Abstract

Background: Prolonged hyperglycaemia can make the pancreatic beta cells work harder and cause fatigue. When this happens, it can trigger oxidative stress reactions, which can produce free radical compounds that can damage pancreatic beta cells. The body compensates by activating protective mechanisms such as the production of antioxidant compounds to reduce the levels of free radicals in the cells. One such compound is glutathione (GSH). Insulin Gaussia Luciferase (iGL) cells are a cell line derived from rat pancreatic β (beta) cells. These cells can be used as a model of oxidative stress in hyperglycaemia to measure GSH levels and there are no studies using iGL cells to measure GSH levels. Therefore, in this study, the iGL cells are used as the object of research.Objective: To investigate the effect of GSH levels on glucose toxicity condition through in vitro experiments on iGL cells.Methods: The study used 5 different glucose concentrations of 11, 16.5, 22, 33, and 44 mM with the addition of iGL cell growth medium exposed for 7 days. We measured the amount of intracellular GSH using a colourimetric method at a wavelength of 405 nm. The analysis used in this study was a one-way ANOVA test. Differences between groups were tested using SPSS.Results: The results of this study showed that there was an increasing trend in total GSH levels on the third and seventh day.Conclusion: there was a daily decrease in GSH/cell in the iGL sample cells exposed to different concentrations of glucose for 7 days. This can be due to an increase in the oxidative stress reactions in the cells, which can lead to a decrease in the levels of antioxidants.

Abstract

Background: Deficiency of vitamin B12 remains a health problem in both developed and developing countries. Many journal studies showed an association between deficiency of vitamin B12 and metabolic disorder, one of which is insulin resistance, one of the most significant metabolic diseases. Resistance to insulin may be assessed by changes in plasma glucose and insulin levels. This study explored a possible relationship between vitamin B12 restriction and glucose metabolism. Materials and Methods: This study used an experimental method of 18 male Sprague Dawley rats (Rattus norvegicus, 300-400 gram, age 7-8 months), divided into three groups: control (G1), four weeks treatment group (G2 ) and 12-week treatment group (G3). In the control group, regular feed, the standard diet was given, while the treatment group was assigned vitamin B12, AIN-93M deficiency feed according to the treatment age. Results: Plasma glucose increased in treatment group after 4 weeks (control vs P1= 104.7 ± 5.04 vs 206.8 ± 4.5 p = 0.000) and 12 weeks (control vs P3 = 99.9±6.3 vs 144.7±6.8 p<0.01). Plasma insulin decreased in treatment group after 4 weeks (control vs P1=63.8 ± 1.5 vs 59.9 ± 0.7 p > 0.05) and 12 weeks (control vs P3 = 61.5±0.64 vs 57.4±1 p>0.05). Conclusion: Increased homocysteine deficiency due to dietary vitamin B12 can cause insulin resistance syndrome. Abstract

Background. Vitamin B12 deficiency is a significant problem, particularly in developing countries. The increasing complexity of the Vitamin B12 molecule, the deficiency condition, causes a complex cell level disorder. The clinical manifestation of vitamin B12 deficiency is anemia, but it is believed that due to the complexity of vitamin B12, there are still other molecular disorders. This study aims to establish the pathway of damage caused by vitamin B12 deficiency in the Nrf2 molecule. Methods. An animal experiment involving 12 male Sprague Dawley rats were randomly divided into control and 16-week treatment. There are two research groups, namely the control group and the treatment group. The control group was given a standard diet, while the treatment group was assigned a vitamin B12 deficiency diet. At the end of treatment, The levels of vitamin B12 in the liver tissue and plasma NRF2 were measured. Statistical Analysis. Nrf2 and vitamin B12 levels were statistically analyzed using the Student's T-test, and the correlation between vitamin B12 and Hb was analyzed using the Spearmen test. Data that cannot use by Independent T-test were tested using the Mann Whitney test. Results/Conclusion. There was no significant difference between the control and treatment groups on plasma Nrf2 levels (4.02 ± 0.59 pg/ml vs 3.34 ± 0.55 pg/ml; p = 0.07) and tissue vitamin B12 (0.0035 ± 0.0016 µg/ml vs 0.0030 ± 0.0006 µg/ml ; p = 0.8). Abstract

Background and Aims. Anemia is a condition when the number of red blood cells or hemoglobin is lower than the normal number. The main cause of anemia is a lack of factors for the formation and maturation of red blood cells. Vitamin B12 plays a role in the synthesis and maturation of red blood cells, a lack of this vitamin will cause the formation and maturation of red blood cells to be disrupted. Method. This was an experimental animal study with twelve rats with frederer's family. The rat was fed a Vitamin B12 Deficient Diet for 16 weeks as the treatment group and standard diet as the control group. Vitamin B12 in the liver was analyzed in rats for 16 weeks. Hemoglobin and MCV were analyzed in plasma for 16 weeks. Results are expressed as means of control, and test animals were established by ANOVA using SPSS 20.0 software Results. In the present study, we found that liver vitamin B12 level was decreased in treatments groups compares to the positive control group at sixteen weeks (p<0.01). Hemoglobin was decreased in the treatment group compare positive control group at sixteen weeks (p<0.001). Mean Corpuscular Volume (MCV) was increased in the treatment group's compare positive control group at sixteen weeks (p<0.01) Conclusions. Our findings demonstrate that vitamin B12 deficiency decreased hemoglobin and increased MCV.

Abstract

One of the most often utilized measures in calculating body proportion criterion is the BMI. The goal of this study is to see if there is a link between BMI and glucose levels in students who are given virtual physical activity. A high sedentary lifestyle is thought to be linked to the high prevalence of metabolic disorders among students. A rise in BMI or waist circumference levels indicates that a person may be at risk for a variety of metabolic disorders as a result of body mass index. An experimental study was utilized to investigate if there was a difference in the respondents' body mass index and waist circumference before and after the intervention. Before and after the treatment, there were significant variations in waist circumference (76.921.51 vs 74.651.47 cm, p0.001) and BMI (23.540.76 vs 23.050.77 cm, p0.001). Furthermore, with a value of r=0.895 and p0.001, there is a high link between BMI and waist circumference.

Abstract

Background: Desa Tambakbayah penyumbang kasus gizi buruk terbanyak di Kecamatan Cibadak, dengan 46 warganya mengalami masalah gizi. Situasi ini akan semakin parah pada tahun 2023, dimana data ePPGBM mencatat adanya peningkatan angka gizi buruk dan stunting. Sekitar 29.867 jiwa hidup di bawah garis kemiskinan ekstrem dengan pendapatan rumah tangga kurang dari Rp 600.000 per bulan. Tujuan dari kegiatan ini adalah untuk mendorong kemandirian kesehatan masyarakat Desa Tambakbayah. Metode: melalui serangkaian kegiatan sosialisasi dan pelatihan untuk meningkatkan pengetahuan dan keterampilan berwirausaha. Hasil: Hasil kegiatan PkM menunjukkan peningkatan pengetahuan peserta mengenai pengetahuan tentang stunting dan pemasaran digital. Dalam kegiatan PkM ini, peserta juga diperkenalkan dengan aplikasi Cek Stunting. Aplikasi ini dapat membantu masyarakat dan petugas kesehatan dalam melakukan deteksi dini terhadap risiko stunting pada anak. Aplikasi ini memberikan informasi mengenai nutrisi yang tepat untuk mencegah stunting, serta memantau tumbuh kembang anak secara berkala. Aplikasi ini juga memberikan rekomendasi dan edukasi kepada orang tua tentang cara mencegah dan mengatasi stunting.  Aplikasi Cek Stunting diharapkan dapat meningkatkan kesadaran masyarakat akan pentingnya pencegahan stunting. Kesimpulan: Sebagai upaya meningkatkan perekonomian keluarga peserta, mereka telah mendapatkan peralatan produksi keripik pisang, serta mendapatkan pelatihan teknis penggunaan peralatan yang baru diterima. Peserta juga dapat berlatih menggunakan alat dalam memproduksi keripik pisang dengan berbagai rasa. Peserta merasakan peningkatan pengetahuan mengenai stunting dan pemasaran digital. Selain itu, peserta merasakan peningkatan keterampilan dalam memproduksi keripik pisang.

Abstract

ntroduction The Thousand Islands, a popular tourist destination in DKI Jakarta Province, is facing a new challenge, namely the increasing concern about the transmission of Sexually Transmitted Infections (STIs) triggered by the increase in tourism visits. In response to this issue, a community service activity was carried out by the Faculty of Medicine at UHAMKA. Objective The aim of this activity is to raise awareness and knowledge in the community, particularly among educators in the Thousand Islands, about the dangers of and prevention against STIs and the importance of Reproductive Health Education. Method The partners in this community service activity are teachers in the Thousand Islands. A total of 30 teachers were involved in this training. The methods used involve training and counseling that incorporate direct training and open discussions. Data were collected through observations and field notes. Results The results of this activity include an increase in understanding and skills of teachers concerning Reproductive Health Education and STI prevention, which they can apply in educating students. Additionally, it was found that community awareness about the dangers of STIs also increased as a direct result of this activity. Conclusion The conclusion that can be derived from this activity is that appropriate education and training on reproductive health and the prevention of STIs for the community, specifically educators, can be an effective strategy in controlling STI transmission in tourist areas like the Thousand Islands. This activity also helps to strengthen the local health system in dealing with the challenge of STI transmission.

Abstract

Connexin-36 (Cx36) merupakan salah satu protein kanal antar sel. Protein ini paling banyak ditemukan pada otak. Gen Cx36 mengekspresikan protein penghubung Cx36 yang membentuk sinaps listrik. Penelitian ini bertujuan untuk memperoleh desain primer terbaik untuk menganalisis ekspresi Cx36 pada otak tikus dengan metode Quantitative Real Time PCR (qRT-PCR). Jenis penelitian ini adalah penelitian true experimental menggunakan mRNA dari 4 otak tikus. Analisis in silico menggunakan tools Primer 3 dan OlygoAnalizerTM by IDTdengan menggunakan database gen pada NCBI. Tiga primer dengan kriteria yang paling optimum kemudian dievaluasi dengan metode qRT-PCR. Berdasarkan hasil penelitian ketiganya mampu mengamplifikasi gen Cx36 dengan menggunakan qRT-PCR. Hasil primer terbaik menunjukkan melt curve tunggal pada primer no.5 gen Cx36 pada kisaran Tm 80-81oC. Hasil perbandingan uji in silico primer Cx36 yang sudah pernah dipublikasi sebelumnya menunjukkan bahwa primer no. 5 juga memiliki nilai optimal pada setiap kriteria. Maka hasil desain primer yang memiliki spesifisitas dan efektifitas yang optimum untuk mengamplifikasi gen Cx36 pada otak tikus Rattus norvegicus ialah primer no.5 (F: 5’-ATTTCCCGCTTCTACATCATCCAAG-3’ dan R: 5’-CACAGCAAACATGAACACCAGAAAG-3’).
