Abstract

Sorghum (Sorghum bicolor L.) is a cereal plant that can adapt to dry area so it has the potential to be developed in Indonesia. Sorghum has many benefits, namely as food, feed and industry. One of the ingredients in sorghum that can be used for industrial materials is lignin which can be used in the manufacture of particleboard and briquettes. In previous studies, selection of lignin content was carried out on 30 genotypes of sorghum and obtained the genotype with the highest lignin content, namely KS (Konawe Selatan). However, the KS genotype has low biomass, so improvements need to be made to increase biomass. Mutation is one way to increase genetic diversity. The purpose of this study was to obtain a sorghum mutant line that had characteristics related to biomass such as plant height, stem diameter and stem weight that were better than the parent. The research was carried out in July – November 2019 at the Citayam Experimental Field. The genetic material used was the M2 mutant lines of 300, 400 and 500 Gy gamma irradiation and 0 Gy control plants. A total of 50 panicles per gray were planted one row per panicle and the agronomic characters were observed. The agronomy observed on the M2 mutant line showed that the population of the 300 Gy mutant line had a wider range of values than the 400 and 500 Gy populations and control. The 300 Gy mutant population also had a higher mean value than the 400 and 500 Gy mutant populations as well as the control on plant height 309.18 cm (control 305.33 cm), stem diameter 24.09 mm (control 19.37 mm), stem weight 860.73 g (control). 507.10 g), panicle length 42.55 cm (control 39.95 cm) and panicle weight 95.52 g (control 64.50 g). Mutations with gamma ray radiation can increase genetic diversity and selection in the M2 mutant lines obtained lines that have plant height, stem diameter and stem weight are better than the control which shows an increase in biomass.

Abstract

Nitrogen-based fertilizer production mainly involves the Haber-Bosch process, which synthesizes Ammonia via a chemical reaction of N2 from the air and H2 under high temperature and pressure conditions in the presence of a catalyst. However, the environmental concern for greenhouse gas emissions and high energy consumption triggers other Nitrogen fixation method development. The non-thermal plasma oxidation of N2 into Nitrate is a promising method since it has lower energy consumption and a more suitable process condition with a better environmental profile. This study tested the designed plasma discharge reactor tube to produce nitrate from air N2 with varying voltage, time, and airflow rate operating conditions. Nitrate concentration was analyzed using a UV-Vis spectrophotometer. The results show that nitrate concentration changes linearly with the applied voltage in a constant airflow rate and length of treatment time. The highest performance of the plasma discharge reactor was achieved on a 6 kV applied voltage with a flow rate of 0.2 L/min and a treatment time of 40 minutes where the obtained nitrate concentration was 7.7 ppm, which indicated the potential plasma discharged nitrate production for green fertilizer application. Further study shall be carried out to enhance the performance.

Abstract

Diabetes is a degenerative disease that can arise due to unhealthy lifestyles. Until now there is no cure for this disease, but this disease can be minimized or prevented by consuming healthy food or functional food. Tea-based functional drinks have long been recognized and developed for their potential as an antidiabetic agent. One of them is by combining it with other ingredients like herbal plants. This study processed a black tea-based functional beverage combined with singkil leaves and stems. The purpose of this study was to determine the inhibitory enzymes and phenol content of black tea products combined with single leaf tea, and stem. Based on the analysis, all samples, both consisting of one composition or from the combination, generally have varying phenol levels. The highest inhibitory ability is possessed by tea samples which are categorized as strong, while the lowest inhibitory value is on black tea and singkil tea (THS2) with an IC50 value of 106,236. Based on the parameters of the observation of phenol levels, samples that have the highest phenol content are black tea (182,586 µg GEA / 100 g sample) while the sample with the lowest phenol content is tea (6,413 µg GEA / 100 g sample). All samples showed the ability to inhibit the alpha-glucosidase enzyme with a range between medium to strong. Phenol content is the only parameter used in this study. The overall sample showed the higher phenol levels have the enzyme inhibitory ability which tends to decrease. There may be a role from other indigenous secondary metabolite compounds that are not yet known from this study apart from compounds that arise due to the effects of the processing, such as. The results of the correlation test analysis showed a strong relationship between phenol contains and enzyme inhibitory properties (R = 0,765). Key Word: black tea, singkil, Premna seratifolia, antidiabetes, phenol

Abstract

The formation of roots (adventive) and shoots as well as a slow dormancy period is an inhibiting factor for the successful implementation of cuttings. Increasing the slow growth of roots and shoots and accelerating plant dormancy is an effort to overcome this. The purpose of this study was to determine the growth response of grape stem cuttings (Vitis vinifera L.) to the immersion time on several types of PGR. This research was carried out for 3 months from January to April 2021 at the yard of Jurangsapi Village, Tapen District, Bondowoso Regency, East Java at an altitude of 226 meters above sea level. This study used a factorial randomized block design (RAK) with the first factor being the type of PGR (Z), with 3 levels, namely: Z1: Rootone-F 300 mg/liter of water, Z2: onion extract 800 ml/liter of water, Z3 : extract of bean sprouts 300 ml/liter of water, and the second factor is the long immersion (L) for 3 levels, namely: L1: 0.25 hours, L2: 6 hours, L3: 12 hours. The results showed that a single factor of PGR had an effect on increasing shoot length (8,10, and 12 WAP), shoot diameter (6 WAP), and number of leaves (10 and 12 WAP) of grape plants with the best PGR of Rootone-F. The long immersion treatment had a significant effect on the percentage of success in planting grape cuttings with the best soaking time of 6 hours. While the interaction between PGR type treatment and long immersion significantly affected shoot length (6,8,10, and 12 WAP), number of leaves (4, 8,10, and 12 WAP), and canopy wet weight of grape cuttings with the best treatment on PGR Rootone-F and 0.25 hours of long immersion. Keywords: auxins; long immersion; stem cuttings; Vitis vinifera L. var. prabu bestari.

Abstract

The aim of this study is to identify the arrowroot morphology characters and its distribution pattern in Jember District, East Java Province. Plant inventory was carried out in 6 sub-districts, i.e. Arjasa, Sumbersari, Silo, Tanggul, Semboro, and Wuluhan from July to October 2021 using survey method. The sampling technique was done by accidental sampling. The results of dendogram analysis based on morphological characters showed that Sumbersari, Wuluhan, and Arjasa were sub-district areas with a lot of arrowroots, if compared to other 3 sub-districts. Three main groups were formed based on the similarity of arrowroot morphological characters, accessions from geographically adjacent sub-district or village had greater morphological similarities. Leaf length has a close relationship with leaf width, leaf sheath length, and leaf petiole diameter. The distance between stem segments has a correlation with stem height. The longest fibrous roots and stem diameter contributed less to arrowroot diversity. The number of internodes positively correlated with tuber circumference, tuber length, and tuber weight. Arrowroot plant accessions grew in groups according to certain morphological characteristics, which was also showed a close relationship between these accessions. Keywords: accession, arrowroot, genetic relationship, morphological characteristic
