Abstract

Centella asiatica is a medicinal plant used for wound healing through increasing of collagen synthesis.This evidence generates a new expectation that it could be used for therapy of tuberculosis infection,especially for healing lung tissue damage. Until now, the effects and mechanisms onC. asiatica to cure thelung tissue damage due to M. tuberculosis infection remains unclear. The aim of this study was to prove theeffect and mechanism of ethanol extract of C. asiatica to repair the rats lung tissue damaged throughexpression of the enzimmatrix metalloproteinase-1(MMP-1)danenzimtissue inhibitor of matrixmetalloproteinase-1 (TIMP-1). The study was conducted in male rats. Twenty four rats were infected withM. tuberculosis through intratrachea and randomly divided into four groups. Group 1, 2 and 3 were thetreatment groups that they were given the ethanol extract of C. asiatica at dose 375mg/kgbw, 750 mg /kgbw, and 1500 mg / kgbw, orally and once a day for fourteen days. The fourth group was a control groupthat given distilled water. On day 15 rats were euthanized and lungs tissue have been taken. Evaluationof lungs tissue damage were assessed by the Dorman’s score in Hematoxylin Eosin and evaluation of the expression of MMP - 1 and TIMP 1 were performed by immunohistochemistry. Data of TIMP-1 wereanalyzed with ANOVA and data of lung tissue damage and MMP–1 were analyzed with Mann WhitneyU (á = 0.05). The results showed that there was a significant differences in the lungs tissue damagebetween the dose groups of 375 mg / kgbw and controls (p = 0.006), the dose groups at dose 750 mg / kgbwand controls (p = 0.004 ), the dose groups of 1500 mg / kgbw and controls (p = 0.043). There wasn’t asignificant difference between the treatment groups and control in the expression of MMP-1. In the expressionof TIMP – 1, there was a significant difference between the treatment group at dose of 750 mg / kg andcontrol. The conclusion of the study is the ethanol extract of C. asiatica has the ability to reduce lung tissuedamage of rats infected with M. tuberculosis, through increasesthe expression of TIMP-1

Abstract

The aim of this study was to evaluate the capability of two human commercial enzyme-linkedimmunosorbent assay (ELISA) kits(DRG International Inc.,Germany and GBC Taiwan) for measuringestradiol (E2) and progesterone (P4) in plasma of kacang goat. Three healthy and non pregnant femalekacang goats aged 2-3 years with regular estrous cycles were used in this study. Blood samples werecollected from the jugular vein using a 21G venoject every two days and it was intensified every day duringthe period before heat. Collected plasma were stored at -20ºC until the analysis. Capability validationwas conducted by measuring accuracy (parallelism test), sensitivity and precision.Parallelism test usingDRG commercial kit showed that sample curvewas parallel with standard curve of E2 and P4. In contrastit was not parallel with standard curve of GBC commercial kit. Sensitivity was measured from the lowesthormones concentration of E2 and P4 at 90% binding that were 25 pg/ml and 0.14ng/mL in DRG kit, whilein GBC kit were 5 pg/mL of E2 and 0.2 ng/mL of P4, respectively. Coefisien of variation of intra- andinterassay for both ELISA commercial kits were less than10%. It can be concluded that DRG commercialELISA kit E2 and P4 can be used to analyse female kacang goat blood plasma, while GBC commercialELISA kit E2 and P4 are not recommended.

Abstract

Diabetes mellitus (DM) is a metabolic syndrome due to pancreas failure to produce insulin, thereforehyperglycemia is the hallmark of DM. There is hyperglycemia in this disease. In the world, there isaround143 million diabetes mellitus patients. One of the deathcause in diabetes mellitus is chronic liver disease.Herbal medicine is thought to give less side effect than sintetic medicine. A traditional plant that hashypoglicemia property is decoction of Indonesian bay leaves. This study was aimed to study effectivity ofIndonesian bay leaf decoction in decreasing blood glucose level, pancreas and liver hemorrhage scores, andKupffer cell count on alloxan-induced Wistar rats. This Control Group Design used twenty four rats (threemonth old). The rats were divided randomly into four groups, i.e.: control, 1 mL of Indonesian bay leafdecoction (0.9 g/kg body weight (BW)), 2 mL of Indonesian bay leaf decoction (1.8 g/kg BW), and 3 mL ofIndonesian bay leaf decoction (2.7 g /kg BW). After one week adaptation the rats were inducedwithalloxan,then pretest for fasting blood glucose test was examined. Treatment was given for ten days and posttestblood glucose test was done afterwards. At the end of the experiment, all rats were sacrificed to collectpancreas, kidney, and liver. Those organs were stained with hematoxylin eosin. Levelof fasting bloodglucose, hemorrhage score, on pancreas and kidney and Kupffer cell count were analyzed by using Anova.In conclusion, Indonesian bay leaf decoction may lower fasting blood glucose level on hyperglycemic rats aswell as Kupffer cell count significantly, but it is not likely to give significant result in lowering of pancreasand kidney hemorrhage scores.

Abstract

A study to evaluate the correlation between the pigments produced by Staphylococcus aureusisolateswith their adhesion capability on epithelial cells and phagocytes activity of macrophages in vitro.Ten S.aureus isolatesfrom bumble foot and arthritis casesin broiler were used in this study. The adhesion assaywas performed using epithelial cell derived from human buccal epithelial cells whereas phagocytic activitywas conducted using mouse peritoneal macrophage cells.The results showed that S. aureus isolates frombroiler produced both yellow and white pigments. Staphylococcus aureus producing white pigment adheredto human buccal epithelial cells with the density of about 24,25 bacteria/cell, which were higher than thoseof Staphylococcus aureus producing yellow pigmentabout 23,78 bacteria/cell. Staphylococcus aureusproducing white pigment was more phagocytosed by macrophages than S. aureusproducing yellow pigment,with an average phagocytic activiry of19,86 bacteria/cellas compared to15,96of bacteria/cell, respectively.

Abstract

The aims of this study were to determine both chemical composition and fraction of the proteincompounds of sumba mare’s milk. Determination of the chemical compositions of sumba mare’s milk havedone by analyzing protein content using the Kjeldahl method, fat content using Gerber method, lactosecontent and the total solids content. Identification of antimicrobial compounds of whey proteins in milkusing high performance liquid chromatography (HPLC) method. The results showed that the average ofsumba mare’s milk contained protein, fat, lactose and total solids were; 1.82%, 1.67%, 6.48% and 11.37%respectively. The average value of protein and fat in sumba mare’s milk was decrease significantly at fifthmonth of lactation period. Based on identification of antimicrobial compounds using HPLC method, thereare six main peaks with different polarities and retention times. In conclusion, sumba mare’s milk havea balance composition that can be used as a source of nutritious food and the milk likely also has six mainantimicrobial compounds in its whey protein.

Abstract

The aim of the present study was to investigate the characteristics of frozen-thawed Garut ramsemen when cryopreserved in extender supplemented with Orvus Es Paste (OEP) as surfactant. Ramsemen were collected and evaluated, then cryopreserved in extender supplemented with OEP at differentconcentrations: OEP-0 (without OEP); OEP-0.25%; OEP-0.50%; OEP-0.75%; and OEP-1.00%. Thecharacteristics observed were motility, viability, and Intact Plasma Membrane (IPM). The results showedthat semen which were cryopreserved and supplemented with OEP had a higher motility, viability, andIPM, respectively compared to those without OEP. No significant differences in the characteristics wereobserved within the different OEP concentrations. It can be concluded that supplementation of OEP in theextender could protect the quality of cryopreserved ram spermatozoa.

Abstract

Study to determine the potential of tempe flour in improving bone quality of ovariectomized rats (ovx)based on calcium (Ca) and phosphorus(P) levels in the plasma and bones; osteoblast and osteoclastprofiles have been conducted for 4 months. A total of 20 female Sprague Dawley albino rats, age of 3months and average body weight of 200 g were used in this study. The rats were divided into 4 groups,i.e.normal rats + diets (K0); normal rats +tempe flour (K1); ovx + diets (OV0); and ovx + tempe flour (OV1),respectively. The Ca and P levels in plasma and bones were analyzed using Atomic AbsorptionSpectrophotometer (AAS) and spectrophotometric methods. In addition, the profiles of osteoblast andosteoclast were also observed. The results showed that Ca and P levels in plasma and bones of normal ratswere 11.08 mg/dL and 7.06 mg/dL; 30.88 mg/dL and 18.63 mg/dL, respectively. The highest Ca and P levelsin both plasma and bones were observed in normal rats which were given tempe flour (K1), 11.25 mg/dLand 7.42 mg/dL; and 31.60 mg/dL and 19.78 mg/dL, respectively. Similarly, Ca and P levels in plasma andbones of ovxwhich were given tempe flour were slightly higher (10.91 mg/dL and 6.96 mg/dL; and 29.89 mg/dL and 17.87 mg/dL) compared to ovx without tempe flour diets (9.38 mg/dL and 6.46mg/dL; and 26.58 mg/dL and 16.75 mg/dL). The numbers of osteblast was found slightly higher both in normal rats and ovxwhich were given tempe flour; whereas slightly decreased of osteoclast was observed only in ovx which weregiven tempe flour. The administration of tempe flour in ovx diets could improved bone quality as indicatedby the Ca and P levels in plasma and bones and decreased numbers of osteoclast.

Abstract

Fresh water and marine fish horticulture in Bali is often harmed by the outbreak of diseases such asthose caused by Aeromonas sp (aeromonad disease).Aims ofstudy were 1) to find out the primary agent ofthe aeomonad disease in the fresh water aquaculture in Bali based on conventional and molecularidentification, 2) to find out the genetic variability of Aeromonas species, 3) to determine the effectiveantibiotic against the agent. Samples of fishes were collected from 5 different locations of fresh wateraquaculture that had high number of morbidity and mortality. Many different fishes which showed clinicalsign such as skin lesion and exophthalmus were collected.Aeromonas hydrophila and A. salmonicida wereisolated and identified from fishes, they were then identified molecularly with DNA extraction, DNAamplification in 16S rRNA gene, purification and sequencing. Sequences of both Aeromonas species fromdifferent location were analysed to create the phylogenetic tree with Maximum Parsimony and NeighborJoining method. Sensitivity of 5 antibiotics to both species of Aeromonas were done to determine the bestantibiotic against the disease. Aeromonad disease were found only in 3 regions in Bali. As many as 10isolates of A.salmonicida and 11 isolates of A.hydrophila were examined. The histopathological examinationshowed dermatitis, epicarditis, retinitis, liver and kidney congestion in fish.There were two clusters ofA.salmonicida, subspecies smithiaand subspecies achromogenes.Aeromomas hydrophyla had a close relationwith A. veronii.Aeromonas salmonicida subspecies salmonicida has not been found in Bali. Enrofloxacineand gentamycin was the best antibiotic for treating the Aeromonad disease which were more effective ascompared to3 other antibiot ics (Ampicillin, Doxycycline, and Eritromycin).
