Abstract

Banana Bunchy Top Disease (BBTD) caused by Banana Bunchy Top Virus (BBTV) is one of the most serious banana diseases, constraint and devastate to the local and regional banana production. Some of banana cultivars were more readily infected by the virus, but considering no cultivars is resistant. The incidences of BBTD and the type and severity symptoms in natural conditions without any artificial inoculation were recorded. The observations results on 2009 showed that 12.14% of the total accessions indicated positively infected BBTD. The BBTD incidences were increased rapidly about twofolds per year, 28.57% in October 2011 and 54.29% in October 2011. The spread and development of BBTD is optimum at humid, high rainfall intensity and temperatures, high soil fertility and also low light intensity or shaded. The latest monitoring on February 2012 (62.14%) comprises of 64 accessions were positively infected and 39 accessions were symptomless. Those symptomless accessions were initially indicated as tolerant banana cultivars to BBTD e.g. Pisang Kepok (BBB), Pisang Sobo (BBB), Pisang Bandung (ABB), Pisang Nangka (AAB), Pisang Candi (AAB) and  Pisang Raja Marto (AAB). These preliminary results are in agreement with several previous studies indicated that genotypes with one or two B genomes tend to be more tolerant to BBTD. However several cultivars which known tend to BBTD tolerant are indicated susceptible to Blood Disease. Some cultivars also showing complication symptoms both BBTD and Blood Disease.

Abstract

Treating gaharu (Aquilaria microcarpa) seedlings with inorganic fertilizers is essential to obtain vigorous seeds with high adaptability for establishment on the field. The aims of this research were to find out the effect of fertilization on the survival and growth rate of Aquilaria microcarpa seedlings and to identify appropriate dosage of fertilizer that could increase the growth optimally. Complete Random Design was employed with two treatments i.e. using root fertilizer (Urea, TSP, KCL) with 5 dosage levels and using leaf fertilizer (Gandasil D and Bayfolan) with 7 dosage levels. Data was analyzed by using statistic descriptive and analysis of variance (ANOVA) method. The parameters used were seedling survival rate, height increase and number of leaves. The result showed that the two fertilizers could increase survival rate of the seedling. However, higher dosage of root fertilizer was followed by lower survival rate. The treatment using root fertilizer had a significant effect on the number of leaves, whereas treatment using leaf fertilizer had a significant effect on the increase of the seedling height. Adding leaf fertilizer is considered more useful in terms of survival and growth rate.

Abstract

Grammatophyllum scriptum is a beautiful wild orchid that has an interesting economic value. However, the existence of this ornamental plant species is at risk because of irresponsible exploitation of the natural population and insufficient cultivation efforts. Currently, the Bogor Botanic Garden is developing a program of orchid propagation for enhancing the conservation of all potential orchid collections. The propagation of G. scriptum was basically carried out by culturing the seeds in a modified Hyponex or Vacin & Went medium, transplanting the plantlets into a more suitable medium and finally acclimatizing the resulting seedlings. A study was done to observe the effect of fertilizer application on the growth of acclimatized G. scriptum seedlings, as a means to improve the propagation technique of the species. The result showed that the application of 1 and 2 m1/I organic fertilizer (Sugih) as well as 1 and 2 g/I inorganic fertilizer (Hyponex) increased the seedling mortality, in which the application of 2 g/ml Hyponex fertilizer was being the most damaging treatment, leaving 12,5 % seedlings to survive. However, in terms of seedling growth, the application of 1 m1/I Sugih fertilizer was slightly beneficial to the formation of root and the elongation of stem of the acclimatized seedlings. It may be suggested, therefore, that G. scriptum seedlings require no (or very little amount of) supplementary fertilizer to boost their vegetative growth.

Abstract

Osmunda angustifolia Ching ex Ching et Wang (Osmundaceae) was, formerly, known to be distributed in China (Guandong), Hainan, Hongkong, and Taiwan. Now, O. angustifolia is reported as a newly recorded species in Sumatra. A complete morphological description with photographs is presented. It is defined as a rare species in Indonesia. A cytological record is also reported for the first time for the Sumatran species. The somatic chromosome number of root tip cells of O. angustifolia is 2n = 44 (diploid).

Abstract

Bengkalis Island is one of the areas with high diversity of durian (Durio zibethinus Murr.). This research was aimed to analyze the diversity of  durian cultivars of Bengkalis Island. It was conducted from April up to August 2010. The morphological data were analysed using NTSYS-pc 2.02 and minitab 14.13 softwares. Genetic similarity value among 36 accesions ranged from 0,28 (between BT 14, BT 15 and SB6) to 0,66 (between BT6 and BT7). The cluster analysis showed that durian diversity from Bengkalis island was 34-58% or had similarity level 42-66% and formed 7 main groups. However, they were ungroupped depend on their growth areas. Based on PCA (Principal Components Analysis) value, durian diversity was 99,7% and formed 6 groups.  Furthermore, the result of pearson correlation analysis among 77 character showed that 10 characters were positively correlated and 4 characters were negatively correlated.

Abstract

Pranajiwa (Euchresta horsfieldii (Lesch.) Benn) is a medicinal plant, which is growing wild and it is regarded as a rare plant. Currently, its existence in the wild increasingly threatened. Pengelengan Hill is one of the natural habitats of E. horsfieldii in Bali. Study of autecology aims to describe the ecology of E. horsfieldii in their natural habitat. E. horsfieldii was found abundant in plots II with 16.02% relative density, 9.68% of relative frequency and 25.69 of Important Value, grow clumped together with other plants (Idk 2.72).

Abstract

Mature seeds of a pitcher plant (Nepenthes grocilis Korth.) were cultured in seven different media, namely half strength MS medium, half strength MS medium supplemented with BAP and NAA, 0.8 KC medium supplemented with coconut water + bean sprout extract, 0.8 KC + H3B03 + CuSO3 + ZnSO4 + coconut water + bean sprout extract, 0.25 KC + Vitamin (MS), HYPONeX + coconut water + sprout bean extract, and HYPONeX + potato extract. Each culture was incubated under light conditions. Young seedlings were subsequently transferred onto fresh media of 0.5 MS and 0.25 KC supplemented with 0, 5, 10, and 15 mg/I GA3 after 15 weeks of sowing. Seed germination and seedling growth were observed weekly. It was noted that the seeds started to germinate in ten weeks after incubation and the highest seed germination was obtained in HYPONeX medium + potato extract. However, the appearance of the seedlings was better on MS media and KC media without organic compound than on the other media. Seedlings in all level of GA3 in half strength MS media and 0.25 KC media grew faster than those in media without GA3.

Abstract

Salah satu lokasi di Taman Nasional Bukit Barisan Selatan (TNBBS) yang belum banyak diteliti adalah di kawasan Gunung Sekincau pada ketinggian 1100–1719 m dpl. Kawasan ini termasuk dalam kategori hutan hujan pegunungan bawah. Penelitian ini dilakukan dengan tujuan untuk mengetahui variasi struktur dan komposisi pohon di kawasan hutan Gunung Sekincau TNBBS. Pengumpulan data dilakukan melalui penarikan petak-petak cuplikan yang dibuat secara acak terpilih (purposive random sampling) pada 5 lokasi sebanyak 10 petak cuplikan berukuran 50 x 50 m2 atau seluas 0,25 ha. Hasil penelitian menemukan 32 suku 59 marga dan 89 jenis dengan kerapatan 253 ind/ha. Kawasan Gunung Sekincau memiliki kondisi yang cukup baik, hal ini diindikasikan dengan nilai rata-rata indeks keanekaragaman jenis (2,67) dan indeks kemerataan jenis (0,47). Pohon yang sering ditemukan di kawasan ini adalah anggota suku Lauraceae, Fagaceae, Euphorbiaceae, Dipterocarpaceae, Myrtaceae, dan Rubiaceae. Quercus blumeana Korth. (kayu pasang) merupakan jenis penting (INP=27.18%) dan memiliki persebaran luas pada kawasan Sekincau. Hutan di kawasan ini mempunyai peranan yang penting bagi penyangga kehidupan dan lingkungan. Maka dari itu, pengelola TNBBS dapat meningkatkan penjagaan dan pemeliharaan kawasan serta vegetasi di dalamnya. Terlebih kawasan Sekincau berbatasan langsung dengan pemukiman penduduk, sehingga diperlukan pengelolaan secara berkelenajutan guna memenuhi kebutuhan masyarakat sekitar.

Abstract

Ekinase (Echinacea purpurea (L.) Moench) a medicinal plant that has immunostimulatory activity. This plant has been cultivating in Tawangmangu region by Medicinal Plant and Traditional Medicine Research and Development Office since 2002. Ten accessions of E. purpurea were found based on their morphological variation, three accessions of them are selected as promising accessions namely BH2, BHU3 dan BHU5. The objective of this research was to observe the genetic diversity of  those  promising accessions and 8 variants accession from mass selection year I using RAPD analysis. Those accessions were amplified using 10 RAPD primers. A total of 64 scorable fragments were generated from 9 RAPD primers, among which 48 fragments (75%) were polymorphic. The Dice coefficient was used to calculated the genetic similarity and UPGMA was used to generate the dendogram. The genetic similarity index among accessions evaluated ranged from 75,49-84,21% thus indicating that low level of genetic diversity. RAPD analysis proved to be efficient for genetic diversity of ekinase accessions from mass selection year I.
