Abstract

Piroxicam solubility in water is very small this cause problem in the process of absorption so that it?s required an optimal formula to establish the physical properties of tablet quality for piroxicam tablets. The purpose of this research was creating formulas with the optimal combination of excipients lactose, Avicel pH 101, and piroxicam amprotab to obtain tablets with good physical properties and disolution. Seven piroxicam tablet formulas were made with a combination of lactose, avicel PH-101, and Amprotab on 2 batches. Method for making tablets piroxicam was established by direct compression. Tests included were the physical properties of piroxicam tablets tablets weight uniformity, uniformity of size, hardness, brittleness, disintegration test, as well as dissolution test, and content uniformity. Based on the simplex lattice design method, lactose is a dominant factor to improved the uniformity of tablet?s concentartions. Interaction of two components avicel PH-101 and amprotab is a dominant factor to reduced vulnerability. Interaction of three components lactose, avicel PH-101 and amprotab is a dominant factor to lowered the value coefisien variation of weight uniformity, increased hardness, decreased the disintegration, and improved dissolution of tablets. Selection of the optimum formula was determined by the method of simplex lattice design through diagrams superimposed contour plots and contour plots for obtaining the optimal proportions of each ingredient as follows: lactose (85,154?100%), avicel PH-101 (0?12,437%), and amprotab (0?5,425%).

Abstract

The existence of pharmacies is needed by the community, considering the higher number of morbidity which has an impact on the need for drugs. These phenomena make the pharmacists have to continue to exist and the community continues to visit and receive pharmaceutical services. The aim of this study was to describe the views and expectations of consumers towards the presence of Pharmacists in pharmacy services in Semarang. The descriptive survey research involved respondents from PKK with age of 40 - 60 years, who could read and write, consumers had visited the Pharmacy to redeem prescriptions or bought medicine and without distinguishing the respondent from the first time or the customer often visited the Pharmacy. The study was conducted in the city of Semarang with a total of 55 respondents. The results showed that almost all respondents (94.5%) visited the Pharmacy more than three times a month, with the biggest goal being to redeem recipes for children or families by 92.7%. Consumers’ views on the presence of mandatory pharmacists in pharmacies are 94.5% and pharmacists' expectation of providing pharmaceutical services 67.2%. Consumer expectations for the presence of pharmacists in pharmacies are 92.7%, expectation of meeting Pharmacists at 70.9%. Consumer experience about pharmaceutical services in the form of information on drug use amounts to (80%), side effects (65.4%) as well as how to store and dispose of drugs (52.7%.). Consumers expect the presence and can meet the Pharmacist at the Pharmacy so that they will get direct services in the form of consultation and counseling about drugs by the Pharmacist.

Abstract

Noni fruit (Morinda citrifolia L) is one of the natural ingredients commonly used as traditional medicine to overcome various diseases, such as cancer. Noni fruit contains anthraquinone and coumarin which have anticancer effects. This study aims to determine the cytotoxic effects of n-hexane, ethyl acetate and ethanol extract of noni fruit and to determine the presence of anthraquinone and coumarin in the extract. Extraction was carried out by maceration method using n-hexane, followed by ethyl acetate and ethanol. The cytotoxic activity on T47D cells was carried out by the MTT method. The presence of anthraquinone and coumarin was identified using thin layer chromatography (TLC). Results showed that n-hexane extract had moderate cytotoxic effect on T47D cells with IC50 value of 582.13 ± 61.64 µg/mL, while the ethanol and ethyl acetate extract did not have cytotoxic effect on T47D cells. TLC results indicated that n-hexane extract of noni fruit contains anthraquinone and coumarin.

Abstract

A study on isolation and antioxidant activity of secondary metabolit of methanol extract of fruit of Senduduk Bulu (Clidemia hirta (L.) D. Don) has been carried out. Senduduk bulu is used for folk medicine to many diseases. Flavonoid and phenolic has been reported to its activity as antioxidant. The aim of this research was to find antioxidant compound and antioxidant activity of this fruit. Isolation works were conducted using Vacuum Liquid Chromatography and Column Chromatography method. Antioxidant activity was determined using DPPH free radical scavenging methods. The isolation works afforded 10 mg of pure compound CHP1 which was obtained as white crystal, with its melting point of 165–167ºC, soluble in chloroform and slightly soluble in methanol. This compound gave positive reaction with Liebermann-Burchard (LB) reagent and concluded as terpenoid. Based on it UV spectrum, this compound showed a λ max of 316 nm. Its FT-IR spectrum showed appearence of some functional groups of OH, C-H aliphatic, C=O ester,C=C alkene, CH bending and C-O. The 1H-NMR spectrum showed the alkene protons and methyl protons which is a characteristic of terpenoid compounds. The pure compound (CHP1) provided a weak antioxidant activity with the IC50  of 327,01µg/mL.

Abstract

Previous research has shown that some compounds in leaves and seeds of sugar apple have a cytotoxic activity. The aim of this research was to determine the cytotoxicity of semipolar fraction of ethanolic extract from sugar apple stem bark (Annona squamosa L.) on T47D cancer cells. The semipolar fraction of ethanolic extract from sugar apple stem bark was collected by fractionation using Vacuum Liquid Chromatography (VLC) with hexane:ethyl acetic (9:1, 8:2, 7:3, and  6:4)  as  mobile  phase.  Cytotoxicity from the fractions of five different concentration namely; 25, 50, 100, 150, and 250, µg/mL was measured by MTT assay. The potency of the cytotoxicity was defined by the ability of the fraction to inhibit the growth of T47D cells indicated by the value of IC50. Qualitative analysis of contained compounds in the fraction was done by Thin Layer Chromatography (TLC) method using silica gel F 254 as a stationary phase and hexane:ethyl acetic (7:3) as a mobile phase. UV 254 and 366 nm lamp also Dragendorff, citroboric, and FeCl3 spray reagents were used to visualize the spots of the secondary metabolites. The result proved that the semipolar fraction of ethanolic extract from sugar apple stem bark showed potential cytotoxicity on T47D cancer cells with IC50value of 70,77 µg/mL. Qualitative analysis showed that the fraction contained flavonoids and alkaloids which is presumably responsible for its cytotoxic activity.

Abstract

Diabetes mellitus is one of the endocrine metabolic disorders that has caused morbidity and mortality worldwide. Α-glucosidase inhibitor which plays an important role in carbohydrate metabolism is needed to avoid postprandial hyperglycemia. A bioactive peptide derived from soy protein was chosen as an alternative treatment for diabetes because of its therapeutic potential. Several bioactive peptides have been shown to inhibit the α-glucosidase enzyme, such as the bioactive peptide LLPLPVLK, SWLRL, and WLRL. This study aims to identify and evaluate molecular interactions that occur between bioactive peptide molecules and α-glucosidase enzyme macromolecules using protein-peptide docking methods through in silico. Bioactive peptide sequencing was first modeled using the PEP-FOLD software. The best conformation was chosen for an interaction study of the α-glucosidase enzyme macromolecule using HPEPDock software. Further exploration was carried out on the molecular interactions formed using BIOVIA Discovery Studio 2020 software. Based on the results of molecular docking, the WLRL bioactive peptide has the best affinity against the α-glucosidase enzyme, with a binding free energy value of −748.12 kJ/mol. Therefore, the bioactive peptide is predicted to be a suitable candidate for the α-glucosidase enzyme inhibitor.

Abstract

Drug interactions are one of part of Drug Related Problem (DRP) which significantly affect the development patient health. This study aims to determine the characteristics of kidney failure patients (age and genitals), an overview of the number of potential drug interactions, an overview of the types and amounts of other drugs interacting, and the severity of drug interactions in kidney failure patients in the Outpatient of “X” Hospital Cirebon City. This study involves 126 patient prescriptions. This type of research is a non-experimental research with descriptive analysis method, and using data collection methods with prospective random sampling technique in the Outpatient of RS “X” Cirebon. The results of the research from 126 samples obtained showed that the characteristics of Kidney Failure patients were predominantly male as many as 64 people (50.79%), the most age was in the age range 61-70 years as many as 38 people (30.16%). drug interactions 78 sheets of prescriptions (61.9%), the most severity of drug interactions was the strict / significant monitoring of 128 events (64.65%), the average of drug interactions per prescription sheet was 3 interactions, the type and number of the most interacting drug was furosemide + folic acid with 32 potential interactions (41.02%).

Abstract

Diabetes mellitus is a chronic metabolic with high prevalence and it can increase the risk of the complications, if it not handled properly. Medication adherence is one of the determining factor to achive therapeutic outcome. This research aims to determine the affecting factors of adherence in  diabetes mellitus type 2 outpatient at RSUD Sukoharjo in 2020. This research type is  cross sectional with a purposive sampling technique. A total of 85 patients diabetes mellitus type 2 outpatient at RSUD Sukoharjo with inclusion criteria received OAD therapy (Oral Antidiabetic Drugs) or a combination of OAD with insulin for at least 1 month, recruited in this study. The data was collected from medical records and questionnaires. Adherence was measured by using the MARS 5 questionnaire (Medication Adherence Report Scale 5). Factors contributing to medication adherence was analyzed by using chi-square test. The results showed that 37 respondents (43.5%) were obedient. The significant factors of medication adherence are monthly income (p-value = 0.018 and OR = 2.887), Medication (p - value = 0.007 and OR = 3,551), the frequency of treatment (p-value = 0.006 and OR = 3,529) and blood glucose level (p-value = 0.006 and OR = 3,529).

Abstract

Diabetes mellitus is a chronic disease that requires treatment for a long period of time. Adherence to consuming drugs is one of the most important things in achieving treatment goals. This study aims to determine the level of adherence and quality of life and determine the relationship to the quality of life of patients with type 2 DM in hospitals in Central Java. This study uses analytic design by asking cross-sectional. Samples were taken at two hospitals in Central Java. Sampling was purposive sampling with the criteria of type 2 diabetes mellitus patients who are undergoing outpatient care in the hospital, have undergone therapy for at least 3 months, and can communicate well. As a measurement used the Medication Adherence Rating Scale (MARS) questionnaire while the quality of life measure used the Short Form 36 (SF-36) questionnaire. Score data received and quality of life that has been obtained was processed with the SPSS (Product Statistics and Service Solutions) chi-square test. Research results in 200 respondents found 9 people (4.5%) had low adherence, 46 people (23%) were moderately related and 145 people (72.5%) have high adherence. While the quality of life found 41 people (20.5%) had a poor quality of life and 159 people (79.5%) had a good quality of life. The lowest aspect of quality of life is physical limitations and the highest aspect of quality of life is mental health. From the results of the analysis on the chi square test, the p value was obtained: 0.00. This shows that there is a significant relationship between adherence and quality of life in patients with Type 2 diabetes mellitus at the hospitals and a Prevalence Ratio value of 23.47.
