Abstract

Radikal bebas merupakan suatu atom atau molekul yang tidak mempunyai pasangan elektron dan dapat merusakmolekul-molekul penting untuk fungsi seluler. Pada kondisi stres oksidatif, radikal bebas akan menyebabkan terjadinyaperoksidasi lipid membran sel dan merusak organisasi membran sel. Pemberian asupan antioksidan berupa vitamin Edapat menurunkan efek radikal bebas dalam tubuh. Tujuan penelitian ini adalah untuk melihat apakah ada pengaruhpemberian vitamin E terhadap kadar estrogen dan stuktur tulang alveolar mencit akibat radikal bebas. Mencit (Musmusculus L.) betina dibagi dalam 6 kelompok, tiap kelompok terdiri atas 5 ekor, P0= tidak diberi perlakuan (kelompokkontrol); P1= latihan fisik maksimal setiap hari selama 30 hari; P2= vitamin E selama 30 hari; P3= latihan fisik maksimalselama 15 hari, selanjutnya 15 hari lagi vitamin E; P4= vitamin E selama 15 hari, selanjutnya 15 hari lagi latihan fisikmaksimal; P5= latihan fisik maksimal dan vitamin E selama 30 hari. Pada akhir perlakuan sesuai dengan kelompok,dilakukan pemeriksaan terhadap kadar estrogen dan terhadap stuktur tulang alveolar mencit. Hasil penelitian inimenunjukkan vitamin E berpengaruh terhadap kadar hormon estrogen mencit (Mus musculus L.) betina dewasa yangmelakukan latihan fisik maksimal secara bermakna (p<0,05), dan vitamin E juga berpengaruh terhadap stuktur tulangalveolar mencit (Mus musculus L.) betina dewasa yang melakukan latihan fisik maksimal secara bermakna (p<0,05).Sebagai kesimpulan, vitamin E berpengaruh terhadap kadar hormon estrogen dan stuktur tulang alveolar mencit (Musmusculus L.) betina dewasa yang melakukan latihan fisik maksimal secara bermakna (p<0,05).

Abstract

Ethanolic leaf extract of Saurauia vulcani Korthor pirdot in Indonesia was evaluated to improve the number of erythrocytes in laboratory rats after being immunized with sheep red blood cells (SRBC). This study was conducted in an experimental manner using a non-factorial Completely Randomized Design (CRD) with four treatments and six replications. The tested animals were 24 rats (Rattus norvegicus) maintained in the laboratory. The experimental designs were designated as follows: G0as a control group treatedwith regular animal feed,G1 was treatedwith 0.1 mLSRBC, G2was treated with 500 mg ethanolic leaf extract of Saurauia (EES)/kg body weight (BW), G3 was treated with 500 EES/kg BW and 0.1 mL SRBC. The immunological effect was monitored by analyzing the level of interleukin-2 (IL-2) using ELISA, lymphocytes using ABX Microx 60, and liver histopathology. The results showed that lymphocyte levels was increased in G2 and G3 but not significant to G0. The IL-2 level in G1 was recorded the highest among groups due to the treatment using SRBC. The percentage of necrotic cells in G1was 5.8%, followed by G3 with 2.3%. The morphologies of liver histology were normal, parenchymatous and hydropic degenerations, and necrosis. The phytochemical constituents in ethanolic leaf extract of S. vulcanipossessed an immunological effect through the increased lymphocytes, reduction of IL-2, and protection through normal liver histopathology in rats.

Abstract

This study aimed to determine film characteristics of chitosan/collagen scaffold for tissue engineering applications. Scaffold prepared using freeze drying method. Surface structure and biological testing chitosan/collagen scaffold crosslinking reagent addition Glutaraldehide studied using Scanning Electron Microscopy test (SEM) and Microscope inverted. Variations in the ratio of chitosan/collagen (10:0, 8:2, 7:3, 6:4, 5:5 4:6, 3:7, 2:8, 0:10), and treated with a crosslinking reagent 0.25% of Glutaraldehyde (GA) of the total weight of the polymer. The next process dissolving and mixing, followed by printing in glass moulds (7,5x7,5) with a thickness of 5 mm. This was followed by the freezing and drying with a freeze dryer. Scaffold chitosan/collagen ratio of 80:20 and a concentration of 0.25% GA showed growth of human skin fibroblast cells most and nonporous surface structure. This study is part of a study of the processing of chitosan/collagen scaffold for applications in tissue engineering

Abstract

A research was conducted to invenstigate antihyperglycemic activity ethanol extract of leaf kembang bulan (Tithonia diversifolia) in mice with alloxan induced diabetes. The experiment used completely randomized design, 6 treatment groups with 5 replicates each. KP mice, P1, P2, P3 and P4 fasted for 16 hours and then injected intraperitoneally alloxan 5.04 mg. On day 4 post induction, KP, P1, P2, P3 and P4 positive diabetes. P1, P2 and P3 were treated 14; 42 and 70 mg/Kg of ethanol extract of kembang bulan while P4 were 0.9 mg/Kg glibenclamide for 12 days. The results showed that all treatment doses did not significantly affect body weight of diabetic mice (P>0.05). P1 obtained with a dose of 14 mg/Kg body weight is able to decrease blood glucose levels (P<0,05). The ethanol extract at a dose of 14 mg/Kg body weight effect on decreasing the number of giant cells of the spleen. It can be concluded ethanol extract of kembang bulan can lower blood glucose levels  and effect  the  microstructure spleen of mice  with alloxan induced diabetes. Abstract

Tujuan dari Penelitian ini adalah mengetahui pengaruh penambahan kuning telur bebek dalam pengencer two-steptm extender terhadap motilitas, viabilitas dan morfologi spermatozoa sapi limousin. Penelitian menggunakan eksperimen laboratorium menggunakan pola rangcangan acak lengkap (RAL) dengan 5 kali ulangan dari 5 ejakulasi dari sapi yang sama. Spermatozoa disimpan selama sepuluh hari. Motilitas spermatozoa diamati dengan mikroskop cahaya (400x) pada suhu 37°C. Viabiltas spermatozoa diamati dengan pewarnaan eosin menggunakan mikroskop cahaya (400x), morfologi spermatozoa diamati dengan pewarnaan metilen blue menggunakan mikroskop cahaya (400x). Hasil penelitian menunjukkan suplementasi kuning telur bebek pada pengencer two-steptm extender berpengaruh terhadap motilitas dan viabilitas, morfologi spermatozoa sapi limousin selama penyimpanan pada suhu 4-5°C. Konsentrasi kuning telur terbaik adalah 15% dalam mempertahankan motilitas (41,00%±2,24), viabilitas (74,44±4,77%) dan morfologi (69,91±1,98%) spermatozoa sapi Limousin setelah penyimpanan empat hari pada suhu 4-5°c.   Kata kunci: two-steptm extender, kuning telur, motilitas, viabilitas, morfologi spermatozoa sapi Limousin .

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh jenis media tumbuh aklimatisasi dan jenis pupuk daun terhadap pertumbuhan pada proses aklimitasiasi anggrek Dendrobium sp. Penelitian ini dilaksanakan di Laboratorium Pemuliaan Tanaman, Fakultas Pertanian, Universitas Sumatera Utara, Medan yang dimulai dari bulan Januari sampai dengan Maret 2009. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan dua faktor. Faktor pertama adalah jenis media tumbuh aklimatisasi (arang sekam, cocopeat, dan campuran antara arang sekam dan cocopeat). Faktor yang kedua adalah jenis pupuk daun (pupuk majemuk foliar I dan pupuk majemuk foliar II). Parameter yang diamati adalah persentase hidup, pertambahan tinggi planlet, pertambahan diameter batang, pertambahan jumlah daun, pertambahan berat total, saat muncul tunas, jumlah tunas, berat akar, volume akar dan jumlah klorofil daun. Hasil penelitian menunjukkan bahwa interaksi jenis media tumbuh aklimatisasi dengan jenis pupuk daun berpengaruh nyata terhadap parameter saat muncul tunas dan jumlah tunas. Abstract

Liver histological features of DDW mice (Mus musculus L.) treated with n-hexane extract of Andaliman (Zanthoxylum acanthopodium DC.) fruit during pre- and post-implantation has been studied in Laboratory of Animal Structure, Department of Biology, Faculty of Mathematics and Natural Science, University of Sumatera Utara, Medan. Experiment used completely randomized design (CRD) consisted of 2 periods of treatment: pre-implantation (day 0-3; group A) and post-implantation (day 6-14; group B). For each group,  mice were treated daily by oral gavage with 0, 1, 2, 4, or 6 % of andaliman extract in 0.3 ml of 1% CMC. The result showed, liver in treatment groups was pale and showed mottled surface texture. Hepatocyte damage in post-implantation group was higher than in pre implantation group (p> 0.05). We concluded that n-hexane extract of andaliman fruit appears to negatively affect liver histology of mice. Abstract

The effects of electric cigarette exposure on lung tissue of mice were studied. The experiments using completely randomized design (CRD) with 3 treatments, i.e.  treatment of electric cigarette with strawberry flavour, tobacco flavour, and control treatment that was not exposed by cigarette smoke. Eight male mice were used for each group. Exposure treatment was done every day for 2 consecutive weeks with dosage 20 times suction using syringe of 60 mL. After receiving treatment for 2 weeks, the mice were killed by cervical dislocation. The lungs were isolated and prepared histologically using paraffin method and then stained with by haematoxylin and eosin  (HE). The results showed that, smoke treatment mice lose weight significantly (p< 0.05). In gross anatomy, electric cigarrete smoke affect lung feature, but histological observation on  membrane damage, damage to the lumen and the relationship between alveoli the smoke effect were statistically unsignificant (p>0.05).. In summary short period exposure to electric  cigarette smoke  tends affect lung damage in anatomy, but statistically has no effect of the damage to the lung tissue.   Keywords: alveolar lumen, alveolar membrane, electric cigarettes

Abstract

This study aimed to determine the effects of vitamin C and E on liver microstructure of mouse exposed to monosodium glutamate (MSG) for 30 days. The experiment used completely randomized design which consists of 6 treatments and  5 replications. Mice were treated daily by oral gavage with 4 mg MSG (P1),  4 mg MSG + 0.26 mg  vitamin C (P2),  4 mg MSG + 0.026 vitamin E (P3), or  4 mg  MSG + 0.26 mg  vitamin C + 0.026 mg vitamin E (P4). All dosages were based on mg/g body weight. Two control groups K- and K+ were treated with water and castor oil respectively. The mice liver tissue slide was prepared by the paraffin method and stained with haematoxylin-eosin (HE). The results showed that MSG caused cell degeneration and necrosis in liver tissue. Vitamin C and E was  able to recover  liver cell damage induced by MSG.  The highest result was found in P3 and combination treatment (P4) where the recovery effect reaches 64% and 80% respectively. Abstract

The aim of this research is to study the effect of vitamin C and E on kidney microstructure of mouse exposed to monosodium glutamate for 30 days. This research used complete randomized design which consisted of 6 treatments and 5 replications. The control consisted of  K-, and K+ (castor oil 0.3 ml). The treatments were MSG 4 mg/g BW, P1 (only MSG), P2 (MSG plus vitamin C 0.26 mg/g BW), P3 (MSG plus vitamin E 0.026 mg/g BW) and P4 (MSG plus vitamin C and E). After treatments were completed, the mice were sacrificed by cervical dislocation, and kidney tissue section was prepared by  the paraffin method and  stained with Haematoxylin and Eosin staining. Microstructure damage of kidneys showed a significant difference between control and treatments. In P1, the proximal tubule damage was 69.95%±5 and in P4 was 47.22%±3. In summary MSG can cause damage in the form of a narrowing of the lumen of the proximal tubules of the kidney. A combination of vitamins C and E can reduce the effect of MSG on the kidney.   Keywords: kidney, Monosodium glutamat (MSG), Vit C, Vit E.
