieldwork in 2021-2022 found new records of L. paradoxa in Uzbekistan and Kyrgyzstan. We calculated the Extent of Occurrence (EOO) and Area of Occupancy (AOO) of the species, which were 5.08 km2 (VU) and 28 km2 (EN), respectively. Therefore, based on the IUCN Red List and Criteria, we recommend the conservation status of L. paradoxa as endangered (EN) in categories B2 ab (ii, iii, iv) + D. Our modeling prediction suggested that compared to the current, suitable habitat of L. paradoxa to reduce significantly in the future with very suitable habitats are expected to expand to the south and high mountain areas. For the first time, the conservation measures of L. paradoxa were carried out at the Tashkent Botanical Garden. The conducted research made it possible to preserve L. paradoxa in ex-situ conditions. The results of this research can be helpful in determining and planning measures for the protection of L. paradoxa in the territory of the Kyrgyz and Tajikistan Republics, both now and in the future.

Mangrove ecosystems have the potential to be used as objects of marine ecotourism, in addition to many ecological functions and one of the main producers in marine fisheries. Bangkalan has a mangrove area of 1500 hectares which has the potential to be used as ecotourism. However, there is no research that analyzes and identifies the suitability index of mangrove tourism along the north coast of Bangkalan, Madura. The purpose of this study was to determine mangrove species, mangrove density, and mangrove tourism suitability index on the north coast of Bangkalan in order to identify the potential of mangrove for eco-tourism. The research was done at 14 stations on the north coast of Bangkalan. Data calculation and analysis were carried out by calculating the thickness of mangroves forest, mangrove species, mangrove density, biota that can be found, tides, accessibility, and infrastucture that were used to calculate mangrove tourism suitability index. The results showed that mangrove thickness in the north coast of Bangkalan were 138 m-864 m. 17 mangrove species were found, with the highest number of species was found in site 13 (Labuhan Education Park) with 14 species. Mangrove density was between 400-3500 trees/ha, with 10 sites in dense category, 2 in average category, and 2 in low category. The results showed that all research sites were included in the sufficient criteria for mangrove tourism. Mangroves in Bangkalan have great potential to develop mangrove tourism since all research sites were included in the sufficient criteria for mangrove tourism.

Abstract. Wahyudi A, Sari MF, Rajamin I, Arsri M. 2023. Evaluation of proximate analysis and yield production of watermelon in Lampung, Indonesia. Biodiversitas 24: 6010-6015. The breeding of watermelon plants is carried out to produce new superior varieties. This study aimed to test the moisture, fat, carbohydrate, ash, and vitamin contents, and the yield potential of a hybrid watermelon line (F1). The materials used were 6 hybrid watermelon and 2 commercial lines as control varieties. A randomized block design was adopted in this study and was analyzed by LSD test, dendrogram, and proximate analysis. The result showed that the 6 hybrid watermelon lines had high yield potential. Furthermore, the moisture content of the fruit of the WM 2110-1204 lines (92.24%) was higher than other lines. Proximate data analysis showed that the WM 2110-0616 line had a higher ash content (0.16%), and the carbohydrate content of the fruit of the WM 2110-0806 line (3.64%) was also higher. The vitamin C-vitamin content of the fruit of the WM 2110-0308 line (0.36 mg.g-1) was higher, while the 4 watermelon lines, namely WM 2110-0308, WM 2110-204, WM 2110-0806, and WM 2110-1606 had advantages in terms of skin color and striation, fruit sweetness level and weight. In a dendrogram analysis, qualitative characters were divided into two clusters, with clusters I and II containing 8 and 1 lines, respectively.

Identification of blaSHV and blaTEM extended spectrum beta-lactamase genes in Klebsiella pneumoniae in the dairy wastewater, East Java Province, Indonesia. Biodiversitas 24: 6092-6099. Klebsiella pneumoniae infection in dairy cows causes various livestock production hazards and threatens public health because it acts as a Multi-Drug Resistance Bacteria (MDRB), especially Extended-Spectrum Beta-Lactamase (ESBL) strains. Livestock wastewater is a repository for commensal isolates including K. pneumoniae and potentially can be a reservoir for ESBL spreaders for the environment. The aim of this study was to identify the blaSHV and blaTEM ESBL genes in K. pneumoniae from wastewater of dairy cattle farms in East Java Province. This study used a sample of 342 dairy farm wastewater isolated on 2% BPW media from 6 cities/regencies with the highest population of dairy cows in East Java, Indonesia, namely Pasuruan District, Malang District, Tulungagung District, Blitar District, Batu City and Kediri District. Samples were identified on MCA media, Gram staining, EMBA, and biochemical tests. Confirmation of ESBL phenotypically used the Double Disc Synergy Test (DDST) method and the Polymerase Chain Reaction (PCR) test to detect the ESBL gene. The result showed that bacterial isolation and identification were 14.32% (49/342), confirmed K. pneumoniae. DDST confirmation results of ESBL-producing K. pneumoniae isolates from the wastewater of dairy farms in East Java obtained were 5.55% (19/342). The sample was continued with the PCR test showing positive for the blaSHV gene 63.1% (12/19) and blaTEM 31.57% (6/19). In conclusion, the ESBL gene blaSHV and blaTEM identification in K. pneumoniae isolates, indicate that wastewater from dairy farms has the potential to be a reservoir of ESBL carriers in the environment and pose a threat to public health in East Java Province, Indonesia.

Abstract

Abstract. Mahulette F. 2023. Microbiological and chemical characteristics of porkfish inasua, traditional fish fermented from Maluku, Indonesia. Biodiversitas 25: 1-8. Porkfish inasua consisted of two types, i.e., with and without coconut sap. The research aimed to analyze the microbiological and chemical characteristics of two types of porkfish inasua. The microbiological characteristics are used to determine the role of microbes and product safety whereas the chemical characteristics, including proximate composition, amino acids, and fatty acids contents can determine the nutritional value for consumption. The sample of porkfish inasua was taken from traditional producers in Layeni, Teon, Nila, and Serua (TNS) Islands, Maluku, Indonesia. The microbiological analysis was done using the plate count method. From the measurement, the total number of halotolerant and coliform bacterial in porkfish inasua without sap were 6.2 log cfu/g and 6.1 log cfu/g, respectively. In comparison, the total of Bacillus and related genera in porkfish inasua with sap was 5.9 log cfu/g at the end of fermentation. The fat content of both types of inasua was around 17%. The total amino acids and fatty acids contents of porkfish inasua without sap at the end of fermentation were 15.31 and 57.08%, while porkfish inasua with sap were 15.85 and 63.81%, respectively. The dominant bacteria found in porkfish inasua without sap was Staphylococcus saprophyticus, while porkfish inasua with sap was Bacillus cereus. These two bacteria play a role in fermentation so porkfish inasua was safe for consumption. Generally, chemical characteristics of porkfish inasua with sap were better than those of porkfish inasua without sap, but statistically, it was not significantly different. This research can improve the quality of porkfish inasua as local fermented fish products in Maluku.
