Abstract

Songi (Dillenia serrata) is a tree endemic to Southeast Sulawesi and its stem bark been used in folk medicine. Nonetheless, only a handful scientific knowledge regarding chemistry and biological activities has been investigated on the plant. The present study aimed to investigate the antioxidant and xanthine oxidase (XO) inhibitory activity of the stem bark of the plant. Methanol extract and organic fractions (petroleum ether, ethyl acetate, and methanol) of the dried powdered stem bark of songi were evaluated for phytochemical screening, total phenolic (TPC), total flavonoid contents (TFC), DPPH, FRAP, and XO in vitro assays. Flavonoids, tannins, terpenoids, steroids, and saponins present in the extract. TPC and TFC in extract were 59.2 mg GAE/g and 23.4 mg QE/g. The contents in organic fractions were in solvent-dependent manner (methanol>ethyl acetate>petroleum ether). Extract and fractions scavenged DPPH radicals (48.2–59.7%) at 100 μg/mL compared to ascorbic acid, trolox, and gallic acid (90.3–93.8%). FRAP values varied from 0.8–3.4 μg/μg equivalent trolox amount (quercetin and gallic acid were 25.7 dan 32.4 μg/μg, respectively). They also inhibited xanthine oxidase (15.3–50.3%) at 100 μg/mL (allopurinol, 98.2%). The study concluded the potential of methanol extract and organic fraction of the stem bark of songi, thus highlighted the prospect of songi to be used in herbal and drugs development from nature.

Abstract

Olae (Etlingera calophrys), an endemic of Southeast Sulawesi, Indonesia, is traditionally used as a culinary spice and  medicine, yet remains scientifically underexplored. This study investigated the phytochemical composition, antioxidant activity, and acute toxicity of infusion extract from the seeds and fruit pericarp. Phytochemical screening was performed using specific color reagents, antioxidant activity was evaluated with the 2.2-diphenyl-1-picrylhydrazyl (DPPH) method, and acute toxicity was assessed by the Brine Shrimp Lethality Test (BSLT) against Artemia salina. The seed infusion contained alkaloids and phenolics, while the pericarp infusion contained tannins and phenolics. Antioxidant testing revealed very strong radical scavenging activity, with SC₅₀ values of 0.05 g/100 mL (seeds) and 0.04 g/100 mL (pericarp). Toxicity testing indicated no significant toxicity, with LC₅₀ values of 1740 g/100 mL (seeds) and 2510 g/100 mL (pericarp). These findings suggest that infusion extract of E. calophrys possess strong antioxidant potential and are safe for use, supporting their prospective application as natural antioxidant sources.

Abstract

Susube (Meistera aculeata (Roxb.) Škorničk. & M.F.Newman) is one of the species of the zingiberaceae family that grows in Southeast Sulawesi, especially in Konawe Regency. The fruit of this plant is often consumed and used as a cooking spice by the native. Meanwhile, the compounds content and bioactivity of the rhizome and fruit which is a characteristic of the Zingiberaceae family, has never been reported. Therefore, this study aimed to identify compounds contained in the rhizome along with their antioxidant and cytotoxic properties. The research method used is experimental research, starting with qualitative phytochemical screening in ethanol exctract and then continued with antioxidant (1,1-diphenyl-2-picrylhydrazyl or DPPH) and cytotoxicity assay (Brine Shrimp Lethality Test or BSLT). The results of the phytochemical screening indicate that the ethanol extract of the Susube rhizome and fruit contains alkaloids, flavonoids, tannins, terpenoids, and saponins. Antioxidant activity was determined based on the ability to reduce DPPH radicals and was calculated by determining the IC50 value, while toxicity was assessed based on shrimp larvae mortality and calculated by determining the LC50 value. The study results show that the IC50 values for the ethanol extract of Susube rhizome, fruit and the positive control ascorbic acid are 4.9 µg/mL, 4,5 µg/mL and 2.2 µg/mL, respectively. Meanwhile, the LC50 values for the ethanol extract of Susube rhizome, fruit, and the positive control potassium bichromate are 578 µg/mL, 958 µg/mL, and 2.8 µg/mL, respectively. Based on these results, it can be concluded that the ethanol extract of Susube rhizome and fruit (Meistera aculeata (Roxb.) Škorničk. & M.F.Newman) exhibits toxic effects on shrimp larvae (≤1,000 mg/L). Additionally, its ethanol extract shows very strong antioxidant effects in scavenging DPPH free radicals. Therefore, this study suggests that the Susube rhizome and fruit extract warrants further investigation as a promising candidate for treatment or prevention of diseases, supporting the potential of Southeast Sulawesi's local natural resources for phytopharmaceutical or herbal medicine

Abstract

Artocarpus elasticus belongs to Moraceae; a family best known for their fruits that support human life. Along with its nutritive components, different parts of A. elasticus are traditionally used to treat fever, hypertension, digestive problems, infections, and female fertility. However, scientific data of this plant, especially its leaves, are still limited, requiring further research. The present study investigated secondary metabolites, antioxidant, and acute toxicity of its leaves using methods on ethanolic maceration, phytochemical screening, determination of phenolics and flavonoids content, qualitative and quantitative DPPH radical scavenging activity, and brine shrimp lethality test. The yield of ethanolic extract of A. elasticus leaves was 6.52 gram extract per 100 gram of dried leaves. The study found the presence of alkaloids, flavonoids, tannins, terpenoids, steroids, and saponins in the extract. The TPC (28.29 ± 1.46 mg GAE/g of extract) and TFC (7.72 ± 0.34 mg QE/g of extract) found high in diluted extract at 10 and 100 µg/mL, respectively. The extract showed moderate free radical scavenging activity with an SC50 value of 8.04 µg/mL and an AAI of 0.99. The extract also displayed toxicity towards A. salina nauplii with an LC50 of 116.24 µg/mL. The study concludes that leaves of A. elasticus could be further explored for antioxidant and cytotoxic agents.
