Abstract

Free radicals are one of the cause of various diseases. The use of synthetic antioxidant compounds could prevent the effect of the free radicals, however may cause adverse effects on the human body such as impaired liver, lung, intestinal and poisoning. Therefore antioxidant from natural resources needs to be developed. The purpose of this research was to determine the antioxidant activity and IC50 value of the ethanol extract of ganitri (Elaeocarpus ganitrus Roxbs.) leaves. Ganitri leaf ethanol extract activity test was carried out using DPPH method with vitamin C as a standard. Antioxidant activity was determined as a decreas in the absorbance of DPPH at 517 nm wavelength after an addition of the extract with the concentrations of 20, 40, 80, and 100 ppm. The antioxidant acitivity measurement of the ganitri leaf extract showed that the linier regression equation obtained was y = 0.3669x + 29.546, r = 0.4573 while the IC50 value was 54,12 ppm. Based on the result, it is concluded that the ethanol extract of ganitri (Elaeocarpus ganitrus Roxb.) leaf showed was categorized as strong antioxidant.

Abstract

Ischemic stroke is a disruption of blood supply to the brain caused by the blockage of blood vessels. Hypertension is a comorbid disease in ischemic stroke. Appropriate treatment can affect the success of therapy and the achievement of the desired blood pressure. This study aims to evaluate the rationale for using antihypertensive drugs to achieve clinical outcomes in the form of blood pressure. This study was designed in an observational analytic manner with a cross-sectional approach, retrospective in nature with a purposive sampling method in ischemic stroke patients who received antihypertensive prescriptions in RSUD Dr. Soegiri Lamongan. The sample used was 100 patients. The results showed that the most widely used antihypertensive drugs were CCB with a percentage of 41.8%, ARB with a percentage of 27.6%. The rationale for prescribing was obtained: 100% correct indication, 100% correct patient, 91% correct drug, and 100% correct dose. The results of the Chi-Square test have obtained an expected value of 5, so that it is followed by the Fisher's Test, which obtained a p-value of 0.021 (p 0.05). The conclusion of this study is that patients who received a rational antihypertensive prescription were 91%, for patients who achieved clinical outcomes as much as 86% and there was a relationship between the rationality of prescribing antihypertensive drugs with clinical outcomes in the form of achieving target blood pressure in ischemic stroke patients at RSUD Dr. Soegiri Lamongan.

Abstract

Mangrove plants as traditional medicine have long been used by the society for the therapy of anticancer diseases. So far, the potency of mangrove plants as anticancer has not been studied intensively. The methanol extract of R. mucronata stem bark has cytotoxic activity on myeloma cells. This study aims to determine the isolates of the ethyl acetate fraction of the methanol extract of the stem bark of R. mucronata and R. apiculata, and to examine the potency of cytotoxic activity against MCF-7 and T47D cells. The powder of the stem bark of R. mucronata and R. apiculata were extracted with methanol by maceration, and then the methanol extract was fractionated consecutively using n-hexane, ethyl acetate, and methanol. Ethyl acetate fraction was then purified by column chromatography with a diameter of 2 cm and a height of 30 cm with a static phase in the form of silica gel 60 (0.063-0.2 mm) Merck 7734 and eluent n-hexane:ethyl acetate (6: 4, 5: 5, 6: 4, 7: 3, 8: 2, 9: 1) and n-hexane: methanol (9,5: 0,5) 10 ml each. The eluates that produce the same number and pattern of stains were combined into one isolate. Then the eluate was evaporated at room temperature. Then the purification results of the extract R. mucronata were isolated using preparative TLC with 0.25 mm GF254 silica gel as static phase and n-hexane mobile phase: ethyl acetate (6: 4), while the apiculata extract with n-hexane mobile phase: ethyl acetate (5: 5). The results of the stain separation of the compound were scraped off and separated from the static phase using a solvent. The purity of isolates compound was examined using TLC in the static phase of silica gel GF254 0.25 mm. The qualitative test results of the ethyl acetate fraction of R. apiculata stem bark with an NMR spectrophotometer showed a composition of cis-3- (3,5-dihydroxyphenyl) acrylic acid, while the ethyl acetate fraction of R. mucronata stem bark contained n-hexan-3-ol compounds. The cytotoxicity test of ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol extract was carried out on MCF-7 and T47D cells using the MTT method. The results proved that the ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol fraction from the ethanol extract of R. mucronata and R. apiculata stem bark, based on cytotoxic activity, were inactive against MCF-7 and T47D cells.

Abstract

An important treatment for type 2 diabetes is controlling blood glucose levels. α-amylase enzyme in the body plays a role in breaking down carbohydrates into starch. Control of the amylase enzyme is needed in diabetes cases Kremah plant (Alternanthera sessillis) is a plant whose leaves can be used to lower blood glucose levels because it contains secondary metabolites, one of which is flavonoids which can inhibit the α-amylase enzyme. The purpose of this study was to determine the activity and inhibition value of kremah leaves against the α-amylase enzyme. The method used in this research is the enzymatic reaction by measuring the intensity of the color using UV-Vis spectrophotometry. The IC50 value obtained by α-amylase enzyme inhibition was 101.89±7,21 μg / ml while the IC50 acarbose value was 127.17±4,42 μg / ml. These results indicate that kremah leaf extract has activity (hyperglycemia) by inhibiting complex carbohydrate hydrolyzing enzymes such as the α-inhibition enzyme α-Amylase which is good compared to acarbose because the IC50 value of the extract is smaller, so that the leaves of kremah has inhibiting of the α-amylase enzyme.

Abstract

The Covid-19 infection has spread to almost all regions of Indonesia. The government has issued a health protocol policy for the public in carrying out their activities to minimize the transmission of covid-19. This study aimed to obtain an overview of public compliance behavior in implementing health protocols as an effort to prevent Covid-19. The questionnaire survey was conducted at one of the pharmacies in the Banjarsari area of Surakarta for the period 14-30 September 2020. The research subjects were determined by purposive sampling method. The questionnaire contains 16 statement items covering the domain of social distancing, use of masks, maintaining hygiene and hand washing, and cough etiquette. Respondent behavior was classified into 2 categories, namely compliance and noncompliance. Respondent characteristic data (gender, age, and occupation) were tested for their relationship with compliance behavior. This study involved 54 respondents who were dominated by women (68.52%), age range 36- 45 years (37.18%), and work as private employees (37.04%). A study on 54 respondents showed that 51.85% of respondents had complied with the implementation of health protocols. Community non-compliance was still high in the domain of maintaining hygiene and cough etiquette. Gender, age, and occupation were not significantly related to respondent behavior.

Abstract

Hyperuricemic is a condition in which uric acid level in blood exceeds its normal level. Empirically, African leaves (Vernonia amygdalina Delile.) were used to treat diabetic, hypercholesterolemic, fever, hypertension and hyperuricemic by drinking boiled water of African leaves. This in vivo study was performed to determine antihyperuricemic activity of ethanolic extract of African leaves on male mice using the POCT (Point of Care Testing) method. Suspension combination of melinjo and chicken liver juice were used as hyperuricemic inductor. It is given orally for consecutive 9 days and the uric acid level were tested on day 1, 3, 6, and 9. The results showed that the ethanolic extract of African leaves (Vernonia amygdalina Delile.) at dosage 100, 200, and 400 mg/KgBW had a significantly different antihyperuricemic activity against negative control.

Abstract

Avocado plants are widely used as traditional medicine in Indonesia. This research aimed to determine the cytotoxic and antiproliferation activity of avocado seed extract and fractions on the T47D breast cancer cell line. Extraction was done by maceration method using 96% ethanol, then fractionation was done by liquid-liquid partition using n-hexane, chloroform, and ethyl acetate. Cytotoxic and antiproliferation tests were done by the MTT method. Apoptosis activity was investigated by the double staining method, using acridine orange-propidium iodide as a staining reagent. Results showed that the n-hexane fraction of avocado seed had a cytotoxic activity with IC50 27.9 µg/mL. N-hexane fraction of avocado seed inhibited proliferation and induced apoptosis of T47D cell line.

Abstract

Hyperuricemia is a disease caused by an increase in uric acid levels in the blood. Currently, various treatments are developed by utilizing natural ingredients as an anti-hyperuricemia. Sungkai (Peronema canescens Jack) is one of the plants that contain a natural compound that can use as an anti-hyperuricemia treatment is the leaves of sungkai (Peronema canescens Jack.). The aim of this study to determine the anti-hyperuricemia activity of the ethanol extract of Sungkai leaves. The test animals used in this study were male white mice Wistar strain. This study used a completely randomized design (CRD) with 5 groups consisting of negative control (Na CMC 0.5%) positive control (allopurinol 10 mg / kg BW), treatment 1 (extract 125 mg / kg BW), treatment 2 (extract 250 mg / kg BW) and treatment 3 (extract 500 mg / kg BW). The parameters seen were uric acid levels measured using the POCT (Point of Care Test) method and analyzed using One Way Anova statistical analysis and Duncan's continued test. Our results showed that the treatment group with a dose of 125-500 mg / Kg BW reduce uric acid levels in mice. The best dose was a dose of 500 mg / Kg BW in reducing uric acid levels in mice with percent decline of 38.66%.

Abstract

Gambas or Oyong fruit has various benefits, namely it can overcome enlarged lymph nodes, diuretics, laxatives, antioxidants, hepatoprotective, inhibiting the growth of fungi and bacteria. The purpose of this study was to determine the levels of total flavonoid compounds in gambas (Luffa acutangula (L.) Roxb). Extraction methode using soxhletation with 96% ethanol as solvent. The extract obtained was used for qualitative and quantitative analysis. Phytochemical analysis of the flavonoid content in the sample using the Shinoda test, 10% NaOH solution, and concentrated H2SO4 solution, obtained positive results containing flavonoids. Quantitative analysis using UV-Vis Spectrophotometry with the maximum wavelength obtained is 429.5 nm and operating time is 30 minutes. The average total flavonoid content obtained was 9.897±0.11 mg/gram extract with %KV 1.14%.
