Abstract

Premenstrual syndrome (PMS) is physical and non-physical symptoms experienced by women of the reproductive age two to fourteen days before menstruation. One of the factors can cause PMS is stress. Objective: To determined the correlation between stress and premenstrual syndrome among year 2021 Faculty of Medicine students at University of Tarumanagara. Methods: This research was an observational analytic study with a cross-sectional research design. The stress variable was measured using the Depression Anxiety Stress Scales-21 (DASS-21) questionnaire and the premenstrual syndrome variable was measured based on prospective questionnaire by the American College of Obstetricians and Gynecologists (ACOG). The number of respondents obtained was 138 and the chi-square test was analyzed as a hypothesis test. Results: There were 56 (62,2%) respondents classified as having stress and experienced PMS, while 15 (31,3%) respondents not having stress experienced PMS. The 34 (37,8%) respondents were classified as being stressed but not experiencing PMS, and 33 (68,8%) respondents were not stressed and not experiencing PMS. The affective and somatic symptoms most often experienced irritability (77,54%) and joint or muscle pain (49,28%). There was a significant relationship between stress and premenstrual syndrome with p-value = 0,001 (p-value <0,05). Conclusion: There is a statistically significant relationship between stress and PMS (p<0,05). Affective symptoms were felt more than the somatic ones. Keywords: DASS-21, diagnostic criteria of PMS, premenstrual syndrome, stress

Abstract

Burns with diabetes mellitus can interfere with the wound-healing process. Mesenchymal stem cells have been studied to accelerate the healing of burns, one of which originates from the bone marrow. Objective: To determined the effect of human bone marrow mesenchymal stem cells (hBM-MSCs) on burn wound healing in granulation tissue formation in Diabetes Mellitus rats. Methods: This study was an experimental study with a post-test-only control group design, using 30 rats divided into two groups, the control group Phosphate Buffer Saline (PBS) and the treatment group (hBM-MSCs). Mice were induced by alloxan to cause hyperglycemia. Burns are made using a heated plate. Tissue collection was carried out after the termination of the experimental animals on the 3rd, 7th and 14th days and then histological preparations were made to assess the formation of granulation tissue. Data were analyzed using the two-way ANOVA test. Results: There was an increase in the number of macrophages, fibroblasts and new blood vessels after being given hBM-MSCs. On day 14 neutrophils and lymphocytes on day 3, there was no increase. Statistical analysis showed a significant increase in lymphocytes, macrophages, fibroblasts and new blood vessels. Conclusion: Human bone marrow mesenchymal stem cells can increase the formation of granulation tissue by increasing the number of macrophages, fibroblasts and new blood vessels in diabetic rats. Keywords: burn wound,  diabetic rat, human bone marrow mesenchymal stem cells, granulation tissue

Abstract

A  decrease in high-density lipoprotein (HDL) happens in type 2 diabetes mellitus. The cytokines TNFa and IL-6 are proinflammatory cytokines that are elevated in the blood in many patients with DM. Turmeric is one of the natural ingredients with antioxidant, anti-cholesterol, and anti-inflammatory properties through its active substance, curcumin. Objective: To prove the effectiveness of the combination of metformin and turmeric extract on HDL levels, TNFa levels, and IL-6 levels in Streptozotocin-Nicotinamide-induced Wistar rats. Methods: The study used a post-test-only controlled group design. The subjects were 24 male Wistar rats which were randomly divided into four groups, namely K1, K2, P1, and P2. The whole group was a type 2 DM model induced by streptozotocin-nicotinamide. Group K2 was given metformin 45mg/KgBW/day, group P1 was given turmeric extract at a dose of 200 mg/KgBW/day, and group P2 was given a combination of metformin and turmeric extract. On day 15, blood was drawn for HDL, IL-6, and TNFa examination. The data were analyzed using the one-way Anova test. Results: The mean HDL level in the P2 group (72,4±33,31) was the highest compared to the other three groups. The lowest mean TNFa level among the four groups was the P2 group (7,56±0,44). The lowest mean levels of IL-6 were obtained in the P2 group (58,82±2,15). One-way Anova test on HDL, IL-6, and TNFa levels showed a significant difference between groups with p-value = 0.000. Conclusion: The combination of metformin and turmeric extract can increase HDL levels and reduce IL-6 levels and TNFa levels better than giving metformin or turmeric extract alone. Keywords:  turmeric extract, HDL, IL-6, TNFa

Abstract

Mesenchymal Stem Cells Wharton's Jelly (MSC-WJ) has become a new hope for Alzheimer's disease due to its capability to suppress Amyloid-beta (Aβ) plaques formation. It is found that people with Alzheimer's disease have a lower expression of Sirtuin-1 because of the oxidative stress that was produced by Aβ plaques. Objective: To observe the Sirtuin-1 gene in Alzheimer's disease rat models that AlCl3 was inducing after being given MSC-WJ. Methods: This was an experimental study with a post-test-only control group design, including three groups with a total sample of 18 samples of Alzheimer RNA rat models. Group-1without AlCl3 and MSC-WJ; Group-2 with AlCl3; Group-3 AlCl3 and MSC-WJ. Mean value of the Sirtuin-1 gene was obtained by the Image-J application using a semiquantitative method which was then tested using the One Way ANOVA test. The study was significant if the p-value < 0,05. Results: This study found that the mean ± standard error value of the Sirtuin-1 gene in three groups were 0.18±0.04; 0.16±0.03; and 0.36±0.06 respectively. The statistical test showed a significant difference between the three groups (p = 0.014). Conclusion: There was an increased Sirtuin 1 gene expression in Alzheimer’s rat Model after being given Mesenchymal Stem Cells Wharton’s Jelly. Keywords:  Alzheimer, MSC-WJ, Sirtuin-1
