Abstract

Kinin adalah senyawa alkaloid dari kulit batang Cinchona sp. yang digunakan sebagai antimalaria. O-desmetil kinin (C19H22N2O2) adalah senyawa turunan kinin baru yang diperoleh dengan menghilangkan gugus metil pada kinin. Selain sebagai antimalaria, kinin juga dapat digunakan untuk mengobati kram kaki. Karena efek samping yang besar, FDA (2009) melarang penggunaan kinin oral dalam pengobatan kram kaki. Oleh karena itu, penelitian ini dilakukan untuk menguji efek kinin dan pada rute pemberian oral dan topikal serta mengetahui efek o-desmetil kinin pada terapi kram kaki. Mencit dibagi menjadi 5 kelompok yaitu kelompok normal, kelompok kinin oral dosis 26 mg/kg bb, kelompok kinin topikal dosis 52 mg/kg bb, kelompok o-desmetil kinin oral dosis 26 mg/kg bb, dan kelompok o-desmetil topikal dosis 52 mg/kg bb. Mencit diletakkan di atas rotarod dengan kecepatan 10-16 rpm selama 4 menit. Waktu yang dapat ditempuh mencit selama berada di atas rotarod diukur. Perlakuan yang sama diulang dengan durasi 45 menit selama 4,5 jam. Setelah menit ke 180, kelompok uji mengalami peningkatan waktu ketahanan di atas rotarod sedangkan kempok normal mengalami penurunan. Pada menit 270, kelompok oral dan topikal memberikan perbedaan yang bermakna dibandingkan dengan kelompok normal. Pemberian kinin dan odesmetil kinin pada rute pemberian oral dengan dosis 26 mg/kg bb dan topikal dengan dosis 52 mg/kg bb sebagai antikram memberikan hasil yang bermakna dibandingkan dengan kelompok normal (p<0,05). O-desmetil kinin memberikan efek yang tidak berbeda bermakna dari kinin base (p<0,05).Kata kunci: kram kaki, kinin, o-desmetil kinin, rotarod.AbstractQuinine is an alkaloid from the bark of Cinchona sp. as an anti-malaria. Because of the great side effects, FDA (2009) banned the use of quinine oral for leg cramps therapy. Therefore, this study was aimed to determine the effect of quinine by oral and topical administration and to determine o-desmethyl quinines effect for leg cramps therapy. Mice were divided into normal group, quinine oral dose 26 mg/kg bw, quinine topical dose 52 mg/kg bw, o-desmethyl quinine oral dose 26 mg/kg bw, and o-desmethyl topical dose 52 mg/kg bw. Mice were taken on the rotarod at 10-16 rpm for 4 minutes. Time to remain on the rotarod was measured. The same treatment was repeated after 45 minutes for 4.5 hours. Over the 180 minutes, the endurance of the test groups were increased on the contrary, the normal group is decreased. After 270 minutes of observations, the oral and topical groups have significant difference compared by the normal group. The administration of quinine by oral dose 26 mg/kg bb and topical dose 52 mg/kg bb as an anti-cramps showed significantly difference compared to the normal group (p<0.05). O-desmethyl quinine has no significantly difference compared by quinine (p<0.05).Keywords: leg cramps, quinine, o-desmethyl quinine, rotarod.

In the gastrointestinal system, gastric ulcers are one of the common serious diseases. The pathophysiology of gastric ulcer is an imbalance between aggressive and mucosal defense factor. The results of study showed that groups which given Bangle rhizome extract have signi cant effect in gastric acidity, number of ulcers, diameters of ulcers, ulcer index, and healing ratio compared to the control group. Histopathological study of the stomach ulcers showed tissues regeneration in group were treated by Bangle rhizome extract compared to the control group. It can be concluded that from the experimental study of Bangle (Zingiber cassumunar (Roxb.)) rhizome extract have potential antiulcer activity in aspirin-induced rats model. Abstract

ABSTRAK: Perbedaan letak geografis suatu tanaman dapat menyebabkan perbedaan pada kandu–ngan kimianya. Kandungan kimia yang berbeda dapat menyebabkan perbedaan aktivitas farmakologitanaman, salah satunya adalah aktivitas antioksidan. Pada penelitian sebelumnya telah dilakukan berbagaipengujian antioksidan terhadap secang yang berasal dari beberapa lokasi baik di luar nege–ri maupun Indonesia. Oleh karena itu pada penelitian ini dilakukan pengujian aktivitas antioksidanekstrak etanol kayu secang (Caesalpinia sappan) yang berasal dari B2P2TOOT Tawangmangu. Metodeekstraksi yang digunakan dalam penelitian ini adalah soxhletasi dengan pelarut etanol 80%. Skri–ning fitokimia dilakukan untuk mengetahui kandungan kimia yang terdapat dalam ekstrak etanolkayu secang. Ekstrak tersebut menunjukkan hasil positif pada golongan glikosida flavonoid, flavonoidbebas, alkaloid, dan polifenol. Pengujian aktivitas antioksidan menggunakan metode peredaman radikalbebas DPPH, ABTS, dan FRAP. Trolox digunakan sebagai standar pembanding untuk menunjukkanaktivitas antioksidan. Hasil pengujian aktivitas ekstrak etanol kayu secang dan trolox menunjukkannilai IC50 masing-masing sebagai berikut: 101,47 dan 76,15 ppm (DPPH); 26,70 dan 19,38 ppm(ABTS); 11,37 dan 11,04 ppm (FRAP). Hal ini menunjukkan bahwa ekstrak etanol kayu secang memilikidaya antioksidan yang kuat. Polifenol dan flavonoid diduga memiliki peranan penting pada aktivitasantioksidan kayu secang.

Abstract

Beta-adrenergic receptor blockers (β-blockers) are an appropriate treatment for patients with hypertension who have concomitant ischemic hearat disease, heart failure, obstructive cardiomyopathy, aortic dissection or certain cardiac arrhythmias. Taya (Nauclea subdita (Korth) Steud) is a local natural potential of Kalimantan that is empirically believed to have antihypertensive activities. This study aims to determine the activity of β-adrenergic receptor inhibitors of stem bark water extract in male wistar rats. Tests were carried out on male Wistar rats by the Non Invasive Blood Pressure method using the CODA® tool. Test animals were divided into 5 groups: normal group, induction group (Epinephrine 1.2 µg/kgBW), 100 mg/kgBW extract group, 200 mg/kgBW extract group, and 4,5 mg/kgBW atenolol group. The induction group showed an increase in systolic blood pressure, diastolic blood pressure, and heart rate that were significantly different from the normal group, the 100 mg/kgBW extract group, the 200 mg/kgBW extract extract group, and the 4.5 mg/kgBW atenolol group (p<0,05). The 100 mg/kgBW extract group was not significantly different from systolic blood pressure, diastolic blood pressure, and heart rate with the normal group, the 200 mg/kgBW extract group, and the 4.5 mg/kgBW atenolol group (p> 0.05). The results of this study showed that taya bark water extracts 100 mg/kgbw had inhibitory activity of β-adrenergic receptors in male wistar rats.

Abstract

Herbal Products of Moringa oleifera was produced by KWT (Farmer Woman’s Group). These product were certified by DINKES (Health Unit Ministry) as PIRT (Pangan Industri Rumah Tangga). These products has been marketed with limited information on the packaging. Based on Indonesian Regulation about Safety Product for Consumen (UU RI No. 8 year 1999), the consumers must be guaranted about safety, comfortability , and security about the product. The  Objective of the Education program was to give the right information  about the Nutrition Content and Safety Aspect From Moringa Tree. Farmer Women’s Group (KWT) “Sri Rejeki” and Farmer Forest Group (KTH) Bogo Village, Kapas Bojonegoro District was the participant of this program. Methods for the Education program was interactive discussion with a competent and expertised  presenter/ trainer. There were two modul program’s book which have been published to improve the knowledge of the partocipant regarding the topics. Result from the questionnaire analysis showed that the knowledge about nutritional content of Moringa oleifera was increased from 77.44 to 88.61 and the knowledge about safety aspect of Moringa oleifera was increased from 37.50 to 69.38.  ABSTRAK Produk olahan kelor yang sudah dipasarkan oleh KWT telah mendapatkan sertifikat PIRT oleh DINKES setempat. Informasi produk pada kemasan belum dituliskan batasan penggunaan dalam sehari, serta informasi nilai nutrisinya. Menurut UU RI No. 8 Tahun 1999 tentang Perlindungan Konsumen, konsumen berhak atas keamanan, kenyamanan dan keselamatan dalam mengkonsumsi barang/ jasa. Tujuan kegiatan sosialisasi ini adalah untuk edukasi terkait Kandungan Nutrisi dan Aspek Keamanan Kelor. Edukasi diberikan pada ibu-ibu KWT Srirejeki dan Bapak-bapak anggota KTH yang menggeluti usaha dibidang produk olahan kelor, agar lebih memahami nilai nutrisi dan keamanan produk yang mereka pasarkan. Metode yang digunakan untuk peningkatan pengetahuan dengan pemberian materi terkait topik, yang diberikan dengan metode ceramah interaktif oleh narasumer yang kompeten dibidangnya. Pada kegiatan ini dihasilkan dua modul pelatihan yakni Modul Kandungan Nutrisi Kelor dan Modul Tinjauan Efektivitas dan Keamanan Kelor. Dari hasil penilaian kuesioner menunjukkan tingkat pengetahuan peserta pelatihan meningkat dari nilai rerata 74.44 menjadi 88.61, sedangkan untuk pemahaman terhadap keamanan kelor pengetahuan peserta masih sangat kurang (rerata skor nilai total 37.50) dan di akhir sosialisasi (rerata skor nilai total 69.38).Kata kunci : pelatihan; edukasi; kelor; nutrisi; keamanan

Abstract

Background: Free radicals consist of one or more unpaired electrons in an outer orbit that causes them to be more reactive. This will lead to oxidative stress, which is one of the major causes of degenerative diseases such as cancer, diabetes mellitus, and stroke. Therefore, consuming sufficient antioxidants can prevent oxidative stress. Faloak (Sterculia quadrifida R. Br.) is one of the endemic medicinal plants in East Nusa Tenggara and is a resource of antioxidants that are supported by scientific evidence. Until now, information about the antioxidant compounds in Faloak was limited. Objective: This study aims to identify active compounds in an 80% ethanol extract from faloak bark as antioxidants based on their TLC-bioautography profiles using an ABTS assay. Method: The methods of this study start with the preparation of simplicia, fractination using kinetic maceration, compound group identification and TLC-bioautography, separation using column chromatography [stationary phase: silica H60; mobile phase: 2-propanol: chloroform: acetic acid glacial (5:1:0,5)], identification using a UV-Vis spectrophotometer, and antioxidant activity determination of the extract and isolate using an ABTS assay. Result: Through this study, a subfraction (SFFE A) with a brown-red color and a blue spot under UV 365 nm and an Rf value of 0.65 was obtained. This subfraction (SFFE A) was predicted to be a polyphenol compound. The inhibition percentages of 125, 250, and 500 ppm successively are 80,93%; 95,54%; and 96,69%.Conclusion: The active isolate of ethanol extracting 80% of the faloak stem bark is suspected to be a polyphenol group and has strong antioxidant activity. Intisari Latar belakang: Radikal bebas merupakan suatu molekul yang terdiri atas satu atau lebih elektron tanpa pasangan pada orbital terluar. Hal ini dapat menyebabkan tubuh mengalami stress oksidatif sehingga memicu munculnya berbagai penyakit degeneratif, seperti kanker, diabetes mellitus, serta stroke. Oleh karena itu, dengan mengonsumsi antioksidan yang cukup dapat mencegah stres oksidatif. Faloak (Sterculia quadrifida R. Br.) merupakan salah satu tanaman obat endemik di NTT (Nusa Tenggara Timur) yang berpotensi digunakan sebagai sumber antioksidan kuat yang didukung oleh studi ilmiah. Hingga saat ini, informasi terkait senyawa yang berperan langsung sebagai antioksidan pada tanaman ini masih terbatas. Tujuan: Penelitian ini bertujuan untuk mengidentifikasi senyawa aktif dari ekstrak etanol 80% kulit batang faloak yang berperan sebagai antioksidan berdasarkan profil KLT-bioautografi dengan metode ABTS. Metode: Metode kerja dimulai dengan preparasi simplisia, fraksinasi dengan metode maserasi kinetik, identifikasi golongan senyawa dan KLT bioautografi, pemisahan dengan kromatografi kolom [(fase diam: silika H60; fase gerak: 2-propanol:kloroform:asam asetat glasial (5:1:0,5)], dan identifikasi senyawa menggunakan spektrofotometer UV-Vis, dan pengujian aktivitas antioksidan pada ekstrak dan isolate menggunakan metode ABTS. Hasil: Melalui penelitian ini, didapatkan subfraksi (SFFE A) berwarna merah-kecoklatan yang memiliki noda berfluoresensi biru pada UV 366 dengan nilai Rf 0,65 dengan pengamatan UV 365 nm. Sub fraksi (SFFE A) yang didapatkan diduga merupakan senyawa golongan polifenol. Persentase inhibisi subfraksi pada konsentrasi 125, 250, dan 500 ppm secara berturut-turut adalah 80,93; 95,54; dan 96,69%. Kesimpulan: Isolat aktif dari ekstrak etanol 80% kulit batang faloak diduga merupakan golongan polifenol dan memiliki aktivitas antioxidan yang kuat. Abstract

Introduction: The herbal medicine business (in Bahasa called as Usaha Jamu Gendong or UJG) "Jamu Seger Bu Mur" is one of the small business engaged in the field of herbal or herbal drinks located around the University of Surabaya campus. This UJG has various obstacles to survive in the midst of changing times and business competition in urban areas. Objectives: This mentoring program is intended to increase the capacity and competitiveness of UJG "Jamu Seger Bu Mur" so that it can continue to run its business, as well as one of the efforts to increase the role of educational institutions in the preservation of Indonesian traditional medicines. Methods: This program was carried out using a mentoring approach model, where mentoring activities were divided into two stages, namely training in making instant herbal medicine (powder form) and assistance in improving product packaging and labels. Results: The results obtained from this program were an increase in the skills of partners to diversify herbal dosage forms, namely instant powder forms. Abstract

Cultural identity may indirectly determine how a person within a particular cultural context takes action to prevent and treat a condition or a disease. Knowledge is one of predisposing factors that can influence both cultural identity and health behavior. Rural communities in Indonesia have their own cultural identity that may influence the health-related behavior of the residents. The Village of Duyung is one of example of this case. The lifestyle and medicine-related behavior of the residents might be influenced by the nature of its collective or communal culture. This report aims to present the level of awareness of the principles of healthy lifestyle and appropriate medicine-related behaviour in the Village of Duyung. A community empowerment program was designed and conducted in July and August 2023. All 23 participants were women with age ranged from. 26 to 47 years. The proportion of participants who were unaware of the principles of healthy lifestyles and medicine-related behavior was as high as 78.3 and 87.0%, respectively. There is a statistically significant difference in the final score of knowledge before and immediately after the training on the principles of healthy lifestyle and medicine-related behavior. The program demonstrated a significant change of the awareness and knowledge of some participants in the Village of Duyung on healthy lifestyle and the appropriateness of medicine-related behaviour. The impact on how this method of education contributes to cultural identity and consistent health-related behavior over longer time needs further study.
