Abstract

Dirofilaria immitis (D. immitis) as a causative agent of heartworm disease is not only caused problems in animals but also zoonoses. For diagnosis of dirofilariasis (disease caused by D. immitis) serologically is needed anti-D. immitis antibodies. The objective of this study was to produce antibodies against excretory-secretory antigens produced by male and female worms for developing the diagnosis of dirofilariasis based on the antigens detection. Base on this study can be concluded that male excretory-secretory antigens (MES), female excretory-secretory antigens (FES), and MES+FES can stimulate BALB/c mouse to produce polyclonal antibodies in the same pattern. Antibodies have been produced at day 21 and the peak titter was at day 35 after first immunization.

Abstract

Centella asiatica (pegagan) merupakan salah satu tanaman obat yang memilikibanyak fungsi, salah satunya adalah sebagai imunostimulator, yang berarti dapatmerangsang tubuh untuk meningkatkan respon imun. Namun efek pegagan terhadapstruktur mikroskopis hati pasca diinfeksi Salmonella typhi (S.typhi) belum pernahdilakukan. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak pegagan terhadapstruktur mikroskopis hati mencit yang diinfeksi S.typhi . Penelitian ini menggunakan 24ekor mencit jantan umur 8-12 minggu, yang dibagi menjadi 4 grup. P0 sebagai kontroldiberikan aquades, dan grup P1,P2, dan P3, masing-masing diberikan 125 mg/bb, 250kg/bb mg, 500 mg kg/bb. perlakuan diulang sebanyak 6 kali. Extrak pegagan diberikansecara oral selama 14 hari, pada hari ke 15 seluruh mencit diinfeksi dengan S.typhi,kemudian pada hari ke 14 semua mencit dinekropsi dan hati diambil untuk diprosespembuatan preparat dengan metode pewarnaan hematoxylin eosin (HE). Pengamatandilakukan terhadap gambaran mikroskopis berupa perdarahan, degenerasi vakuola, dannekrosis. Hasil analisis menunjukkan mencit yang tidak diberikan pegagan dengan mencityang diberikan pegagan dosis 125 mg/kg bb., memberikan hasil yang tidak berbeda nyataNamun mencit yang diberikan pegagan dosis 250 mg/kg bb, dan 500 mg/kg bb memberikangambaran mikroskopis yang berbeda nyata.

Abstract

An observational study in order to determine the distribution Enterobacter spin Bali cattle at different geographical area (highland and lowland) and according to the animal maturity (calves, heifers, adult) have been undertaken. Isolation and identification of bacteria based on the method of Carter and Cole with slightly modification and the antibiotic sensitivity test were determined using the disc diffusion method by Kirby and Bauer. The Enterobacter sp. was isolated from 16 (13.3%) out of the 120 nasal swab samples,where 8 (6.7%), 6(5.0%), and 2 (1.7%) was isolated from calf, heifers, and adults, respectively. Meanwhile, 6 (5.0%) Enterobacter sp. was isolated from the highland and the remains (8.3%) from the lowland. Antibiograms showed that 13 of the 16 isolates (81.2%) weresensitive to sulfamethoxazole and the remains (18.8%) intermediate. While 12 (75.0%), 1 (6.2%), and 3 (18.8%) isolates weresensitive, intermediate, and resistant to ampicillin, respectively. To oxytetracycline 8 (50.0%) isolates were sensitive, 4 (25.0%) isolates were intermediate, and 4 (25.0%) isolates were resistant. Generally, the Enterobacter sp isolates were sensitive to all the antibiotics tested.

Abstract

This study aims to identify the species Staphylococcus grouper in Karangasem Regency.Faeces samples were collected and grown on media Blood order and identified by Gram staining.Suspected staphylococcal bacterial culture, included in the 10% Chelex media. The 16S rRNAgene fragments amplified from DNA (deoxyribonucleic acid) by Polymerase Chain Reaction(PCR) with specific primers that is Tsta G422 and Tstag 765. PCR (Polymerase Chain Reaction)products were sequenced in Barkley Sequencing Facility in the USA The result of sequencing wasedit and sorted using MEGA 5.0. Final sequence from each isolate was aligned using database in GenBank by fitur of BLAST. Several of standard sequences were downloaded to construct thefilogenetik tree. Species which can be confimed was Staphylococcus warneriwith homology valueof 100% with the data in GenBank.

Abstract

The research has been conducted to five kinds of traditional herbs which were collectedfrom Tajun village, Kubutambahan District, Buleleng Regency, Bali. It was conductedthrough extracting process and antibacterial bioactivity test to the five traditional herbs,naely kamboja (Plumeria rubra), kedondong (Spondias pinnata), kembang sepatu(Hibiscus rosa sinensis), mangga (Mangifera indica) dan manggis (Garnicia mangostana).The bioactivities antibaterial of these herbs had been exsperimented toward Micrococcusluteus and Escherichia coli. As the result, the leaves of the herbs which actively hamperedthe growth of Micrococcus luteus and Eschericia coli bacteria was manggis leaves (Garnicia mangostana). The herb which only hampered the growth ofEscherichiacoli actively was kamboja leaves (Plumeria rubra), kembang sepatu (Hibiscus rosasinensis), mangga (Mangifera indica). Mean while kedongdong leaves do not haveactivities to both kins of bacteria. Manggis had a goods potential to hamper the growthof Micrococcus luteus and Eschericia coli. It could be seen from the wide area of obstacleproduced by manggis leaves (Garnicia mangostana), namely 4,63 mm2 towardMicrococusluteus and toward Eschericia coli 5,63 mm2 in 105,5 part of point concentration.

Abstract

This study aim was to determine the total bacteriacountthat contaminated broiler farming water in Mengesta Village, Penebel District, Tabanan Regency. The water sample was collected from the source of the spring, water reservoir and chicken drinking pot. Each sample was taken as many as 100 ml and placed in the erlenmeyer glass. Water sampling was repeated 9 times infour-day sampling period. The data obtained were tested by analysis of variance and continued with least significant difference test. The results showed that total bacteria in chicken drinking pot(389,78 CFU / ml) was higher than in springs (247,89 CFU / ml), and shelter (171,33 CFU / ml). Its concluded that bacterial contamination occurred along the water flow.

Abstract

The aims of this study were to determine the viability and motility of Landrace pig’sspermatozoa in the Beltsville Thawing Solution (BTS) diluent with addition vitamin C which werestored at a temperature of 15ºC. During the storage process, metabolic activity of spermatozoaproduce free radicals that can reduce the viability and motility of spermatozoa thus it needs theaddition of antioxidants. Vitamin C is an antioxidant that can be used to scavengethe free radicals.This study used vitamin C added into the BTS diluent with scavenge dose is 0.1 mg/ml, 0.2 mg/mland a dose of 0.3 mg/ml. The results showed that a dose of 0.1 mg/ml is the best dose to maintainthe viability and motility of Landrace pigs spermatozoa were storage at a temperature of 15ºC.

Abstract

Penelitian ini bertujuan untuk mengetahui prevalensi,  intensitas infeksi dan predileksi parasit crustacea pada ikan sulir kuning (Caesio cuning) dan Ikan Pisang-pisang (Pterocaesio diagramma) yang dipasarkan di Pasar Ikan Kedonganan, Kabupaten Badung. Penelitian ini dilakukan dengan menggunakan 35 sampel Ikan Sulir Kuning (Caesio cuning) dan 35 sampel Ikan Pisang-pisang (Pterocaesio diagramma) yang diperoleh di Pasar Ikan Kedonganan. Metode penelitian yang digunakan adalah metode observatif dan analisis data dengan uji Chi-Square. Hasil penelitian yang telah dilakukan, didapatkan prevalensi infeksi parasit Crustacea pada Ikan Sulir Kuning (Caesio cuning) adalah 22,86 %, dan pada Ikan Pisang-pisang (Pterocaesio diagramma) adalah 31,43 %. Parasit Crustacea yang menginfeksi Ikan Sulir Kuning (Caesio cuning) dan Ikan Pisang-pisang (Pterocaesio diagramma) adalah dari kelompok Isopoda dan Copepoda. Identifikasi lebih lanjut, ternyata kedua ikan ini terinfeksi jenis parasit Crustacea yang sama, yaitu Isopoda jenis larva praniza dan Copepoda jenis Sagum folium. Rerata intensitas infeksi  Sagum folium pada Ikan Sulir Kuning (Caesio cuning) adalah 1,5 ± 1,46 dan larva praniza adalah 1,5 ± 1,40. Rerata intensitas infeksi Sagum folium pada Ikan Pisang-pisang (Pterocaesio diagramma) adalah 2 ± 1,77 dan larva praniza adalah 1. Parasit Crustacea yang ditemukan berpredileksi pada insang, operculum dan mata. Setelah dilakukan analisis dengan Uji Chi-Square, ternyata tidak terdapat hubungan yang nyata antara jenis ikan dengan prevalensi infeksi parasit Crustacea.

Abstract

This study aims to determine the profile of immunoglobulin M (IgM) of bali cattle in Nusa Penida. The serum samples of 54 bali cattle that were kept in Nusa Penida based on geographical criteria consisted of highlands (Klumpu and Batumadeg villages) and lowlands (Ped villages), age covering young bali cattle (12-18 months ) and adults (24 months and over), while sex (male and female). The serum obtained was tested using the ELISA (Enzyme Linked Immunosorbent Assay) method. The results showed that 54 samples of serum of bali cattle in Nusa Penida were detected in the presence of levels of immunoglobulin M (IgM) with values ranging from 2.565 ng/mL – 8.834 ng/mL. The mean rate of serum IgM cattle raised in the lowlands (4,837 ± 1,385 ng / mL) was descriptively higher than in the highlands (4,761 ± 1,353 ng / mL), while for female bali cattle (5.018 ± 1.370 ng / mL) higher than male (4,477 ± 1.290 ng / mL), while adult bali cattle (4,869 ± 1.417 ng / mL) was higher than young bali cattle (4,707 ± 1.305 ng / mL), but statistically comparable from all categories showed no significantly (P> 0.05). The average rate of IgM of bali cattle maintained in Nusa Penida can be used as a data base of IgM profile of bali cattle that is maintained in Nusa Penida.

Abstract

Purpose of this research is to learn the level of effectiveness of ethanol extract from red betel leaf (Piper crocatum) within increase weight of male white mouse in condition for diabetes mellitus. 20 male white mice (Rattus novergicus) in 3 mounth with weight between 190-250 gram which adapted during 2 weeks. It classified to be 5 groups random, each group consisty by 4 mice. The first treatment as a negative control (healty mouse) whitout giving treatment. The second treatment as alloxan positive control, the third treatment alloxan added by 2% suspension of ethaol extract from red betel leaf (dose 100 mg/kg bb) per oral. The fifth treatment is alloxan added by 0.02% suspension glibenclamide (dose 1 ml/kg bb) per oral. Giving extract betel leaf is done by oral with used sonde. Giving treatment at treatment III, IV and V is done everyday start from day 3 until the end of the day (21 day). The weight of mouse is measure at day 7, 14 and day 21 used analytical balanced. The result of this research show up that giving 2% ethanol extract from red betel leaf (Piper crocatum) with dose (50 mg/kg bb), dose (100 mg/kg bb) and glibenclamide 0,02% ( 1 ml/kg bb) with significant can increase weight (P<0,05) of white mice which induce alloxan. Result of this research could be conclude that ethanol extract of red betel leaf (Piper crocatum) can used for increase the weight.
