Abstract

AbstrakWereng Batang Coklat (WBC) (Nilaparvata lugens Stal.) merupakan serangga hama yang dapat merusak tanaman padi (Oryza sativa L.). Tanaman kipait (Tithonia diversifolia) berpotensi sebagai insektisida nabati karena memiliki senyawa toksik terhadap serangga. Penelitian ini bertujuan untuk mengkaji aktifitas makan akibat pemberian ekstrak daun kipait. Konsentrasi ekstrak daun kipait yang digunakan adalah 0% (kontrol), 1%, 3%,5% dan 7% dalam RAL (Rancangan Acak Lengkap) dengan 5 kali ulangan. Hasil penelitian menunjukkan bahwa pemberian ekstrak daun kipait berpengaruh terhadap penghambatan daya makan WBC. Penghambatan makan tertinggi terjadi pada konsentrasi 7% setelah 24 jam.Kata kunci : penghambatan daya makan, Nilaparvata lugens Stal., Tithonia diversifoliaAbstractBrown planthopper (BPH) (Nilaparvata lugens Stal.) Is an insect pest that can damage rice plants (Oryza sativa L.). Kipait (Tithonia diversifolia) is potential as a bioinsecticide because it is toxic to insects. This study aimed to evaluate the feeding activity that was influenced by kipait leaf extract. The concentration of kipait leaf extract were 0% (control), 1%, 3%, 5% and 7%. The experiment design was CRD (completely randomized design) with 5 replications. The results showed that the kipait leaf extract influenced the feeding inhibition of BHP. The highest inhibition occurred in the concentration of 7% after 24 hours treatment.Keywords : brown planthopper, feeding inhibition, Tithonia diversifolia

Abstract

ABSTRAKBakteri endofit merupakan salah satu sumber penghasil senyawa ekstraseluler yaitu enzim. Enzim dari bakteri endofit lebih menguntungkan dan produksinya lebih cepat. Penelitian ini bertujuan untuk menganalisis kemampuan aktivitas enzim ekstraseluler dari bakteri endofit tumbuhan mangrove Avicennia marina. Metode penelitian yang digunakan yaitu eksploratif eksperimental, melakukan isolasi bakteri dari tumbuhan mangrove A. marina dan selanjutnya dilakukan uji potensi aktivitas enzim ekstraseluler dari isolat bakteri endofit. Hasil isolasi memperoleh tujuh isolat bakteri endofit dari tumbuhan A. marina, isolat endofit tersebut mampu menghasilkan aktivitas enzim ekstraseluler yaitu empat isolat menghasilkan enzim amilase, enam isolat menghasilkan protease, satu isolat menghasilkan selulase dan dua isolat menghasilkan gelatinase.Kata kunci: bakteri endofit; Avicennia marina; amilase; protease; selulase; gelatinase ABSTRACTEndophytic bacteria are one source that can produce extracellular compounds, namely enzymes. Enzymes from endophytic bacteria are more profitable and can produce faster. This study aims to analyze the ability of extracellular enzyme activity from endophytic bacteria in mangrove plants Avicennia marina. This research used experimental explorative method, isolating bacteria from mangrove plant A. marina and then testing the potential of enzyme extracellular activity from endophytic bacteria isolated. Isolating result obtained seven endophytic bacterial isolates from A. marina plants,this endophytic isolates are able to produce extracellular enzyme activity is four isolates can produced amylase enzyme, six isolated can produced protease, one isolated can produced cellulase and two isolated can produce gelatinase. Secara langsung, salah satunya yaitu bakteri dapat memproduksi dan menyekresikan zat pengatur tumbuh indole-3-acetic acid (IAA, auksin). Penelitian ini bertujuan untuk menguji kemampuan bakteri endofit ubi jalar dalam menghasilkan IAA. Metode analisis IAA dilakukan dengan metode kolorimetri. Analisis produksi IAA dilakukan dengan penambahan dan tanpa penambahan L-triptofan. Hasil penelitian menunjukkan bahwa tanpa penambahan L-triptofan, diperoleh sejumlah 19 jenis yang menghasilkan IAA dengan kisaran konsentrasi 0,29-7,21 mg/L. Dengan penambahan L-triptofan, jumlah jenis positif dan konsentrasi IAA yang dihasilkan meningkat signifikan. Jumlah jenis positif 20 jenis (91%) dan konsentrasi IAA yang dihasilkan mencapai kisaran 0,96-115,63 mg/L.Kata kunci: bakteri endofit; IAA; ubi jalar; L-triptofan ABSTRACTPlant-bacteria associations, can promote plant growth by both direct and indirect mechanisms. One of direct mechanisms is that bacteria can produce and secrete indole-3-acetic acid (IAA, auxin) growth regulators. This study aims to examine the ability of sweet potato endophytic bacteria to produce IAA. The detection of IAA production was conducted by colorimetric technique. IAA production analysis was carried out with addition and without addition of L-tryptophan. Without the addition of L-tryptophan, a total of 19 species produced IAA with a concentration range of 0.29-7.21 mg/L. With the addition of L-tryptophan, the number of positive species and the concentration of IAA produced increased significantly. The number of positive species was 20 species (91%) and the concentration of IAA produced reached a range of 0.96-115.63 mg/L.Keywords: endophytes bacterial; IAA; sweet potato; L-tryptophan

Abstract

(Article History: Received March 4, 2021; Revised March 31, 2021; Accepted April 7, 2021) ABSTRAKPemanfaatan kembali limbah pertanian sebagai pupuk organik memiliki prospek yang baik dalam meningkatkan produktivitas lahan melalui perbaikan sifat biologi tanah. Tujuan penelitian ini adalah untuk menganalisis efek pemberian amelioran pupuk organik dari limbah jagung (Zea mays L.) terhadap peningkatan jumlah populasi mikroba tanah dan pertumbuhan tanaman jagung. Metode penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) yang terdiri dari 4 perlakuan dengan tiga kali ulangan: AA (Amelioran  pupuk organik jagung 40 kg/ha), AB (Amelioran pupuk organik jagung 80 kg/ha), UR (Urea 40 kg/ha), dan K (tanpa amelioran atau urea). Perhitungan jumlah koloni mikroba tanah menggunakan metode hitungan cawan. Hasil penelitian menunjukkan perlakuan amelioran pupuk organik berpengaruh signifikan terhadap populasi mikroba tanah dan pertumbuhan tanaman jagung pada parameter tinggi tanaman dan jumlah daun. Perlakuan amelioran pupuk organik jagung tidak menunjukkan pengaruh terhadap diameter batang. Kata kunci: Amelioran; mikroba tanah; pupuk organik; Zea mays L. ABSTRACTThe reuse of agricultural waste as organic fertilizer has good prospects in increasing land productivity by improving soil biological properties. The aims of this study was to analyze the effect of using ameliorant organic fertilizer application from corn waste to increase the number of soil microbial populations and the growth of maize plants (Zea mays L.). The method was using CRD (Complete Random Design), which consist of 4 treatments with three repetitions: AA (Ameliorant corn organic fertilizer 40 kg/ha), AB (Ameliorant corn organic fertilizer 80 kg/ha), UR (Urea 40 kg/ha), and K (without ameliorant or urea). Calculation of the number of soil microbials colonies was using the plate count method. The results showed that the treatment of ameliorant organic fertilizer had a significant effect on soil microbials population and growth of maize on the parameters of plant height and number of leaves. The treatment of organic fertilizer ameliorant showed no effect on stem diameter.Keywords: Ameliorant; Soil microbes; Organic fertilizer; Zea mays L.

Abstract

AbstrakAndrographis paniculata diketahui mengandung senyawa andrografolid, yaitu suatu metabolit sekunder yang memberikan efek farmakologi berupa hepatoprotektif, antiviral dan antikanker. Dalam penelitian ini dilaporkan studi awal prosedur transformasi genetik A. paniculata dengan perantara Agrobacterium tumefaciens. Eksplan daun A. paniculata diinkubasi dengan Ag. tumefaciens strain LBA4404 yang mengandung vektor ganda pCAMBIA1304 dengan gen hpt sebagai gen penanda untuk resistensi higromisin dan gen gusA sebagai gen pelapor. Setelah kokultivasi, eksplan daun dikultur pada medium seleksi yang mengandung higromisin 20 mg L-1 dan sefotaksim 400 mg L-1. Hasil uji histokimia GUS pada potongan daun setelah tiga hari kokultivasi menunjukkan ekspresi transien GUS mencapai 18,83%. Sebanyak 64,44% jaringan A. paniculata yang telah berhasil ditransformasi menunjukkan regenerasi sel dengan menghasilkan kultur kalus transforman pada medium yang mengandung 20 mg/L higromisin.Kata kunci: transformasi genetik, Agrobacterium tumefaciens, Andrographis paniculata, asai GUS.AbstractAndrographis paniculata is known to contain andrographolide, a secondary metabolite which shows pharmacology effects such as hepatoprotective, antiviral and anticancer. We established an Agrobacterium tumefaciens-mediated transformation procedure for A. paniculata. Leaf explants of A. paniculata were incubated with Ag. tumefaciens strain LBA4404 containing a binary vector pCAMBIA1304 with the hpt gene as a selectable marker for hygromycin resistance and an gusA gene as a reporter gene. Following co-cultivation, leaf explants were cultured on selective medium containing 20 mg L-1 hygromycin and 400 mg L-1 cefotaxime. GUS assays showed that only 18.83 % transformation frequency was obtained in leaf disk tissues after 3 days co-cultivation. As much as 64.44 % of the transformed tissue on MS medium containing selection agent 20 mg/L hygromycin showed cell regeneration to produce calluses.Keywords: genetic transformation, Agrobacterium tumefaciens, Andrographis paniculata, GUS assay.

Abstract

Commercial sterility analysis is a crucial step in quality control of Ultra High Temperature (UHT) products such as coconut cream liquid products produced by PT Sasa Inti Minahasa Selatan, to ensure the safety and stability of shelf life. Stability incubation testing takes a long time, which is 7-14 days. Development of commercial sterility methods using Chromogenic Agar selective media is needed to cut the incubation time that is too long. Agar Chromogenic Media is tested for its ability to detect common contaminants in UHT coconut cream products, in particular thermophilic and mesophilic microorganisms that cause spoilage, with a short incubation time. UHT coconut cream samples are prepared by artificial and natural inoculation. Validation was carried out by comparing the results on standard media based on sensitivity, specificity and Time-to-result (TTR) parameters. The results showed a real difference between NA/TGEA media and chromogenic media so that the reading of bacterial colonies growing on chromogenic media was clearer and easier to read due to the change in color and short incubation time where the bacteria causing decay had grown on day 5 with a reading of results that took only 24 hours. Based on the validation that has been carried out, chromogenic selective media can accelerate product release time and improve quality control efficiency in the UHT coconut cream industry.
