Abstract

Cancer was ranked first as the cause of death in all over the world. Chemopreventive sought from traditional preparations for example, various types of plant parasites. This study aims to determine the chemopreventive activity of aqueous extract of leaves of parasites jackfruit ( Macrosolen cochinchinensis ( Lour. ) van Tiegh ) on the growth of lung tumors of mice ( Mus musculus ) female strain Balb / c induced benzo [a ] pyrene . This research was conducted using the method Newborn Mice. Test animals used female mice (Mus musculus) strain BALB / c and benzo [ a] pyrene as carcinogenic compounds. Female mice were divided into 5 groups. Cancer control group were injected with B [ a] P were dissolved in dimethylsulfoxide ( DMSO ) . Solvent control group was given the same volume of DMSO in the control of cancer. The treatment group was divided into three dose groups are a dose of 250, 500, and 750 mg / kg . Extract made after female mice induced B [a] P. Chemopreventive intensity expressed as percentage inhibition of tumors in each group by counting the number of tumor nodules on average per lung in each mouse organs within each group. Statistical analysis used is the analysis of Test homogenity of variances and continued with Mann- Whitney Test. Parasite jackfruit leaf aqueous extract (Macrosolencochincinensis (Lour.) van Tiegh) dose of I, II, and III were able to inhibit development into lung of female mice with the percentage inhibition respectively 50 %, 36.87 %, and 84.57 % .

Abstract

Background. Periodontal disease is a disease of the tissues supporting the teeth. Clinical signs of periodontitis is the presence of gingival inflammation, swelling interdental papillae, formation of pocket / gingival pocket and gingival recession. The increase of periodontal disease prevalence in smokers caused by poor oral hygiene and late diagnosis.Method. Reseach objective was to prove the factors that related to periodontal pocket. Study design was cross-sectional. The study population were 85 labours of loading and unloading at the Port of Tanjung Emas Semarang, the collection of data through interviews and clinical observations. Data were analyzed by chi-square test with 95 % Confidence Interval and Prevalence Ratio were calculated. Results. The results showed all respondents have periodontal pocket with an average depth of pocket were 1.4 mm. The proportion of periodontal pocket depth more than 1.4 mm (50.6%) and less than 1.4 mm (49.4%) were almost comparable. Factors significantly associated with periodontal pocket were the duration of smoking (p-value 0.04, 95% CI = 1.037 to 5.941; PR = 2.482), oral hygiene (p-value 0.023, 95% CI = 1.143 - 13.311; PR = 3.9), and saliva buffer capacity (p-value 0.011, 95% CI = 1.283 to 7.984; PR = 3.2). Conclusion. Smoke have a greater risk of periodontal pocket, therefore smokers should be given health education on dental and oral health maintenance in the form of brushing and regular dental plaque cleaning (scaling). Smokers need to reduce the number of cigarettes that smoked or quit smoking.

Abstract

A novel 1-(3-chlorobenzoyl)-1,3-dimethylurea has been designed, synthesized, structurally determined, and the in vitro cytotoxic activity was evaluated. Docking simulation was performed to position this compound into the Checkpoint kinase 1 (Chk1) active site to determine the probable binding model. Synthesis of 1-(3-chlorobenzoyl)- 1,3-dimethylurea was completed by acylation reaction between 1,3-dimethylurea and 3-chlorobenzoyl chloride. The purity of synthesized product was determined by Thin Layer Chromatography and melting point measurement. Structure identification was performed by UV spectrophotometer, FT-IR and NMR spectrometer. Antiproliferative assay result demonstrated that this compound possessed good cytotoxic activity against HeLa cells, which is comparable to the positive control. This compound with potent cytotoxic activity might be a potential anticancer agent.

Abstract

Development of methods to improve delivery of drugs for treating dangerous disease like cancer and viral infections are very important today. Selectivity in therapy is needed, hence capability to deliver drugs at specific site being studied in order to reduce toxicity and unwanted side effect at non target site. Many drugs delivery from passive targeting to active targeting delivery will be improved to reach that purpose. Because this system is very important for cancer therapy, many cytotoxic agent have been approved by clinical examination. Besides that targeting drug delivery is very useful for neurological diseases such as alzhemier, liver diseases, kidney, lung, and colon diseases.

Abstract

Nicotine which is one of toxic component in cigarette smoke had been reported to inhibit androgen biosynthesis and Leydig cell growth. Cigarette smoking alters plasma testosterone concentrations inmen. The effect of cigarette smoke can spoil sperm viability and spermatogenesis. It can cause hormonal condition and trigger the emergence of toxic substance on sperm that it can harm male fertility.The research design employed in this study was Post Test Only Control Group Design. The samples were adult male musmusculus. There were six groups of research subject chosen randomly after homogenization. Three groups were the control group and the other three were the experimental group. The subjects in the experimental group were injected with nicotine subcutaneously for one week, two weeks and three week with the same dosage namely 5 mg/kg bodyweight/day. The final observation was aimed at calculating the number of Leydig cell. The result of the one way Anova showed that the cell Leydig had a statistically significant difference on some experimental groups (p < 0.05). LSD test showed that there was significant difference between Leydig cells of P3 and K3; P2 and K3; P1 and K3. The conclusion of this research was that nicotine caused a decrease in the number of Leydig cell.

Abstract

HIV / AIDS patients are prone to get opportunistic infections because of their low immunity level. The unappropriate treatment of opportunistic infections may result in morbidity and mortality. The research was carried out to analyze the use of anti-infective in the hospital. The study used a non-experimental and descriptive methods retrospectively from patient medical records. A total of 79 patients who met the inclusion criteria were analyzed descriptively. Suitability analysis based on the CDC and WHO guideline. Suitability analysis of drugs according to WHO and CDC in the case of TB / tuberculosis, candidiasis, pneumonia, PCP / Pneumocystis carinii pneumonia, (hepatitis C, toxoplasmosis, hepatitis B, meningitis, CMV infection / Cytomegalovirus consecutively were 95.2%, 96.8%, 64.3%, 81.8%. Suitability candidiasis dose and frequency according to the CDC : nystatin: underdose (81.8%), overdose (2.3%); fluconazole: appropriate dose (20.5%), underdose (4.5%), overdose (72.7%); itraconazole: underdose (100%), according to WHO: nystatin: appropriate dose (40.9%), overdose (43.2%); fluconazole: appropriate dose (50%), underdose (50%); ketoconazole: appropriate dose (100%). Suitability choice of drugs on pneumonia, according to the CDC: beta-lactams (78.6%), macrolides (100%), quinolones (50%). Suitability of doses and frequency of the PCP: cotrimoxazole, underdose (100%).

Abstract

Gene therapy involves the introduction of DNA or ribonucleic acid (RNA) into the target cell either to express or suppress the biosynthesis of proteins. The success of gene therapy is highly dependent on a suitable carrier system that can efficiently deliver genes in to specific desired cell with minimum cytotoxicity on target cells. Chitosan is interesting to be used as a gene carrier because it has a high positive charge and low toxicity to cells. Positive charge chitosan can form complexes with the plasmid. DNA complexes provide protection against enzymes degradation and promote internalization of plasmids that have been condensed. In this study the complex formation of chitosan-pEFneo-GFP and chitosan / TPP-pEFneo-GFP is by complex coaservation method. The results of complex analysis with a 0.8% agarose gel electrophoresis for 30 minutes 100Volt showed that the complex was stayed in the well and the plasmids did not migrate like plasmids which were dissolved in the water and buffer solvent. Stability evaluation in storage at room temperature for 14 days showed that the complex with chitosan concentration 0.02%-0.04% were the most stable, so that transfection analysis performed for the complex with chitosan concentration of 0.02%-0.04%. Transfection results showed that chitosan is able to protect plasmid and promote the internalization of plasmid into the nucleus and then expressed as a green luminescence. From these results chitosan potentially be developed as a carrier system in gene delivery.

Abstract

Cleft palate is a congenital deformity that causes a multitude of problems and represents a special challenge to the medical community. Speech production, feeding, maxillofacial growth, and dentition are just a few important developmental stages that may be affected. Beside that, these children and their families often experience serious psychological problems. A broad spectrum of variations in clinical presentation exists. Several subtypes of cleft palate can be diagnosed based on severity. The correct diagnosis of a cleft anomaly is fundamental for treatment, for further genetic and etiopathological studies, and for preventive measures correctly targeting the category of preventable orofacial clefts. Abstract

One of dental treatments on gangrene radix is an extraction. However, tooth extraction on the chronic gangrene radix needs particular procedure since this condition frequently emerges periapical disorder like dental granuloma or radicular cyst. Both dental granuloma and radicular cyst may rest and grow to be jaw cyst leading to the damage of jawbone although the causing tooth has already been removed. The procedure of tooth extraction with chronic gangrene radix covers extraction accompanied with enucleation and curettage of periodontal tissue using both close method and open method

Abstract

Diabetes mellitus (DM) is a metabolic disease with characteristics of chronic hyperglycemia and glucosuria. This occurs because of abnormalities of insulin secretion, insulin’s function or both. DM almost always followed by the increasing possibility of chronic complications. One complication encountered is hypercholesterolemia. Therefore we need a comprehensive DM therapy. Bitter melon fruit extracts (Momordica charantia lynn) shown to control the metabolism of glucose (hypoglycemic), This is because the extract contains pinsulin working a case of the hormone insulin. In addition there are several opinions and the results of studies suggest that the extract can lower blood cholesterol levels (hypolipidemic). This research carried out aiming to prove the presence or absence of decrease in blood cholesterol levels in diabetic male wistar rats (injected with alloxan) after given a bitter melon fruit extract. The samples in this study were divided into three groups: group I, group II and group III. All samples were injected alloxan to make the mice became diabetic. Blood cholesterol levels after administration of alloxan and three days later were measured and compared. On 3rd day until 6th day Group I was given 1% CMC Na as controlnegative, group II were given extracts of bitter melon fruit and the third group is given glibenclamide as a positive control. On ay-7 cholesterol levels were measured again. The results of data analysis showed that no significant differences in all outcome measures from all groups (Kruskall Wallis> 0.05). From this it was found that 62 hours after injection of alloxan did not have elevated levels of cholesterol and no significant decrease in cholesterol in male rats fed extracts of the fruit of bitter melon 700mg/200gr BB in 4 days.
