Abstract

Glibenclamide is a second-generation sulfonylurea that used for type II diabetes mellitus. Glibenclamide is included in the biopharmaceutical classification system (BCS) class II, which has a low solubility and high permeability. One effort that can be done to improve the solubility of drug is inclusion complexation. The aims of the study was to determine temperature and heating time on inclusion complexation of glibenclamide-β-cyclodextrin using sealed-heating method. Optimization done using a factorial design for two factors with 22 number of experiments. Inclusion complexes were determined its miosture content, solubility and in vitro dissolution. Moisture content study showed that F1> FB > FA > FAB. The results of solubility study were analyzed statistically using One-Way ANOVA and least significantly different (LSD). Statistical analysis showed that each formula had a significant difference solubility with FAB > FB > FA > F1>Control. Optimization on in vitro dissolution showed that FAB was an optimum formula with drug release of 87.24% within 180 min. This formula was characterized by DSC to observed for broadening, shifting and appearance of new peaks or disappearance of certain peaks. DSC analysis was indicated that inclusion complexation between glibenclamide and β-cyclodextrin was formed. Keywords: glibenclamide, β-cyclodextrin, inclusion complexe, sealed-heating

Abstract

1-(4-nitrobenzoyloxymethyl)-5-fluorouracil (4-NFU) is a derivative of anticancer drug 5-fluorouracil (5-FU) which has been substituted by alkyl, ester, benzene, and nitro groups at N1 position. 4-NFU has been synthesized by Kurnia but anticancer study has not been reported. This research aims to compare the cytotoxicity and antiproliferative activity between 4-NFU and 5-FU on MCF-7 cells. The cytotoxicity and antiproliferative activity were studied by MTT method. The cytotoxicity study was done for 24 hours. The antiproliferative study was done for 24, 48, and 72 hours. The result showed that cytotoxicity activity of 4-NFU (IC50 = 134.039 μM) was higher than 5-FU (IC50 = 4211.508 μM) on MCF-7 cells. Based on the unpaired t test, there was a significant difference between IC50 of 4-NFU and 5-FU (α<0.05). The antiproliferative study showed that there was a growth inhibition on MCF-7 cells. The doubling time value of 4-NFU was 290.51 hours at ½ IC50; 350.97 hours at IC50; and 418.2 hours at 1½ IC50. The doubling time value of 5-FU was 185.99 hours at ½ IC50; 214 hours at IC50; and 220.63 hours at 1½ IC50. The doubling time value of 4-NFU was higher than 5-FU so that antiproliferative activity of 4-NFU was higher than 5-FU on MCF-7 cells. Abstract

Background: Recurrent Aphthous Stomatitis (RAS) is a disorder characterized by recurring ulcers confined to the oral mucosa with no other signs of disease. RAS is classified according to three classification, there are RAS minor, RAS mayor, and RAS herpetisformis. RAS prevalence of general population ranges from 5%-66%. Purpose: This study aimed to obtain the prevalence and distribution of patients suffering from RAS in Oral Medicine Departement of Dental Hospital, Dentistry Faculty, University of Jember in 2014. Method: This study used descriptive study with retrospective approach and purposive sampling. The population of this study is all patients of Oral Medicine Departement of Dental Hospital, Dentistry Faculty, University of Jember in 2014 as many as 1073 peoples. Result and Conclution: The result showed that 146 patients suffer SAR (14%) from 1037 patients in Oral Medicine Departement of Dental Hospital, Dentistry Faculty, University of Jember on January until December 2014. There were 97% of patients had RAS minor, 3% of patients had RAS mayor, and 0% of patients had RAS herpetisformis from 146 patients with RAS. RAS can occur in women or men and children or adults, the result of this research showed that RAS was most common in women, that was 70% and the age groups of 12-24 years was 54%. Abstract

Pain is a multidimentional experiences. Chronic pain is caused by inflammation. Nowdays, the treatment of chronic pain is still a challenge. Plants that known to have analgetic activity is red ginger (Zingiber officinale var. rubrum). This research observed the effect of red ginger oil on inflammatory pain. Inflammatory condition was induced by intraplantar injection of CFA (Completed Freund's Adjuvant). Twenty mice were divided into 4 groups i.e sham, negative control, red ginger oil in two different doses (100 and 200 mg/kg BW). Red ginger oil was administrated per oral once a day for seven consecutive days, a week after CFA injection. Latency time toward thermal stimulus was measured on days 0, 1, 3, 5, 7, 8, 10, 12, and 14 after CFA injection. Paw thickness at the ipsilateral site was also measured on days 0, 1, 3, 5, 7, 8, 10, 12 and 14 after CFA injection. The result showed that the administration of red ginger oil significantly increased latency time toward thermal stimulus compared to negative control. It also significantly reduced paw thickness especially at dose of 200 mg/kg BW. These findings suggested that administration of red ginger oil could treat the inflammatory pain, caused by CFA injection. Keywords: red ginger oil, inflammatory pain, CFA, hyperalgesia

Abstract

Newborn (BBL) has a high risk of mortality. It happened because the public’s knowledge and practiceabout simple treatments such as prevention of hypothermia, giving colostrum and exclusivebreastfeeding is still very poor. Cadre is one closest person of newborn‘s mother who can affectmaternal behavior, so she can provides a good information to newborn‘s mothers and families whenher knowledge has been good. The aim of this research is to obtain information from the analysis ofthe effect in newborn care education by Syndicate Group methods on cadres’ knowledge inSumberdanti village. This research used pre-experimental method with one group pretest-post testdesign that used 24 cadres’ of Sumberdanti Village who registered by the village midwife. Beforegetting intervention, there were 50% cadres who belong to category of having enough knowledgeand there were 58.3% cadres who belong to the category of having good knowledge after gettingintervention. Analysis of hypothesis used Wilcoxon Signed Rank test with α = 0.05 which obtained pvalue = 0.001, indicating a significant effect of newborn care education by Syndicate Group methodon cadres’ knowledge. Hopefully, the Sukowono Public Health Center and the midwives can continueimproving the cadres’ knowledge by interactive methods such as Syndicate Group and othermethods, so that their role can be more optimal.Keywords: Posyandu Cadres, Knowledge Of Newborn Care, Syndicate Group Method

Abstract

Burns is a serious global health problem. Annually, 265.000 dead occured because of burns. Local burns wound treatment ideally uses material that can act as antimicrobial and antiinflamation, as well conditions moist environtment. Cucumber contains flavonoid and saponin that enhance Vascular Endothelial Growth Factor that can increase angiogenesis. The purpose of this research was to investigate the effect of cucumber extract gel and cucumber powder gel on number of vascular lumen angiogenesis of IIB-burn wound healing in wistar rat. This research was true experimental laboratories with post test control only design by using wistar rats and divided into 8 groups. Rats were burns-induced and then terminated on the 3rd and the 10th day, each group consist of negative control group (normal saline), positive control group (gentamycin eye ointment), treatment group 1 (cucumber extract gel), treatment group 2 (cucumber powder gel). Data analysis using One Way ANOVA test for the 3rd day and the 10th day on vascular lumen number showed 0,031 and 0,036 (p<0,05) significance respectively. In conclusion, there was significant difference for cucumber extract gel and cucumber powder gel towards vascular lumen number in IIB-burns wound healing in wistar rat. Abstract

Mango is one of the plants which have a lot of benefits in pharmacology. Total phenol in its peel is higher than in its flesh with the main component is mangiferin and quercetin which known have activity to inhibit cyclooxygenase 2 (COX-2) enzyme. The inhibition of COX-2 leads to the decrease of the production of prostaglandin (PgE2) so it can reduce pain which marked by the decrease of paw licking time on mice induced by formalin. Formalin-induced pain is divided into two phase: early phase at 0-5th minutes and late phase at 15-30th minutes. The aim of this research was to know the effect of Arumanis mango peel extract to decrease the paw licking time in white male mice. The research design was true experimental. The result of analyses used One Way Anova shows that there were significant difference in each treatment groups with significance p < 0,05. The LSD analysis on late phase showed no significant difference between the fifth treatment group and the positive control group in LSD test. It can be concluded that arumanis mango peel extract has equal effect with natrium diclofenac in decreasing paw licking time on late phase in dosage 26 mg/g. Abstract

Background: Deflection of orthodontic wire can be changed by oral condition, for exampleacidic condition caused by carbonated drink consumption. Objective: to analyze the differencein deflection of Nickel-Titanium archwire and NiTi epoxy resin coated immersed by artificialsaliva and carbonated drink. Materials and Methods: This research used four sample groups:NiTi non coated archwire immersed by saliva, NiTi non coated archwire immersed by salivamixed carbonated drink, NiTi coated archwire immersed by saliva, and NiTi coated archwireimmersed by saliva mixed carbonated drink. The samples were immersed for 3.5 hours at 37° Cin incubator. The deflection was measured with Three Point Bending method using UniversalTesting Machine. Result and Conclusion: One Way Anova test result showed significantlydifference between coated and non coated group that immersed by saliva with the groupimmersed by saliva mixed carbonated drink. Abstract

Paederus dermatitis is an acute skin irritation caused by skin contact with toxins in the rove beetle (Paederus sp). Pederin poison was previously thought derived from the rove beetle's body. Based on other studies, poison pederin obtain from the symbiotic rove beetles and Pseudomonas aeruginosa which is able to secrete the matrix metalloprotease (MMP). Doxycycline is an antibiotic act as an inhibitor of MMP. This study aimed to verify the effect of Doxycycline gel to the number of macrophages in healing process Paederus dermatitis due to poison of rove beetle (Paederus sp.). The study design was a quasy experimental design using 40 mice (Mus musculus), were divided into 4 groups. Normal control group, negative control group, P1 group was given doxycycline 1%, and P2 group was given doxycycline 5%. Experimental animals were terminated on day 1, 3, 5, 7, and 15. The result of One Way Annova test on the number of macrophages on day 1, 3 and 5 showed a significance of 0.00 (p<0.05). This study showed that there was a significant difference in the effect of doxycycline gel to the number of macrophages in healing process of Paederus dermatitis. Abstract

DMBA (7,12-Dimethylbenz(α)antracene) is a prototype of Polycyclic Aromatic Hydrocarbons (PAHs) which is present in automobile exhaust, cigarette smoke, and wood stoves. DMBA is converted into its reactive metabolites DMBA-DE and cause liver cell membrane lipid peroxidation, which results in an increased of liver malondialdehyde (MDA) levels. Broccoli contains flavonoid that able to prevent lipid peroxidation. The current study was conducted to find out the different hepatoprotective effect of ethanolic extract of broccoli against DMBA-induced hepatotoxicity in wistar rats using liver MDA as a marker. A total of 24 male wistar rats were divided into six groups, consisting of normal control (distilled water), negative control (DMBA), and treatment (ethanolic extract of broccoli in doses of 250, 500, 1000, and 2000 mg/kgBW). Rats were treated for 7 days prior to a single dose of DMBA on the 8th day of treatment. Liver tissue samples were taken on the 12th day for measurement of liver MDA levels. Data were analyzed using One Way ANOVA followed by Post Hoc Test LSD. The present study showed significant differences on liver MDA levels between groups (p<0.001). This study concluded that there were different hepatoprotective effects of ethanolic extract of broccoli treatment on liver MDA levels in wistar rats induced by DMBA.
