Abstract

The histological changes of the liver in does with pregnancy ketosis were characterized. Twenty pregnant does at day 80 of pregnancy were used for this experiment. A total of 10 does were fed on grass (napier) and goat concentrate with water ad libitum consider as healthy pregnant goat, and another 10 ten were showed clinical and subclinical signs of ketosis consider as unhealthy pregnant doe. Liver biopsies were performed when clinical signs appear. Beta-Hydroxybutyrate (BHBA), free fatty acid (FFA), and glucose were dosed. Histological preparation revealed similar incidence and intensity of mild liver steatosis with lower cellular vacuolation in hepatocyte presence in healthy late pregnant does. Almost all of pregnant does with ketosis state (n=8/10) had large amount of small lipid droplets in almost every hepatocyte over the whole liver acinus with higher number of cellular vacuolation, and related with higher BHBA and FFA levels while low in glucose levels.Key Words: Beta-Hydroxybutyrate, Does, Free Fatty Acid, Liver Biopsies, Pregnancy Ketosis

Abstract

Vaccines developed from certain membrane proteins lining the lumen of arthropod’s gut have been demonstrated effective in the control of some arthropod ectoparasites. A similar approach could also be applied to Sarcoptes scabiei since this parasite also ingests its host immunoglobulins. To evaluate immune protection of the membrane proteins, insoluble mite proteins were fractionated by successive treatment in the solutions of 1.14 M NaCl, 2% SB 3-14 Zwitterion detergent, 6 M urea, 6 M guanidine-HCl and 5% SDS. Five groups of goats (6 or 7 goats per group) were immunised respectively with the protein fractions. Vaccination was performed 6 times, each with a dosage of 250 μg proteins, and 3 week intervals between vaccination. Group 6 (7 goats) received PBS and adjuvant only, and served as an unvaccinated control. One week after the last vaccination, all goats were challenged with 2000 live mites on the auricles. The development of lesions were examined at 1 day, 2 days, and then every week from week 1 to 8. All animals were bled and weighed every week, and at the end of the experiment, skin scrapings were collected to determine the mite burden. Antibody responses induced by vaccination and challenge were examined by ELISA and Western blotting. This experiment showed that vaccination with the insoluble-protein fractions resulted in the development of high level of specific antibodies but the responses did not have any protective value. The severity of lesions and mite burden in the vaccinated animals were not different from those in the unvaccinated control.     Key Words: Sarcoptes scabiei, Insoluble Protein, Goat, Vaccination

Abstract

The aims of this research was to obtain the quality of Spotted buffalo epididymal sperm in different kind of extender in the three stages of cryopreservation (after dilution, post equilibration and post thawing). Spermatozoa was collected with combination of slicing and pressure method into the epididymal tissue in Andromedâextender. Soon after diluted and equilibrated, epididymal spermatozoa was cryopreserved in liquid nitrogen (-196°C).  The result showed that the percentage of motility after thawing in Andromedâ + raffinose 0.4% (47.0 ± 2.4%), was significantly different (P<0.05) from that of control (41.0 ± 2.0%), but there was no significantly different (P>0.05) from that of Andromedâ + raffinose 0.2% (46.0 ± 2.0%). The percentage of live sperm after thawing in control (52.2 ± 2.5%), was the lowest and significantly different (P<0.05) from that of Andromedâ + raffinose 0.2% (59.2 ± 2.6%) and Andromedâ + raffinose 0.4% (58.8 ± 3.1%). Moreover, the percentage of membrane integrity after thawing in control, Andromedâ + raffinose 0.2% and Andromedâ + raffinose 0.4% was 68.0 ± 1.1%; 67.2 ± 1.6% and 67.6 ± 1.2%, respectively. There was no significantly different (P>0.05) in the percentage of membrane integrity from all treatments. In conclusion, the addition of 0.2 and 0.4% raffinose into Andromedâ extender could improve the percentage of motility and viability of post thawing spotted buffalo epididymal spermatozoa. Abstract

Data of body weight of Garut sheep and its crossbreds of 488 head, consist of  149 head of Garut sheep, 115 head of St. Croix X Garut (HG), 68 head of Mouton Charollais X Garut (MG), 101 head of MG X HG (MHG) and 55 head of HG X MG (HMG) which is collected from Indonesian Research Institute for Animal Production at Bogor station were used in individual growth curve analysis. Three growth curve non linier model were used in this study i.e Logistik, Gompertz and Von Bertalanffy models. Comparisons were made among these models for goodness of fit, biological interpretability of parameters and computional ease and effect of genotype and environment in them. Least square means growth curve parameters which have biological interpretability were used to compare effect the genotype, interse mated procces and estimated heterosis effect. The result indicated that Von Bertalanffy was the best model in fitting the data from Garut and Crossbreds although the model needed more iteration than others in computations. All models have good biological interpretability especially for parameter mature size (A) and rate of maturity (k). Genotype, year of birth, sex and type of birth reared were important effects (p<0.01) in mature size (A) for all models except effect of type of birth rearing (P<0.05) in Logistic model. Year of birth had important effect (p<0.01) in rate of maturity (k) for all models. Genotype, year of birth, sex, parity and type of birth reared also had important effect (p<0.01) in parameter b/M; except b parameter in Von Bertalanffy was affected significantly by sex (P<0.05). Abstract

The aim of this experiment was to investigate the effectiveness of santoquin (Sq) and vitamin E (VE) as feed additives to improve performance of male local ducks. The experiment was conducted using a completely randomized design. Two hundred and eighty male day old ducks (dod) of male Mojosari Alabio crosbred ducks were allocated to ten treatments with four replicates and seven ducks/replicate. The experimental diets were: Basal-diet (RO) without antioxidant (RB) (control); RB + 50 ppm santoquin (Sq) + 100 IU vitamin E (VE) (R1); RB + 50 ppm Sq + 200 IU VE (R2); RB + 50 ppm Sq + 300 IU VE (R3); RB + 100 ppm Sq + 100 IU VE (R4); RB + 100 ppm Sq + 200 IU VE (R5); RB + 100 ppm Sq + 300 IU VE (R6); RB + 150 ppm Sq + 100 IU VE (R7); RB + 150 ppm Sq + 200 IU VE (R8); RB + 150 ppm Sq + 300 IU VE (R9). The ducks were fed ad libitum for 8 weeks. Parameters observed were: feed consumption, live weight, body weight gain, feed conversion rate (FCR) and mortality. The results showed that Sq and VE supplementation did not significantly affect (P > 0.05) the feed consumption, live weight, body weight gain, feed conversion ratio and mortality rate of the ducks. This experiment shows that santoquin and  vitamin E supplementation did not affect the performance of male local MA ducks. Abstract

The process of freeze drying caused detrimental effect on plasma membrane and acrosome of the spermatozoa, even it potentially could alter the chromatin and DNA integrities. On the other hand, DNA integrity is essential for spermatozoa to participate in pronucleus formation during fertilization event. Therefore the evaluation of DNA integrity should be carried out to study the effect of freeze drying process. EDTA, EGTA, and PBS were used as dilution media of spermatozoa prior to freeze drying process to protect the DNA. Toluidine blue staining and comet assay methods were used to evaluate the alteration on chromatin and DNA integrities of spermatozoa, respectively. The results revealed that the highest compacted chromatin after 6 months storage of freeze-dried spermatozoa were observed from EGTA-3 (98%) and EGTA-1 (97%) treatments that had significant differences compared to all PBS treatments (90-92%), but not for fresh spermatozoa (100%). Whereas, the highest compacted DNA integrity of freeze-dried spermatozoa were observed from EGTA-2 (92%) and EGTA-3 (92%) but had no significant differences compared to other treatments including fresh spermatozoa (97%). These results demonstrate that EDTA and EGTA tend to be able to protect chromatin and DNA integrities of ram spermatozoa during freeze-drying and storage compared to PBS. Abstract

This study was intended to determine the effects of increasing the ratio of banana trunk to soybean meal on intake, nitrogen excretion and lambs performance. Sixteen lambs (averaging 21.21 + 1.69 kg) were pen individually and fed a basal diet consisted of chopped fresh king grass and commercial concentrate and unlimited access of water. Lambs were randomly assigned to one out of four different ratio of banana trunk to soybean meal as feed supplements, i.e. (R0) 0:1; (R1) 1:1; (R2) 2:1 and (R3) 4:1. Results showed that the inclusion of banana trunk had no adverse effect on lambs performance. As it is expected, altering the ratio of banana trunk to soybean meal increased (P < 0.05) dry matter intake of forage as well as total dry matter intake. The digestion coefficient of dietary ration was not affected (P >0.05) by feed supplement. Despite of no difference was observed in the coefficient of nitrogen digestibility, apparent fecal and urine N-output were significantly (P < 0.05) affected. The highest ratio of fed supplement (R3) tended to decrease nitrogen retention. Overall data indicated that lambs offered basal diet supplemented with R2 treatment accumulated more N per-day, than those fed other treatments, and consequently lambs performance was the best on those fed R2 treatment.   Key words: Banana trunk, protected protein, lambs

Abstract

A research was done to investigate the advantages of Tithonia diversifolia (Hemsl.) A. Gray and Moringa oleifera Lamk  leaves as protein source in ruminant concentrate on in-vitro rumen metabolism. Randomized Block Design with 4 treatments and 5 replications was applied in this experiment. The treatments were: A = Tithonia diversifolia (TD) 75% + Rice Bran (DD) 25%: B = TD (56.25%) + Moringa oleifera (MO) 18.75% + DD 25%; C = TD 37.50%) + MO 37.50% + DD 25%; D = TD 18.75% + MO 56.25% + DD 25%. Samples were weighted 375 ± 5 mg, placed into syringe glass 100 ml, 30 ml rumen liquor with bicarbonat buffer media was added and incubated in 390C for 48 hours. Parameters measured were gas production after 0, 2, 4, 6, 12, 24, and 48 hours incubation, degradability of organic matter (DBK) and dry matter (DBO), NH3, total VFA concentration, and microbe biomass production (mg) after 48 hours incubation. Results showed that Moringa oleifera replaced some Tithonia diversifolia in concentrate could improve gas production, total VFA concentration, DBK and DBO significantly (P < 0.05). The highest gas production was obtained from treatment D (52.98 ml/375 mg DM) and the lowest was from treatment A (41,02 ml/375 mg DM). The highest VFA total was produced by treatment D (88.20 mM) and the lowest was from treatment A (78.86 mM). The highest DBK dan DBO were obtained from treatment D (73.74 and 73.32%) and the lowest was from treatment A (68.77 and 67.54%). Treatment D produced the highest NH3 and microbial biomass (38.09 mg/100 ml and 89.50 mg), the lowest was obtained from treatment A (35.84 mg/100 ml and 84.66 mg). Abstract

The aim of this present study was to investigate the quality of canine epididymal spermatozoa during storage at 4°C. Spermatozoa was collected by flushing technique with physiological saline (NaCl 0.9% w/v) and diluted in modified Tris based extender containing 20% (v/v) of egg yolk for three days. The result showed that mean concentration of spermatozoa from cauda epididymal was 95.29.106 spz/ml. The percentage of progressive motility and membrane integrity of spermatozoa on time of collection was 70.71% and 72.85%, respectively. Quality of epididymal spermatozoa was decreased significantly (P<0,05) during storage at 4°C. The percentage of progressive motility during storage were 70.71% on day 0 (H-0, after diluted), 60.71% (H-1), 45.71% (H-2), and 33.57% (H-3). The percentage of membrane integrity during storage were 72.85, 68.88, 61.06 and 47.47% on H-0, H-1, H-2 and H-3, respectivelly. In conclusion, quality of canine epididymal spermatozoa was decreased during three days of storage at 4°C. Key Words: Epididymal Sperm, Storage, Canine

Abstract

This study was aimed to determine the right level of vitamin C in extracted pineapple rind to prevent heat stress effects so it does not interfere with the performance of broiler chickens. This study was done in a series of field experiments. Experiments in the field/cage was biological test of the effect of vitamin C from pineapple rind on production responses (percentage of body weight gain and carcass) and physiological responses (thyroxine hormone levels) in broiler chicken aged 3 weeks as many as 360 of Strain Arbor acress. The basic design used was CRD 3 x 4 factorial models and 3 replications with 10 chickens for each box, as factor 1: Room temperature (21 : 27 and 33ºC), and factor II: level of vitamin C in the pineapple rind (0:500:1000 and 1500 ppm). The data obtained were statistically analyzed using SAS program package, and if it showed any significant effect then followed by Duncans test/DMRT. Variables measured were body weight gain, carcass percentage and levels of thyroxine hormone of broiler. Results showed that addition of pineapple rind containing 500 ppm vitamin C in the drinking water reduced heat stress in chicken that were kept at temperature of 27ºC, while at 33ºC needed 1000 ppm vitamin C. Both treatments increase level of thyroxine hormone, produce weight gain equal to control, more over, there was no effect on the percentage of carcasses. Key Words: Ananas comosus L.
