Abstract

Brucellosis is a zoonotic disease cause by pathogenic bacteria of the genus Brucella. The disease infects livestock mammals such as cows, goats, pigs, and including humans. Brucella abortus has several potential virulence factors, i.e. Proteins VirB. Type IV secretion system (T4SS) which is a combination of 12 proteins from VirB1-VirB11 and VirD4. Brucella can survive for long periods in the environment despite the limitations of nutrients and oxygen. This study aims to characterize the protein VirB4 of local isolate of B. abortus using SDS PAGE (Sodium Dodecly sulfate-polyacrylamide gel electrophoresis). The results showed that the protein contained 10 protein bands with a molecular weight of 158.93; 110.89; 99.931; 70.60; 64.61; 59.25; 45.32; 42.35; 23.63; and 16.70 kDa, respectively. Protein VirB4 of the local isolate of B. abortus have a molecular weight of 59.25 kDa. ABSTRAK Brucellosis merupakan salah satu penyakit zoonosis yang disebabkan oleh bakteri patogen genus Brucella. Penyakit ini menyerang hewan ternak mamalia seperti sapi, kambing, babi, dan dapat menular ke manusia. Bakteri Brucella abortus memiliki faktor virulensi potensial yaitu protein VirB. Type IV Secretion System (T4SS) merupakan gabungan dari 12 protein yaitu VirB1–VirB11 dan VirD4. Brucella dapat bertahan hidup lama di dalam lingkungan meskipun memiliki keterbatasan nutrisi dan oksigen. Penelitian ini dilakukan untuk karakterisasi protein VirB4 B. abortus isolat lokal dengan metode Sodium Dodecly Sulfate-Polyacrylamide Gel Electrophoresis (SDS PAGE). Hasil karakterisasi protein B. abortus isolat lokal dengan teknik SDS-PAGE terdapat 10 pita protein dengan bobot molekul 158,93; 110,89; 99,931; 70,60; 64,61; 59,25; 45,32; 42,35; 23,63; dan 16,70 kDa. Simpulan penelitian ini adalah terdapat protein VirB4 B. abortus isolat lokal yang mempunyai bobot molekul 59,25 kDa.

Abstract

Banyuwangi is still listed as areas not completely free from hemorrhagic dengue fever outbreak. Changes in the genome of Aedes aegypti mosquito as the main vector of dengue virus is estimated to be the cause of difficulty in controlling the disease. Expression of Voltage Gated Sodium Channel (VGSC) genes can be used as an important indication of Aedes aegypti mosquitoes resistancy to certain groups insecticides. The purpose of this study was to detect the Voltage Gated Sodium Channel (VGSC) genes coding in Aedes aegypti using Polymerase Chain Reaction (PCR); as it is related to the mechanism of resistance to organophosphate insecticides groups. Results of resistency by susceptibility test showed that Aedes aegypti mosquito samples from three district ie. Tegaldlimo, Purwoharjo, and Banyuwangi were resistant to malathion 0.8% insecticide (organophosphates group). PCR results detected gen band with 250 bp length, which confirmed that insecticide resistance gene has been detected in the tested samples. Based on this study results, it is recommended to use alternative insecticides apart from organophosphate group for dengue vector control in Banyuwangi. ABSTRAK Kabupaten Banyuwangi masih tercatat sebagai daerah yang belum sepenuhnya bebas dari wabah demam berdarah dengue (DBD). Perubahan genom nyamuk Aedes aegypti sebagai vektor utama virus dengue diperkirakan menjadi penyebab sulitnya pengendalian penyakit DBD. Ekspresi gen Voltage Gated Sodium Channel (VGSC) dapat dijadikan indikasi penting sejauh mana perkembangan nyamuk Aedes aegypti resisten terhadap insektisida golongan tertentu. Tujuan penelitian ini adalah untuk mendeteksi gen penyandi VGSC pada nyamuk A. aegypti yang berkaitan dengan mekanisme resistensi terhadap insektisida golongan organofosfat menggunakan teknik Polymerase Chain Reaction (PCR). Hasil uji resistensi susceptibility test menunjukkan sampel nyamuk A. aegypti dari tiga kecamatan di Kabupaten Banyuwangi yaitu Kecamatan Tegaldlimo, Purwoharjo, dan Banyuwangi resisten terhadap insektisida malathion 0,8 % (kelompok organofosfat). Hasil deteksi gen melalui PCR didapatkan band yang muncul dengan panjang 250 bp. Hal tersebut menunjukkan bahwa gen pembawa resistensi insektisida (VGSC) telah terdeteksi pada sampel yang diuji. Berdasarkan hasil penelitian, disarankan untuk pengendalian vektor DBD di Kabupaten Banyuwangi dengan menggunakan alternatif insektisida selain dari golongan organofosfat.

Abstract

Infectious disease which caused by parasite is the most serious problem in tortoise breeding. This study identifies gastrointestinal endoparasites infection on Red Foot (Chelonoidis carbonaria) during November 2020 to February 2021 in Surabaya City. Fecal samples from a tortoise, a total of 14 tortoises were collected and examined by native, sedimentation and floatation methods for parasitic eggs and oocyst. Fecal samples for this research were collected from breeding farm in Surabaya City. Data obtained from this study were presented descriptively and analyzed by Chi-Square test. Based on examination results, the occurrence of gastrointestinal endoparasite infection on Red foot was recorded at 100% from the total samples. The gastrointestinal parasites were identified as helminths and protozoa namely which is dominately by Tachygonetria spp. (14%), Angusticaecum spp. (3%), and Balantidium coli (2%) respectively. The provision of deworming medication as a curative measure by the owner is carried out regularly and continuously on tortoises that are positively infected with gastrointestinal endoparasites and prevention can be done by paying attention to the management and cleanliness of the cage to prevent potential transmission of endoparasites from the environment to the tortoise.

Abstract

Rousettus spp. is one of the wild animals that may transmit various diseases including zoonotic. Rousettus spp. or known as fruit bat is found in Java island and belongs to the Pteropodidae family. Generally, Rousettus spp. consumes fruit and floral products. This case report aims to detect ectoparasites in fruit bats (Rousettus spp.) found at Djawatan Banyuwangi, East Java. The samples in this case report are seven fruit bats (Rousettus spp.). Based on the results of laboratory identification using the whole mount method, from 7 samples of fruit bats (Rousettus spp.), there are five fruit bats (Rousettus spp.) were infected with Spinturnix spp.. Spinturnix spp. that obtained from fruit bats (Rousettus spp.) were examined using a binocular microscope (Olympus CX-23, Tokyo Japan) with a magnification of 100 times. Images of ectoparasites and bats were taken using a Nikon d5300 camera. Total amount of Spinturnix spp. that was successfully obtained from 5 fruit bats (Rousettus spp.) are 25 individuals.

Abstract

The purpose of this study was to determine the type of insecticides which Aedes aegypti mosquitoes are resistant to, so that other susceptibleinsecticides still can be used. The study was a cross sectional epidemiological study with cluster sampling in Sub-district Wongsorejo,Banyuwangi, Muncar, Tegaldlimo, Kalibaru which is considered sufficient to represent Banyuwangi Regency. Mosquito samples appropriate tothe characteristics were isolated and tested for insecticide-resistance primer namely voltage gated sodium channel (VGSC) to determine thespecific resistance expressed in mosquitoes. The result of resistance test using WHO standard method showed that Aedes aegypti mosquito fromBanyuwangi Regency were resistant to cypermethrin 0.25% and malathion 0.8%. There was a VGSC coding gene with 250 bp band detectedusing polymerase chain reaction (PCR) technique which was associated with the resistance of Aedes aegypti mosquitoes in Banyuwangi Regencyto organophosphate insecticides (malathione) and pyrethroid insecticides (cypermethrin).

Abstract

This research aimed to characterize VirB4 protein of local isolate Brucella abortus with Western blotting method. The result showed that there were four protein bands with molecular weights of 64.61, 59.25, 21.63, and 16.70 kDa by triggering a reaction between the whole Brucella abortus and anti-Brucella abortus serum. The results also revealed that there was only one protein band with a molecular weight of 59.25 kDa triggering a reaction between the whole Brucella abortus and anti-VirB Brucella abortus serum. Finally, it can be concluded that VirB4 protein can affect the virulence factor of Brucella abortus, successfully characterized with the appearance of one band with a molecular weight of 59.25 kDa by using Western blotting method.

Abstract

The potential of Pulau Santen, which has been known as a tourism beach, is increasingly visible with the large amount of infrastructure and tourism built by the local government. This was done as an effort to promote Pulau Santen as one of the best natural tourism centers in Banyuwangi. The community in Pulau Santen must be prepared as a group that is aware of the potential of their village and the sustainability of its natural tourism. Pulau Santen is also categorized as a location which every year frequently used for sea turtle nests that land on the Banyuwangi coastal areas. This is actually very contrary to the natural habits of sea turtles which prefer to make nests in quiet places without any disturbance from sound, light, and humans. Because the place used as a natural nest is a busy tourist spot, local residents must also be given insight regarding the protection of sea turtles that land on the coast of Pulau Santen as one of the wild animals protected by the Conservation Law in Indonesia. The purpose of this activity is to strengthen the awareness of Pulau Santen tourism awareness groups regarding the important role of communities around the coast to participate in wildlife conservation and their ecosystems. Through the community service activities that have been carried out, the sustainability of the program is needed so that a model of a conservation education tourist spot is formed that has the potential to improve the economy of the surrounding communities and the conservation of biodiversity in the area.

Abstract

The javanese keelback water snake (Fowlea melanzostus) is a semi-aquatic reptile that often found in Indonesia and endemic to Java island. Water tiger snakes are usually kept as exotic pets. Wild caught javanese keelback water snakes have risks of spreading several disease agents that can be zoonotic, which is nematodiasis caused by Ophidascaris spp. This study aims to determine the prevalence level of Ophidascaris spp. infections in javanese keelback water snakes from Banyuwangi district. This study used a descriptive method with accidental sampling. The total sample in this study was 33 wild-caught javanese keelback water snakes. The identification method used is snake samples that have been collected are then euthanized and necropsied to check for nematode infections in the subcutan, muscular, and visceral part. The result showed that 28 samples were positive for nematode infection and 5 samples were negative with a prevalence rate of 84.84%. The nematodes that have been identified come from the genus Ophidascaris spp. Therefore, further research is needed to determine the incidence of nematode transmission in other wild caught snakes in Indonesia.

Abstract

Penelitian ini bertujuan untuk mengetahui angka prevalensi kasus infeksi cacing saluran cerna dan mengidentifikasi jenis cacing pada kucing liar di wilayah Kota Banyuwangi. 126 sampel feses kucing yang berasal dari wilayah Kota Banyuwangi dikoleksi dan disimpan dalam larutan formaldehide 10%. Seluruh sampel feses tersebut diperiksa dengan dua metode berbeda yaitu metode natif dan metode pengapungan untuk mengidentifikasi telur cacing yang terdapat pada feses kucing. Pada penelitian ini teridentifikasi tiga genus cacing yang menginfeksi kucing liar di wilayah Kota Banyuwangi yaitu Toxocara, Ancylostoma, dan  Diphyllobothrium. Hasil penelitian ini menunjukkan bahwa prevalensi kasus infeksi cacing saluran cerna pada kucing liar wilayah Kota Banyuwangi masih tinggi, yaitu Toxocara Spp (43,65%), Ancylostoma Spp (3,96%), dan Diphyllobothrium Spp (52,38%). Untuk itu diperlukan program pengendalian yang tepat terhadap kasus infeksi cacing saluran cerna pada kucing liar di wilayah Kota Banyuwangi.

Abstract

Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Aspergillus Spp pada paru-paru ayam kampung. Sampel yang digunakan dalam penelitian ini adalah 20 paru-paru ayam kampung yang diambil secara acak dari Pasar Banyuwangi yang terletak pada wilayah tengah Kota Banyuwangi, Jawa Timur. Sampel dicuci dengan aquades steril yang berisi antibiotik selanjutnya ditanamkan pada media spesifik Sabouraud's Dextrose Agar (SDA) kemudian diinkubasikan pada suhu kamar selama 3-7 hari. Pertumbuhan morfologi Aspergillus diamati secara makroskopis. Koloni yang diduga Aspergillus Spp diperiksa secara mikroskopis. Data yang diperoleh dianalisis secara deskriptif. Hasil pemeriksaan menunjukkan bahwa Aspergillus Spp dapat diisolasi pada 9 dari 20 sampel paru-paru. Dapat disimpulkan bahwa 45% sampel paru-paru yang diperiksa positif terinfeksi Aspergillus Spp dan 55% bebas dari infeksi Aspergillus Spp.
