Abstract

Free radicals released from metabolism processes in the body were reactive and toxic to cells and tissues because it cause protein, cell membrane and nucleic acid damages which lead to cancer. It could be overcame if antioxidant system in the body is in good function.Nevertheles antioxidant system could be destructed by variety of conditions. Modern lifestyle that frequently consumed of high saturated fat, additives, and low fiber content in foods were potential risk for cancer and hypercholesterolemia. To overcome that risks with specific drugs were high cost. Therefore this research was conducted to take advantages of natural sources which potential for antioxidant and antihypercholesterolemia activities that could be fast, easy and inexpensive processing. One of the natural sources that meet the criteria was angkak resulted from rice fermented by Monascus purpureus. The result revealed that angkak contained lovastatin showed antioxidant and antihypercholesterolemia activities and increased glutathion peroxidase activity of optimal dose 5 g/day. The histopathologi observation of rat’s liver showed that administered of angkak on rats feed high level of cholesterol inhibited accumulation of fat in rat’s liver.

Abstract

To increase domestic and international demand of Kencur (Kaempferia galanga L.) makes this plant potentially develop.Is traditionally used to keep the body warm, as analgetic and expectorant. In the attemp of providing adequately and qualitatively uniformed supply, in vitro experiment has been conducted at BB-Biogen (Indonesian Center for Agricultural Biotechnology and Genetik Resources Research and Development).The selected rhizomes was used as explant. The experiment was orthogonally arranged consisting of MS vitamin and B5, and BA ( 0, 3 and 5 mg/1) and thidiazuron 0,1 mg/1. This experiment comprised three activities, they were shoot initiation, shoot multiplication and acclimatization. The result showed that MS + BA 3 mg/1 + thidiazuron 0,2 mg/1 could induce shoot formation. From the applied media, it was shown that the addition of MS vitamin at the MS basic media and BA 3 and 5 mg/1 added with thidiazuron could result the most optimum shoot, leaves and roots and was not significantly different from the addition of B5 vitamin at basic media of MS + BA 3 and 5 mg/1, 6.9 shoot was averagely produced in this media. The shoot could generate such an adequate number of root that it could be directly acclimatized. The acclimatized plantlet in the green house uses the mixture of soil and manure with the ratio of 1:1 can optimally grow.

Abstract

Research on vegetation, where Hoya purpureofusca Hook.f. grows, has done in the Selabintana Resort, Mount Gede-Pangrango National Park in September 2011. Purposive random sampling was set up for a total of 18 square plots of 10m x 10m, where placed on two different sites. Nine plots were placed in the growing sites of H. purpureofusca, otherwise were placed in other sites. Observation was made in each sampling plots on the number of species at three stages, i.e. seedling, sapling and tree in each sampling plots. The diversity indices (including species richness, heterogeneity, and evenness) were analyzed using Jakcknife index, Margalef index, Menhinick index, Simpson index and Shannon-Wiener index. Species dominance was analyzed by important value index and similarity index whereas species associations was analyzed by chi-square. Based on the analyses, there were differences on vegetation composition and diversity between growing site of H. purpureofusca and non growing site. Schima wallichii (DC.) Korth. is the dominant species that has an important role for the growth of H. purpureofusca.

Abstract

Fertilization with organic and inorganic fertilizers increase the availability of nutrients in the soil and improve soil physical and chemical properties, The nutrient must fulfill crop requirement regarding to its effect to seed quality. The object of experiment was to study the effect of a combination organic fertilizer and inorganic fertilizer to resilience of soybean {Glycine max (L.) Merr.} seed var. Anjasmoro. Storage of seeds, were conducted in the Post Harvest Laboratory of Balai Penelitian Pertanian Lahan Rawa-Balittra (Indonesian Swampland Agriculture Institute), Banjarbaru from October 2009 until April 2010. The experiment was designed in completely randomized with 4 replications. Seed water content, seed germination, vigor index, electrical conductivity and seedling development were observed. The results showed that during storage, water content of seed increased 3-4% and limit the water content of seeds was still able to be maintained at <13%. Organic matter and NPK fertilizer declined vigor and germination of seed. Vigor is still above 70% and germination as high as >77%; As lower as seed germination generate as higher as electrical conductivity and lower the seedling dry weight.

Abstract

Keladi tikus (Typhonium flagelliforme (Lodd) BI.) of the Araceae generally grows wild in the open or in slightly shaded areas. The active compounds of the plant have not yet been known, however its potential for cancer medication has been reported by several media. Since the plant is being hevaly removed in their habitat, research for adoption of in vitro technique for production of their planting materilals is needed. Lateral shoots of a 2-cm tuber, were sterilized with 30% sodium hypochlorite for 15 minutes. After several rinses with sterile distilled water, the shoots were cultured on a soild Murashige & Skoog (MS) medium supplemented with BAP (0,25;0,5;0,75;1,0 mg/l) in combination with IBA (0,25;0,5;0,75;1,0 mg/l). All cultures were incubated at 26 C under fluorestcent lights of 16 hours/day. Rooting of shoots was induced on MS medium without growth regulators and the plantets were established in a potting medium of sand,compost and soil (1:1:1). The result showed that the highest rate of shoot proliferation and growth was on MS medium supplemented with 0,5 mg/l 1 BAP and 0.25 mg/l IBA. Using this medium,  a sixteen-fold increase in shoot multiplication can be achieved with survival rate of 95%.

Abstract

This study was aimed at studying the effectiveness of alcohol, formaldehyde and their mixture in inhibiting fungal growth.This study contained two step. The first step was fungal isolation from specimen.One isolate was then selected to be inoculated into alcoholat various concentration (15%, 30%, 50% and 70%) with or without formaldehyde mixture.The second was fungal inoculation intoalcohol at various concentration (15%, 30%, 50%, dan 70%) with or without formaldehyde.The effectiveness of these subtances as specimen preservatives for shrimp were studied after six months incubation time.From the fisrt step, only genus group Monascus was isolated from seven specimens(Mabuia multifasciata, Myotis muricola,Chironax melanocephalus,Naya naya sputratrix, Miniopterus schreibersi blepotis,unidentified shrimp, unidentified Coelenterata).From the second step, the best growth of isolate Monascus sp.MM which isolated from the fisrt step was achieved in 70% alcohol.This fungus was not able to grow in both formaldehyde and its mixture with alcohol (1:1, 1:2, 1:3 and 1:4). The results also showed that Monascus growth was observed in alcohol with not sterilized shrimps without inoculation with Monascus sp. MM. It is recommended to apply mixture alcohol 70% with formaldehyde 1% at ratio 4:1 as animal specimen preservative to inhibit fungal growth.

Abstract

Genetic crossing of white oyster mushroom to introduce longer storage life trait can only be done within individuals in this particular species. However, longer storage life trait is possessed by brown oyster mushroom (Pleurotus cystidiosus) which is other species within this genus. Thefeore, protoplast fusion between white oyster mushroom (Peurotus floridae) and brown oyster mushroom (Pleurotus cystidiosus) was conducted to hopefully obtain an oyster mushroom strain that has higher production and longer storage life. Protoplast fusion was done by isolating protoplast from 5-days old monokaryotic mycelia grown in PDB. As much as 3.15 x 10 protoplasts/ml were harvested using mixture of cellulase Onozuka R-10 (1%) and macerozyme R-10 (1%) from brown oyster mushroom with 80.61% viability. Similarly, 3.71 x 10 protoplasts/ml were harvested using lysing enzyme (2%)from brown oyster mushroom with 83.68% viability. Protoplast fusion were conducted using 0% (control), 30%, 40% and 50% of PEG6000. Fusion periods were done at 10, 20 and 30 minutes. The candidate fusants were then screened using MRM (minimum regeneration media) media. Based on this experiment, the optimum conditions for protoplast fusion is 10 minutes incubation using 40% PEG6000 that yielded 121 colonies grown on MRM media as candidate fusants.

Abstract

Stachytarpheta jamaicensis (L.) Vahl. belongs to family Verbenaceae which has potential as medicinal and ornamental plant. Research on determination of the effect of storage duration on seed germination of Stachytarpheta jamaicensis (L.) Vahl. was conducted in the glass house of Purwodadi Botanic Garden from July 2012 - July 2013. Experiment used a completely randomized design with treatments storage duration namely: S0 = no stored ; S1 = stored for 2 months ; S2 = stored for 5 months ; S3 = stored for 8 months and S4 = stored for 12 months. The treatments was replicated 5 times with 100 seeds for each replication. The seeds were sowed on river sand medium sifted with 2 mm sieve mess at about 0.5 cm deep, in polybags ( 15x10 cm ). The polybags were placed in a plastic box 38 x 28 x 13 cm and covered by transparent plastic and black paranet. Variables measured were germination percentage and daily germination rate. The results showed that the seed storage duration had significantly affected seed germination rate of Stachytarpheta jamaicensis (L.) Vahl.. The highest seed germination (96.8 %) was recorded for the treatment of eight months storage duration (S3). Whereas the fastest germination rate (5.53 days) was observed on the treatment of two months storage period.

Abstract

Various biological aspects o/Telenomus sp.(Hymenoptera: Scelionidae) an eggs parasite of the rice stem borer Scirpophaga incertulas (Lepidoptera: Pyralidae) was conducted in the Pest Laboratory for Crop Institute, Bogor, Indonesia.The results showed that the parasite is solitary in habit and mainly live in eggs of the rice stem borer.The arfu Telenomus could maintain longer life span (3.4 days) in the cage and produced a large number of progenies (29.4 individuals) when they were given food of 10% honey solution compared to those which were given only water (2.1 days of life span, and 22.4 individual progenies).The parasite life span and the number of progenies were the least (1.8 days of life span, and 18.8 individual progenies) when they were treated without any food.The number of progeny was also the greatest when they were provided with one day old of stem borer eggs (45.87 individuals), while the number of progeny declined when they were provided with 2 days (36.93 individual progenies) and 3 days (24.87 individual progenies) old stem borer eggs respectively. Pairs of day old parasites are able to parasite more host eggs compared to that of successive days old parasites.The sex ratimf 2$ : 2$ produced more progenies compared to that with the sex ratio IS : 2$ and 2$ : lQ.Parthenogenetic reproduction in arfu Telenomus parasite being tested resulted in the male progenies only.

Abstract

Efficiency of water use in paddy rice irrigation system is necessary to increase cropping intensity. Development of rice cultivars that are responsive to adequate soil moisture with high yield and efficient use of water is required for tropical paddy fields. The research was conducted in Sleman district of Yogyakarta province, from April to August 2011. The study was conducted on some selected rice cultivars to be cultivated on soil saturated culture (SSC) system to asses the response of cultivars that can produce high grain yield. Experiment was using randomized block design (RBD) and environment factors as replications. Control treatment was rice field. Rice cultivars used are IR.64, Ciherang, Inpari-1, Inpari-2, Inpari-3, Inpari-4, Inpari-10, Inpari-6 Jete, Cigeulis, Ciliwung, Luk Ulo, Way Apo Buru, Widas, Bondoyudo, Silugonggo, Tukad Petanu, Batang Piaman, Cibogo, Dodokan, Sarinah, OM.5637, WS.63-22-1-1-3-3, Inpari-8, Situ, Bagendit, and Cimelati. The results showed that the cultivars Cimelati and Sarinah was a cultivar response to the SSC system compared to other cultivars. The Cimelati cultivar grain yield potential was 25.02 g/hill and Sarinah was 21.45 g/hill. The yield using SSC system was higher compared to rice fields system.
