Abstract

Tuberculosis (TB) remains a significant and deadly infection among pulmonary diseases caused by Mycobacterium tuberculosis, a highly adaptive bacterium. The ability of M. tuberculosis to evade certain drugs has been linked to its unique structure, particularly in the cell envelope, where the Ag85 complex proteins play an essential role in this part.  The aim of this study was to utilize a drug repurposing strategy targeting the Ag85 complex proteins. This study utilized a computational approach with 120 selected drugs experimentally identified to inhibit Tuberculosis. A virtual screening molecular docking with Autodock Vina was used to filter the compounds and identify the strong binders to the Ag85 Complex. Molecular dynamics simulations employed the Gromacs Packages to evaluate the stability of each complex, including root mean square deviation (RMSD), root mean square fluctuation (RMSF), and radius of gyration (RoG). Additionally, absorption, distribution, metabolism, excretion, and toxicity (ADMET) assessments were conducted to gather more information about the drug-likeness of each hit compound. Three compounds, selamectin, imatinib, and eltrombopag were selected as potential drugs repurposed to inhibit the activity of the Ag85 complex enzyme, with binding affinities ranging between -10.560 kcal/mol and -11.422 kcal/mol. The MD simulation within 100 ns (3 replicas) showed that the average RMSD of each Ag85A complex was 0.15 nm–0.16 nm, RMSF was 0.09 nm–0.10 nm, and RoG was 1.80 nm–1.81 nm. For Ag85B, the average RMSD was 1.79 nm–1.80 nm, RMSF was 0.08 nm–0.09 nm, and RoG was 1.79 nm – 1.80 nm. Then, for Ag85C, the mean RMSD was 0.16 nm–0.18 nm, RMSF was 0.09, and RoG was 1.77 nm. The study highlights that these promising results demonstrate the potential of some repurposed drugs in combating the Ag85 complex.

Abstract

BACKGROUND: Acute ischemic stroke (AIS) is usually treated with thrombolysis, however the percentage of patients receiving this therapy is not quite low. Therefore, it is necessary to find alternative therapy using neuroprotective agent such as Moleac (MLC) 901. Heat shock proteins (HSP)-70 and matrix metalloproteinase (MMP)-9 are usually related to AIS due to the triggered stroke-induced physiological stress. However, the effect of MLC 901 on Hsp70 mRNA expression, HSP-70 and MMP-9 remains unclear. This study was conducted to determine the effect of MLC 901 on those three parameters in relation to cerebral infarction volume and functional outcomes in an AIS model.METHODS: Rats were induced with AIS using unilateral common carotid artery occlusion (UCAO) and received three different treatments: 43.2 mg/200 gBW MLC 901, 21.6 mg/200 gBW MLC 901, and sodium carboxymethyl cellulose (CMC-Na), that were administered orally for 14 days. HSP-70 and MMP-9 protein levels were assessed using enzyme-linked immunosorbent assay (ELISA), and Hsp70 mRNA expression was assessed using quantitative real-time polymerase chain reaction (qRT-PCR). Foot fault scores for evaluation functional outcome and infarction volume were assessed by ImageJ.RESULTS: AIS-induction increased HSP-70, MMP-9, and Hsp70 mRNA expression within 24-48 h. MMP-9, HSP-70 , and Hsp70 mRNA expression were reduced by MLC 901. MLC 901 at dose of 43.2 mg/200 gBW and 21.6 mg/200 gBW were effective in reducing these levels compared to the control. MLC 901 improved functional outcomes and decreased cerebral infarction volume. Moreover, a dosage of 43.2 mg/200 gBW was more effective in reducing Hsp70 mRNA expression and HSP-70, improving functional outcomes, and reducing cerebral infarction volume than a dosage of 21.6 mg/200 gBW, but not MMP-9 protein.CONCLUSION: MLC 901 effectively decreased Hsp70 mRNA expression, HSP-70 and MMP-9 protein levels, infarct volume, and functional outcomes. MLC 901 could be a potential therapeutic agent for AIS.KEYWORDS: MLC 901, HSP-70, MMP-9, acute ischemic stroke

Abstract

Counter electrodes are essential in dye-sensitized solar cells (DSSCs) for facilitating charge transfer and catalyzing the regeneration of the electrolyte, impacting overall efficiency. Common counter electrode materials include platinum (Pt), poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS), and graphene, each with distinct advantages and challenges. Pt, a traditional choice, offers excellent catalytic activity but is expensive and scarce. PEDOT:PSS, a conductive polymer, is cost-effective and easily deposited but often suffers from high recombination losses and lower efficiency. Graphene, known for its high conductivity and large surface area, is emerging as a promising alternative. However, a lack of comparative studies on how different counter electrode materials influence recombination dynamics limits the understanding needed for optimizing DSSC performance. This study addresses this gap by examining Pt, graphene, and PEDOT:PSS -based counter electrodes, analyzing their effects on charge transfer, recombination behaviour, and efficiency through J-V measurements, charge extraction, and transient photocurrent (TPC) as well as transient photovoltage (TPV) analyses. Graphene-based DSSCs show superior performance, achieving the highest photocurrent density and power conversion efficiency up to 5.12% at an intensity equivalent to 1 sun (100 mWcm-2), due to enhanced charge extraction and minimized recombination. TPC data reveal that graphene supports faster charge transport, while TPV analysis shows longer electron lifetimes than PEDOT:PSS-based DSSCs. In contrast, PEDOT:PSS-based DSSCs exhibit high recombination losses, lower photocurrent, and s-shaped J-V curves, indicating high resistance of limited charge transfer efficiency. These findings highlight graphene’s potential as an optimal counter electrode material for efficient, high-performance DSSCs.

Abstract

The increasingly massive spread of Wahhabi ideology in Indonesia has given rise to reactive reactions among society, this is because Wahhabi preaching emphasizes the labeling of Bid'ah and Infidels towards other communities who have different understandings, and this has implications for the many horizontal conflicts that occur during public. Even though Indonesian society is known as a harmonious society amidst diversity and religion. This research is descriptive research with a qualitative approach, while the data collection technique used is a literature study. Then the data collected through the study of the text was compiled, then analyzed using the descriptive analysis method with a deductive mindset. The results of this research are that the Wahhabi ideology pioneered by Muhammad bin Abdul Wahab is an understanding that is oriented towards purifying the creed, which at that time was considered to have experienced many setbacks along with the development of the tarekat's teachings. However, the movement for this understanding is often accompanied by doctrines of heresy, syrk, and infidelity which create polarization in society. In this position, the state constitutionally regulates that every society has the right to believe in beliefs and express thoughts and attitudes following their conscience, but if these beliefs and statements of belief are carried out by causing divisions in society, then this is where the role of the State is needed to ensure peace. and the harmony that is the identity of the Indonesian nation is maintained, meaning that there needs to be a firm stance from the government regarding the spread of Wahhabi ideology, and the Perppu is an alternative as well as a legal instrument that is very possible for the Government to implement to stop the spread of Wahhabi ideology in Indonesia and at the same time reflects this attitude. The government is firm against anyone who destroys the diversity of Indonesian society Keywords: Wahabi, Labeling, Horizontal Conflict
