Abstract

The decrease in fertility is considered to be the main cause of reproductive loss in dairy cattle and beef industry. Many candidate genes that play an important role in fertility and embryonic development. The purpose of this study was to detect genetic variations of the growth hormone gene family (GH|MspI, GH|AluI, GHR| AluI, GHRH|HaeIII and Pit-1|HinfI) and its association with superovulation response, ovulation, fertilization and transferable embryos rate. A total of 45 blood samples taken from cows that have been superovulated Angus, Brahman, HF, Limousin and Simmental. DNA was extracted with phenol-chloroform protocol followed by polymerase chain reaction technique (PCR) using specific primers for GH, GHR, GHRH and Pit-1 gene. PCR product was cut with restriction enzyme MspI, AluI, HaeIII and HinfI and electrophoresed on agarose gel and stained with ethidium bromide (EtBr). Superovulation is done by injecting a totally of 20 ml FSH for 4 days. Injecting the prostaglandin hormone (PGF2α) was performed on the eleventh day of CIDR implantation. Artificial insemination (AI) performed two or three days after the injection of PGF2α and Flushing was done on the seventh day after the AI.  The results showed that the Angus, Limousin, Brahman and Simental GH loci diversity of GH|MspI, GH|AluI, GHR|AluI, GHRH|HaeIII and Pit-1|HinfI was not associated with superovulation response, ovulation, fertilization and transferable embryo rate. In HF dairy cattle, genotype on Pit-1|HinfI AA has higher percentage of superovulation response (P < 0.05) when compared to AB genotype, but did not differ to BB genotype. Dairy cattle HF AA genotype also had higher ovulation rate (P < 0.05) when compared to AB and BB genotypes, but AB and BB have the same ovulation rate. Abstract

Indigofera sp is a tree leguminous forage species known to be adaptable to the drought climate and saline soil and so it has potential as alternative feed resource to support the ruminant animal production. This study aimed to study the  responses of goats fed Indigofera sp as the sole foliages in their diets. Twenty male Boer x Kacang crossbred goats  (6 months of age;16 ± 2,1 kg BW) were randomly assigned to four dietary treatments containing fresh or wilted leaves of Indigifera sp. as the sole foliages and supplemented with high carbohydrate (HCC) or protein (HPC)concentrates. Consumable parts i.e. leaves and twigs of about 8 to 10 months of age of Indigofera sp. were harvested by hand plucking every day in the morning. The experiment was a 2 x 2 factorial design arranged in a completely randomized design. The ruminal pH ranged from 6.14-6.85 and were  not affected (P > 0.05) by wilting nor by the type of concentrates. Wilting did not affect (P > 0.05) the concentration of  ruminal NH3-N, but  it was significantly higher (P < 0.05) in HPC group (32.36-32.54 mg dL-1) than those in HCC group(17.94-23.57 mg dL-1). Total VFA concentration were not affected by wilting  nor by the type of concentrates  (P > 0.05). Goats in the HCC group,however,numerically have higher total VFA concentration (178.5-183.75 mmol L-1) than those in the HPC group (142.21-174.64 mmol L-1). The apparent digestibility coeficients  of DM, OM, CP and energy of the diet were not different (P > 0.05) when contained wilted or fresh Indigofera foliage.  Significant increases (P < 0.05)   in the DM and  OM, but not in the CP and energy  apparent digestibility coeficients were observed in the HPC goats. Dry matter intakes were not different (P > 0.05) among dietary treatments, but the ADG of goats in the HCC group (60-63 g d-1) were significantly lower (P < 0.05) than those  of goats in the HPC treatment (80-87 g d-1). Blood glucosa concentration was not affected by wilting nor by type of concentrates (P > 0.05).  BUN was not affected by wilting process, but providing high protein concentrates significantly (P < 0.05) increased the BUN concentration. It is concluded that foliage of Indigofera sp could be used as the sole forage in intensive production of goats. Wilting the foliage prior to feeding seemed to be unnecessary, since this process does not improve the animal productivity in term of daily gain and efficiency of feed utilization. Abstract

Quality of eggs might decrease when hens under heat stress. A further study found that a specific plant extracts could reduce the impacts of heat stress. The aim of this study was to determine effects of Salix tetrasperma plant extract in combination with extract of turmeric and neem to improve egg quality and productivity of laying hens under heat stress. Sixty laying hens strain Isa Brown of 6 months old were used and reared in individual cages. The feed and drinking water were supplied ad libitum. This study was conducted in a completely randomized design with five treatments (two controls and three treatments) and each treatment consisted of 12 replication. Treatment consisted of with (KP) and without (KP) commercial anti-stress supplement. Formulations of extract were S. tetrasperma 1.000 mg / l water (EJ), S. tetrasperma 1.000 mg / l + Turmeric 250 mg / l + neem 250 mg / l (EJ+K1), and S. tetrasperma 1.000 mg / l + Turmeric 500 mg / l + neem 500 mg / l drinking water (EJ+K2). The hens were exposed to heat stress for 5 hours per day at a temperature range of 34.0±1.0°C. Supplements were dissolved in drinking water and were given for 30 days in the morning and noon. Results showed that a single extract of S. tetrasperma or the combination of an extract of turmeric and neem were significantly increased thickness of eggshell (P<0.05), but did not affect color of egg yolk, height albumin, egg weight, and HU value. Extract of S. tetrasperma combined with turmeric and neem extract dissolved in drinking water for 30 days in laying hens reared under heat stress could not improved quality of the eggs, but may increase thickness of the egg shell and cause decreased water consumption.

Abstract

biotic and catalytic supplement, on the productive performances of sheep fed Pennisetum purpuphoides (King grass). The concentrate was made of rice bran, molasses-coated palm kernel cake, minerals and salt. Probio-catalytic supplements were added either at 0.5% or 1.0% of the concentrate. The probiotic in the probio-catalytic supplements was either Probion (produced by Balitnak) and assigned as probio-catalytic supplement A, or a mixture of rumen microbes of buffaloes which was assigned as probio-catalytic B. The catalytic supplement consisted of gelatinized sago, Zn, Co, urea and sulfur. Twenty heads of male young sheep with an average liveweight of 18.8 ± 1.7 kg were divided into 4 groups based on the bodyweight and allocated to 5 feeding treatments. The treatments were (1) Control (without probio-catalytic supplement), (2) R1 addition of probio-catalytic supplement A at 0.5%, (3) R2 addition of probio-catalytic supplement A at 1.0%, (4) R3 addition of probio-catalytic supplement B at 0.5%, and (5) R4 addition of probio-catalytic supplement B at 1.0%. The experiment was carried out by a randomized block design. A four-week adaptation period was then followed by a 12-week feed intake data collection and growth trial. An intake and digestibility study of the feed was carried out for 7 days. Rumen fluids were taken for analysis of pH, ammonia and volatile fatty acids and microbial population. Results indicated a significantly greater weight gain in the group received R1 than that of the other treatments. Feed dry matter intakes were less in the groups supplemented with probio-catalytic resulting in a better feed conversion ratio. The pH and concentration of ammonia of rumen fluid were not significantly different among treatments. While the acetic acid and propionic acid concentrations were not different among treatments, the butyric acid was significantly lower (P<0.05) in the groups received R3 and R4, whereas those received R2 and R3 was not significantly different than that of the control group. The molar proportion of volatile fatty acid components was within the normal values. Based on the results of the present experiment, it was concluded that addition of probio-catalytic supplement A at 0.5% was advantageous and it could be suggested to be included in the diet to improve the productive performance of sheep.     Key Words: Probio-Catalytic Supplement, Productive Performance, Sheep

Abstract

In attemp to increase the productivity of Garut sheep, Indonesian Research Institute for Animal Production has crossed Garut sheep (GG) with St. Croix sheep (HH) that has high frame body size and adaptable to the hot climate (in 1995) and with Moulton Charollais sheep (MM) that has high body weight gain and good milk production to raise multiple birth (in 1996). The objective of this research was to evaluate the ewe productivity of Garut sheep and its crosses with St. Croix and Moulton Charollais. This research was conducted at Animal Research Station, Bogor from 1995 to 2002. In this study the crossing was done using frozen semen of M. Charollais and ram of St. Croix so that the real performance of these sheep under Indonesian condition is not known. So that the relative superiority of these crosses is calculated from the percentage of the differences between traits mean of crossbred and purebred divided by trait means of purebred Garut, except for the threeway crosses (MHG and HMG) is calculated from the difference between the means of threeway crossbred trait with the means of two parents (MG and HG). It is concluded that HG and MHG show higher dam productivity than GG, it can be seen from their litter weight at birth and weaning. In poor feed condition GG showed higher productivity than the crossbred sheep (MG and HG), but MHG/MHG showed higher relative superiority compare to their parents (MG and HG). In good feed condition HG and MHG/HMG sheep showed higher productivity than Garut sheep. The relative superiority of HG sheep is 26.40% over GG and for MHG/HMG is 11.24% over their parents (MG and HG).       Key Words: Garut Sheep, St. Croix Sheep, M. Abstract

Feather meal contains more than 90% protein that most of this protein (70%) are ruminally undegradable, therefore it is a potential source of by pass protein. The study was done to evaluate the response of young rams to hydrolised feather meal (HBA) substitution in ration. Twenty five young rams aged 7-8 months, with average body weight of 21.16 ± 2.47 kg were used in this study. This study was conducted based on ramdomized block design and the rams were grouped into 5 groups based on body weight. The ration consisted of 30% grass and 70% feed supplement on dry matter basis. Feed supplement of the control diet (R0) contained 72% of total digestible nutrient (TDN) and 15% of  total crude protein (CP), whereas R1-R4 were the improved rations that contained 75% of TDN and 18% of CP. HBA was used to substitute feed protein and as by pass protein source. The HBA substitution was done in 5 levels, namely: R0 = grass + feed supplement with 0% HBA; R1 = grass + feed supplement with 1.1% HBA; R2 = grass + feed supplement with 2.2% HBA; R3 = grass + feed supplement with 4.4% HBA and R4 = grass + feed supplement with 8.5% HBA. The study was conducted for 12 weeks. The results showed that the increase of CP in ration and HBA substitution did not affect dry matter (DM), organic matter (OM) consumption, as well as DM, OM and CP digestibility. The substitution of HBA in ration significantly increased CP consumption of ram fed R1, R2, R3 and R4 compared to R0 (143.86; 142.58; 147.46; 134.43 vs 109.98 g/head). The level of NH3-N in rumen fluid was significantly (P<0.01) affected by CP in the ration and HBA substitution. On the other hand total VFA and molar volatile fatty acids were not different except for molar isobutirat. The value of N retention and average daily gain (ADG) were affected (P<0.05) by level of CP and subtitution HBA in ration. The highest ADG value was resulted by rams received R2 diet (133.77 g/head). It was concluded that 2.2% HBA substitution in ration gave the best response on consumption, digestion, rumen fermentation, N retention and ADG. Key Words: Hydrolised Feather Meal, Substitution, Ram

Abstract

The success of freezing chicken semen is the hope for preserving Indonesian native chickens. Semen from twenty Arab roosters were collected using massage technique once in a week. Cryoprotectant DMA (dimethyl acetamide) or DMF (dimethyl formalmide) of 7 or 9% and thawing A at temperature of 30oC for 30 seconds or in B at 5oC for 5 minutes. The volume of fresh semen was 0.3 ± 0.072 ml/ejaculate, white colour, rather thick to thick, with 2200 ± 372 millions sperms/ml and pH 6.95 ± 0.32, 4+/3+ mass movement, 80% motility, 84 ± 4.48% and abnormality of 14.75 ± 1.28%. There were not statistically significant (P>0.05) effect of interaction of treatments (kinds and concentrations of cryoprotectant, and thawing methods) on motility and live-sperms. Sperm motility preserved with DMA (34.69%) significantly higher than with DMF (29.84%). Sperm motility was also significantly higher (34.53%) when preserved with 7% cryoprotectant than with 9% (30%). Thawing-A significantly gave higher motility (35.31%) than thawing-B did (29.22%). Live-sperms of semen preserved with DMA (46.75%) was significantly higher than with DMF (41.72%). Cryoprotectant concentration of 7% gave higher live-sperms (46.98%) than of 9% did (41.48%). Thawing-A left live-sperms of 47.14%, which was significantly higher than thawing-B did (41.30%). Abstract

The population of anoa, which is an endemic fauna to Indonesia, was getting decrease caused by the illegal hunting and deforestation. Anoa is included in endangered species by IUCN, and Appendix I by CITES. The experiment aimed to characterize the seminal plasma contents and to cryopreserve the anoa semen for artificial insemination application in captivity. The experiment was carried out in Taman Safari Indonesia (Bogor). Semen was collected from 2 anesthetized males (4-10 years) by electroejaculation. Seminal plasma gained by centrifugation of ejaculate (3000 rpm, 20 minutes), and then was evaluated the biochemical contents. Other ejaculates were evaluated macroscopically and microscopically, and then extended in Tris and Na-citrate media to a total concentration of 100 billion cells mL-1. Extended semen was stored at 4oC, and evaluated the motility and viability every 12 h. Frozen semen was made in Tris medium added with 5% of glycerol. The seminal plasma of anoa contained total lipid, Na, Ca and Mg higher than the buffalo, but its total protein, K and Cl were lower. Electrophoresis of seminal plasma using by SDS-PAGE method showed 10 bands of proteins (17-148 kDa). The motility and viability of chilled-extended semen in Tris and Na-citrate media were not significantly different (P > 0.05) during 72 h of evaluation. Extended semen in both of media may applicable for AI program for 24-48 h. Post thawing motility of frozen semen was still low, 26.00 ± 9.62%. Therefore, it is necessary to improve each stages of semen processing, so the motility will increased and resulted high pregnancy in AI program. Abstract

The study was aimed to evaluate several nutritional quality parameters (chemical composition, intake and digestibility) in goats of S. secundatum and B. humidicola known to be highly and moderately shade tolerant species. The species of Pennisetum purpureuphoides was used as a control diet. These forages were offered to 21 male Kacang goats with an average body weight of 15.2 kg. The animals were divided into three groups based on the body weight, and were randomly allocated to one of the three forages species in a Completely Randomized Design. During the adaptation period (14 days) and the intake monitoring period (5 days) after the adaptation period the forages were offered ad lib. During the feces and urine collection period (7 days) feed was offered at 90% of the maximum  intake to minimize the intake fluctuation. The dry matter content of S. secundatum (332 g kg-1DM) and B. humidicola (321 g kg-1DM) were relatively higher than P. purpureuphoides (211 g kg-1DM), while the organic matter content were relatively equal among the three forages species. The crude protein content of S. secundatum (83 g kg-1DM) and B. humidicola (85 g kg-1DM) was almost identical, but were lower than that of P. purpureuphoides (111 83 g kg-1 DM) The NDF and ADF contents were relatively similar among the forage species. The dry matter (DM) intake of goats fed S. secundatum (588 g d-1) or fed B. humidicola (577 g d-1) did not differ (P>0.05), but it was higher (P>0.05) than dry matter  intake of goats fed P. purpreuuphoides. The similar trend was shown when DM intake was expressed in the percentage of body weight (3.60%, 3.55% and 2.77% BW, respectively) or when expressed as gram per kg BW0.75 which were 35.5, 35.6 and 27.7 g/kg BW0.75 respectively. The digestion coefficient of DM and OM were not different (P>0.05) between S.secundatum and B. humidicola, but it was higher than that in P. purpureuphoides. The digestion coefficient of energy were not different among these forages. The N balances were positive (3,0–3,6 g d-1) in goats offered these forages. It is concluded that the nutritional qualities of S. secundatum and B. humidicola were comparable, and it is relatively higher that the P. purpureuphoides when offered to goats as the sole feed. Abstract

Crossbreeding between two different strains or breeds is often done in a livestock production system to take advantage of the heterosis (hybrid vigour) which may resulted from the crossing. The level of heterosis largely depend on the genetic distance betweeen the two parental groups, and is also affected by the environmental condition in which the heterosis would be more significant under less optimal condition. The aim was to evaluate the heterosis level of the crosses between Tegal and Mojosari ducks under stressful environment of low nutritional content (14% protein) in the diet. The experiment was conducted in the animal facilities of the Indonesian Research Institute for Animal production, using 40 Tegal (TT) ducks, 44 Mojosari ducks (MM), 46 crossbreds between Tegal drakes and Mojosari ducks (TM), and 42 crossbreds between Mojosari drakes and Tegal ducks (MT). Results showed that heterosis was not significant for the average age at first laying (AFL) and weight of first egg (WFE), in which AFL of TT (171.8 d) was not significantly different from that of MT (163.4 d) and both were shorter than the AFL of MM (182.3 d) or that of TM (182.8 d). Heterosis also did not exist on egg production to 68 weeks old (EP-68), in which the EP-68 of TT was 141.3 eggs and that of MT was 122.6 eggs, and both were lower than EP-68 of MM (156.8 eggs)  or of TM (154.0 eggs). For egg qualities, heterosis also did not show any significant level and difference between genotypes only existed for the Haugh Unit values. It seems that even under stressful environment heterosis did not exist among crosses between Tegal and Mojosari ducks, and therefore it can be concluded that the crosses between these two breeds will not produce any beneficial effects in a production system under any condition.
