Abstract

Aspergillus niger is pathogen fungi that can live in various locations and can live contiguous with many hosts, one of them is queen termite’s nest. The aims of the study were to isolated the secondary metabilite of A.niger. Extraction proccess of secondary metabolite compounds was carried out by maceration method using methanol solvent. Based on that proccess, methanol extract was be yield 4,32% sample weight. Fractination proccess was carried out in the separating funnel using ethyl acetate solvent, which ethyl acetate fraction was be yield 14.39% methanol extract. Separation of the compounds was carried out by column chromatography method using n-hexane and ethyl asetate eluents. Purification of the compounds were done by recrystallization method using n-hexane and ethyl asetate. Two secondary metabolite compounds were successfully isolated from ethyl acetate fraction of the methanolic extract of fungus A. niger “In-Habiting” queen termite’s nest M. gilvus Hagen. Based on organoleptic examination, the compound signed AM-12-22-01 is 35 mg, white needle crystals, melting point 151-153 oC. While, the AM-12-60-01* is 15 mg, white needle crystals, melting point 91-93 oC. Based on the chemical analysis, thin layer chromatography, ultraviolet and infrared spectra data it was identified that AM-12-22-01 and AM-12-60-01 were a phenolic compounds.Key words: isolation, A. niger, In-Habiting, M. gilvus Hagen.

Abstract

Diabetes mellitus (DM) is a chronic disease which becoming health community problem in the world. DM patients can be treated by antidiabetic drugs throughout their lifetime. Therefore, to alleviate the concern for the side e ect, it is important to have an alternative therapy which has same e ect but less side e ect. One of alternative therapies which has been known for a long time ago is Mulberry leaves (Morus alba). For something to be clinically used as an alternative therapy, e ective and lethal dose should be known. Here we tried to discover the lethal dose (LD50) and the liver morphological changes, using the e ective dose that had been known from previous study. In our experiment, we used Morus alba leaves infusion and 25 male Wistar rats. These rats were divided into 1 control group and 4 groups treatment dosage, 0.3, 3, 30, and 300 grams/kg bodyweight. We observed the mortality in 7 days and examined the liver morphological changes. From this study, we conclude that 300 grams/ kg bodyweight or 48 grams/kg bodyweigtht in human was pseudolethal dose in male Wistar rats. We did not found any signi cant liver morphological changes. Therefore, we can categorize Morus alba as a harmless alternative therapy. Key words: Alternative therapy, diabetes mellitus, liver morphological changes, lethal dose, Morus alba.

Abstract

Bawang Tiwai (Eleutherine americana (L.) Merr bulbus. is a typical plant from Kutai tribes which commonly used as an anti-bacterial and food coloring. The stability test is performed to determine the stability of the extract. Stability test was conducted in two solvents are distilled water and ethanol include the effects of temperature (25°C, 50°C, 80°C), pH (3, 5, 7), an oxidant for 6 hours, conditions of storage at room temperature and the temperature of the refrigerator for 24 hours and storage for 2 weeks. The absorbance of extract in ethanol and aquadest was decreasing due to the change of temperature, time, the presence of oxidator but due to pH changes, the absorbance of extract was increasing. Meanwhile, on the storage condition, showed the highest absorbance on dark, and room temperature storage compared to other condition.Key words: Eleutherine americana (L.) Abstract

Glimepirid merupakan obat antidiabetika oral golongan sulfonilurea generasi ketiga yang termasuk dalam Biopharmaceutical Classification System (BCS) kelas II dengan sifat kelarutan praktis tidak larut dalam air sehingga dapat ditingkatkan kelarutannya dengan pembentukan kompleks inklusi menggunakan senyawa turunan siklodekstrin yaitu betasiklodekstrin yang memiliki rongga toroidal dengan bagian dalam bersifat hidrofobik dan bagian luar bersifat hidrofilik. Penelitian ini dilakukan untuk menentukan harga tetapan stabilitas kompleks berdasarkan parameter termodinamika (ΔH, ΔG, dan ΔS) pada proses pembentukan kompleks inklusi glimepirid-betasiklodekstrin. Penelitian dilakukan dalam dapar asetat pH 6,2 dan dapar fosfat pH 7,4 pada suhu 32°, 37° dan 42°C. Hasil penelitian menunjukkan bahwa glimepirid dapat berinteraksi membentuk kompleks inklusi dengan betasiklodekstrin. Interaksi antara glimepirid dengan betasiklodekstrin pada pH 6,2 berlangsung secara eksotermik (ΔH<0), proses terjadi secara spontan (ΔG<0) dan terjadi peningkatan ketidakteraturan sistem (ΔS positif). Pada pH 7,4 interaksi berlangsung secara eksotermik, proses terjadi secara spontan (ΔG<0) dan terjadi penurunan ketidakteraturan sistem (ΔS negatif).Kata kunci : glimepirid, betasiklodesktrin, kompleks inklusi, termodinamika.

Abstract

Nypa fruticans is a standout amongst the most broadly appropriated and helpful palm in the mangrove backwoods in the Southeast Asia. The sap of this plant is produced at the fruit stalks or called as ower stalks or in orescence. Nipa sap contains a lot of chemical constituents that can be used other alter- native to produce sugar that give a lot bene ts. The crystallization process of this sap was conducted in controlled temperature, pressure and humidity. As the crystal formed, remaining liquid was tested for biological studies which are antidiabetic, antioxidant and toxicity test. The antidiabetic properties were determined by inhibition of α-amylase and the result was compared to the synthetic drug which is acarbose. The sample showed that a high property of antidiabetic (50 mg/mL= 78.50% inhibition) compared to the acarbose (50 mg/mL= 80.90% inhibition). Meanwhile, the antioxidant properties of the sample were determined by2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity. The sample also showed a positive result which is EC50 112.90 mg/mL. Lastly, the toxicity test was tested to brine shrimp (Artemia salina) to get the LC50. The result (LC50=271.7 mg/mL) showed low toxic- ity at different concentration of the sample. From this study, the sample had potential antioxidant and antidiabetic properties, with low toxicity. Abstract

Telah dilakukan kokristalisasi glimepirid (GMP) dengan asam oksalat (AO) menggunakan metode penggilingan dan pelarutan (menggunakan pelarut aseton). Diagram fase sistem biner GMP-AO digunakan untuk identifikasi awal pembentukan interaksi antar kedua komponen serta ditegaskan kembali dengan analisis mikroskopik menggunakan alat pemanas (hot stage) yang dihubungkan dengan mikroskop polarisasi. Padatan hasil kokristalisasi dikarakterisasi dengan metode analisis termal (Differential Scanning Calorymetry), difraktometri sinar-X serbuk (Powder X-Ray Diffraction), spektrofotometri inframerah (Fourier Transform-Infra Red) dan mikroskopi (Scanning Electron Microscope). Hasil identifikasi dan karakterisasi menunjukkan interaksi eutektik antara kedua fase kristalin GMP-AO dalam keadaan padat pada perbandingan molar 3:7, dengan titik eutektik pada temperatur 128,7°C. Selanjutnya, uji kelarutan dan laju disolusinya menggunakan media dapar fosfat pH 7,4. Kelarutan dan laju disolusi GMP hasil kokristalisasi meningkat dibandingkan dengan campuran fisika dan senyawa tunggalnya.Kata kunci : glimepirid, kokristalisasi, eutektik, kelarutan dan laju disolusi.

Abstract

MOCAF (modified cassava flour) is a high potential food product. This product can be used to replace wheat flour in the manufacture of various food products. However, there is no characterization of starch properties of MOCAF. The efforts to improve the quality of starch from cassava starch by fermentation process using a combination of enzymes and microbes  from Lactic acid bacteria.The purpose of this research is to determine the influence of fermentation time to yield and moisture content. Single factor randomized block design with 3 (three) levels i.e fermentation time (0, 12, and 24 hours) with two group namely varieties of cassava Cimanggu and Kaspro. Cimanggu and Kaspro cassava varieties were used to compare the properties of starch due to different fermentation time (0, 12 and 24 hours) with triplicate. The results showed the yield and moisture content were affected by fermentation time. Average moisture content ranged from 13.3 to 13.8% with the starch yield is 38-59%. Application of starch MOCAF can be used for food, cosmetics, and medicines industry.Keywords : modified starch, cassava, fermentation, yield

Abstract

Kulit batang trengguli (Cassia fistula L) mengandung senyawa yang bersifat antioksidan yang dapat ditentukan dengan metode peredaman radikal bebas 1,1-Difenil-2- Pikrilhidrazil (DPPH). Penelitian ini dilakukan untuk mengetahui adanya peredaman radikal bebas DPPH oleh ekstrak etanol kulit batang trengguli (EEKBT), fraksi Etil asetat (FEAKBT), fraksi n-Heksan (FnHKBT), fraksi air (FAKBT) dan vitamin C sebagai pembanding. Identifikasi golongan senyawa dilakukan dengan penapisan fitokimia. Fraksinasi dilakukan dengan metode ekstraksi cair-cair dengan tingkat kepolaran yang berbeda. Aktivitas antioksidan dilakukan dengan peredaman warna radikal bebas DPPH menggunakan spektrofotometri UVVis. Penelitian menunjukkan bahwa Fraksi etil asetat memiliki aktivitas antioksidan paling baik dengan nilai IC50 3,980 μg/ml dibandingkan vitamin C 4,716 μg/ml. Sedangkan nilai IC50 ekstrak etanaol, fraksi n-heksan, fraksi air adalah 10,613 μg/ml, 38,904 μg/ml dan 7,636 μg/ml terhadap peredaman warna DPPH.Kata kunci: Antioksidan, Cassia fistula, Fraksinasi, metode DPPH

Abstract

In the gastrointestinal system, gastric ulcers are one of the common serious diseases. The pathophysiology of gastric ulcer is an imbalance between aggressive and mucosal defense factor. Imbalance between aggressive and mucosal defense factor have contribution against developed of gastric ulcer disease. The objective of the research was to evaluate gastric ulcer healing effect of Bangle (Zingiber cassumunar (Roxb.) rhizome extract in aspirin-induced rats model. Antiulcer activity of Bangle rhizome extract was evaluated by several parameters involving gastric acidity, number of ulcers, diameters of ulcers, ulcer index, and healing ratio. Doses level of Bangle rhizome extract which used in this study such as 50 mg/kg and 100 mg/kg, respectively. Histopathological of the stomach was performed using hematoxylin-eosin stained. The results of study showed that groups which given Bangle rhizome extract have signi cant effect in gastric acidity, number of ulcers, diameters of ulcers, ulcer index, and healing ratio compared to the control group. Histopathological study of the stomach ulcers showed tissues regeneration in group were treated by Bangle rhizome extract compared to the control group. It can be concluded that from the experimental study of Bangle (Zingiber cassumunar (Roxb.)) rhizome extract have potential antiulcer activity in aspirin-induced rats model. Abstract

The aim of this study was to investigate the effects extract of Kenikir in male rats parameters blood glucose, cholesterol and histopathological of the pancreas. Thirty rats were divided into six groups, and each group consists of five individuals with the details of groups I, II, and III as control groups and groups IV, V, and VI for the experiments. Group I: normal control was given NaCMC 0,5%; Group II: control negative was given Streptozotocin 30 mg/kgBW i.p, high feed fat 25 g/day, and the positive control was given met form in 45 mg/kgBW, and the experimental groups were each given extract of the kenikir leaves with dose 100 mg/kg BW, 200 mg/kgBW , and 400 mg/kg BW. The results showed that there were secondary metabolites contained in the extract of leaves of kenikir. Giving the extract of kenikir leaves at the dose of 200 mg/kg is effective in lowering the blood glucose levels, extract of leaves of kenikir with the dose of 400 mg/kg is effective to reduce the total cholesterol levels and regenerate pancreatic tissue, and extract of leaves of kenikir with the dose of 100 mg/kg BW did not give any maximum impacts to decrease the blood glucose.
