Abstract

This study aimed to assess genetic structure of Calophyllum inophyllum using simple sequences repeats (SSR) markers to control genetic diversity at a provenance seed stand. Leaf samples were collected for DNA templates and 6 SSR primers were used. Genetic data of 280 trees were analyzed by FSTAT software, while PCoA was analyzed by GeneAlex. Results showed that number of allele (NA) per SSR primer between 3 and 4, the allelic richness (AR) ranged 2.805 to 4.000, the gene diversity (HE) ranged 0.018 to 0.631, coefficient inbreeding (FIS) of overall SSR primers were insignificant deviate from Hardy-Weinberg Equilibrium, excepted NY3. The mean HE values in the TBP was in low (mean HE= 0.272). However, the mean coefficient inbreeding values was not significant. PCoA analysis showed that the seed stand consisted genetically related trees; the 280 trees originated from only 5-6 mother trees. This is because the stand was originated from a conservation stand with narrow genetic differentiation. Management of the seed stand was discussed.

Abstract

The tree improvement research of nyamplung has entered the selection stage at the individual levels having high quality and oil yield properties. In order to maintain the superiority of this characteristics, a proper vegetative propagation techniques is required to multiply selected trees.The purpose of this study is to determine the effect  of cutting materials from sprouts on seedlings and mother trees. The study was arranged in a completely randomized design with factorial pattern. The factors tested were cutting materials (namely sprouts on seedlings and on the mother tree branches) and application of growth hormone types (control, pasta and  powder). The results showed that the survival rates and the growth of cuttings from seedlings and mother trees  were relatively similar. However, the rooting percentages and the growth of cuttings were significantlyinfluenced by the interaction between the two factors. The survival rates of shoot cuttings varied from 51.92 to 82.30% and rooting percentages range from 32 to 70%. The best result of rooting percentage and shoot cuttings growth were obtained at shoot cuttings from seedlings with growth hormone in the pasta type.

Abstract

Kayu merah (Pterocarpus indicus Willd), a high quality and multiple purpose tree, has been categorized as endangered species. Only a few know that kayu merah has two subspecies (forma) that are differentiated based on the surface structure of the central seed case. This study aims to know the differences on morphological and growth characters of two populations/subspecies kayu merah i.e. Pulau Seram, Mollucas (smooth pods, forma indicus) and Pulau Flores, East Nusa Tenggara (prickly pods, forma echinatus). The genetic relationship ofthese two populations/subspecies was analyzed using PBSTAT program, while their growth parameter analyzed using SAS program. The study found differences on morphological and growth characters of kayu merah from Pulau Seram and Pulau Flores. Kayu merah from Pulau Seram has many smooth spots and little grooves on the surface of its stem and has tapered leaf tip shape, while kayu merah from Pulau Flores has many grooves and little smooth spots, and has splitting leaf tip form. The two populations/subspecies were clearly clustered into their own group. Kayu merah from Pulau Seram has better growth than that of Pulau Flores. The result of this research has expected can support conservation and further utilization of the species in Indonesia by providing information on preliminary identification of the origin of kayu merah i.e. Pulau Seram and Pulau Flores as well as early indication of indicus and echinatus subspecies differences in the nursery.

Abstract

High demand of kayu kuku (Pericopsis mooniana Thw) has threatened the existence of its populations in natural distribution. Therefore, ex-situ conservation need to be carried out to protect them from extinction. Preparation genetic materials in nursery stage is very important to support the success of establishing ex-situ conservation plot. The objective of this research is to observe the role of genetic factor in the growth characteristics of kayu kuku seedlings in which the seed was originally collected from South East Sulawesi provenance. This research was arranged in Completely Randomized Complete Bock Design with 40 families in three replications. The result showed that genetic difference among the tested families affected the growth characteristics of seedling for height, diameter and sturdiness index. Genotype and phenotype correlation between height and diameter traits were 0.95 and 0.74 respectively. It revealed that high genetic diversity of kayu kuku could be observed in seedling stage, and indicated an important of genetic materials collected from Southeast Sulawesi provenance for further conservation and tree improvement program.

Abstract

Vulnerable status of kayu kuku (Pericopsis mooniana) is due to heavily wood harvesting. Aim in this study is to asses genetic diversity and structure of kayu kuku at Lamedai forest, Southeast Sulawesi using RAPD markers. Leaves samples were collected from 4 natural populations at Lamedai forest i.e. Lamedai natural reserve, Lamedai village, Balijaya and Tangketada. The 22 polymorphic loci of 5 RAPD markers were subjected to the genetic analysis, binary data were calculated by GenAlex software to obtain parameters in genetic diversity within population and analysis molecular variant (AMOVA), while PopTree software was used to construct a dendrogram. Results showed that genetic diversity of the 4 populations was in moderate level (mean He: 0.361 ± 0.017) and the values was similar each other. There was no private allele at any populations, indicating that there was no genetic specification. A dendrogram analysis showed that Lamedai natural reserve genetically closed to Tangketada population and Balijaya, while Lamedai was separated into another cluster. AMOVA analysis showed 13% genetic diversity were observed among populations. In conclution, kayu kuku at Lamedai forest might be a large and continuous population in ancient period.

Abstract

Matoa (Pometia Pinnata J.R. Forster & J.G. Forster) is one of potential multipurpose tree species for forest and land rehabilitation program in North Sulawesi. Limited information on the technique of cultivation of matoa often become an obstacle for development, especially seed storage techniques because of all the seed are not always directly sown after collection. This study aims to determine the effect of time and storage media on the viability of matoa seeds. The design used in this study was complete randomly design arranged in factorial, consisting of (two) main factors : 1) storage time and 2) storage media. The storage time consists of three  levels : 5 days, 10 days and 15 days. Storage media consists of four levels : aluminium foil, calico cloth, cocopeat and without storage media. The results showed that the aluminium foil is the best storage media that can be used to store the matoa seeds up to 15 days.

Abstract

Timoho (Kleinhovia hospita L) is a rare plant that has high economic and cultural value for the Javanese people, especially in Yogyakarta. Timoho wood has a beautiful pattern called pelet and is used as a raw material for keris sheath. In Javanese culture, the keris is a masterpiece culture that is used as a weapon, spiritual object and complementary traditional events such as marriage. In Yogyakarta, Timoho trees are difficult to find, currently, so that plant propagation in a generative way also has difficulty due to scarcity of seeds. Therefore, it is necessary to conduct Timoho propagation research through vegetative methods. The purpose of this study was to determine the relationship between the origin of stem cuttings from three Timoho mother trees and the stem section (base, middle, and end) to the success of Timoho stem cuttings. Stem cuttings come from three Timoho trees in the Arboretum of the Center for Forest Research and Development of Forest Biotechnology and Tree Improvement Yogyakarta. The research method used a nested experimental design using 3 mother trees and 3 stem parts (base, middle and end) which were nested in the mother tree. The characters observed were percentage of shoots, number of shoots, length of shoots, and number of leaves. The results showed the percentage of live cuttings varied from 10.9-60.0%, number of shoots 1-7 pieces, number of leaves 3-26 strands and length of shoots 2.00-17.7 cm. The mother tree treatment has a significant effect on the percentage of sprouts and shoot length, while the cuttings material has a significant effect on the length of shoots at the cuttings measurements of the age of 2 months since planted in polybags.

Abstract

ABSTRACT The study was carried out in the Dusung Community Forest (Agroforestry) area of Toisapu Negeri Hutumuri Hamlet, South Leitimur Sub-District, Ambon City in August - October 2018 to determine the relationship between stand volume and stored biomass for scale plots in dusung agroforestry areas in Toisapu Hamlet. For this reason, it begins with an inventory of potential at seedling, sapling, pole and tree levels so that information and data on the actual potential of carbon content in the dusung system are obtained based on the value of the diversity of stand volume and biomass content. The results of the three measurement plots showed that the highest biomass was seen to be dominated by Durian, Pala and Langsat and Duku and Clove plants. Based on the calculation of the biomass value of the total carbon content in the three plots, it can be seen biomass potential for a total 400 m² plot area or 0.04 ha for the three plots with an area of 1,200 m² or 0.12 ha having a total biomass of 50,783.77 Kg / m² multiplied by the assumption of 0.5% of the total amount of tree biomass and poles so that the total carbon content stored in the three plots is 25,391.88 kg / m² or 2,539.19 tons / ha. Keywords: Agroforestry, Shelter Biomass, Dusung ABSTRAK Penelitian dilaksanakan pada areal Hutan Rakyat (Agroforestry) Dusung di Dusun Toisapu Negeri Hutumuri Kecamatan Leitimur Selatan Kota Ambon pada Bulan Agustus 2018 untuk menentukan hubungan antara volume tegakan dan kandungan biomasa tersimpan untuk skala plot pada areal agroforestry dusung di Dusun Toisapu. Untuk itu, diawali dengan menginventarisasi potensi pada tingkat semai, pancang, tiang dan pohon sehingga diperolehnya informasi dan data potensi aktual kandungan karbon pada sistem dusung berdasarkan nilai keragaman volume tegakan dan nilai kandungan biomassa. Hasil penelitian pada tiga plot pengukuran menunjukan bahwa biomasa tertinggi terlihat didominasi oleh jenis tanaman Durian, Pala dan tanaman Langsat serta Duku dan Cengkih. Berdasarkan perhitungan nilai biomassa terhadap jumlah kandungan karbon pada ketiga plot, terlihat potensi biomassa untuk total luas petak ukur 400 m² atau 0,04 ha untuk ketiga plot dengan seluas 1.200 m² atau 0,12 ha memiliki jumlah total biomassa 50.783,77 Kg/m² yang dikalikan dengan asumsi 0,5 % dari total jumlah biomassa pohon dan tiang sehingga jumlah kandungan karbon yang tersimpan pada ketiga plot sebesar 25.391,88 kg/ m² atau 2.539,19 ton/ha. Kata Kunci : Agroforestry, Biomassa, Dusung.

Abstract

Mulberry is an important feed for increasing the productivity in cocoons on silkworm cultivation. Mulberry having a high nutrition is very important. This is because the nutritional value is the most important factor influencing the quality and quantity of cocoons and silk. Therefore, the development of improved mulberry accompanied with an appropriate cultivation technique is necessary. This study aims to determine the effect of fertilization and trimming on the leaves productivity of three species of mulberry planted in a demonstration plot in Rurukan, Tomohon, North Sulawesi. The three species of mulberry used in this study are mulberry of SULI 01, Morus cathayana and M. canva which were tested under three types of fertilizer: Taikam, Petroganik and manure. The trimming test was only practiced for the two species of M. cathayana dan M. canva with three trimming height of 30 cm, 45 cm and 60 cm above the ground. All the tests were arranged under a completely randomized factorial design. The genetic materials of mulberry were supplied from Forest Productivity Research and Development Center in Bogor. The measured parameters were diameter and height of plant, and leaves production. The results showed that in general the fertilization had no effect on leaves productivity for all of the tested species of mulberry. The application of Taikam and manure only affected the growth of diameter and height of mulberry plant. The trimming treatment provided a difference of leaves productivity between the two species of mulberry. The highest leaf production for M. canva (639.61 gr) was found at trimming of 45 cm, while for M. cathayana was found at trimming of 60 cm (482.40 gr).

Abstract

Arbuscular mycorrhizal fungi (AMF) is a symbiosis between fungi and plants. This association is mutualism, AMF contributes to increased plant growth while fungi obtain energy sources from plant assimilation. The purpose of this study was to determine the association of AMF with durian plants (Durio zibethinus) in Manokwari, West Papua. The survey was conducted in 10 villages located in 4 districts known as the center for producing durian. Observation of the presence of AMF spores in the plant's rhizosphere was carried out by the wet sieving method. Furthermore, to find out the association between the two symbionts is done by observing AMF colonization in plant roots. The types of AMF found in the plant's rhizosphere are morphologically identified based on spore characteristics. The results showed that durian was associated with AMF. The percentage of AMF colonization ranged from 39.29 to 80.00%, while the number of spores was 112-336 spores per 100 grams of soil sample. Based on the spore morphological characteristics, AMF found is dominated by the genus Glomus, Scutellospora, and Acaulospora.
