Abstract

Penelitian ini bertujuan mengetahui keberadaan antibodi dari bakteri brucella pada sapi perah di Kecamatan Purwoharjo Kabupaten Banyuwangi menggunakan Rose Bengal Test (RBT). Metode yang digunakan dalam penelitian ini adalah metode analisis diskriptif dengan pendekatan kuantitatif, yaitu metode yang menggambarkan keberadaan antibodi Brucella pada sapi perah dengan cepat. Informasi pendukung diperoleh dari survei peternak dengan teknik wawancara. Sampel ditentukan dengan rumus slovin dengan ukuran sampel sebanyak 75 sampel dari 294 ekor sapi pada populasi sapi perah di Kecamatan Purwoharjo. Hasil penelitian menunjukan 2 sampel dinyatakan positif Brucella dari 75 sampel. Hasil penelitian ini dapat disimpulkan bahwa reaktor Brucella ditemukan pada sapi perah di Kecamatan Purwoharjo Kabupaten Banyuwangi yang berpotensi menyebabkan penyakit Brucellosis. Harapan dari penelitian ini adalah data yang diperoleh dapat diuji ulang dan diuji lebih lanjut dalam penelitian selanjutnya sehingga hasil dapat dikonfirmasi.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui potensi ekstrak metanol daun Afrika (Vernonia amygdalina Delile) sebagai antibakteri terhadap bakteri Escherichia coli ATCC 25922. Isolat E. coli dibiakan pada media Muller Hinton Agar (MHA). Uji antibakteri menggunakan metode difusi sumuran. Kontrol negatif (K-) pada lubang sumuran hanya diberikan pengencer ekstrak CMC Na 0,5%. Kontrol positif (K+) pada lubang sumuran diberi dengan eugenol. Perlakuan (P1, P2, P3, P4) diberikan ekstrak metanol daun Afrika (Vernonia amygdalina Delile) dengan masing - masing konsentrasi 100%, 75%, 50% dan 25%. Rancangan penelitian menggunakan metode Rancangan Acak Lengkap (RAL), dengan enam perlakuan dan empat pengulangan. Data dianalisis dengan ANOVA, dilanjutkan dengan uji Duncan. Hasil analisis zona hambat menunjukkan, K+ memiliki perbedaan yang nyata dengan semua perlakuan K-, P1, P2, P3, dan P4. Serta K- ditemukan tidak berbeda nyata dengan perlakuan P1, P2, P3 dan P4. Hasil penelitian menunjukkan bahwa setiap perlakuan tidak menghasilkan zona hambat sehingga tidak berbeda nyata dengan kontrol negatif (CMC Na 0,5%).

Abstract

Oksitetrasiklin diketahui sebagai salah satu antibiotik yang sering digunakan dalam menangani kasus mastitis pada hewan ternak. Penelitian ini merupakan penelitian deskriptif untuk mengetahui ada atau tidaknya residu oksitetrasiklin pada susu kambing Peranakan Etawah (PE) di Kelurahan Kalipuro, Banyuwangi. Sampel susu di koleksi kemudian dilakukan pengujian residu antibiotik dengan metode uji bioassay menggunakan bakteri standar Bacillus cereus ATCC 11778. Penelitian yang dilakukan pada 26 sampel susu yang di ambil secara acak berasal dari ternak kambing perah PE di dapatkan hasil yang negatif. Hal ini di tunjukkan dengan tidak terbentuknya zona hambat di sekitar well yang diisi sampel susu. Berbeda dengan kontrol postif oksitetrasiklin dengan konsentrasi 1 µg/ml yang menghasilkan zona hambatan sekitar 18 mm. Dapat disimpulkan bahwa 100% dari 26 jumlah sampel susu kambing PE yang di ambil secara acak dan di periksa dari Kelurahan Kalipuro, Banyuwangi tidak mengandung residu antibiotik oksitetrasiklin.

Abstract

The aim of this research was to determine the level of stability of Progesterone-retention time on Eluen Phase Mobile (EPM) solution in storage period for six, eight and 12 hours using High Performance Liquid Chromatography (HPLC). The research method used posttest-only control group design by using three treatments and six repetitions. The three repetitions consist into six hours, eight hours and 12 hours. The data were obtained analyzed by Summery Independent T-Test with SPSS 24 for windows. The results showed that the progesterone treatment in EPM solvents with six-hour storage (P1) and eight hours (P2) retention time progesterone are stable but the progesterone treatment in EPM solvent with 12-hour storage (P3) showed an unstable due to storage time. Besides that there are irrelevant peak which show that many pure molecular fractions are separated in the stationary phase so that the molecular structure of the substance begins to break down. Based on these results, it can be concluded that the storage time of Progesterone in Eluen Phase Mobile began to be unstable at 12 hours of storage.

Abstract

The problem on food health is the high amount of bacterial contamination of food served by food industries. The quality of beef can be affected by the level of bacterial contamination in meat. Coliform is a group of bacteria used as a sanitary hygiene indicator. The purpose of this study is to measure the value of MPN Coliform in beef at Krian's abattoir of district Sidoarjo. Thirty samples were taken from the abattoir of Krian, Sidoarjo. Most Probable Number was used to determine the level of Coliform's contaminate beef. The results showed that all beef samples at abattoir were contaminated with Coliform. The average contaminant levels was 706,76 MPN/g. Twenty six samples exceed the maximum limit of Coliform bacteria contamination in accordance with Indonesian National Standard (SNI). The highest Coliform MPN value is ≥ 1600 MPN / g, whereas the lowest MPN Coliform value is 8 MPN/g.

Abstract

The purpose of this study was to determine how Staphylococcus aureus responded to Kembang Bulan (Tithonia diversifolia) leaf extract's antibacterial properties. Eight S. aureus bacterial isolates from three Surabaya dairy farms were used in this study. Tithonia diversifolia leaf extract at concentrations of 10%, 5%, 2.5%, 1.25%, and 0% were employed in the study's samples. The antibacterial activity of Tithonia diversifolia leaf extract was assessed in this work using the disk diffusion method and an inhibition zone estimated in millimeters. Based on the findings of this study, it was determined that Tithonia diversifolia leaf extract at 5% and 10% concentrations had 100% antibacterial activity against S. aureus in eight samples. As with S. aureus, Tithonia diversifolia leaf extract at concentrations of 0%, 1.25%, and 2.5% did not exhibit any antibacterial action.

Abstract

This study was aimed to determine the effect of urea in maturation medium on in vitro oocyte maturation rate. The medium used was TCM-199 added with Hepes, NaHCO3, Kanamycin 0.15 IU/mL, PMSG, 0.15 IU/mL hCG, and 10% FBS. Cumulus oocyte complexes (COCs) of cows derived from follicle aspiration were divided into three groups. In control group (P0), the COCs were matured in vitro in a maturation medium without urea addition, meanwhile in the P1 and P2 groups, the medium was added with urea 20 and 40 mg/dL, respectively. Each petri dish contained three drops of maturation medium (300 µl/drops) according to the groups. Microdrops were coated with mineral oil and then incubated in a 5% CO2 incubator, at 39 ËšC with maximum humidity. Aceto-orcein staining was conducted to evaluate the maturation of oocytes based on the achievement of metaphase II phase that is indicated by the presence of metaphase plate and/or first polar body. The result showed that the oocyte maturation rates of P0, P1, and P2 were 51.25, 52.43 (p >0.05), and 46.88 % (p <0.05) respectively. It could be concluded that the presence of urea at 40 mg/dL in maturation medium reduced the percentage of bovine oocyte maturation in vitro.
