Journal of Aquatropica Asia (JoAA) is an open access scientific periodical managed by the Department of Aquaculture, Bangka Belitung University. JoAA Journal involvement is carried out 2 (two) times a year, namely in July and December in the form of the main manuscript is an article that contains the results of research (research articles). Other manuscript contributions can be in the form of short articles (short communication), articles review, and also special issues. Articles received will be reviewed by reviewers managed by the Editor in Chief before the manuscript is accepted and approved for publication in JoAA. The Journal of Aquatropica Asia (JoAA) accepts articles written in Indonesian or English with regard to aquaculture studies and aquatic ecology in the broadest sense covering aspects of reproduction, nutrition and feed, genetics, physiology, morphology, health of aquatic organisms, water quality, plankton, conservation, and other aspects relevant to the field of aquaculture.

Abstract

Extremophilic fish require genetic characterization to determine the identity of their species, their kinship, and the potential for genetic variation due to extreme environmental adaptation. DNA barcoding using Fish-F2 and Fish-R2 primers at an annealing temperature of 52oC for 15 seconds showed qualitative failure results in most fish species. For this reason, optimizing the annealing temperature in the PCR process is necessary to obtain DNA bands that correlate with successful identification. Fish samples were obtained from Ruai Silip, Bangka Island, Membalong, Belitung Island, and Way Kanan, Lampung, Sumatra Island. The annealing temperature optimization was set at 50, 50.4, 51.1, 52.3, 53.7, 54.8, 55.5, and 56°C. Five species of extremophile fish were tested, namely Brevibora sp, Barbodes binotatus, Rasbora bankanensis, Anabas testudineus, and Aplocheilus panchax. Extremophile fish showed differences in the appearance of DNA bands in PCR with different annealing temperatures. Brevibora sp showed a visualization of DNA bands at 54.8°C, Barbodes binotatus at 53.7, 54.8, 55.5, and 56°C, Anabas testudineus at 50 and 50.4°C, and Aplocheilus panchax produced clearly visible band on annealing temperatures.

Abstract

The Selincah fish (Belontia hasselti) is a local Indonesian fish that is widely consumed and has the potential to become ornamental fish. This fish must be domesticated to suppress its exploitation from natural catches. The domestication process requires biological information including eating habits. For this reason, in this study, the identification of phytoplankton contained in the stomach contents of the agile fish was carried out. Fish samples were obtained from Rawa Metukul, South Bangka Regency. The types of phytoplankton in the stomach and intestines of lively fish are the classes Bacillariophyceae, Chlorophyceae, Cyanophyceae, Chlorophyceae, Chrysophyceae, Chrysophyceae. The most dominant species were found in the Chlorophyceae class with an Index of preponderance value of 42.42%, while the Chrystophyceae class was found at least 2.99%.

Abstract

Grouper production in Indonesia can be supported by hybridization technology which can produce grouper species with relatively faster growth and more resistance to disease. Cantang grouper is a type of grouper fish that was engineered by crossbreeding between female tiger grouper (Epinephelus fuscoguttatus) and male grouper Kertang (Epinephelus lanceolatus). The purpose of this study was to determine the stages and technology of nursery grouper Cantang which was carried out at BPBAP Situbondo. The method used in this research is the method of observation, participation, and interviews. The primary data needed in this study are data on infrastructure, data on the number and size of seeds, feed management, water quality management, and harvesting. The data is presented with a descriptive method by discussing the data that has been obtained compared to the theories that have developed at this time. The results of this study indicate that cantang grouper nursery activities start from tank preparation, water filling, seed selection and distribution, feed management, water quality, fish pest and disease control, grading, and harvesting of cantang grouper seeds. During the practice period, the seeds were spread from hatcheries with a stocking density of 3000 to 5000 individuals in one tank with a size of 4 x 2.5 x 1 m, or the equivalent of 300 to 500 individuals/m3. The distribution of seeds is carried out after the acclimatization process, namely the adaptation process before releasing the seeds. After the seeds are removed, the tub is given an antiseptic to prevent disease in the seeds. There are several factors that can affect the growth rate of seeds, namely water quality management and feeding. During the maintenance period, a flowing water system is used, so that the water used is continuously renewed. In the morning, the seeds are flushed and soaked using fresh water, with the aim of releasing mucus on the seeds so as to reduce the potential for disease outbreaks. Nursery grouper at BPBAP Situbondo can run optimally by paying attention to the stages of cultivation, especially in the feed management and water quality management.

Abstract

Blue swimming crab is a fishery product with high economic value in Indonesia. Betahwalang Village, Demak, is one of the crab fisheries locations in the province of Central Java. High selling prices and processing industry demand encourages fishing activities. One issue brought on by increased fishing activity is the smaller crab size. It will impact the crab fishing industry in Betahwalang. The majority of the fisherman in Betahwalang Village catch crabs using arad nets, simple gill nets, and traps. This study aims to evaluate the size of crab catch results on fishing gear used by fishermen and recommend proper fishing gear for catching crabs. This research was conducted in April 2019. The catch size of crab was the primary data collected for this study, also with data on fishing grounds, methods, and characteristics of fishing gear. Data were collected by observation, interviews, and measurements of fishing gear and the carapace width of the crab. The results of data collection and processing were analyzed descriptively. The results showed that the highest proportion of legal size crabs (carapace width ≥10 cm) was caught in traps, while the proportion of unlegal size crabs caught was mostly in bottom gillnets and mini trawl.

Abstract

Pearl Catfish (Clarias gariepinus) is a type of freshwater fish that is widely cultivated by many people because of its economic value. Pearl catfish is a new strain of African catfish Clarias gariepinus Burchell, 1822 superior breeding results of the Sukamandi Fish Breeding Research Agency (BPPI) which has been declared passed in the Type / Variety Release Assessment on October 27, 2014, under the name MUTIARA (Peerless High Quality) catfish. This study aims to determine the effect of the addition of duckweed on feed on the growth and survival of pearl catfish fry. The test fish is a pearl catfish measuring 7-10 cm long and weighing 6 gr that is kept in a sized aquarium (120 cm x 50 cm x 40 cm), as many as 9 aquariums, three aquariums as a control, three aquariums for treatment A, three aquariums for treatment B, with each treatment A(5% dry duckweed), treatment B (10% dry duckweed) and treatment K (control) without treatment. . The results showed that dry dukweed can be used as an addition to fish feed, of the three feeds that provide the best growth, namely in the B treatment (10% dry duckweed) has a daily growth rate (GR) of 0.41 g, Specific Growth Rate 2,57%, absolute weight 19 g, absolute length is 11.31 cm with a survival of 90%.

Abstract

Artikel ini bertujuan untuk menjelaskan dan mengambarkan filogenetik genus Channa di Indonesia menggunakan gen COI (Chytocrome C Oxydase Sub unit I). Sebanyak sembilan (9) spesies Channa yang tersebar di Indonesia dan terdata di NCBI Genbank terdiri dari Channa limbata,Channa bankanensis,Channa lucius,Channa marulioides,Channa melasoma, Channa micropeltes,Channa pleurophthalma, Channa gachua dan Channa striata telah dianalisis filogenetik dan jarak genetik antar spesiesnya. Jarak genetik terdekat yaitu pada Channa pleurophthalma dengan Channa striata yaitu 0.000. dengan komposisi nukleutida pada keduanya adalah timin sebanyak 26,98%, sitosin sebanyak 29,80%, adenin sebanyak 25,16% dan guanin sebanyak 18,04%.

Abstract

Barbodes sellifer merupakan merupakan ikan air tawar dari keluarga cyprinid. Spesies ini pertama kali dideskripsikan pada tahun 2021 oleh Kottelat dan Lim. Saat ini spesies Barbodes sellifer dapat ditemukan di Singapore dan malaysia. Di Indonesia sendiri barbodes sellifer ditemukan di Sumatera, Kepulauan Riau hingga pulau Natuna. Kehadiran Barbodes sellifer di Kepulauan Bangka merupakan catatan baru. Meskipun spesies ini dapat ditemukan di Pulau Bangka, namun jumlahnya tidak banyak, sehingga perlu dilakukan upaya-upaya domestifikasi untuk mempertahankan populasi. Agar kegiatan domestifikasi dapat berjalan dengan baik, perlu melakukan beberapa penelitian mendasar yang salah satunya adalah studi morfometrik dan meristik untuk memastikan validasi spesies dan sebagai dasar untuk penelitian-penelitian selanjutnya. Dalam penelitian ini kami melakukan analisis karakter morfometrik dan meristik spesies Barbodes sellifer yang ditemukan di Pulau Bangka dan sebagai langkah awal dalam proses domestikasi. Hasil studi karakter morfometrik menujukan panjang total (TL) 7,4 cm, panjang standar (SL) 6,0 cm, tinggi kepala (HD) 1,0 cm, diameter mata (ED) 0,5 cm, tinggi badan (BD) 2,0 cm, panjang sirip ekor bagian atas (LUCL) 1,2 cm, panjang sirip ekor bagian tengah (LMCL) 0,8 cm, panjang sirip ekor bagian bawah (LCLL) 1,6 cm. Sedangkan meristik spesies Barbodes sellifer yaitu jari-jari sirip punggung memiliki rumus D.I-II.7, jari-jari sirip anal memiliki rumus A.6, Jari-jari sirip dada memiliki rumus P.I-II.10, Jari-jari sirip perut memiliki rumus V.I.7. Ikan barbodes sellifer memiliki tipe mulut terminal,bentuk tubuh compressed, tipe sisik ctenoid dan bentuk ekor forked. Berdasarkan karakter morfologi dan ciri khusus jenis Barbodes sellifer yang ditemukan dialiran sungai Jada Bahrin juga termasuk spesies Barbodes sellifer yang ditemukan di Singapura.

Abstract

Binahong leaves (Anredera cordifolia) are a species from the Basellaceae family that has many benefits as an antibacterial, antitumor, wound healing, intestinal inflammation, and others. This is due to the presence of active ingredient compounds owned by Binahong leaves. This study aimed to screen for the active ingredient compounds present in Binahong leaf extract using water as a solvent. The method used in this study was fresh Binahong leaves mashed and then tested for active ingredient compounds including flavonoids, saponins, phenols, tannins, terpenoids, and steroids. The results obtained from this study were saponins and steroid compounds, while there were no flavonoids, terpenoids, tannins, or phenols.

Abstract

Sea urchins a biota that is often associated with seagrasses were often found in seagrass beds. The objectives of this research were to discover the condition of sea urchins and seagrass, such as diversity, uniformity, dominance in sea urchins, density, and cover on seagrass, and related to variables in this research. This research was conducted on Derawan Island. The research method that was used is a 5m x 5m (sea urchin) quadrate and 0.5m x 0.5m (seagrass) placed intentionally on 2 stations with a total of 30 quadrats. The results found 3 species of sea urchins and 4 species of seagrass. At station 1 abundance of sea urchins is 2.94 individual/m2 whereas at station 2 is 0,28 individual/m2. At station 1 seagrass density 117,33 shoot/m2 whereas station 2 is 120,13 shoot/m2. Seagrass cover at station 1 is 58,75% and at station 2 is 60,28%. The dominant sea urchin species is Diadema setosum with the following diversity values H'=0,36, E=0,3, and C=0.81. There was no significant difference between variables at each station except temperature (0.020<0.05) and DO (0.027<0.05). Variables that were correlated with sea urchins in the seagrass ecosystem are nitrate, salinity, phosphate, temperature, and pH. The association with seagrasses in Derawan Island is positive.

Abstract

Green mussels (Perna viridis) in Banyu Urip Village, Ujung Pangkah Gresik District, have great potential to be developed due to the abundant availability of seeds in nature, easy cultivation methods, namely using floating net cages and step nets, and high market demand. One of the issues with green mussels is the high content of the heavy metal Lead (Pb) in their flesh, which is caused by heavy metal accumulation in the waters. The goal of this study was to isolate, characterize, and test the ability of heavy metal Lead-Pb bacteria to biosorb heavy metal Pb in post-harvest green mussels from sediments and green mussels in the Banyu urip Ujung Pangkah Gresik Sea. The stages of the research included sampling, isolation, macroscopic and microscopic characterization, in vitro testing of Pb heavy metal absorbent. The results showed 6 bacterial isolates, namely two from green mussels coded K1 and K2 and 4 bacterial isolates from marine sediments coded S1, S2, S3 and S4. The six bacterial isolates obtained macroscopic and microscopic characteristics and have been shown in vitro to be able to grow in Nutrient agar enriched 0.8 mg/L Pb(NO3)2.
