Abstract

Abstract. The objectives of this research was to study the macroscopic, the microscopic and phytochemicals characterization of raw materials, total polyphenol content and the antibacterial activity of nut grass extracts (Cyperus rotundus L.) against Shigella dysenteriae, Salmonella typhimurium, Staphylococcus epidermidis, and Pseudomonas aeroginosa. The macroscopic and the microscopic characterization of raw materials were done with the determination of the water contents, the water soluble contents, the ethanol soluble contents, total ash and acid soluble ash. The extract was prepared by percolation using n-hexane, ethanol, and ethyl acetate as solvents. Phytochemical screening was done on the raw materials as well as n-hexane, ethyl acetate, and ethanol extracts. Total phenol contents were tested with the Folin-Ciocalteu reagent method. The antibacterial activity test was done by measuring the diameter of the inhibition zones using the diffusion agar method. The results showed that raw material retrieved 7.33% water content, 21.85% water soluble contents, 7.9% ethanol soluble contents, 1.53% total ash, and 0.67% total insoluble-acid ash. The phytochemical study showed that the nut grass contains various secondary metabolites including flavonoids, glycosides, saponins, anthraquinone glycosides, and steroid/triterpenoids. The total polyphenol contents of ethanol extracts were expressed as catechin equivalents 1.1616 mg/g extract. All of the nut grass extract showed antibacterial activity. Ethyl acetate extract of nut grass indicated the highest antibacterial activity against Shigella dysenteriae, Salmonella typhimurium, and Pseudomonas aeroginosa. Abstract

ABSTRACT Objective: The purpose of this study was to analyze minerals in gadung (Dioscorea hispida Dennst.) tuber. Method: The analysis of minerals was conducted by using atomic absorption spectrophotometry. Results: The results showed that the concentration of iron, cadmium, potassium, calcium, sodium, magnesium, copper and zinc were 0.6445 + 0.0029 mg/100g, 0.0011 + 0.0083 mg/100g, 265.2780 + 0.4652 mg/100g, 5.1015 + 0.0490 mg/100g, 8.3011 + 0.0974 mg/100g, 29.0237 + 0.0706 mg/100g, 0.1110 + 0.0019 mg/100g, and 0.6113 + 0.0026 mg/100g, respectively. Conclulsion: The gadung (Dioscorea hispida Dennst.) tuber contains iron, cadmium, potassium, calcium, sodium, magnesium, copper and zinc as the minerals.

Abstract

The tuber of Dioscorea hispida Dennst. has potential for development as a synthetic pharmaceutical ingredient due to the presence of secondary metabolites such as diosgenin and flavonoids, which exhibit biological activity within the body. Further exploration of this potential requires the isolation and identification of these compounds. Diosgenin was isolated from the ethanol extract using thin-layer chromatography, while flavonoids were isolated from the ethyl acetate fraction using paper chromatography. The pure isolates of both compounds were identified using UV-Vis spectrophotometry and FT-IR spectroscopy. The UV-Vis spectrophotometric analysis of diosgenin revealed maximum absorption wavelengths at 205 and 453 nm. Flavonoid isolates exhibited maximum absorption wavelengths of 264 nm for the first isolate, 265 nm for the second, 276 nm for the third, and 272 nm for the fourth. FT-IR spectroscopic analysis of the diosgenin isolate indicated the presence of functional groups such as -OH, aromatic C=C, and aromatic C-O, while the flavonoid isolate showed the presence of O-H, aromatic C-H, aliphatic C-H, C=O, C=C, and C-O. These identification results confirm that the isolates obtained indeed contain diosgenin and flavonoids.

Abstract

Rumput teki (Cyperus rotundus L.) family Cyperaceae dikenal sebagai gulma yang mengandung metabolit sekunder yang berpotensi sebagai antibakteri. Data ilmiah pada bagian herba rumput teki masih terbatas. Penelitian ini menganalisis aktivitas antibakteri ekstrak etanol terhadap Escherichia coli dan Staphylococcus aureus. Ekstraksi herba rumput teki (Cyperus rotundus L.) dilakukan dengan cara maserasi menggunakan pelarut etanol. Uji aktivitas antibakteri dilakukan secara in vitro menggunakan metode difusi dengan konsentrasi 400, 300, 200, 100, 50, 40, 30, 20, 10, 5, 2.5, dan 2 mg/mL serta penentuan konsentrasi hambat minimum (KHM). Hasil menunjukkan kadar air pada simplisia herba rumput teki (Cyperus rotundus L.) sebesar 6,63%. Skrining fitokimia menunjukkan ekstrak etanol mengandung alkaloid, flavonoid, saponin, tanin, glikosida, serta steroid/triterpenoid. Hasil uji menunjukkan ekstrak etanol herba rumput teki memiliki aktivitas antibakteri yang efektif pada konsentrasi 400 mg/mL dengan zona hambat 14.6 ± 0.07 mm untuk Escherichia coli dan 13.2 ± 0.07 mm untuk Staphylococcus aureus. Konsentrasi hambat minimum (KHM) ekstrak etanol herba rumput teki terhadap terhadap Escherichia coli dan Staphylococcus aureus diperoleh pada konsentrasi 2,5 mg/mL. Penelitian ini mendukung bahwa herba rumpu teki (Cyperus rotundus L.) berpotensi sebagai sumber antibakteri alami.
