Abstract

Gene therapy involves the introduction of DNA or ribonucleic acid (RNA) into the target cell either to express or suppress the biosynthesis of proteins. The success of gene therapy is highly dependent on a suitable carrier system that can efficiently deliver genes in to specific desired cell with minimum cytotoxicity on target cells. Chitosan is interesting to be used as a gene carrier because it has a high positive charge and low toxicity to cells. Positive charge chitosan can form complexes with the plasmid. DNA complexes provide protection against enzymes degradation and promote internalization of plasmids that have been condensed. In this study the complex formation of chitosan-pEFneo-GFP and chitosan / TPP-pEFneo-GFP is by complex coaservation method. The results of complex analysis with a 0.8% agarose gel electrophoresis for 30 minutes 100Volt showed that the complex was stayed in the well and the plasmids did not migrate like plasmids which were dissolved in the water and buffer solvent. Stability evaluation in storage at room temperature for 14 days showed that the complex with chitosan concentration 0.02%-0.04% were the most stable, so that transfection analysis performed for the complex with chitosan concentration of 0.02%-0.04%. Transfection results showed that chitosan is able to protect plasmid and promote the internalization of plasmid into the nucleus and then expressed as a green luminescence. From these results chitosan potentially be developed as a carrier system in gene delivery.

Abstract

Rheumatoid arthritis  (RA) adalah  penyakit  autoimun  yang menyebabkan  peradangan  kronis  pada sendi. Penatalaksanaan RA harus agresif dan sedini mungkin sehingga mampu meningkatkan hasil jangka pendek maupun panjang penderita. Rheumatoid arthritis akibat reaksi autoimun dalam jaringan sinovial yang melibatkan proses fagositosis. Tujuan dari pengobatan rheumatoid arthritis tidak hanya mengontrol gejala penyakit, tetapi juga penekanan aktivitas penyakit untuk mencegah kerusakan permanen. Penderita RA memulai pengobatan mereka dengan DMARDs (Disease Modifying Anti-Rheumatic Drugs) seperti metotreksat, sulfasalazin dan leflunomid. Alternatif pengobatan yang dapat dijadikan salah satu pilihan dalam penanganan RA yaitu senyawa kurkumin dan analognya. Sistem nanopartikel mampu meningkatan efektifitas dalam pengobatan terutama keadaan RA. Kata kunci : rheumatoid arthritis, Disease Modifying Anti-Rheumatic Drugs, kurkumin, nanopartikel.

Abstract

Losartan merupakan obat antihipertensi poten dengan bioavailabilitas rendah dan waktu paruh eliminasi cepat. Penelitian ini bertujuan untuk mengetahui komposisi formula optimum, karakteristik dan profil transpor  in vitro  patch transdermal kalium losartan. Rancangan formula berdasarkan metode simplex lattice design menggunakan software Design Expert. Evaluasi  karakteristik meliputi  ketebalan, bobot, moisture uptake, loss on drying,  folding enduranc,  dan drug content. Uji transport in vitro menggunakan  sel difusi vertikal, penetapan kadar transpor losartan menggunakan instumen HPLC dan analisis data menggunakan software WinSAAM. Komposisi formula optimum patch transdermal  losartan adalah 44,4% propilen glikol, 29,3% asam oleat,  dan 26,3% isopropil alkohol,  dengan karakteristik  tebal 0,6 mm, bobot 82,2 mg, loss on drying12,8%, moisture uptake 6,4%, folding endurance 300 lipatan dan drug content 98,9%. Profil transpor in vitro losartan menghasilkan model lima kompartemen dengan kinetika orde pertama.

Abstract

Brown marine algae are the prominent source of marine natural products that have bioactive metabolites. Sargassum turbinarioides and Sargassum ilicifolium were dominated in Indonesia as brown marine algae that well known as a source of fucoidan. The samples were collected from Gunungkidul, Yogyakarta, Indonesia. In this study, we investigated and identified the yield of aqueous crude and purified extracts using different extraction temperatures (60°C, 70°C, 80°C, 90 °C). The highest yield of S. turbinarioides crude extract (7.36%) was obtained at temperatures 90 and 80 °C while the highest yield of S. ilicifolium was 3.49 % at 80 °C. The presence of sulfate polysaccharide in Sargassum turbinarioides is 3,78 % and Sargassum ilicifolium is 2,93 %. Each of the extract was screened using phytochemical detection, Thin Layer Chromatography (TLC) and Fourier transform infrared spectroscopy (FT-IR) analysis. The phytochemical detection indicated that Sargassum ilicifolium has bioactive metabolites such as carbohydrates, proteins and amino acids, terpenoid, phenolic compounds, and flavonoids. The FTIR spectrum of the S. turbinarioides and Sargassum ilicifolium extract refer to the presence of ester sulfate groups through showing  peaks at 1300 to 1200 cm-1 and 980 to 950 cm-1. The result indicated that Sargassum turbinarioides and Sargassum ilicifolium contain of sulfate polysaccharide were prospect a biological activities to use for the development of marine nutraceutical drugs especially as antioxidant.

Abstract

Furosemide is a diuretic having low solubility and low bioavailability thus the diuretic activity is not optimal. The purpose of this study was to determine the effect of furosemide SNEDDS formation to  the diuretic activity. SNEDDS was made with a mixture of 66% tween 80, 26% propilene glycol, 8% oleic acid and furosemide 40 mg/mL. A total of 20 test animals were divided into 4 groups, namely the negative control 1 (CMC 0,5% suspension), negativ control 2 (a mixture of tween 80, propilene glycol and oleic acid), furosemid suspension and SNEDDS furosemid. Tests on the diuretic activity is done by measuring the volume of urine released for 6 hours.Furosemide SNEDDS  could enhance the diuretic activity of furosemide.

Abstract

Abstract Nanoparticles are non-viral vectors with biodegradability and controlled release abilities that are widely used in gene transfer. Nanoparticles are said to be able to effectively combine and concentrate DNA and RNA in products to be delivered to various cells. Based on changes in microRNA expression in cancer, delivery using microRNA nanoparticles from outside the body is expected to be one of the therapeutic solutions. Irradiated chitosan is assumed to undergo cutting off the main polysaccharide group to produce a smaller molecular weight compared to nonirradiated chitosan. The desired irradiation process to reduce chitosan particle size so that it has better absorption efficiency into the cell needs to be further evaluated including looking at its effect on the final potential zeta. Zeta potential is important to ensure that the nanoparticles are positive enough to be able to enter the negatively charged cell membrane. Chitosan used was irradiated at a dose of 5kGy mixed with 1: 1 nucleic acid ratio. The results of zeta potential measurements showed the highest zeta potential values ​​of 15.27 mV and 13.63 mV obtained in the formula with chitosan-microRNA composition only. The measurement result of zeta potential of irradiated chitosan nanoparticles decreases the surface charge of the particles, this can reduce the potential of nanoparticles for the process of delivery into the cell. Abstract

Background: Vitamin D is an essential fat-soluble vitamin that is required daily by the body. Deficiency in vitamin D is experienced by most of the world's population. Vitamin D plays a role in maintaining the skin barrier by regulating keratinocyte proliferation and differentiation, promoting filaggrin formation, and intraepidermal lipids. Discussion: The vital role of vitamin D, non-compliance with supplement consumption, and poor nutritional status of adult patients and the elderly with deficiency and insufficiency of vitamin D encourage studies regarding vitamin D delivery via the transdermal route. The lipophilicity of vitamin D allows it to penetrate through the skin barrier. Conclusion: Research indicates that topical application of vitamin D3 is not only safe but also effective in increasing serum vitamin D levels. Thus, topical delivery is a promising alternative method for providing vitamin D supplementation or as a therapeutic option for specific conditions that leads to improved skin hydration and serum vitamin D levels.

Abstract

Indonesian aquaculture cultivation production of catfish has been growing faster than other fish. It is a potential source of fish oil and contains polyunsaturated fatty acids that have health benefits. The objective of this study was to perform physicochemical characterization and fatty acid profile of catfish oil. This study used fish oil extracted from the head and the flesh of catfish using dry rendering with a mechanic press. The result showed that catfish flesh oil (CFO) and catfish head oil (CHO) were significantly different parameters (p<0.05) in terms of physicochemical characteristics including acid value, peroxide value, iodine value, and saponification value. The best characteristics are catfish flesh oil than catfish head oil for acid value, peroxide value, iodine value, and saponification value following the number is 2.17 mg KOH/g, 5.15 meqO2/kg, 76.13 I2/100 g, 175.86 mg KOH/g. The predominant fatty acids were oleic, palmitic, and linoleic acids. These catfish oils are found suitable for consumption for beneficial health effects.

Abstract

Fish is a product with high nutritional content, and the development of fish oil has made it easier for consumers to consume. High-quality fish oil can be a target for adulteration, such as with vegetable oils like palm oil. FTIR spectroscopy was developed for the authentication of bawal fish oil (BFO) from palm oil (PO). Bawal fish oil and palm oil were prepared in binary mixtures with concentrations from 0-100%, resulting in 21 mixtures. The oil was directly measured by attenuated total reflectance (ATR) spectral measurement in the mid-infrared region (4000-600 cm-1). The results were combined with linear discriminant analysis (LDA) and multivariate calibration, such as PLSR or PCR. The results showed that LDA could make clear discrimination between bawal fish oil, palm oil, and their mixtures without any misclassification observed. Multivariate calibration with PLSR using the first derivative spectrum provided the best model for the relationship between actual and predicted FTIR values. At this conditions, the R2 values for the calibration and validation models obtained were 0.9966 and 0.9969, with RMSEC and RMSEP values of 0.0249 and 0.0248, respectively. Therefore, FTIR spectroscopy combined with LDA and PLSR is an effective method for authenticating bawal fish oil from palm oil.
