Abstract

The Peucang Island is situated just off westernmost tip of West Java and is a portion of the Ujung Kulon Nature Reserve.It covers a total area of 450 hectares,comprising the level land and a ridge in the central portion with the highest point of about 30 m above sea level.Geologically this island is composed of Tertiary materials and alluvial materials on southern and eastern coastal areas (Verbeek and Fennema 1896).From the southeastern shore towards the hills, there are three types of soil, i.e. the sandy regosols, the tuffaceous over sandy regosols and the latosolized tuffaceous regosols over podsolized grumusols (Soerianegara 1969).

Abstract

Kentang hitam [Plectranthus rotundifolius (Poiret) Sprengel] of family Lamiaceae is a minor root crop known only for people living in some parts of Java, Bali and Madura. It was rarely found in its natural habitat, thus it was assumed to have low level of genetic diversity. This present study aimed to assess genetic diversity of 63 accessions of kentang hitam from provenances of Java based on Inter Simple Sequence Repeats (ISSR) and Random Amplified Polymorphic DNA (RAPD) fingerprints. Ten primers of ISSR and RAPD were initially screened and eight were selected for the analysis. These eight primers (OPA13, OPB10, OPB13, OPD8, OPN14, UBC 807, 834 and 835) generated 61 bands with an average of 7.63 polymorphic fragment per primer. Percentage of polymorphism ranged from 8.20% (UBC 807 and 834) to 16.39% (OPB 10) with an average of 12.50%polymorphism.Clustering analysis was performed based on ISSR and RAPD profiles using the neighbour joining method and Principle Coordinate Analysis (PCO). The range of genetic similarity among accessions was 51-100% to which most of the accessions were clustered with more than 80% similarity. This confirmed our hypothesis of the low level of variation existed among accessions.

Abstract

M. DJAJASASMITA. 1985. Molluscan fauna of the two stream rivers of Riau. Berita Biologi 3(3): 121 - 124. - A preliminary study on the composition and population density of the molluscan fauna in the streams of the rivers Tiwi (stony and sandy bottom) and Tandun Kecil (clayed muddy bottom), in Riau, East Sumatra, has been carried out, in July 1977. The molluscs collected consisted of Brotia costula, Clea bocki and Corbicula moltkiana in Tiwi River and B. costula, Pseudodon vondembuschianus and Rectiden graeilis in Tandun Kecil River. B. costula dominate in both localities, 86.08% and 72.58% respectively; the other species range from 2.42% to 25%. In general both localities have low molluscan population density, 41.2 m" and 26.8 m" respectively. The food condition is rather sufficiently available,the water quality may support the molluscan life, the physical condition, however, seems to be less suitable.

Abstract

Beauveria bassiana produces several metabolites that are toxic to insects so that it can be used as a biological insect control agent as an alternative to synthetic pesticides. The aim of this study was to determine the larvicidal activity of ethyl acetate and hexane extract from B. bassiana filtrate culture against Aedes aegypti 2nd instar larvae. This research was it cooked by determining the optimum age of spore inoculum of B. bassiana on the Potato Dextrose Agar (PDA) based on the number of spores and its viability. Afterwards, we determine the incubation time of B. bassiana in the Potato Dextrose Broth (PDB) in order to obtain filtrate culture which have highest mortality effects against Ae. aegypti 2nd instar larvae. B. bassiana filtrate culture was extracted with hexane and ethyl acetate and tested aegypti for larvicidal activity with a concentration range of 50, 100, 200, 300 ppm. The LC50 value was carried out by probit analysis. The results showed that ninth day old culture in the PDA was the optimum age of spore inoculum with the spore number and viability were 2.54 x 107 spore/mL and 93.46% respectively. The filtrate of sixth day old culture in PDB medium gave 100% mortality against 2nd instar Ae. Aegypti larvae. LC50 values of ethyl acetate and hexane extract were 117.28 dan 287.09 ppm. These results showed that the ethyl acetate and hexane extract of B. bassiana filtrate culture have biopesticide potential against 2nd instar Ae. aegypti larvae.

Abstract

YUN INDIARTO, MULYADI & RAZALI YUSUF, 1989.The structure and composition of mangrove in Bikoen-Kupang,Timor.Berita Biologi 3(9): 455 - 457.Mangrove forests in EastNusa Tenggara cover a total areas of about 1930 ha(Darsidi, 1984). Little is known about the mangrove forest in that island, especially in Bikoen, Kupang Region.Therefore, the study has been conducted at the mangrove forest of Menipo Nature Reserve in order to analyse the structure and composition of the species.Four transects perpendicular to the coast line were used for data collection.At least, there were 12 mangrove species found in the forest, dominated by Ceriops tagal, Rhizophora stylosa and Bruguiera exaristata. Seedlings dominated the forest by 49,92%, where as the sapling were 45.43%.

Abstract

ELIDAR.1987.The preparation of inoculum by using various flour as subtrates or carriers Suppl. Berita Biologi. 3: 51:55. for 2 mold and 2 yest strains,the rice,glutinous rice and tapioca wates flour colud be used as a substrate better then others flour.The survival of the inoculated microorganisms in the rice,glutinous rice and tapioca waste inoculums were not significantly difference.The viable spore count in those three inoculums decreased due to th duration of storage The viability of the spores count were  104- 106 propagule/g in the early storage and then it descreased 104 propagule/g after 3 months storage

Abstract

The study aimed to determine the effect of storage temperature and time reconditioning to maintain the quality of potato cultivars Margahayu. The experiments were conducted in the Laboratory of Postharvest of Vegetable Crops Research Institute (BALITSA) from July to October 2012. Altitude research is ± 1200 masl with temperatures between 14.94 - 24.61°C and the relative humidity (rH) 85.56%.Experimental design used in this study was factorial a randomized complete design consisting of 2 factors. Treatments were factorial combinations of storage temperature (4 °C, 7 °C and 10 °C) and time reconditioning (0 days, 3 days, 6 days and 9 days).The number of replications for each treatment was 3. The results of research showed that there is interaction between storage temperature and reconditioning on reducing sugar content and vitamin C. Storage temperature and reconditioning treatments were positive affect on Total Soluble Solid (TSS) and organoleptic chips. The treatment can maintain the quality of the potato tuber is the storage temperature 7 °C, 10 °C with a time of 6 to 9 days reconditioning.

Abstract

An extracellular p-glucosidase[EC 3.2.1.21]derived from Asvereillus vulverulentus were separated and partially purified by successive chromatographies and its some characterization and transglucosylation capacity were studied.The purification protocol included precipitation with ammonium sulphate,gel filtration,ion exchange chromatography and native polyacrylamide gel electrophoresis.The enzyme readily hydrolysed cellobiose to form transglucosylation products in the present of primary alcohol acceptors.This P-glucosidase was stable at temperatures up to 70 °C and from pH 2.5 to 8.5, while its highest activity was in the pH 4.5 at 65°C.

Abstract

D.I. HARTOTO, N. TRISNANINGSIH & I.RACHMATIKA. 1985. Fish niche of Ci Siih and CiJaralang estuaries, Sunda strait, West Java. Berita Biologi 3 (3) : 77 - 83. - A study of the fish community of Ci Silih and Ci Jaralang estuaries, Sunda Strait, West Java was conducted during the peak of dry season, August 1982. This study was aimed to apply Hutchinsonian niche theory into operational level and to reveal the stage of succession processes on the two estuarine fish communities.The study shows that type of refuges were more variate in Ci Jaralang than in Ci Siih and diversity of physical structure of spatial habitat resources affected a more diverse type of refuges. Means of absolute spatial niche breadth as a measure of concentration (?i) and uncertainty(?i) are larger in Ci Siih than in Ci Jaralang.Those facts indicate that fish species in Ci Siih were more dispersed in occupying the spatial habitat resources available. The larger (?i) value indicate that these two fish communities were still in early stages in their succession. The significantly similar means of absolute spatial niche overlap shows that the intensity of resources sharing on these two communities are nearly similar and are quite intensive.
