Abstract

Belimbing wuluh (Averrhoa bilimbi Linn.) has antihypercholesterolemic activity. This study aims to determine antihypercholesterolemic potential, optimal dosage and effect of water extracts of Belimbing wuluh (Averrhoa bilimbi Linn) on the index of rat organs. Test animals used were male Wistar rats, divided into 4 groups: normal control, negative control, positive conrol (simvastatin 0.18 mg/kg), and the aqueous extracts of fruit Belimbing wuluh dose of 63 mg/kg. The parameter which was measured in this study were total cholesterol levels which obtained using CHOD-PAP method, the weight of rats for 50 days, and organ indexes. Those parameters were further tested with One Way ANOVA. In parameter decrease blood cholesterol levels of rats between groups positive and group aqueous extract of the fruit starfruit  dose of 63 mg/kg did not experience a statistically significant difference. Meanwhile, the group aqueous extract of the fruit starfruit dose 63 mg/kg and a negative control experiencing significant difference (p<0.05). Conclusion, aqueous extract of the fruit starfruit dose of 63 mg/kg had the effect antihypercholesterolemic seen from the decrease in total cholesterol levels from day 30 and day 50, as well as statistically significant different from the negative control, and can affect the index spleen and pancreas but does not affect the index of the kidneys, liver, heart, and lungs.

Abstract

The role of TGF-β1 is known as the main immunosuppresor associated with tumor, but on the other opinion, it is associated with proliferation and tumor invasion. The increase and decrease of the secretion of TGF-β is to regulate the proliferation, differentiation, and death of various cell types. Now we all know the extract of Jatropha curcas L seed serves as antitumor. Allegedly, it can regulate the expression of TGF-β1 in control of cell number. The purpose of this study is to determine the effects of Jatropha seeds to the regulation of gene expression of TGF-β1 as a tumor marker. The method is performed by giving a dose groups the extract of jatropha seed (0, 5, 25, 50, 250 mg/BB) in mice. Then measurement of mRNA expression (RT-PCR), the protein of TGF-β1 levels (ELISA), and qualitative observations of liver histology were done. The expression of TGF-β1 mRNA is significantly 4.39 to 7.34 times higher than (ANOVA, p 0.05) than the control. Histological observation of liver showed the extract of jatropha seed induces damage nucleus of hepatocytes cell and sinusoidal. The effects extract of jatropha seed increased the level of TGF-β1 mRNA but not followed by increasing protein of TGF-β1 levels, and it was stimulated necrosis and apoptosis of hepatocytes cell.

Abstract

This study aims to determine the bioactivity of L.domesticum as cosmetic ingredient. The extraction was prepared by maceration with ethanol 96 % and ethyl acetate to obtain ethanolic extract of fruit flesh (BLE), ethanolic extract of fruit peel (KLE), ethyl acetate extract of fruit flesh (KLE) and fruit peel (KLEA). Then, determination of compounds was conducted by thin layer chromatography (TLC) and analysed of total phenolic and flavonoid content with colorimetric method. The antioxidant capacity was measured using radical scavenging assay of 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching assay (BCB) and ferric reducing antioxidant power assay (FRAP). The inhibitory effect of skin degradation enzymes (anti-aging) was carried out using elastase and collagenase assay. The tyrosinase inhibitory effect was determined using mushroom tyrosinase. Based on TLC result, extract of L.domesticum containing phenolic, flavonoids, terpenoids and steroids compounds. The largest of total flavonoid content was obtained in KLE (4,76±0,26 %b/b EQ) and phenolic total content in BLEA (6,4±0,15 %b/b EG). Sequence of antioxidant activity measured using DPPH method was BLEA> KLE> BLE> KLEA; while using BCB method and FRAP method were respectively KLEA>BLEA> KLE> BLE; and BLEA> KLE> BLE> KLEA. KLE and KLEA extracts showed the most potent to remaining activity of the enzyme elastase and collagenase. In addition, BLE extract has the higest tyrosinase inhibition than others. Based on these results it can be concluded that the extracts of the fruit flesh and fruit peel of L. domesticum can be used as a cosmetic active ingredient for anti-aging and anti-tyrosinase.

Abstract

Siam citrus peel (Citrus nobilis L. var. Microcarpa) is a plant derived from Sambas Regency, West Kalimantan Province. Bioavailability of herbal active compounds can be enhanced by formulating extract into nanoparticle. The polymer used was chitosan with crosslinker Na-TPP. Stirring speed and stirring time play an important role to produce small particle size in forming nanoparticle using ionic gelation method. Enhancement of stirring speed and stirring time could reduce particle size. Nanoparticles were prepared using ionic gelation method by mixing Na-TPP, extract and chitosan (1:1:6) with varying the stirring speed 500 rpm, 1000 rpm, 1500 rpm and stirring time 1 hrs, 2 hrs, 3 hrs. The particle size of nanoparticle was found to be 85.3 nm at 1000 rpm of stirring speed and 3 hrs of stirring times, with polidispersity index 0.287, zeta potential +32.37 mV and entrapment efficiency 87.12 %.

Abstract

Antioxidant is an interesting topic due to their capability to inhibit free radical and prevent damage because of oxidative processes. Endophyt fungi is one of antioxidant compound resources in nature. The low yield to gain antioxidant compound from fungi challenges to look for the composition of media and optimalization of growth conditions. This research aimed to know the effect of medium condition in different carbon and nitrogen sources as well as initial pH towards antioxidant activity of endophyt fungi Bo.Ci.Cl.A3. Shaker fermentation was used on 120 rpm at room temperature for 14 days. The carbon sources were glucose, sucrose, and starch and nitrogen sources were NaNO3, NH4NO3, and yeast extract with initial pH at 5, 7, and 9. Ethyl acetate was used as extractor. The results showed that endophyt fungi can produce secondary metabolite as antioxidant at all variation of fermented media. The nitrogen source of yeast extract could increase antioxidant activity of endophyt fungi Bo.Ci.Cl.A3, while other sources such as nitrogen source, carbon sources, and different initial pH on the basal medium that were used did not give increasing antioxidant activity. The conclusion of this research was the substitution of nitrogen source with yeast extract (3 g/L) on the basal medium Czapek Dox’s Broth could increase antioxidant activity of endophyt fungi Bo.Ci.Cl.A3.

Abstract

Costus speciosus has been reported to have biological activity among antidiabetic, antihyperlipidemia, antioxidants. One of the chemical constituents that is responsible for some of the biological activity is diosgenin. This study aimed to determine diosgenin content of Costus speciosus water extract (CS). Water extract of CS was obtained with infundation method. The extract was dried using freeze dryer. The assay of diosgenin was performed by HPLC method. HPLC system used: LiChrospher® 100 RP-18 endcapped (5 µm) column length 25cm, colimn id 4 mm as stationary phase, acetonitrile-water (9: 1 v/v) as mobile phase, flow rate 2mL/min and UV detection at 205 nm. HPLC method developed was linear in the range 20-200 μg mL with R = 0.9999 ; slope = 6423.7; and intercept = -4280.5; LOD = 2.14 ug/mL and LOQ = 6.50 mg/mL; and recovery 100.63 %. Diosgenin levels in CS was 0.279 %. The developed HPLC method is relatively simple, rapid, sensitive, and accurate to determine the content of diosgenin in water extracts of Costus speciosus.

Abstract

Torbangun (Plectranthus amboinicus (Lour.) Spreng) is a medicinal plant that has been traditionally used in tropical countries to cure various illnesses. The objective of this study was to identify the active compounds in the chloroform fraction which have effect on the apoptosis-related genes expression of breast cancer MCF-7 cells. Apoptosis was observed morphologically using Hoechst nuclear staining. Expression of the genes was analyzed using Real-Time PCR. Chemical compounds of the plant fractions were determined using LC-MS. Result of cell morphology observation clearly indicated apoptosis after the treatment of the plant fraction. Increased expression of anti-apoptotic gene Bcl-2 could not prevent the cells from apoptosis. Expressions of p53 and p21 genes were increased significantly. The expressions of caspase 9, caspase 7 and caspase 1 were increased at concentration-dependent manner. Most of the compounds in the chloroform fraction are identified as diterpenoids which may contribute to the apoptosis inducing activity of the fraction.

Abstract

Kandidiasis adalah penyakit infeksi jamur yang masih menjadi masalah kesehatan di Indonesia, kandidiasis terutama oral sebagian besar dikarenakan pertumbuhan abnormal Candida albicans pada rongga mulut. Minyak jeruk purut dan serai telah terbukti memiliki aktivitas antijamur terhadap Candida albicans. Komposisi limonen, α-terpineol, sitronelil assetat, terpineol, β-pinene, pada minyak jeruk purut dan citronelal pada minyak sereh membuat kombinasi keduanya minyak memiliki potensi sebagai terapi alternatif untuk kandidiasis oral. Kedua minyak ini dapat diformulasikan dalam bentuk nanoemulsion obat kumur. Nanoemulsion dipilih karena memiliki beberapa keuntungan seperti yang jelas, stabil, dan meningkatkan aktivitas antijamur minyak sereh dan minyak jeruk purut. Penelitian ini dimulai dengan determinasi tanaman, uji karakterisasi minyak atsiri (kelarutan, indeks bias, dan berat jenis), dan analisis kandungan minyak atsiri dengan GC-MS. Nanoemulision diformulasikan dengan metode titrasi air. Optimalisasi campuran minyak dan VCO dilakukan untuk mendapatkan nanoemulsion yang jernih dan stabil. Formula terbaik dikarakterisasi (ukuran partikel, viskositas, potensi zeta, dan stabilitasnya), dilakukan analisis senyawa kimia minyak atsiril dalam formula optimum dan menguji efek penghambatan formula oleh metode mikrodilusi. Analisis data daya hambat dilakukan dengan One Way ANOVA. Berdasarkan hasil penelitian, 0,4% minyak campuran, 3,6% VCO, 17,3% tween 80, 8,7% PEG 400 dan 70% air merupakan formula terbaik yang dapat membentuk sistem nanoemulsion dengan rata-rata ukuran partikel 21,4 nm, viskositas rendah, potensi zeta yang rendah dan stabil selama penyimpanan freez-thaw. Nanoemulsi secara signifikan memiliki efek penghambatan lebih efektif daripada kontrol positif.

Abstract

Muna tribe in Kota Wuna Settlement has utilized various types of medicinal herbs. However, until nowthere has been no documented information about the scientific names and their utilization of the medicinal plant species in the area. The objective of this study was to determine the number and types of medicinal herbs used by the society in the study area, efficacy and organs of the plants are utilized. The research was conducted from January until November 2013, in Wuna City Settlement Tongkuno districts, Muna regency, Southeast Sulawesi Province. This research method used was survey and interview. Identification of the types of medicinal herbs was done by matching the characteristics that exist with images referring to the plant identification book. The results showed that there were 34 collections of plants that have medicinal properties traditionally utilized by the Muna tribes at Wuna City Settlement. Based on 34 types of plants that used, Based on 34 types of plants used, 31 species have been identified and 3 plants species have not been identified.

Abstract

Jatropha multifida L. (jarak tintir) was a shrub, annual, and had ± 2 m high. Empirically jarak tintir sap was used as traditional medicine by Indonesian people for a long time. Only limited studies were conducted regarding its chemical compound. It was reported that multifidone (diterpenoid compound from the stem) had an activity against cancer cells in vitro. This study aimed to test the toxicity of various extracts (n-hexane, ethyl acetate, and methanol) with Brine Shrimp Lethality Test (BSLT) of Jatropha multifida L. stem. A Fraxidin (8-hidroxy-6,7-dimethoxy coumarin) has been isolated from ethyl acetate fraction based on highest cytotoxic with LC50 value 3.69 µg/mL. The isolated compound was elucidated to gain chemical structure base on spectroscopic data (UV-Vis Spectrofotometric, IR Spectrofotometric, and NMR). Toxicity of fraxidin was tested on BSLT and showed no potential activity with LC50 value > 500 μg/mL.
