Antibiotic production by an endophytic Streptomyces isolated from the medicinal plant Poikilospermum suaveolens. Biodiversitas 25: 2221-2229. This study investigates the antibiotic production potential of endophytic actinomycetes isolated from Poikilospermum suaveolens plants. Samples were collected from various parts of the plant in karst ecosystems. Endophytic actinomycetes were cultured and screened for antimicrobial activity using agar diffusion and liquid microdilution methods. One of the 11 isolated strains exhibited significant antibiotic activity against clinically relevant organisms identified as belonging to the Streptomyces genus, specifically Streptomyces mirabilis strain NRRL ISP-5169. Submerged fermentation of this strain over 9 days showed strong antagonistic effects against Gram-positive bacteria. Spectroscopic analyses confirmed the presence of actinomycin D as the bioactive compound. This study provides on the previously unexplored potential of endophytic actinomycetes from P. suaveolens as a promising resource for antibiotic discovery.

Water quality assessment of river based on phytoplankton biological integrity index in rural areas of the upstream Citarum River, West Java, Indonesia. Biodiversitas 25: 881-889. The deterioration of water quality in the upstream Citarum River is one of the consequences of agricultural practices characterized by fertilizer and pesticide runoff that disrupts the ecological balance of aquatic ecosystems. The research area was on agricultural land use in the Cihawuk segment, Kertasari, Bandung District. The present study attempts to community structure and assess the water quality using the biological indexes of rural areas in upstream Citarum River through phytoplankton studies. A purposive sampling technique was used to collect data on physico-chemical parameters and phytoplankton community structure from stations selected on the basis of agricultural land use. Furthermore, biological indexes such as pollution levels based on the Shannon Wienner diversity index (H'), Palmer algae index, and Diatomeae index are examined using the data community structure of phytoplankton. The findings show that the abundance of phytoplankton in rural areas is 4,105 cells/L. The community structure of phytoplankton was dominated by Coelastrum sp. and Trachellomonas sp. The Palmer algae index ranges from 23-26, indicating that the water was a highly organic pollutant. The pollution level of upstream Citarum River based on the diversity index shows moderately polluted water. Meanwhile, the diatomeae index showed less than 0.2, which sites water bodies were mesotrophic. Phytoplankton and their indexes serve as valuable tools for evaluating the quality of the water environment.

Rose apple (Syzygium samarangense) exhibits a rich diversity in its varieties, metabolic compounds, and biological activities. Genetic diversity studies are crucial in understanding plant species taxonomy and evolutionary history. They provide a roadmap for researchers to trace the relationships between different varieties and offer insights into the evolutionary processes that have shaped the diversity within the species. Many traditional or indigenous varieties of plants may harbor unique traits adapted to specific local conditions. This study, therefore, holds immense value as it aims to evaluate the genetic relationship among rose apple varieties in Vietnam using morphological characteristics and ISSR markers. Sixteen local varieties of rose apple were collected from provinces in Vietnam for this purpose. The morphological characteristics of leaves and fruits were compared among samples, and the genetic diversity was evaluated using ISSR markers. The results revealed significant morphological diversity in the leaf base, blade shape, leaf color, and fruit shape and size. Based on the DNA profiles of 15 ISSR markers, 102 amplicons were produced, and the percentage of polymorphism ranged from 57 to 100%. The dendrogram constructed using the UPGMA method classified these varieties into two main clusters with a genetic diversity of 64%. Such comprehensive information underscores the genetic diversity among S. samarangense, thereby contributing to the germplasm resources for future plant breeding programs.

The relationship between habitat quality and the diversity of phytoplankton, zooplankton, and nekton in mangrove ecosystems of Bawean Island, Indonesia. Biodiversitas 25: 3017-3026. Mangrove ecosystem consists of biotic and abiotic components that interact with each other to form a food chain. This study aims to analyze the relationship between the diversity and abundance of phytoplankton, zooplankton, and nekton and water quality, human activities, and carbon stock in the mangrove ecosystem in Bawean Island. Data was collected from 11 sampling locations across the island. At each location, we observed the diversity of phytoplankton, zooplankton, and nekton and carbon stock of mangroves along with data of water quality (salinity, pH, dissolved oxygen/DO, biochemical oxygen demand/BOD, nitrate, orthophosphate, dihydrogen sulfide/H2S, dissolved lead/Pb) and human activities based on the hemeroby index. The research results showed that the level of DO, H2S, nitrate, and orthophosphate did not meet the quality standard according to the Indonesian government. The level of disturbance from human activities varied from ?-mesoherobic to metahemerobic categories. Human activities and higher levels of H2S in waters had a negative relationship with mangrove ecosystem carbon stock and the diversity and taxa richness of phytoplankton, zooplankton, and nekton. Phytoplankton diversity had a positive relationship with zooplankton, while zooplankton diversity had a positive relationship with nekton. High carbon stock in the mangrove ecosystem will likely increase the diversity of phytoplankton, zooplankton, and nekton. The result of this study implies that anthropogenic activities need to be controlled to improve the Bawean Island mangrove ecosystem services.

Abstract. Fathurrahman F, Ulpah S, Sodiq NAM, Mahadi I, 2024. The effect of Colchicine treatment on phenotype and genotype characteristics of Detam-2 variety of soybean Glycine max. Biodiversitas 25: 1230-1238. The enhancement of soybean plant mutagenesis to obtain phenotypic and genotypic diversity is essential in agriculture. This study aimed to analyze the effect of colchicine concentration on phenotypic and genotypic characteristics based on RAPD markers of detam-2 variety. The experiments were carried out from January to June 2023 in the experimental farm of the Agriculture Faculty, Universitas Islam Riau, Pekanbaru, Indonesia. Therefore, 600 plants were used, consisting of 200 controls and 400 samples treated with 3,500 ppm of colchicine. The growth parameter was analyzed using a DMRT statistical test at p<0.05, and stomata densities were compared using T-test. The mutant-1 produced the highest yield, a total of 235 pods (with 233 phity pods), seed weighed 93.20 g, which was significantly different from those of controls. Mutant-2 produced the highest dry weight of 100 seeds, which was 12.80 g, with a dry seed weight per plant of 63.30 g. Leaf types in mutant samples varied, including normal, purple leaf stems, and curly leaves. The genotypic study showed that primer OPAA-01 produced a different number of DNA bands. The highest genetic similarity was 0.961 among the controls, while the lowest was 0.451 between mutant-1 and the controls. Based on the results, the dendrogram produced two groups: control and mutant-1.

Biodiversitas 25: 2828-2838. The Klutuk banana plant (Musa balbisiana colla) is recognized as a resistant plant against biotic and abiotic stresses. The resistance trait of the cultivar is not exactly revealed but is most likely related to the presence of endophytic bacteria. This study aimed to determine the variation of banana cultivars and their organs on endophytic bacterial populations and their character as plant growth promoters (PGP). The study involved isolating endophytic bacteria from the root, corm, and petiole of cv. Klutuk and cv. Ambon, aged 4-5 months. Isolated endophytic bacteria were evaluated based on plant-growth-promoting (PGP) criteria. Subsequently, 16S rRNA sequences were amplified using 16S primers, 27f and 1492r, then sequenced. The obtained isolates were also identified by comparing the 16S sequences to the NCBI gene bank. The results showed that the population and number of isolates with the most PGP characters were in both banana cultivars' roots, petiole, and corm. Endophytic bacteria with multiple traits as PGP are composed of Phylum Proteobacteria (gamma, alpha, beta), Firmicutes, and Actinobacteria members. Several species are potential as they have multiple PGP traits with high-quantity test results, namely Microbacterium testaceum DSM 20166 strain, Simplicispira sp. CPCC 100842 strain, Klebsiella grimontii SB73 strain, Enterobacter mori LMG 25706 strain, Pantoea sp. CoA11 strain, Agrobacterium tumefaciens IAM 12048, Variovorax guangxiensis GXGD002 strain, Serratia nematodiphila DZ0503SBS1 strain, Bosea thiooxidans BI-42 strain, Erwinia tasmaniensis Et1/99 strain, Methylobacterium fujisawaense DSM 5686 strain, K2 isolate, K8 isolate, and K18 isolates. This study also revealed the endophytic bacterial population and character as plant organs influenced PGP. The roots and petioles became the banana plant organs colonized by endophytic bacteria with multiple traits as PGP. Overall, several isolates were discovered in cv. Klutuk were suspected of being new species with potential as PGP agents, and these were not identified in cv. Ambon.

Abstract. Labajo-Villantes Y, Cootes J, Luo Y-B, Nuneza OM. 2024. Orchid diversity across different forest types on Mt. Malindang, Philippines. Biodiversitas 25: 605-615. Mt. Malindang Range Natural Park is one of the most significant biodiversity and conservation areas and is considered a biodiversity hotspot. There have been many biodiversity studies on Mt. Malindang, but information on orchid taxonomy and distribution is limited. This study aimed to conduct a comprehensive inventory of orchids using an exploratory method. Samplings were conducted in different forest types: montane, mossy, dipterocarp, mixed dipterocarp, and almaciga. A total of 114 orchid species with 55 endemics were recorded. Three of these (Phalaenopsis x intermedia Lindl., Paphiopedilum hennisianum (M.W.Wood) Fowlie, and Paphiopedilum haynaldianum (Rchb.f.) Stein) were under Appendix I (can only be exported under specific circumstances). At the same time, 78 were under the Appendix II (follow controlled trades) categories of the Convention on International Trade in Endangered Species of Wild Fauna and Flora. P. hennisianum and P. haynaldianum, listed as endangered species globally, were observed in montane and mossy forests, respectively. Among the six sampling sites, the montane forest was the most species-rich, with the most endemics, followed by the mossy forest. Almaciga and mixed dipterocarp forests were closely related with 100% similarity. The results of this study are valuable to Mt. Malindang's ecological profile for more effective protection and conservation plans.

Abstract. Yuhendri R, Nurainas, Maideliza T, Meriko L, Alponsin, Abdul Wahab IR. 2024. Anatomy and powder microscopy of Curcuma sumatrana Miq. (Zingiberaceae). Biodiversitas 25: 900-908. Curcuma sumatrana is a wild plant endemic to West Sumatra and is believed to possess various therapeutic values that are beneficial in treating multiple diseases. This research aimed to characterize the anatomy of leaves, roots, and rhizomes through permanent preparations and powder microscopy to contribute to its phytochemical analyses. Observations of anatomical characteristics were carried out using a light microscope and photographed using the DP2-BSW application. The anatomical study of the leaf revealed its upper and lower epidermis thickness to be 49.8 ?m and 31.7 ?m, respectively. Furthermore, its mesophile thickness was 48.9 ?m, and the leaf type was unifacial, containing prism-shaped crystals, and covered with non-glandular trichomes. Its trichome height was 146.3 ?m, aerchyma area was 198.1 ?m, and vessel area was 284 ?m. On the other hand, the anatomical structure of the root showed cortex thickness of 292.7 ?m, endodermis thickness of 5.9 ?m, xylem diameter of 34.7 ?m and pith thickness of 244.6 ?m. Meanwhile, the anatomical structure of the rhizome consisted of eccentric starch, sand-shaped crystals, cortex thickness of 957 ?m, and endodermis thickness of 7.37 ?m. The powder analyses of its leaf showed clear structure of the xylem, epidermis, stomata, and trichomes. In contrast, its root and rhizome showed eccentric starch, spiral-type xylem vessels, fiber, oil glands, and cortex. In conclusion, the anatomical characteristics of C. sumatrana do not show significant differences from other species in the Curcuma genus, except in terms of the color of the rhizome. The rhizome of C. sumatrana is pale purple, while C. longa has a deep yellow rhizome, C. aeruginosa has a blue rhizome, and C. heyneana has a light yellow rhizome.

Biodiversitas 25: 2696-2704. The Zerafshan Valley, Uzbekistan, characterized by a warm and arid climate, has declined vegetation and soil quality over recent decades, marking a stark departure from its once lush past. Understanding the nuanced relationship between species and their habitat is imperative to unraveling this ecological transformation. This research delves into ferns, which are highly responsive to environmental factors, seeking to elucidate their circumstances within the changing ecosystem. Through strategically planned intensive transects and plots as proxies for fern growth and habitat analysis, statistical methods like the Shannon-Wiener Index (H) and Bray-Curtis index were deployed. The climate-related information analysis was conducted using the CRU TS 4.07 tool. We made use of average temperatures and precipitation over 120 years. The results of analyses have been acquired from 490 fern individuals belonging to 13 species, 7 genera, and 6 families. There are four species: Equisetum arvense L., Equisetum ramosissimum Desf., Cystopteris fragilis (L.) Bernh. and Adiantum capillus-veneris L. were employed as indicators of habitat. The findings underscore a substantial decline in fern-populated areas, resulting in diminished diversity and abundance and a relatively high rarity level. Furthermore, a latitudinal correlation with ferns along the stream direction is exhibited by four indicator species, offering valuable insights into the intricate dynamics of the Zerafshan Valley's ecosystem.

Abstract. Rahmawati LA, Afiati N, Putranto TT. 2024. Diversity of soil macrofauna in traditional oil mining of Wonocolo Geosite, Bojonegoro Geopark, East Java, Indonesia. Biodiversitas 25: 2148-2160. Wonocolo traditional oil mining has been going on for 130 years and has led to soil pollution. This study aims to characterize the composition of macrofauna in the soil affected by Wonocolo's traditional oil mining. The existence of macrofauna is important as a bioindicator of soil quality and its role in maintaining soil fertility levels. Mining areas are grouped based on the density of the oil well (rare, medium and dense), then three sample points are selected in each group. Macrofauna were collected using quadrat-hand sorting, pitfall trap, and flannel trap methods, and then the community structure index was analyzed. The results showed that 709 individuals were collected consisting of 17 orders and 71 species, with 64.8% of individuals found in category-dense areas. Hymenoptera and Polyrhachis sp. as the most abundant order and genus, while Odontomachus brunneus Patton 1894 is the most frequently encountered species. The average value of biodiversity, dominance, and evenness index ranged H'=1.28-1.57 (low), C=0.29-0.38 (low), and E=0.78-0.90 (high), respectively. Indications of environmental conditions in traditional oil mining areas cannot be seen solely from the value of the macrofauna biodiversity index but are apparent in the abundance of the Hymenoptera order in all areas categories and significant presence of Polyrhachis sp. species in the dense category.
