Abstract

Osteoarthritis (OA) is the most common cause of joint pain, but there’s still no satisfying treatment that can inhibit degradation of articular cartilage in OA. One approach that is being widely studied is through activation of PPAR-γ. Many studies have shown effect of PPAR-γ on OA either in vitro or in vivo in animal model. On the other hand, the main risk factor of OA is obesity. Obesity generally followed by increased of leptin level due to leptin resistance. The aim of this study is to examine the effect of pioglitazone as PPAR-γ agonist in reducing the level of MMP-9 and MMP-13 in OA chondrocyte with hyperleptinemia. MMP-9 and MMP-13 are key role enzymes that degrades cartilage in OA. This study use IL-1β-induced chondrocyte exposed to pioglitazone. Pioglitazone 0,1μM, 1μM, and 10μM reduce MMP-9 and MMP-13 level. Whereas pioglitazone together with leptin 10μg/ml reduce the decrease level of MMPs compared to the group without leptin. Leptin inhibit the effect of pioglitazone in reducing the level of MMPs.Keywords: OA, leptin, pioglitazone, MMP-9, MMP-13

Abstract

Coronary heart disease is an important health problem. It is estimated that the death rate from coronary heart disease by 23,4 millions in 20130 (WHO, 2009). Excessive physical activity can cause oxidative stress which lead to atherosclerosis. Mung bean sprouts (Vigna radiata (L)) contain vitamin E, vitamin C, fenol, flavonoid, fitosterol, and other minerals. The aim of this study is to determine the effect of mung beans sprout extract on on the thicknes of the tunica intima-media on physical stress-induced atherosclerosis of male wistar rat. This research is true experimental study using 20 male wistar rat were divided into five groups: negative control, positive control and treatment groups P1, P2, P3 (physical stress + bean sprout extract 50 mg/day, 100 mg/day, 200 mg/day for 10 days). Abstract

BackgroundDiazinon is a widely used pesticide to eradicate agricultural pests globally with the potential to cause harmful effects on humans by generating free radicals and depleting endogenous antioxidants. Soy flour possesses high antioxidant effect. This study evaluated the nephroprotective effect of soy flour  on diazinon-induced nephrotoxicity in rats. MethodsAn experimental laboratory study was carried out involving 30 male Wistar rats were randomly divided into five groups of 6 rats. The normal control (Kn) and the negative control (K-) groups received normal saline, the treatment groups (K1, K2, K3) received 10%, 15%, and 20% soy flour in distilled water, respectively, on day 1 to day 28. Subsequently, for five days, all groups received diazinon 40 mg/kg, except for the normal control group. Blood urea nitrogen (BUN) and creatinine levels were measured at the end of the study. The Kruskal Wallis test with the post hoc Mann Whitney U test were used to analyze the data. ResultsBlood urea nitrogen and creatinine levels were lower in all treatment groups (K1, K2, K3) compared to the negative control group (K-), while the K3 level was the lowest (24.28 ± 1.56 mg/dL and 0.93 ± 0.14 mg/dL).  There was a significant decrease in BUN and creatinine levels in the experimental rats of the 15% and 20% soy flour groups (K2, K3) compared to the negative control group (K-).ConclusionAdministration of soy flour (Glycine max (L.) Merr.) had a nephroprotective effect in preventing increases in BUN and creatinine levels of diazinon-induced nephrotoxic male Wistar rats.

Abstract

Mixed liquor is a liquor mixed with other substances, mainly ethanol and methanol. Mixed liquor can cause kidney destruction. The aim of this study was to investigate the effect of mixed liquor toward kidney's microscopic and macroscopically changes. This study used 24 rats which were divided into 4 groups consist of 1 control group and 3 experimental groups. The first, second, and third experimental groups were given ethanol 1,296ml/2 days; methanol 0,2592ml/2days and aquades 1,4448ml/2 days for 5 days; 11 days and 17 days, respectively. Afterward rats were terminated and the right kidney mass was measured. The kidney tissue was prepared for histological examination and the number of necrotic cell was calculated. According to Kruskal Wallis test, kidney mass was not significantly different (p>α) while total necrotic cells was significantly different (p<α). This study showed that rats induced with mixed liquor for 5 , 11, and 17 days resulted in kidney microscopically change.

Abstract

Mung bean (Vigna radiata (L.)) sprout contains antioxidants especially vitamin E that can prevent oxidative stress and maintain cell membranes from lipid peroxidation, so the development of atherosclerosis can be inhibited. This study investigated the effect of mung bean sprout on the aorta abdominalis tunica intima-media thickness of male wistar rats induced by physical stress. It was a true experimental study with Post-test Only Control Group Design. Twenty five male wistar rats aged 2-3 months, divided into 5 groups: K1, K2, P1, P2 and P3. K1 was a normal group without treatment. The other groups were given physical stress such as swimming test for ten days. P1, P2, and P3 group were treated by 25, 50, and 100 mg/200g/day of mung bean sprout extract simultaneously. At the end of treatment, the aorta abdominalis were taken and prepared for histopathology examination. The result showed the average of tunica intima-media thickness in K1 30,25±1,65 µm; K2 32,45±5,02 µm; P1 31,65±2,63 μm; P2 30,35±4,73 μm and P3 30,45±3,21 μm. Data analysis using One Way ANOVA showed a significancy of p=0,839. In conclusion, there was no effect of the mung bean sprout extract on tunica intima-media thickness of male wistar rats induced by physical stress.   Keywords: Mung bean, physical stress, tunica intima-media

Abstract

Physical stress is a condition that caused by severe physical activities which can increase free radicals production causing oxidative stress. Oxidative stress causing the organs failure include the kidney. It damage glomerulus causing decrease in GFR and tubular cell damage causing cumulation of creatinine and raise creatinine serum. This study investigated the difference of serum creatinine of male Rattus norvegicus with physical stress and without physical stress. Ten male Rattus norvegicus aged 2-3 months were divided into two groups : control and treatment group. Control group without physical stress but treatment group was given physical stress 30 minutes of swimming stress in the morning everyday for 10 days. The creatinine serum measured by Jaffe reaction. The normal range of serum creatinine for Rattus norvegicus was 0,578-1,128 mg/dl. The result of serum creatinine for control group was 0,52 mg/dl and treatment group was 0,62 mg/dl,both still in normal range. The analysis using Mann Whitney showed the significance of 0,23 (p>0,05). We can conclude that there was no difference of serum creatinine of male Rattus norvegicus with physical stress and without physical stress. Abstract

Osteoarthritis is a chronic joint disorder with weakening and disintegration of the joint cartilage followed by the growth of new bone and cartilage. TNF-α is one of the inflammatory mediator that plays an important role in the progression of osteoarthritis. In the others hands, lemuru (Sardinella longiceps) fish oils that contain EPA and DHA has an anti-inflammatory effect. The purpose of this study was to investigate the effect of lemuru fish oil on decreasing the expression of TNF-α in cartilage that induced by Complete Freunds Adjuvant. This was a experimental design study with randomized post test only control group design. The sample was divided into 2 groups: the negative control group (K-) and the treatment group (P), each group was divided into three subgroups based on duration of lemuru fish oils treatment, 7th, 14th and 21st days. TNF-α expression data was obtained from immunohistochemical observations on tibiofemoral articular cartilage. The result showed that the expression of TNF-α in the treatment group is lower than the negative control group and One Way Anova test showed that there is significant difference between groups (p = 0.000). Abstract

Dyslipidemia is a condition of abnormal blood lipid profiles, including hypertriglyceridemia. One of antidislipidemia agent is niasin, that can be found in mungbean (Vigna radiata (L)) sprout. This study aimed to determine the difference in triglyceride serum after treatment with mungbean sprout extract on male wistar rats induced by egg yolk. This was an experimental study with randomized Post-test Only Control Group Design. Twenty five male wistar rats aged 2-3 months, divided into 5 groups: K(-), K(+), P1, P2 and P3. K(-) was treated with standard diet. The other four groups were injected with 0,006 mg/ 200 gram weight IV adrenalin on the first day followed by egg yolk diet for six weeks. P1, P2, and P3 group were treated with 50, 100, and 200 mg/ 200 g/day of mungbean sprout extract simultaneously with the diet. At the end of the treatment, the serum was taken to examine the triglyceride serum. Average levels of triglycerides serum in group K(-) 69,2±15,9; K(+) 92±14,9; P1 63,2±14,1; P2 53±6,9; P3 84,8±23,9. There was significant different of triglyceride serum means between group K(+) and P2 (p=0,014). In conclusion, 100 mg of mungbean sprout extract showed a significant difference in triglyceride serum with control group of male wistar rats induced with egg yolk.   Keywords: Mungbean sprout, egg yolk, triglyceride

Abstract

Burns is a serious global health problem. Annually, 265.000 dead occured because of burns. Local burns wound treatment ideally uses material that can act as antimicrobial and antiinflamation, as well conditions moist environtment. Cucumber contains flavonoid and saponin that enhance Vascular Endothelial Growth Factor that can increase angiogenesis. The purpose of this research was to investigate the effect of cucumber extract gel and cucumber powder gel on number of vascular lumen angiogenesis of IIB-burn wound healing in wistar rat. This research was true experimental laboratories with post test control only design by using wistar rats and divided into 8 groups. Rats were burns-induced and then terminated on the 3rd and the 10th day, each group consist of negative control group (normal saline), positive control group (gentamycin eye ointment), treatment group 1 (cucumber extract gel), treatment group 2 (cucumber powder gel). Data analysis using One Way ANOVA test for the 3rd day and the 10th day on vascular lumen number showed 0,031 and 0,036 (p<0,05) significance respectively. In conclusion, there was significant difference for cucumber extract gel and cucumber powder gel towards vascular lumen number in IIB-burns wound healing in wistar rat. Abstract

Aluminum is widely used in life but includes non-essential metals toxic to the human body that most accumulates in the femur. Aluminum is toxic because it increases the number of free radicals resulting in oxidative stress that causes osteoblast apoptosis. Tamarindus indica has the most potent antioxidant effect on the seed. The polyphenol content in Tamarindus indica seed are myricetin, procyanidin B2, and caffeic acid. Antioxidants in Tamarindus indica seed have mechanism of donating electrons from the -OH group in the phenolic ring to stop oxidative chain reactions and prevent the formation of hydroxyl radicals and lipid peroxidation that play a role in cell apoptosis. The purpose of this study to determine the effect of giving Tamarindus indica extract on the total osteoblasts of femur bone in male wistar rats induced by aluminum. This research is a true experimental in vivo with randomized post-test only control group design using 25 male Wistar rats divided into five groups. Aluminum is given orally dose 300mg/kgBB and Tamarindus indica extract dose 25, 50, 100mg/kgBB for 10 weeks. The results of this study indicate the average total of osteoblasts in groups K, P1, P2, P3, P4 are 15.21 ± 1.71; 18.48 ± 3.65; 17.8 ± 7.05; 17.13 ± 1.16; 16.74 ± 5.71. Data analysis used the Kruskal Wallis test. The results of data analysis showed that there was no significant differences in all groups (p = 0.581). Based on the results of this study, Tamarindus indica extract that was given to male wistar rats induced by aluminum for 10 weeks had no effect on the total osteoblasts.
