Abstract

Usaha untuk meningkatkan produktivitas ternak kambing Boerka di antaranya dengan melakukan inseminasi buatan (IB). Untuk meningkatkan kualitas spermatozoa yang akan digunakan untuk IB maka diberikan hormon prostaglandin F2 alfa (PGF2α). Penelitian ini bertujuan mengetahui pengaruh pemberian PGF2α terhadap peningkatan motilitas spermatozoa kambing Boerka. Dalam penelitian digunakan 3 ekor kambing Boerka yang berumur ±2-3 tahun. Pelaksanaan perlakuan dirancang menggunakan pola latin square 3 x 3 sehingga hewan akan menerima suntikan P1 (1,5 ml NaCl fisiologis), P2 (37,5 μg PGF2α), dan P3 (75 μg PGF2α) dengan interval waktu perlakuan adalah 30 menit sebelum koleksi semen. Sampel semen dikoleksi dengan menggunakan vagina buatan dan diamati warna, konsistensi, volume, konsentrasi, motilitas, viabilitas, dan motilitas spermatozoa. Motilitas spermatozoa diamati setelah 4 jam di dalam refrigerator. Data warna dan konsistensi semen dilaporkan secara deskriptif, sedangkan volume, motilitas semen segar, dan motilitas spermatozoa setelah 4 jam di dalam refrigerator dianalisis dengan analisis varian pola bujur sangkar latin (RSBL) yang dilanjutkan dengan uji Duncan. Hasil pengamatan menunjukkan bahwa warna dan konsistensi semen yang dikoleksi pada semua kelompok perlakuan adalah krem dengan konsistensi kental. Rataan (±SD) volume semen; konsentrasi spermatozoa (106/ml); motilitas semen segar (%); dan motilitas semen setelah penyimpanan pada P1 vs P2 vs P3 masing-masing adalah 0,90±0,4 vs 0,70±0,3 vs 0,90±0,3 ml (P>0,05); 2303,33±327,15 vs 2336,67±332,91 vs 2576,67±261,02 (P>0,05); 84,00±5,1 vs 73,33±11,54 vs 80,00±0,0% (P>0,05); 63,67±4,5 vs 53,33±4,7 vs 66,67±2,2% (P<0,05). Disimpulkan bahwa pemberian 75 μg PGF2α dapat meningkatkan motilitas spermatozoa kambing Boerka setelah penyimpanan dalam regrigerator selama 4 jam.

Abstract

This study was aimed to observe correlation between gestational age and fetal growth of Kacang goat for 60 days after mating with transcutaneous ultrasonography visualization. Three female goats of productive age with normal estrus cycle and one male goat were used in this study. The female goats were synchronized with double injection of prostaglandin F2á (PGF2á) intramuscularly with dosage of 1 mL for each female goat with 11 days’ time interval. Observation of oestrus was conducted using male goat after being synchronized. When the sign of estrus were prominent, the female goats were naturally mated. The result was pregnant positive for one female goat. Detection of early pregnancy was observed on the 24th days after mating, with the result of 6.77 mm length embryo by isoechogenic visualization. On 35th days of pregnancy, fetal could be seen clearly in isoechogenic to hyperechogenic visualization with head diameter and fetal length were 17.4 mm and 36.2 mm, consecutively. The observation also found the placentom. On day 45 of pregnancy, head diameter and fetal length were 21.8 mm and 40.6 mm. Later on day 49 of pregnancy, the size of gestational saccus was 44.1 mm, with head diameter of 25 mm, and average placentom diameter size of 12.4 mm. On day 53 of pregnancy, head diameter was 25.2 mm with fetal length of 63.6 mm and placentom diameter of 15 mm. On day 56 of pregnancy, vertebrae of fetal were observed using hyperechogenic visualization and placentom was measured 17.9 mm in diameter. On day 60 of pregnancy, we observed that the fetal length was 79.8 mm and the organs such as eyes, heart, liver, os costae, and fetal extremities, could be observed clearly. The size of developing fetal and organs would grow along with the addition of gestational age.

Abstract

The aim of this research was to determine the percentage of estrus, performance of estrus, and percentage of pregnancy in local goat which synchronized with seminal vesicles extract. The seminal vesicles used in this research were collected from the waste of Banda Aceh slaughter house. The goats were allotted into 2 groups. Goats in group were injected with 0.5 ml PGF2á intramuscularly, and goats in group II were injected with 5 ml seminal vesicles extract intrauterine. The injection was done twice with the interval of 11 days. The goats which perform estrus sign are mated naturally once in every observation. Pregnancy diagnosing was done using chemical urine method 2 months after mating. The estrus  percentage  and  pregnancy data were analyzed with chi-square  and  the estrus  performance (onset  and  estrus duration) were analyzed  using  student T test. The estrus onset of group I and II were 29.33±4.62 and 24.00±0.00 hours (P0.05). Estrus duration of group I and II were 26.67+4.62 and 20.00+11.97 hours respectively (P0.05). The  estrus percentage  of  group  I and  II did not  show  any significant differences (P0.05), those are 60.0 and 40.0%  respectively,  whereas   the percentage of estrus  from  both  groups were 100.0 %.

Abstract

Penelitian ini bertujuan mengkaji keragaman genetik gen penyandi ND4 pada Tarsius bancanus, T. b. borneanus, T. dianae dan T. spectrum dan untuk penegakan taksonominya. Deoxyribonucleic acid (DNA) diisolasi dari biopsi jaringan masing-masing spesies Tarsius dengan cara diekstraksi untuk digunakan sebagai DNA cetakan dalam proses amplifikasi dengan metode polymerase chain reaction (PCR). Primer yang digunakan dalam penelitian ini didesain untuk mengamplifikasi gen ND4 dan dilanjutkan dengan elektroforesis. Produk PCR hasil amplifikasi yang telah dimurnikan, selanjutnya dipergunakan sebagai DNA cetakan untuk reaksi penentuan runutan nukleotida. Runutan nukleotida gen ND4 hasil pengurutan dilakukan penjajaran berganda dengan primata lain yang diambil dari Genbank menggunakan Clustal W. Selain berdasarkan runutan nukleotida, gen ND4 dianalisis berdasarkan runutan asam amino dari basa-basa yang diterjemahkan mengikuti vertebrate mitochondrial translation code yang ada pada program MEGA versi 4.1. Konstruksi pohon filogenetika menggunakan metode neighbor joining. Hasil penelitian menunjukkan dari 1378 nukleotida ditemukan 119 situs yang bersifat beragam. Jarak genetika berdasarkan nukleotida gen ND4 yang dihitung menggunakan model dua parameter Kimura, terdapat nilai paling kecil 0,6%, nilai terbesar 13%, dan nilai rata-rata sebesar 6,1%. Filogram berdasarkan hasil runutan nukleotida gen ND4 yang menggunakan metode neighbor joining, dapat mengidentifikasi dan membedakan percabangan antar spesies Tarsius.

Abstract

This study aimed to determine protein content of depik (Rasbora tawarensis) fish collected from fish culture in Laut Tawar Lake, Aceh Tengah. Protein content in fish was analyzed using Kjehdal method with two replications. The result showed that average protein content of depik fish was 15.29%. Based on the result, it could be concluded that protein content of fish depik from Laut Tawar Lake is 15.29%.

Abstract

The aim of this research is to know the effect of siamih leaf ethanol extract (Ageratum conyzoides) on the growth of Staphylococcus aureus bacteria. The bacteria which is  used are obtained from the Microbiology Laboratory of the Faculty of Veterinary Medicine of Syiah Kuala University on isolate bacteria of the honey bear wound (Helarctos malayanus). Bacterial identification is done by culturing the isolate on lateral NA media then it is planted on MSA media and  staining Gram, catalase test and hemolysis test. The extraction is done by maceration with 96% ethanol solvent. Extract of Siamih leaf  is done by phytochemical test. The results of pythritic test of Siamih leaf show that Siamih leaf contains alkaloid compounds, steroids, terpenoids, flavonoids and tannins. The extract concentration which is used is 20%, 40%, 60% and 80%. Nergative control by using aquades and positive controls using Amoxicillin with three repetitions. The method used is disc diffusion or Kirby-Bauer method. The results show that Siamih leaf ethanol extract inhibit the growth of Staphylococcus aureus bacteria at concentrations of 20% (0,7 mm), 40% (1,4 mm), 60% (2,2 mm), and 80 % (3,0 mm). Thus it can be concluded that the extract of siamih leaf ethanol has inhibitory power to Staphylococcus aureus bacteria with low category.

Abstract

This research aimed to investigate the inhibition activity of ethanolic extract of binahong leaf (Anredera cordifolia (Ten.) Steenis) on Staphylococcus aureus. Bacteria were obtained from the Microbiology Laboratory of Faculty of Veterinary Medicine Unsyiah, isolated from the honey bear (Helarctos malayanus) wound area. This experiment was conducted with re-identification of the bacteria before treatment. Afterward, the bacteria were treated with ethanolic extract of binahong leaf with a concentration of 20%, 40%, 60%, and 80% by using the disc diffusion (Kirby-Bauer) method. The data were analyzed descriptively by measuring and comparing the inhibition zone among treatments. The results showed that the treatment could inhibit the growth of Staphylococcus aureus, however, the inhibition zone was narrow with a mean diameter of 20% was 6.8 mm, 40% was 7.4 mm, 60% was 8.2 mm, and 80% was 8.1 mm. It can be concluded that ethanolic extract of binahong leaf had comparative inhibition activity toward Staphylococcus aureus.

Abstract

This research aim was to determine the number of erythrocytes and hematocrit value of broilers  that infected  by Eimeria tenella and given neem leaf extract combined with jaloh extract. Sample used in this study is 20 strains Cobb broiler chickens aged 15 days old. Sampled broilers were separated into 5 treatment groups consisting of 4 chickens, each treatment were chicken that infected by oocysts Eimeria tenella and not given other treatments (P1), chicken infected by oocysts Eimeria tenella and given the anti-stress commercial (P2), chicken infected by oocysts Eimeria tenella and given the leaf extract jaloh 1000mg/L (P3), chicken infected with oocysts Eimeria tenella and given extracts of neem leaf 250mg/L (P4), and a chicken infected with oocysts Eimeria tenella and given the leaf extract jaloh 1000mg/L combined with neem leaf extract 250mg/L (P5). Administration of each treatment were dissolved into the chicken water and administered for 10 days in the morning and evening. The design used in the study was completely randomized design with 5 treatments, each treatment consisted of 4 repetitions. Parameters observed were the number of erythrocytes and hematocrit values. Results from this study showed that neem extracts combined with jaloh given for 10 days had no effect on the number of erythrocytes and hematocrit values of broilers that infected by Eimeria tenella

Abstract

ABSTRAK. Penelitian ini bertujuan mengetahui jenis bakteri Gram negatif pada uterus sapi Aceh yang mengalami kawin berulang. Dalam penelitian ini digunakan enam ekor sapi Aceh betina yang terdiri atas tiga ekor sapi fertil (K1) dan tiga ekor yang mengalami kawin berulang (K2, repeat breeding, RB). Pengambilan sampel bakteri dalam uterus sapi dilakukan dengan menggunakan swab steril yang disimpan dalam media nutrient broth (NB) untuk isolasi dan identifikasi jenis bakterinya. Berdasarkan hasil penelitian, baik K1 dan K2, positif terinfeksi bakteri dan diperoleh 16 isolat bakteri di antaranya enam isolat berasal dari K1 dan sepuluh isolat berasal dari K2. Dari 10 isolat bakteri Gram negatif yang ditemukan pada sapi RB terdiri atas Escherichia coli (E. coli) (30,00%), Pseudomonas sp. (30,00%), Enterobacter sp. (20,00%), Klebsiella sp. (20,00%). Disimpulkan bahwa bakteri yang paling banyak menginfeksi uterus sapi RB adalah E. coli dan Pseudomonas sp.(Isolation and identification of gram negative bacteria in uterine Aceh cow with repeat breeding) ABSTRACT. The study determined the type of Gram negative bacteria of the Aceh cattle uterus which has been diagnosed with repeat breeding. The study was conducted on six Aceh cows consists of 3 normal fertile cows (K1) and 3 repeat breeding cows (K2, RB). The bacteria in uterine samples were collected using sterile swab, kept into nutrient broth (NB) for further isolation and identification of bacteria. The result of the study showed that the total of 16 isolates of bacteria was observed in all cows : 6 isolates from group K1 and 10 isolates from group K2. A total of 10 isolates from cattle with repeat breeding consist of Escherichia coli (E. coli) (30.00%), Pseudomonas sp (30.00%), Enterobacter sp (20.0%), and Klebsiella sp (20.0%). It can be concluded that the most bacteria that infect uterine cows with repeat breeding are E. coli and Pseudomonas sp.
