Abstract

This study was aimed to observe correlation between gestational age and fetal growth of Kacang goat for 60 days after mating with transcutaneous ultrasonography visualization. Three female goats of productive age with normal estrus cycle and one male goat were used in this study. The female goats were synchronized with double injection of prostaglandin F2á (PGF2á) intramuscularly with dosage of 1 mL for each female goat with 11 days’ time interval. Observation of oestrus was conducted using male goat after being synchronized. When the sign of estrus were prominent, the female goats were naturally mated. The result was pregnant positive for one female goat. Detection of early pregnancy was observed on the 24th days after mating, with the result of 6.77 mm length embryo by isoechogenic visualization. On 35th days of pregnancy, fetal could be seen clearly in isoechogenic to hyperechogenic visualization with head diameter and fetal length were 17.4 mm and 36.2 mm, consecutively. The observation also found the placentom. On day 45 of pregnancy, head diameter and fetal length were 21.8 mm and 40.6 mm. Later on day 49 of pregnancy, the size of gestational saccus was 44.1 mm, with head diameter of 25 mm, and average placentom diameter size of 12.4 mm. On day 53 of pregnancy, head diameter was 25.2 mm with fetal length of 63.6 mm and placentom diameter of 15 mm. On day 56 of pregnancy, vertebrae of fetal were observed using hyperechogenic visualization and placentom was measured 17.9 mm in diameter. On day 60 of pregnancy, we observed that the fetal length was 79.8 mm and the organs such as eyes, heart, liver, os costae, and fetal extremities, could be observed clearly. The size of developing fetal and organs would grow along with the addition of gestational age.

Abstract

Vector infectious diseases such as Dengue Hemorrhagic Fever (DHF) are still a major health problem in Indonesia including Cirebon City. Cirebon seaport as one of the country’s entrances has the potential to transmit dengue disease. The aim of the study was to measure the population density of Aedes aegypti larvae and analyze the relationship of habitat characteristics with the presence of Ae. aegypti larvae in the perimeter region of Cirebon seaport. Analytic descriptive observation with a cross sectional study approach was conducted in the study. The sample were all the houses/ buildings in the perimeter region of Cirebon seaport are used as breeding habitats for Ae. aegypti. The results of the study, the types of positive containers of Ae. aegypti larvae are water closet (WC) tanks, bathtubs, drums, pails, ponds and used tires. Calculation results of the House Index (HI), Container Index (CI), Breteau Index (BI) and Density Figure (DF) (HI: 8.74%; CI: 9.05%; BI: 12.02; DF: 3). Based on chi square test, habitat characteristics that have a significant relationship with the presence of Ae. aegypti larvae were container type, container material, water volume, water source, water pH and air temperature (p<0.05). The results of the analysis with binary logistic regression had a significant effect on the presence of Ae. aegypti larvae, namely in pails (p=0.003; OR=18.972) and water pH (p=0.022; OR=2.489). Conclusion of the study is Cirebon seaport area has a risk of dengue transmission at a moderate level based on its vector density.

Abstract

Kerbau lumpur merupakan ternak multiguna yang memiliki kedekatan dengan kehidupan masyarakat di kawasan lumbung ternak khususnya di Kabupaten Brebes, Jawa Tengah. Infeksi parasit saluran pencernaan pada kerbau lumpur bedampak negatif terhadap kesehatan ternak sekaligus menyebabkan kerugian ekonomi. Data epidemiologi infeksi saluran pencernaan pada kerbau lumpur di Kabupaten Brebes belum tersedia. Penelitian ini bertujuan untuk mengukur prevalensi, derajat infeksi serta analisis faktor risiko infeksi parasit saluran pencernaan pada kerbau lumpur di Kabupaten Brebes. Sebanyak 240 sampel tinja diperoleh dari kerbau di empat kecamatan, yaitu; Salem, Bantarkawung, Tonjong dan Brebes. Hasil pemeriksaan mikroskopik dengan teknik Mc Master menunjukkan bahwa 27.5% kerbau terinfeksi oleh parasit saluran pencernaan. Infeksi disebabkan oleh Toxocara vitulorum (1.67%; telur tiap gram tinja [TTGT] 174.4), cacing Strongyle (Cooperia, Nematodirrus, Trichostrongylus) (4.17%; TTGT 140.18), Trichuris spp. (3.33%; TTGT 64.84), Moniezia expansa (9.17%; TTGT 122.47) dan Eimeria spp (21.25%; TTGT 162.37). Infeksi lebih tinggi tercatat pada kelompok jantan, umur muda (>12–30 bulan), kerbau di Kecamatan Salem dan yang dipelihara pada daerah bertipe iklim D4. Variabel umur merupakan satu-satunya faktor risiko infeksi yang ditunjukkan dengan ­P-value 0.03. Nilai odd ratio kelompok muda dan pedet adalah 3.424 dan 2.529, artinya, kelompok muda dan pedet memiliki risiko 3.424 dan 2.529 lebih tinggi dibandingkan kerbau dewasa (referensi). Dari penelitian ini dapat disimpulkan, baik prevalensi maupun derajat infeksi parasit saluran pencernaan di Kabupaten Brebes terjadi dalam taraf yang rendah dan dipengaruhi oleh faktor umur ternak.

Abstract

Ovitrap is an alternative mosquito control method that used to reduce the population of Aedes spp, vector of dengue hemorrhagic fever. This study aims to evaluate pyriproxyfen in ovitrap to control Aedes spp through the mechanism of autodissemination by these mosquitoes on a semifield scale. The research was carried out from August to October 2018 in the outdoor environment of Braja Mustika Bogor Hotel. Ovitrap used consist of two types, namely In2Care traps the which contained insecticide of pyriproxyfen, Beauveria bassiana and yeast, (2) the standar ovitraps (without insecticide). In2Care trap (30 pieces) was installed with a distance of 400 m2/trap, while the standard ovitrap (10 pieces) with a distance of 4 m randomly around In2Care trap. Observations were conducted once a week for two months. The parameter observed in In2Care trap was the number of larvae and pupae Aedes spp, which live and dead. The observations on standard ovitrap were the presence of Aedes spp eggs, the number of larvae pupae and adults that eclosed. The results showed that the In2Care traps effectively attracted Aedes spp to laid eggs and breed (86.7-100.0%). The number of larvae observed in In2Care trap ranged from 10-50 larvae (33.380.0%) and all larvae that developed in In2care trap (100%) died after being kept in the laboratory for two months. The results of observations on standard ovitraps showed that there were dead pupae (22.7-80.8%) and adults that failed eclotion (22.6-83.6%) during 2 months of observation. The results showed that there was the effective mechanism of pyriproxyfen autodissemination by Aedes spp from In2Care traps to standard ovitraps when laying their eggs.

Abstract

Meat is one of the livestock products producing from abattoir, which in its implementation must be able to maintain good quality including of cleanliness, health, and halal for consuming by coommunity. This study aims to determine the community’s perception on the existence of abattoirs in the Tahoa Village, Kolaka District. This research is quantitative descriptive using a Likert scale. The samples used in this study ware 34 people. The results of the study showed that of the odor variable has a value of 357 and categorized as in the disturbed category. As well as for, water pollution with a value of 207 was categorized as quite disturbed and also for waste with a value of 233 was categorized as quite disturbed. The total of all three sub-variable values (odor, water pollution and waste) was 797, and it was categorized as moderately disturbed. In conclusion of this study shows that the community’s perception of the existence of abattoirs in the Tahoa Village, Kolaka District, is quite disturbed.

Abstract

Simulium (black flies) are vector of Onchocerciasis in humans and animals. Preimaginal Simulium has most typical breeding habitat in clear water with fast-running water. This study aims to analyze the relationship between distribution of preimaginal Simulium with the water quality and the rivers physical characteristics. The study was conducted on October 2018 until January 2019 in three locations namely Cilember 1 and Cilember 2 (forest areas), and Pamijahan (rural area), Bogor Regency, West Java. Preimaginal Simulium collections, water quality, and rivers physical characteristics measurements were carried twice a month during four months. Identification was carried out under a microscope, and the data was analyzed by canonical correspondence analysis (CCA). The results showed that the distribution of preimaginal Simulium species in the forest areas (Cilember 1 and 2) were more diverse than in the rural area (Pamijahan). Seven species of black flies were found in Cilember 1, four species in Cilember 2, and two species in Pamijahan. The most abundance of black fly species found in Cilember 1 was S. (S.) eximium (43.25%), in Cilember 2 was S. (N.) feuerborni (88.71%), and in Pamijahan was S. (S.) nobile (99.12%). Based on CCA preimaginal Simulium species with high diversity were found in the rivers that have high dissolved oxygen (9.35±0.32 mg/L), low temperature (19.94±0.24ºC), low total dissolved solid (17.45±1.90 ppm), low conductivity (25.48±2.34 ?s), and low concentration of Coliform (0.43×103±0.25 cfu/ mL), and the physical characteristics of the rivers were wide (3.68 m), fast running-water (1.00±0.09 m/s), depth more than 0.1 m, and boulder streambed particles.

Abstract

The feeding system greatly influences the chemical values and blood parameters of livestock. Biochemical determination of serum constituents can provide valuable information about nutritional status and environmental factors that affect animal health. The purpose of this study was to determine the effect of animal feed formation which consisted of four types of rations with multilevel protein energy levels for total cholesterol, triglycerides and other routine blood parameters of the weaning female balinese cattle. The study was carried out by administering a ration formula for three months in 12 weaning female balinese cattle, aged nine months, body weight 101.3-104.3 kg. Randomized Block Design with four treatment was used in this research , e.g. A = PK 12% and ME 2000 kcal/kg; B = PK 13% and ME 2100 kcal/kg; C = PK 14% and ME 2200 kcal/kg; D = PK 15% and ME 2300 kcal/kg. Blood sampling was collected at the end of the experiment followed by examination of total cholesterol, triglycerides, and other routine hematological parameters in the laboratory. Examination of total cholesterol and triglycerides using the enzymatic colorimetry method. Measurement of routine blood parameters such as total erythrocytes, hemoglobin levels, hematocrit values, total leukocytes, neutrophils, eosinophils, basophils, lymphocytes, and monocytes using an automatic hematology analyzer. The results obtained were total cholesterol: 122.0-146.33 mg/dl; triglycerides: 122.0-146.33 mg/dl. Meanwhile, the total erythrocytes are (6.6-7.0) x106 ìl; hemoglobin (10.3-11.3) g/dl; hematocrit (27.6-28.8)%; total leukocytes (7.8-10.3) x 103ìl; neutrophils (20,7-27,6%); eosinophils (0.08-0.3%); basophils (0.3-1.4%); lymphocytes (66.6-72.2%); monocytes (3.65.5%). All results are still in the normal cattle reference range. There were no significant differences (P>0.05) between treatments. It was concluded that the four ration formulas had no effect on total cholesterol, triglycerides, and other routine hematological parameters from the blood of the weaning female Bali cattle.

Abstract

Complex dermatitis is a disorder of the skin caused by the combination of several agents such as fungi, ectoparasites, bacteria, and metabolic diseases with combined clinical symptoms of primary lesions and secondary lesions. This study aims to determine the histopathology of dog skin with complex dermatitis. There were 15 samples of dogs with complex dermatitis, skin biopsy and histopathology. The results showed that the incidence of complex dermatitis in young dogs was 60%, long-haired 67%, male sex 73%, and domestic race 73%. Macroscopic examination found a change in hyperkeratosis. Microscopically observed that infiltration of polymorphonuclear cells, monomorphonuclear cells and macrofag from moderate to severe in the dermis. In hair follicles and epidermis occured akantosis, and keratin proliferation. There were segment fragments from infectious agents such as Demodex canis, Sarcoptes scabiei, and yeast from Malasezia sp. Other changes were observed in the presence of hydrophic degeneration, necrosis, and ulcers. Dogs suffering from complex dermatitis have chronic infections with more than one infectious agent. Disabilities based on histopathological observation of dogs with complex dermatitis with moderate severity of 33%, and severe 67%.

Abstract

Cases of ascariosis due to infection with Ascaris suum are still prevalent in the world, including Indonesia. Apart from causing economic losses, ascariosis can also threaten human health because it is zoonotic. Case studies of ascariosis with typical white spot lesions in the landrace pig liver is described in this paper. Pigs aged 60 days old, sick for 10 days, thin and coughing gathered from the village of Suwung, Denpasar City. After necropsy and post mortem examination were done, tens of A. suum worms were found in the intestine and white spot lesions observed on the livers. Tissues of small intestine, large intestine, liver, lungs, kidney, spleen, urinary vesicles and brain were taken and put into 10% of formalin neutral buffer before they then processed in making histopathological preparations. Histopathological preparation was carried out using paraffin block method and hematoxylin eosin (HE) staining technique. The results of histopathological examination of the small intestine, large intestine, liver and lungs were found to be infiltrated by eosinophil cells. Eosinophil cells infiltration indicates that there are parasites A. suum or the larvae migrant in the tissues. Some of the tissues in livers were also found to have fibrosis, which is suspected that the infection has been chronic. It can be concluded that cases of the landrace pigs were chronically infected by A.suum and accompanied by migration of the larvae to the livers and lungs. More attention is needed to prevent the increase of ascariosis in pigs in order to minimize economic losses and transmission to humans.

Abstract

Primordial germ cells (PGCs) are precursors for gamete cells. The totipotency of PGCs allows them to be used as a model for studying cancer and infertility. The study aimed to examine the characteristics of the mice fetus as a source of PGCs, proliferation rate of PGC and the role of LIF in vitro culture of PGCs. This study used genital ridges from 26 fetuses at 13.5 days post-coital (dpc) to isolate the PGCs. Genital ridges dissociation using 0.1% of trypsin and in vitro culture was carried out using the Dulbecco Modified Eagle Medium (DMEM) and incubated at 37 0C and 5% CO2 atmosphere. The fetus was measured and weighed to determine the normal development of the fetus and continued with the identification of the genital ridges after laparotomy performed under a stereomicroscope. Proliferation rate was measured by calculating Population Doubling Time (PDT), and cell viability was observed after in vitro culture for six days. The effect of adding 1000 IU/ml of leukemia inhibitory factor (LIF) was evaluated from two types of treatment in the medium, 1) DMEM added with 15% of fetal calf serum (FCS) (DMEM + S15%) and 2), DMEM was supplemented with 15% of FCS and 1000 IU/ml LIF (DMEM + S15% + LIF1000 IU/ml). Immunohistochemistry staining was carried out on the third-, sixth- and ninth-day of culture to detect the expression of Oct-4 in the PGCs, then cells were counted. The results showed that the fetus as a source of PGCs had normal development. The fetal sizes were 11 mm, and male and female genital ridges could be distinguished by morphology at the age of 13.5 dpc. The proliferation of PGCs was relatively slow with a 1.3 day PDT value with the viability of around 85%. Culture of PGCs with DMEM + S15% treatment showed the percentage of PGCs that expressing Oct-4 decreased from the third day of culture to the ninth day of culture. The culture of PGCs in DMEM + S15% + LIF 1000 IU / ml treatment showed that the percentage of PGCs that expressed Oct 4 increased on the sixth day of culture and decreased on the ninth day of culture. It can be concluded that the addition of LIF can maintain the number of PGCs until the sixth day of culture. LIF is thought to play a role in the regulation of proliferation of PGCs through receptors of LIF (RLIF) and glicoprotein (gp) 130 receptors.
