Abstract

yphoid fever is an acute infectious disease in the intestinal tract caused by Salmonella typhi. The prevalence of typhoid fever in Indonesia in 2013 was 81% per 100,000 population. Ciprofloxacin is a large-spectrum antibiotics, but in some countries ciprofloxacin has been known to be resistant to S. typhi. Therefore, other methods are needed to improve the effectiveness of ciprofloxacin by combining with chitosan. Chitosan is a polysaccharide which known to be effective in some positive and negative Gram bacteria and able to overcome the resistance of some antibiotics. The research objective was to determine the effect of antibacterial and Minimum Inhibitory Concentration (MIC) of the combination of Litopenaeus vannamei chitosan and ciprofloxacin against S. typhi. The method used to obtain the value of the MIC was microdilution method with quasy-experimental research design and posttest only control group design. MIC which already been obtained then used to find the Fractional Inhibitory Concentration Index (FICI) using a checkerboard test to interpret the effect of the combination. The MIC combination showed different results in each repetition that chitosan: 15.63 to 62.5 µg/mL and ciprofloxacin from 0.03 to 0.5 µg/mL and known to have a smaller value than the MIC single. While the value of FICI from 5 repetitions was 0.476. So, it could be concluded that the combination of chitosan and ciprofloxacin has an antibacterial effect on S. typhi and have a synergistic combination effect. Keywords: Salmonella typhi, chitosan, ciprofloxacin, microdilution method, checkerboard test.

Abstract

Pain is a multidimentional experiences. Chronic pain is caused by inflammation. Nowdays, the treatment of chronic pain is still a challenge. Plants that known to have analgetic activity is red ginger (Zingiber officinale var. rubrum). This research observed the effect of red ginger oil on inflammatory pain. Inflammatory condition was induced by intraplantar injection of CFA (Completed Freund's Adjuvant). Twenty mice were divided into 4 groups i.e sham, negative control, red ginger oil in two different doses (100 and 200 mg/kg BW). Red ginger oil was administrated per oral once a day for seven consecutive days, a week after CFA injection. Latency time toward thermal stimulus was measured on days 0, 1, 3, 5, 7, 8, 10, 12, and 14 after CFA injection. Paw thickness at the ipsilateral site was also measured on days 0, 1, 3, 5, 7, 8, 10, 12 and 14 after CFA injection. The result showed that the administration of red ginger oil significantly increased latency time toward thermal stimulus compared to negative control. It also significantly reduced paw thickness especially at dose of 200 mg/kg BW. These findings suggested that administration of red ginger oil could treat the inflammatory pain, caused by CFA injection. Keywords: red ginger oil, inflammatory pain, CFA, hyperalgesia

Abstract

Meatball (bakso) is one of the favorite foods in Indonesia. The adulteration of pork in beef meatball occurs frequently. This study was aimed to develop a fast and non destructive technique for the detection and classification of pork in beef meatball using Near infrared (NIR) spectroscopy, combined with chemometrics, e.g. partial least square (PLS) and linear discriminant analysis (LDA). The spectral bands associated with pork protein (PP), beef protein (BP), and their mixtures in meatball formulation were scanned, interpreted, and identified by relating them to those spectroscopically representative to pure PF and BF. Study was done in four phases successively; (1) preparation of trained sample; (2) analysis of meatball powder by NIR Spectrophotometer. NIR spectrum were used to form three data sets, i.e. (a) spectrum in full region, (b) spectrum with first derivative, and (c) spectrum with second derivative; then (3) classification model of chemometrics to PLS and LDA was formed using the Unscrambler X 10.2 Software. (4) the NIR method and the best model of chemometric were applied in meatball samples for the prediction in compared with Xematest Pork result. Then prediction from Xematest Pork compared with The result from NIR-Chemometric. Xematest Pork is simillar with the result of NIR-Chemometric. Keywords: beef meatball, pork, adulteration, NIR, LDA.
