Abstract

Belimbing wuluh (Averrhoa blimbi) fruit is commonly used traditionally as antifertility. The bioactive compounds of A. blimbi have been known could inhibit spermatogenesis and development of testes, as well as decrease the quality and quantity of spermatozoa, thus, caused seminiferous tubular atro- phy. This study aimed to determine the reversibility time in testicular damage after the treatment of A. blimbi fruit extract on male Wistar rat. The method employed a completely randomized design (CRD) consists of nine treatments with three replications each. The treatment of the extract at a dose 1140 mg/ kg BW and control group was given the solvent. After 14 days of treatment, the reversibility time was examined in testis histological section by interval of seven days each until 49 days. The results showed that the treatment of A. blimbi fruit extract caused disruptions in seminiferous tubules, i.e. decreased the number of spermatogonium and spermatid, as well as the diameter of lumen and seminiferous tu- bules. The reversibility time was observing at 21 days after the end extract treatments, by increasing the number of of spermatogonium and spermatid, as well as the diameter of lumen and seminiferous tubules that signi cantly different with infertile rat (p<0.05). In conclusion, the reversibility time was 21 days after the treatment of A. blimbi fruit extract as antifertility. Key words: Averrhoa blimbi, rats, reversible time, tubulus seminiferous

Abstract

Pretrombin merupakan prekursor dari trombin yang memiliki aktivitas proteolitik. Trombin merubah fibrinogen menjadi benang fibrin yang salah satu aplikasinya adalah dapat digunakan sebagai lem untuk menggantikan teknik jahitan pasca bedah. Aplikasi trombin untuk pembuatan lem fibrin menuntut diproduksinya trombin rekombinan. Tujuan dari penelitian ini adalah ekspresi gen pretrombin-2 (PT2) manusia rekombinan menggunakan sistem ekspresi Pichia pastoris. Gen pengode PT2 dirancang sesuaidengan kodon preferensi P. pastoris. Fragmen PT2 diamplifikasi dengan metoda PCR dengan penambahan sisi restriksi EcoR1 pada ujung 5’ dan sisi restriksi SacII pada ujung 3’. Produk PCR yang berukuran 924 pb diligasi dengan vektor ekspresi pPICZaB untuk P. pastoris dan disubkloning dalam inang Escherichia coli. Urutan nukleotida dikonfirmasi dengan metoda dideoxy Sanger. Plasmid rekombinan pPICZaB-PT2 kemudian digunakan untuk mentransformasi P. pastoris SMD1168 defisien protease dengan metoda elektroporasi. Hasil penelitian menunjukkan bahwa gen PT2 berhasil diamplifikasi dan dikloning dalam E. coli. Analisis restriksi dan penentuan urutan DNA menunjukkan bahwa PT2 rekombinan 100% homologi dengan hasil rancangan. Hasil ekspresi PT2 oleh P. pastoris menggunakan metanol sebagai inducer memperlihatkan bahwa PT2 dengan berat molekul 35 kDa berhasil diekspresikan. Optimasi kondisi ekspresi melalui variasi konsentrasi metanol sebagai inducer dan sorbitol sebagai sumber karbon tambahan menunjukkan bahwa metanol 2% dan sorbitol 2% merupakan kondisi optimum ekspresi PT2.

Abstract

Microcrystalline cellulose is the most used material for medicine, which able to be found in fibrous plants. Microcrystal celluloses are being used as filler or binder in dosage formulas in tablets and capsules. This research aimed to produce microcrystalline cellulose from ramie based on pharmaceutical grade parameters. Research method include hemp fiber preparation, α-cellulose isolation, microcrystalline cellulose production, and microcrystalline cellulose characterization which compare with Avicel® PH 102 . Result shown microcrystalline cellulose yield is 57.26%. The result of physicochemical characterization that can comply the specifications of pharmaceutical grade as a pharmaceutical excipient.  Keywods :  Microcrystalline cellulose, Ramie, physicochemical, pharmaceutical grade.

Abstract

Body odor of human skin usually occur due to excessive production of sweat glands couples with the presence of bacteria. The active ingredient to prevent body odor are chemicals materials from alum or sodium in order to have negative effects such as skin irritation, Alzheimer disease, prostate and breast cancer. Natural materials technology has been development as an active agent by using cucumber (Cucumis sativus .L). The purpose of this research to conduct a deodorant roll on a stable and qualified physical quality and also develop the use of extract cucumber as an antibacterial Staphylococcus epidermidis. Cucumber extraction has been done by maceration with ethanol 70% for 4 days and then divided into three formulation extract (5%, 10%, and 20% (b/v)). All of the formulations has been tested by zone inhibition, organoleptic, homogenity, pH, viscosity and irritation test. The results given that all of the formulations meet the quality requirements of physical and the best formulation extract 20% able to inhibit bacterial by 26 mm as a largest diameter of the inhibitory zone. In addition the stability of deodorant roll on form were performed accelerated stability test for 35 days at room temperature (28°C) and 40°C. The test results indicate that the stability of the extract 10% quali ed physical quality during storage at room temperature (28°C) and high temperature (40°C) for 35 days. While in the extract 20% at high temperature (40oC) physical changes characterized by the deposition. Keyword : cucumber, deodorant roll on, antibacteria

Abstract

Free radicals in the body can lead to wrinkle. The use of antioxidants can prevent wrinkle. Black rice known to have an antioxidant effect. The purpose of this study is to get a microemulsion preparation of black rice extract that stable and has antiwrinkle effect. Research was started by antioxidant power test in vitro against DPPH, then the optimization of the microemulsion preparation base. The optimum base formula then formulated with black rice extract. Then tested the chemical and physical stability test, also antiwrinkle effect. The results showed that black rice extract microemulsion has composition as black rice extract of 4%, VCO of 28.8%, Croduret-50-SS of 28.8%, glycerol of 28.8%, and citrate phosphate buffer pH 3.0 of 9.6%. The antiwrinkle effectivity of black rice extract in microemulsion form was more than in emulsion form. The average decrease of wrinkle level by black rice extract microemulsion was 44.46%±19.7%, and by black rice extract emulsion was 36.6%±19.5%.       Keywords: Antiwrinkle, black rice, microemulsion

Abstract

Escherichia coli is a gram negative bacterium which is a normal flora in the digestive tract. In birds, this bacterium causes a disease known as colibasillosis. Antibiotics are generally used, but excessive use will cause residues and antibiotic resistance. To avoid resistance or residue, an alternative treatment is needed. The combination of propolis and ginger extract is very promising to develop because both have a synergistic effect as antimicrobials. The research aims to determine the effect of the combination extract on Escherichia coli bacteria in vitro. We used 8 groups which contain combinations of propolis and ginger extract. We used amoxicillin disk as a positive control, 1% Na CMC as a negative control. The results showed a combination of ginger and propolis extract showed good inhibitory activity against Escherichia coli in all groups> 6mm., and the highest inhibitory activity was K3 (5% propolis combination and 15% ginger extract) with 8.7 mm. The combination of propolis and ginger extract has the potential to be used as an alternative antibiotic to prevent antibiotic resistance from synthetic antibiotics.Keywords: alternative antibiotics, combination extracts, Escherichia coli, ginger, in vitro, propolis

Abstract

Kulit buah delima (Punica granatum L.) diketahui memiliki kandungan asam elegat dan asam galat yang menghambat enzim tirosinase, serta punicalagin yang menghambat reaksi oksidasi L-tirosin dan L-DOPA dalam mekanisme pembentukan melanin sebagai penyebab dari hiperpigmentasi. Penelitian ini bertujuan untuk membuat formulasi sediaan krim antihiperpigmentasi yang mengandung ekstrak kulit buah delima. Metode penelitian meliputi ekstraksi kulit buah delima, formulasi sediaan krim antihiperpigmentasi, evaluasi fisik sediaan, pengujian aktivitas penghambatan tirosinase, pengujian cemaran mikroba, dan ujian iritasi sediaan. Hasil penelitian menunjukkan bahwa ekstrak kulit buah delima dapat diformulasikan menjadi krim antihiperpigmentasi yang baik, efektif, dan aman. Namun, krim yang mengandung ekstrak kulit buah delima 1% menunjukkan ketidakstabilan fisik. Sediaan krim dengan berbagai konsentrasi ekstrak kulit buah delima (0,5% dan 1%) efektif menghambat enzim tirosinase dengan nilai IC50 berturut-turut sebesar 363 ppm dan 290 ppm. Kata kunci: Antihiperpigmentasi, ekstrak kulit buah delima, krim

Abstract

Nano-fitosom merupakan suatu teknologi nano yang digunakan untuk meningkatkan bioavailabilitas bahan aktif yang terkandung dalam tanaman dengan cara mengikat bahan aktif dengan fosfolipid yang memiliki sifat mirip dengan membran sel. Nano-fitosom dibuat dengan mencampurkan fitokonstituen dan fosfatidilkolin dalam perbandingan molar tertentu. Penelitian ini bertujuan untuk mengembangkan formula nano-fitosom serbuk liofilisasi seduhan teh hitam (Camellia sinensis L. Kuntze). Nano-fitosom diformulasikan dengan membuat tiga variasi perbandingan katekin:fosfatidilkolin:kolesterol mulai dari 1:1:0,2 (F1), 1:2:0,2 (F2), dan 1:3:0,2 (F3) dengan menggunakan metode refluks. Evaluasi nano-fitosom meliputi ukuran partikel, indeks polidispersitas, dan efisiensi penjerapan. Hasil evaluasi menunjukkan bahwa ukuran partikel nano-fitosom sekitar 52,1-101,9 nm dengan nilai indeks polidispersitas 0,309-0,404. Efisiensi penjerapan berkisar antara 6,7266%-11,4317%. Formula nano-fitosom dengan efisiensi penjerapan yang paling baik yaitu F3 dengan perbandingan molar katekin:fosfatidilkolin:kolesterol 1:3:0,2 dengan ukuran partikel sebesar 52,1±12,5 nm, indeks polidispersitas 0,320 dan efisiensi penjerapan 11,4317±0,3153%. Analisis data menggunakan One-Way Anova menunjukkan bahwa terdapat pengaruh konsentrasi fosfatidilkolin terhadap efisiensi penjerapan secara signifikan (p < 0,05).Kata kunci: nano-fitosom, fosfatidilkolin, serbuk liofilisasi, teh hitam (Camellia sinensis)

Abstract

Nypa fruticans Wurmb which belongs to the family of Arecaceae is one of the most widely distributed and useful palm in the mangrove forests in the Southeast Asia. Nypa palm sugar or commonly known as “Gula Apong” was processed using nypa palm sap as raw material. The nypa palm sugar was expected to benefit wider community as source of alternative sweetener. In this study, 2, 2-diphenyl1-picryhydrazyl (DPPH) scavenging activity and cytotoxicity test of methanol extract of nypa palm sugar were conducted. The antioxidant activity was determined based on DPPH scavenging activity of extract. The cytotoxicity test was conducted against brine shrimp Artemia salina, and the LC50 value of the extract was calculated. The antioxidant activity showed that the EC50 of nypa palm sugar was 1304 mg/mL while the value of EC50 for ascorbic acid was 0.6112 mg/mL. The results of the cytotoxicity showed that the methanol extract of nypa palm sugar was interpreted as non-toxic as the value of LC50 was 184.0 mg/mL.Keywords: 2,2-diphenyl-1-picryhydrazyl, antioxidant, cytotoxicity, Nypa fruticans, nypa palm sugar

Abstract

Chondroitin sulphate (CS) in oral dosage form has widely used to help manage osteoarthritis, but regret its bioavailability was very low at only 13%, It was due to the first passed effect in the liver and to avoid it CS is needed to be formulated into another forms of preparation like patch. This study was aimed to gure out in-vitro possibility of chondroitin sulphate (CS) to be formulated in a transdermal patch and characterized by its physical properties such as permeation profile through the Franz’s cell and partition coefficient studies. The study showed that 150 mg CS in transdermal patch given the constant permeation within 19 hours with the partition coefficient of 2.22. Based on the permeation profile and the partition coefficient study, the CS patch would become an effective preparation to be considered.Keyword : Chondroitin Sulphate (CS), Transdermal patch, Permeation profile, Partition coefficient
