Abstract

Plasmodium  resisten terhadap artemisinin menjadi salah satu permasalahan kesehatan di dunia karena belum ada obat baru pengganti artemisinin. Resistensi P. falciparum terhadap obat antimalaria  artemisinin dapat terjadi karena dipengaruhi oleh faktor internal dari P. falciparum, antara lain induksi ekspresi gen yang mengekspresikan protein. Salah satu gen tersebut adalah gen Triptophan-rich Protein (PArt). Fungsi Triptophan-rich Protein penting dalam membrane-spanning protein dan berperan dalam folding protein untuk menjaga kontak hidrofobik. Penelitian ini bertujuan membuktikan overekspresi gen Triptophan-rich Protein P. falciparum galur Papua 2300 yang disebabkan oleh paparan artemisinin berulang in vitro. Waktu penelitian dilaksanakan bulan Februari sampai November 2013. Tempat penelitian di Rumah Sakit Penyakit Tropik dan Infeksi Universitas Airlangga. Desain penelitian yang digunakan adalah experimental design dengan post test only control group design. Kultur  in vitro P. falciparum galur Papua 2300 dibagi dalam kelompok kontrol (K) dan kelompok perlakuan paparan artemisinin berulang, yaitu paparan artemisinin ke-1 (PO1), paparan artemisinin ke-2 (PO2) dan paparan artemisinin ke-3 (PO3) menggunakan konsentrasi IC50. Ekspresi gen Triptophan-rich Protein (PArt) diukur dengan qRTPCR. Hasil menunjukkan paparan artemisinin berulang pada P. falciparum  dapat meningkatkan level ekspresi gen Part (2??CT) relatif terhadap kontrol. Simpulan, paparan artemisinin in vitro menyebabkan overekspresi gen  Tryptophan-rich Proteins(PArt) oleh promoter P. falciparum galur Papua 2300. Even though the mechanism of artemisinin resistance has not been clearly understood, the resistance of P. falciparum towards the antimalaria artemisinin may occur due to the influence of by the internal factors of P. falciparum, including the induction of the protein-expressing gene expression. One of the genes is the Triptophan-rich Protein (PArt) gene that is important in the membrane-spanning protein and plays a role in protein folding to maintain hydrophobic contact.. This study aimed to prove that  Triptophan-rich Protein overexspression in P. falciparum Papua 2300 strain may cause repeated artemisin exposure in vitro. This study was performed in a period from February to November 2013 in Infection and Tropical Diseases Hospital, Airlangga University. The design used was experimental study with post-test only control group design. In-vitro culture of P. falciparum Papua 2300  strain were divided into a control group (K) and treatment groups that were treated regularly with artemisinin, i.e. artemisinin exposure I (PO1), artemisinin exposure 2 (PO2) and artemisinin exposure 3 (PO3)  using IC50 concentration. The Tryptophan-rich Protein gene expression level was detected using  qRTPCR. The result showed that in vitro repeated artemisinin exposure in P.  falciparum  Papua 2300 strain  relatively increased the expression level of the Tryptophan-rich Protein (PArt) genes (2??CT)  when comparedwith control. Abstract

Penyakit diabetes melitus merupakan salah satu penyakit degeneratif yang hingga kini masih belum tuntas terapinya dan masih menjadi ancaman serius bagi dunia kesehatan di Indonesia dan dunia. Tujuan penelitian ini adalah mengetahui kadar insulin dan C-peptide tikus putih penderita diabetes melitus tipe I yang diberikan transplantasi allograf pancreatic stem cell dengan laparotomi intrapankreatik. Penelitian ini dilakukan selama 6 bulan (Juli?Desember 2014) di lab. Stem Cell, Institute of Tropical Diseases, Universitas Airlangga, Surabaya. Dua belas tikus putih jantan Rattus novergicus galur Wistar dibagi secara acak menjadi dua kelompok . Kelompok pertama (P0) disuntik aloksan 150 mg/kg bobot badan tanpa terapi stem cell . Kelompok kedua disuntik aloksan dengan dosis 150 mg/kg bobot badan dan diterapi dengan 1x106/kg bobot badan stem cell pankreas secara laparotomi intrapankreatik (P1). Akhir penelitian adalah pada hari ke-31 percobaan. Hasil penelitian menunjukkan bahwa kadar glukosa darah pada akhir penelitian berbeda sangat nyata (p <0,01) antara kelompok perlakuan yang menerima terapi stem cell (P1) dengan P0 kontrol positif, meskipun nilai kadar glukosa darah rata-rata tidak senormal seperti pada hari ke-1. Tingkat C-peptide dan insulin P0 dan P1 berbeda sangat nyata (p<0,01). Dapat disimpulkan bahwa terapi stem cell secara laparotomi intrapankreatik dapat menurunkan kadar glukosa darah, serta meningkatkan kadar C-peptide dan insulin. The aim of this study was to describe the level of insulin and C-peptide in diabetes mellitus type I white rats treated with  pancreatic stem cell allograft through intrapancreatic laparotomy. This study was conducted at the Institute of Tropical Diseases, Universitas Airlangga, Surabaya in a 6 month period (July?December 2014). Twelve male white rats Rattus novergicus Wistar strain, were randomly divided into two groups. The first group (P0) was injected by alloxan, 150 mg/kg body weight, without stem cell therapy. Another group was injected by alloxan, 150 mg/kg body weight, and was treated with 1x106/kg body weight pancreatic stem cell throughintrapancreatic laparotomy (P1). The experiment was finalized on the 31th day of the experiment. The results showed that the blood glucose levels at the end of experiment were highly significantly different p<0.01 between the treatment group that received stem cell therapy (P1) and P0 positive control, although the average value of blood glucose levels was not as normal as on the first day. C-peptide and insulin levels of P0 and P1 group differed significantly (p<0.01). It can be concluded that stem cell therapy through intrapancreatic laparotomy can reduce blood glucose levels and increase the levels of C-peptide and insulin. Abstract

Background: Justicia gendarussa Burm. f., has been used traditionally in Indonesia for antifertility. Nowadays, a capsule containing 70% ethanol extract of J. gendarussa leaves has been studied for safety. Objective: This study aimed to determine the acute and sub-chronic toxicity of 70% ethanol extract of J. gendarussa leaves in vivo. Methods: In the acute toxicity study, a single dose of 2,000"‰mg/Kg BW was orally administered to mice (n = 10), which were monitored for 24"‰days. For the subchronic toxicity study, rats were randomly divided into four groups (n = 10). The control group received distilled water, while the treatment groups received a repeated dose of 40, 200, and 1,000 mg/Kg BW orally for 90 days. Blood samples were collected for hematological and biochemical evaluations. Gross pathology and histopathology of liver and kidneys were assessed. Results: No mortality and non-observed adverse effect level (NOAEL) were observed in the acute toxicity study. The hematological analysis did not show significant differences in the subchronic toxicity study. The SPGT, SGOT, and creatinine values showed no change in groups 2 and 4, but the level of SGPT increased in groups 3. The increasing level of BUN was observed in all treated groups. Abnormalities or histopathological changes were observed in the liver and kidney in groups 3 and 4. Conclusions: Using 70% ethanol extract of J. gendarussa leaves at a therapeutic dose is safer, but it needs attention at a higher dose.

Abstract

Training on non-alcohol based hand sanitizer (HS) was conducted for assisted youth in five districts, namely Gresik, Bojonegoro, Lamongan, Probolinggo and Nganjuk for three days. In the training, competency strengthening was carried out, namely theory and practicum, ending with evaluation of activities. During the implementation of theory and practicum, training materials were provided using social media facilities in the form of WhatsApp and e-mail. Specifically for coordination with participants, a training group was created using social media. The results of the training evaluation turned out that in terms of strengthening the competence of knowledge in making HS and working mechanisms of HS, it was increasing. In terms of straightening the meaning of HS production to be more focused according to the basis of science or good manufacturing practices (GMP). Thus, youth assisted with criteria include youth who are able to pass on knowledge to all young people in their respective districts. Furthermore, the knowledge and skills of the assisted youth in making HS based on non-alcoholic substances will be more agile and targeted. Thus, in reality, the training on making non-alcohol based HS has made the assisted youths become pioneers at the end of the line to pass on their knowledge. The conclusion in the implementation of this training is that the assisted youth are capable and ready to make non-alcohol-based HS as well as being ready to produce for a small scale while showing real work to be transmitted (98%). abstrakTelah dilakukan pelatihan pembuatan hand sanitizer (HS) berbasis non-alkohol terhadap pemuda binaan di lima kabupaten yaitu Gresik, Bojonegoro, Lamongan, Probolinggo dan Nganjuk selama tiga hari. Dalam Pelatihan tersebut dilakukan pemberian penguatan kompetensi yaitu teori dan praktikum diakhiri dengan evaluasi kegiatan. Selama berlangsung pelaksanaan teori dan praktikum, materi pelatihan diberikan menggunakan fasilitas media sosial berupa whatsapp dan e-mail. Khusus koordinasi dengan peserta dilakukan pembuatan grup pelatihan menggunakan media sosial. Hasil evaluasi pelatihan ternyata dari segi penguatan kompetensi pengetahuan pembuatan HS dan mekanisme kerja HS, makin meningkat. Segi pelurusan pengertian produksi HS menjadi makin terarah sesuai basis ilmu pengetahuan atau cara pembuatan obat yang baik (CPOB). Dengan demikian pemuda binaan dengan kritetria termasuk pemuda yang sanggup menularkan ilmu ke seluruh masyarakat muda di kabupaten masing-masing. Lebihlanjut pengetahuan dan ketrampilan pemuda binaan dalam membuat HS berbasis bahan non-alkohol makin cekatan dan terarah. Dengan demikian seacara nyata pelatihan pembuatan HS berbasis non-alkohol telah menjadikan pemuda binaan sebagai pelopor di ujung garis untuk menularkan ilmunya. Kesimpulan dalam pelaksanaan pelatihan ini adalah para pemuda binaan telah sanggup dan siap membuat HS berbasis non-alkohol sekaligus siap berproduksi untuk skala kecil sambil memperlihatkan karya nyata untuk ditularkan (98 %).

Abstract

The aim of this research was to investigate the number of prevalence of Trypanosomiasis of wild rats in Banyuwangi. Sixty wild rats were trapped from human residence, markets and rice fields in Banyuwangi. Rat`s  blood smear was stained by Giemsa. The result show that one (1.67%.) of 60 blood sample was appear Trypanosoma sp..  The prevalence of Trypanosomiasis of wild rats in Banyuwangi was 1.67%.

Abstract

This was a Community Service activity that aims to share knowladge about utilization of Curcuma Molasses Block (CMB) to increase performance of goats that infected gastrointestinal (GI) parasites to The Goat Breeder Group at  Wonorejo, Wates District, Kediri Regency. The activities include: counseling about raising goat management, presentasion on worm diseases that infect goats and CMB as anthelmentic, workshop and practice to make CMB, detection egg worm in goat`s feces, treatment on infected goats and filling out questionnaires evaluating the performance of goat before and after giving CMB. The result show that all of feces samples were positive contain egg worms of  Haemonchus contortus, Chabertina ovina, Trichuris globulosa, Strongyloides papillosus, Fasciola gigantica, Trichostrongylus spp., Toxocara sp., and Oesohagustomum columbianum even also protozoans  Eimeria sp., and Blastocystis sp. Administration CMB increase performance of worm-infected goats which indicated by increase in appetite, soft and shiny hair and the body more well-fed.

Abstract

The purpose of this study was to determine the prevalence of gastrointesatinal parasites in cats at Surabaya shelter. Eighty two samples of cat feces used in this study were taken from Griya satwa shelter and Kiro cat shelter. Feces samples were examined using the native, sedimentation and floating methods. Feces samples gastrointestinal parasite were identified by using a microscope with 400x and 1000x magnifications. Faces samples that are diagnosed positively if there is at least one of the three methods. Feces samples has observed, obtained prevalence rate of 43.9% or 36 positive samples from eighty two samples. The results of chi square analysis showed that p > 0.05, which showed that the parasites taken in the two shelters had clear differences in the prevalence of the gastrointestinal tract parasites. The parasites observed were Isospora sp., Ancylostoma sp., Toxocara cati and Toxocara leonina.

Abstract

ABSTRACT             The purpose of this study was to identify ectoparasites that attack pearl catfish and the incidence of ectoparasite infestation in 100 samples of pearl catfish aged one and three months. This research was carried out from January to March 2021 in Makalele's cultivation, Tuban's District, Tuban's Regency. The method used in this research is skin scraping and gill installation. Data analysis using Chi-Square Test with SPSS version 24.0 for windows. The results showed that there were 3 types of ectoparasites found, Trichodina nobilis, Trichodina pediculus, Chilodonella sp., Dactylogyrus sp. and the highest incidence of ectoparasites occurred in young fish compared to old fish with a percentage of 35% and 25%, respectively, and based on the chi-square test there was no significant difference (p️≥0.05). Keywords: Clarias gariepinus, infestation level, ectoparasites.

Abstract

This study aims to evaluate the expression of BAX, BCL-2, and BAX/BCL-2 ratio in maturation media of cow oocytes which supplemented with Urea in vitro because BAX and BCL-2 are the main regulators of apoptosis. A total of 263 oocytes from follicle aspirations originating from ovaries taken from slaughterhouses and were saturated with 3 addition of urea which was divided into three groups. The control group (P0) was control group without the addition of urea, P1 group was added with urea 20 mg/dL, while P2 group was added with urea 40 mg/dL. The results of in vitro oocyte maturation were continued with identification using immunocytochemical staining with the addition of BAX and BCL-2 antibodies. Positive results showed a brownish color on the oocyte and its cumulus. The results of this study indicated that there were significant differences (P0.05) in BAX and BCL-2 expression, although both curves were equally increase. The increase in BCL-2 was more significant than BAX, while the BAX BCL-2 ratio did not show a significant difference (P0.05) in whichthe curve of BAX/BCL-2 ratio was decreased. It can be concluded that the addition of urea does not affect the level of apoptosis.

Abstract

This study aimed to determine the occurences of mice brain ventricles dilatation that congenitally infected with Toxoplasma gondii (T. gondii) as a marker of hydrocephalus and cellular changes in the brain. A total of twenty pregnant mice (11.5 days pregnacy) were divided into 2 groups, which were control (P1) group and treatment (P2) group. The mice in the treatment group were infected with 101 tachyzoites of T. gondii. All mice were maintained until delivery. The newborn mice were sacrificed and their brain were removed and fixed in 10% buffered formalin to prepare histology slides with HE staining for observation of ventricular width, TUNEL assay for apoptosis observation, and immunohistochemistry for the expression of transforming growth factor beta (TGF-β) observations. The data were analyzed using t test and linear regression. The results showed that ventricular width and apoptosis index significantly increased (P0.01) in the treatment group compared to control group, but there was no difference in the expression of TGF-β (P0.05) in both groups. Dilatation of ventricle correlated with the apoptotic index of brain cells but did not correlated with the expression of TGF-β.
