Abstract

Furazolidone (FZD), furaltadone (FTD), nitrofurantoin (NFT) and nitrofurazone (NFZ) are veterinary drugs that belong to the nitrofurans (NFs) group and employed as feed additives for growth promotion and theurapetic treatment of gastrointestinal infections caused by Eschericia coli and Salmonella spp. The occurrence of NFs in animal products will end to cause health problem in human consumed such food. This research conducted to study the analysis of NF residues in chicken meat by a high performance liquid chromatography (HPLC) and to study the occurrence of NFs residues in samples collected from traditional markets and supermarkets in Bandung, Bogor and Depok. The results of validation method on several parameters for each NF showed that the average of the relative standard deviation (RSD) from the precision study were 2.15 to 2.38%, the R2 values of the linearity study were 0.9964 to 0.9995; recoveries were 75.90 % to 91.50 % and the detection limits were 12.01 to 37.25 ng/g. The residual level of NFs for 42 field samples showed that 2 samples positive for NFZ (9.09 and 10.74 ng/g), 1 positive for NFT (10.46 ng/g), 4 positive for FTD (16.44 up to 27.21 ng/g) and none positive for FZD. Present results showed that analysis of NFs in broiler chicken meat can be done using an HPLC and the analysis results from field samples showed that these types of drugs were being used for broiler chicken production both as single and/or combination drugs, therefore it is necessary to raise public awareness to monitor the use of NF in livestock production in Indonesia. Abstract

Digestibility of ruminant feeds is greatly determined by ruminal fermentation which in turn is greatly affected by micro-ecology of the rumen. Manipulation of micro-ecology such as supplementing with various yeasts or micro elements could improve digestibility of the feed. The effect of Comin+ (R1) and Zn-biocomplex (R2) alone or their combination (R3) on performance of Ettawa Crossbred kids, fed concentrate containing Aspergillus oryzae (R0) was studied. Twenty four Ettawa Crossbred kids (6-8 month of age; initial liveweights =16.63±2.10 kg) were divided into 4 groups. Feeds given were King grass (Pennicetum purpuroides) ad libitum and concentrate. The observations were carried out for 16 weeks with 2 weeks adaptation period and the experiment was conducted in a completely randomized design with 4 treatments and 6 replications. The results showed that supplementations significantly (P < 0.05) increased total DM intakes from 685 g/d (R0) became 748, 711 and 858 g/d; and grass DM intakes from 304 g/d (R0) became 373, 331 and 479 g/d for R1, R2 and R3, respectively. Digestibilities of DM, NDF and DE/GE were not affected by supplementation, but supplementations significantly (P < 0.05) affected digestibilities of ADF and CP. Supplementation also significantly improved (P < 0.05) ADG from 65.18 g/d (R0) became 94.64, 83.04 and 90.77 g/d; and FCR from 10.51 (R0) became 7.82, 8.36 and 9.46 for R1, R2 and R3, respectively. It was concluded that supplementation of Comin+ (R1) gave the best growth performance of Ettawa Croosbred goats, where ADG and FCR improved by 45.20% and 34.40%. Abstract

Fiber-digesting bacteria are the main rumen bacteria that play an important role in digesting feed. These bacteria are adapted to low quality forage from agricultural byproduct. The aim of these study was to determine the potency of fiber-digesting bacteria consortium obtained from buffalo rumen inoculated to Frisian Holstein calves during preweaning on feed consumption, utilization, mineral uptake and physiological status. This study used 14 isolates of bacteria obtained from collection of Faculty of Animal Science, Bogor Agricultural University. The experimental unit consisted of six Frisian Holstein calves at two week old with the average body weight of 38.00 ± 6.23 kg. Calves were inoculated by 20 ml of fiber-digesting rumen bacterial isolates [4.56 x 109 cfu/ml] every morning for four weeks. Experimental design used was based on a completly randomized design with three calves received the respective inoculation (treatment group) and three calves without any inoculation (control group). Data were analyzed statistically using t-test method with α = 0.05 and 0.01. The results showed that fiber-digesting bacteria (FDB) from rumen buffalo have adapted in the calves rumen since preweaning periode. Inoculation FDB increased the number of rumen bacteria, digestibility of protein and P uptake calves at eight weeks old. Increased feed intake, uptake of Mg and cobalt calves at 14 weeks old. Abstract

The aim of this research was to identify wool characteristic of Priangan sheep (G) and its crossbred with St. Croix (H) andM. Charolais (M). The parameters observed included yield, fiber length, fiber diameter, percentage of shrink during processing,yarn production, strength and elasticity of the yarn. Eighteen rams of Priangan, HMG and MHG crossbred were used.Completely Randomized Design with 6 replications and One Way analyses were used in data analysing, except for strength andyarn elasticity, 10 replication were used. The results show that breed had no significant effect (P>0.05) on yield, diameter of finefiber, and shrinking percentage during separation and carding process. In contrast, breed had significant effect on fiber length(P<0.01) and on coarse fiber diameter, the shrink, strength and yarn elasticity (P<0.05). In general, the wool of HMG and MHGcrossbred had better quality compared to Priangan sheep, although yarn production was higher in Priangan sheep.Key words: Wool, Priangan sheep, crossbred

Abstract

Ánthrax is an endemic zoonotic disease distributed in many parts of Indonesia. Although vaccination program has been implemented in many areas, cases are still frequently reported. Farmers are reluctant to vaccinate their livestock since spore vaccine used in the field often cause side effects and death of the animals. To overcome this problem, an inactive vaccine composes of Bacillus anthracis toxins, cell wall and capsule subunits was developed. B. anthracis Sterne strain (34F2) was selected to produce toxins and cell walls. Local Bacillus anthracis isolated from Citaringgul was used to produce capsule as the Polymerase Chain Reaction (PCR) revealed that this isolate poses cap gene encoding for capsule. Two vaccines compose of 15 μg toxoid, 30 μg of capsule, 15 μg of cell wall and 30 μg toxoid, 60 μg of capsule, 15 μg of cell walls were designated as vaccine I and vaccine II respectively. For each experiment, 10 mice were nasally immunized by placing 5 μl of vaccine into each nare 3 times at 2-week intervals. A group of 10 mice were unvaccinated and used as control. Blood was collected fortnightly to monitor antibody responses. All mice were challenged with 2 x 105 B. anthracis Sterne spores injected subcutaneously two weeks after the last vaccination. Two weeks after vaccination of antibodies to B. anthracis toxin, capsule and cell wall were detected in dot-blot assay. Mice that were immunised intranasally with chitosan adjuvanted vaccine developed high IgG responses in sera as detected by ELISA, and the response was dose dependent. Vaccine II gave better response than vaccine I. Vaccine I and II protected mice from challenge at a rate of 60 and 80% respectively. This results showed that intranasal B. anthracis vaccine composes of toxin, capsule and cell wall with chitosan as an adjuvant gave a good protection against B. anthracis Sterne spores challenge in mice. Jurnal fntuTernak dan Veteriner 3 (2): 87-93 .Pengukuran konsentrasi asam amino dalam darah arteri dan perbedaan konsentrasi asam amino arteri-vena dari otot kakidilakukan pada 4 tifk kurva pertumbuhan, yaitu: sebelum pembatasan pemberian pakan (PI), 8 minggu setelah pembatasanpemberian pakan (P2), 3 minggu (P3) dan 15 minggu (P4) setelah ternak diberi pakan kembali secara ad libitum. Enam belasdomba lepas sapih dibagi dalam 2 kelompok, Kelompok I dan II diberi pelet lucern (Medicago sativa) secara ad libitum selama3 minggu; kemudian Kelompok I terus-menerus diberi pakan secara ad libitum, sedangkan Kelompok If diberi pakan yang samasebanyak 1/2 kebutuhan pokokaya selama 8 minggu, kemudian diberi pakan secara ad libitum sampai akhir percobaan.Pembatasan pakan (P2) menurunkan konsentrasi asam amino berantai cabang (AABC) dan fenilalanin 43% dan histidin 30%.Pada periode P3 dan P4 konsentrasi AABC dalam Kelompok If naik lebih cepat daripada Kelompok I, tetapi pembatasan pakandan pemberian pakan secara ad libitum fdak berpengaruh pada konsentrasi lisin, arginin, treonin dan asam amino yang nonesensial. Karena variasi yang besar antar ternak, perbedaan konsentrasi semua asam amino pada arteri-vena tidak berbeda padaternak Kelompok I dan II pada semua periode (PI sampai P4) Kenaikan konsentrasi asam amino AABC dalam peredaran darahdari ternak yang mengalami pertumbuhan kompensatori diduga karena sintesishya lebih besar daripada penggunaannya. Karenaitu, disarankan penambahan energi dalam pakan untuk menaikkan penggunaan asam amino untuk sintesis protein.Kata kunci : Pertumbuhan kompensatori, asam amino, sintesis protein

Abstract

Microencapsulation of spermatozoa is a process to entrap a number of spermatozoa in microcapsule. Alginate, as a natural polymer polysaccharide is commonly used in cell microencapsulation. Tris Yolk Citrate buffer is a good buffer for spermatozoa dilution, therefore this experiment aimed to determine optimal concentration of alginate and egg yolk to sperm quality in bull spermatozoa microencapsulation. Concentration of egg yolk and alginate in media of encapsulation were determined in applications of sperm microencapsulation. Four bulls were used as semen source and only semen with good quality were used in this study. Poolled semen was diluted using the medium to get final concentration 100 x 106 cell/ ml. The first study was conducted to determine the effect of concentration of alginate (0, 1, and 1.5%) on viability of spermatozoa. The second study to determine the effect of alginate concentration, egg yolk and its interaction was done by comparing two levels of alginate (1 and 1.5%) with four levels of egg yolk (5, 10, 15 and 20%). Viability of spermatozoa, motility (M), live spermatozoa (L) and Intact Apical Ridge (IAR) were observed at 0, 1, 2 and 3 h incubation at room temperature. Results indicated that alginate concentration increased the osmolality and viscosity but did not affect pH of the medium. The osmolality and viscosity of medium were 275, 325, 425 and 1.12, 26.62, 47.98 for concentration of alginate 0, 1 and 1.5% respectively. Percentage of motility is significantly lower (P<0.05) in alginate medium than those of control, and 1.5% alginate could produce more uniform beads. Concentration of alginate, egg yolk and its interaction did not significantly affect viability of sperm. It is concluded that the combination of 1.5% alginate with 5, 10, 15 or 20% egg yolk can be used as media for sperm encapsulation. Abstract

This study was carried out to investigate genetic, non-genetic affecting factors and estimate genetic parameters for milk production and reproductive traits of Holstein cows via animal model. The data was obtained from a commercial farm (Safi Masr for Developing the Animal Resources), located in the Nile Delta, Dakahlia, Egypt. Data included 4791 records of 1797 cows, 794 dams and 67 sires that represented the period from 2002 to 2012. The means and coefficient of variability (CV%) of milk traits as total milk yield (TMY), 305 day milk yield (305-dMY), lactation period (LP) and dry period (DP) were 5787.8 kg (31.1%), 4695 kg (22.1%),  332 day (14.9%) and 72.3 day (27.7%), respectively. Also, the means (CV%) of reproductive traits as days open (DO) and age at first calving (AFC) were 157.9 day (22.6%) and 30.5 month (16.8%), respectively. Sire, dam, parity of cow, year and season of calving had significant effects on traits studied. Heritability estimated were 0.223, 0.184, 0.112, 0.118, 0.105 and 0.285 for TMY, 305-dMY, LP, DP, DO and AFC, respectively. Estimated rG and rP among milk production traits were positive but it takes negative trend with DP and DO. Moderate heritability estimates and positive genetic correlation for most of traits studied suggested that genetic improvement of these traits would be achieved via multi-trait selection. Abstract

A study was conducted to evaluate the effect of feeding different levels of rumen undegradable protein (UDP) in concentrate on performance of five breeds of sheep. Namely: Compass Agrinak (CA); Garut Composite breed (KG); Barbados Cross breed (BC); St Croix breed (SC); and Local Garut (GL) breed. Ten heads of sheep were used from each breed, in which each breed was grouped into 5 groups according to their body weight, and each sheep in each group was assigned to one of two treatments diet. The diet treatment consisted of concentrate containing different levels of undegradable protein (UDP). The levels of undegradable protein were 4.5% and 7.5%. Concentrate were formulated in iso nitrogen (CP content 16.7%) and iso energy (ME content 2.5 Mcal/kg). During feeding trial the sheep were kept in individual pen for three months. Study was conducted in randomized complete block design in factorial 5 x 2 arrangement, 5 levels of breeds sheep and 2 levels of UDP content. Results shows that there was no interaction effect of breed and levels UDP on feed consumption, average daily gain and feed conversion. These variables were significantly (P<0.05) affected by breed of sheep. The highest DMI (dry matter intake) was in KG sheep, the highest DMI caused by highest crude protein and gross energy intake, which in turn lead to highest average daily gain (ADG) of this breed. However the ADG of KG was not significantly different from GL. Apparent nutrient digestibility was not affected (P>0.05) by interaction between breed of sheep and UDP levels in the diet except for OM digestibility. While breed of sheep did not affect nutrient digestibility and UDP levels only affected NDF digestibility. From this study, it is concluded that increasing UDP in the diet did not improve growth performance of sheep. At similar quality of feed the growth performance was affected by breed of sheep in which KG and GL sheep had similar average daily gain (86.01 vs 82.38 g/day). Abstract

In term of availability, cacao pod is potential for ruminant feed. According to the nutrients content, cacao pod can be used as fiber source feed. Protein sources materials must be combined when cacao pod was ensilage due to low protein content of this material. The aim of study was to investigate digestibility value and end products of rumen fermentation of goat fed by grass or cacao pod based ration. The study used randomized block design to 20 lambs (16.95 ± 2.36 kg) to evaluated 5 type of rations: R (50% grass + 50% concentrate); S (50% cacao pod silage + 50% concentrate); SG (50% cacao pod-gliricidia silage + 50% concentrate); SK (50% cacao pod-calliandra silage + 50% concentrate) dan SC (50% cacao pod-mixture of gliricidia-calliandra silage + 50% concentrate). Feeding trial was conducted over 15 weeks. Measurements were undertaken on feed digestibility and rumen-fermentation end-products after 3 weeks of the animals treated by treatmets rations. Results shows that nutrients digestibility was different significantly among the groups of treatments (P<0.05). Digestibillity of organic matter, NDF and energy of R ration was higher significantly (P<0.05) than those of other groups. N-ammonia of rumen from goat feed R ration was higher (P<0.05) than other groups. Total VFA and each component were different among the groups (P<0.05), however the value was similar among the groups of cacao pod silage rations. It is concluded that cacao pod silaged based rations enriched by Gliricidia and Calliandra leaves did not produced  similar digestibility value and end products of rumen  fermentation with grass based ration.Key Words: Cacao Pod Silage, Digestibility, Ruminal Fermentation
