Abstract

Abstract

Stunting is a significant global public health problem caused by long-term dietary deficits that affect many children worldwide. Both environmental and genetic factors, including variants in the GHSR gene, play a crucial role in stunted growth. This study used an artificial recombinant plasmid DNA method to evaluate two primer set combinations for identifying DNA variants in the GHSR gene. Selecting suitable primer sets for identifying GHSR genetic variants linked to stunting is essential, as evidenced by PCR and sequencing techniques. The target gene, based on the GHSR reference sequence, consists of eight DNA variations (ΔQ36, G57G, P108L, L118L, R159R, C173R, D246A, and A277P). A recombinant plasmid was created by inserting a 1000 bp fragment of the GHSR gene into the pUC57 backbone. Primer sets were chosen based on their capacity to amplify these eight genetic variations and were optimized and validated using PCR methods. PCR and bi-directional sequencing verified the existence of surrounding DNA and specific single nucleotide variants (SNVs). In our study, we discovered four changes in the DNA sequence (R159R G>A, C173R T>C, D246A A>C, and A277P G>C) using the E1_F2/E1_R3 primer pair. Additionally, a new combination of primers (E1_F1/E1_R3) effectively detected seven DNA sequence mutations (ΔQ36 del CAG, G57G C>T, P108L C>T, L118L C>T, R159R G>A, C173R T>C, and D246A A>C). We have developed a new combination of forward and reverse primers to identify seven SNVs in the GHSR gene, which could serve as a diagnostic tool in clinical laboratory settings.

Abstract

Stunting, one of the malnutrition in the prenatal period up to the first two years of life and often exacerbated by recurrent gastrointestinal infections after birth. This study explores from various databases including Pubmed, Scopus, Science Direct, Sage Journal, Springer Link, with keywords: "stunting, gut microbiota, toddler". In this study, 10 open access articles published in 2016-2024 were used as sources of information using PRISMA 2020 guideline. It was found that the presence of gut microbiota plays a role in stunting. Gastrointestinal infections have specific mechanisms that can lead to impaired growth and postnatal growth. Deficiency of non-pathogenic gut microbiota is considered to be the main cause. Changes in gut bacterial composition in children under five are associated with increased gastrointestinal frequency, which in turn leads to chronic growth disorders.

Abstract

Anemia is a condition of the body when the hemoglobin (Hb) level is lower than the normal level or less than 11 gr/dl. In pregnant women, anemia has a high influence on the quality of human resources and can endanger the safety of the mother and child. Anemia has various causes, including lack of knowledge about anemia, foods containing iron and Fe tablet supplements. This research aims to determine the effectiveness of nutritional education media calendars, animated videos, and live Facebook in increasing knowledge and attitudes towards preventing anemia in pregnant women in the Cilincing District area. The method used in this research is quasi experimental with a pre-posttest with control group design. The data obtained was then analyzed and processed using Shapiro Wilk normality followed by the paired sample t test for the knowledge variable because the data was normally distributed and the Wilcoxon test for the attitude variable because the data was not normally distributed. The number of respondents who took part in this research was 45 respondents from pregnant women in the Cilincing District, North Jakarta. The results of this study showed that there were significant differences in the level of knowledge and attitudes of pregnant women towards preventing anemia in the three media provided with a p-value of 0.00 (p<0.05).

Abstract

Wasting is still a major nutritional problem in Indonesia with a prevalence of 8.5%, exceeding the WHO threshold of 5%. Household food security plays an important role in preventing wasting, especially in areas with high economic vulnerability such as Cipicung and Sukatani Villages. This study aims to analyze the relationship between household food security and the incidence of wasting in toddlers. This study was an observational analytical study with a cross-sectional design involving 110 toddlers aged 6–59 months. Data were collected through anthropometric measurements and the Food Insecurity Experience Scale (FIES) questionnaire. The results showed that 93.6% of families were in the category of severe food security, and as many as 13.6% of toddlers experienced wasting. Bivariate analysis using gamma correlation test showed a significant relationship between food security and wasting with a value of p = 0.014. Poor food security increases the risk of wasting due to limited access to nutritious food. The conclusion of this study is that household food security has a significant relationship with the incidence of wasting in toddlers, so there is a need for interventions that focus on improving food security at the community level.

Abstract

The aim of the study is to perform a systematic review and meta-analysis to determine whether the Fat Mass and Obesity-associated (FTO) polymorphisms influenced obesity and which Single Nucleotide Polymorphisms (SNPs) raised the risk of obesity. We used PUBMED, SCOPUS, and Web of Science to identify studies investigating the association between genetic polymorphisms in the FTO gene and obesity. We discovered 16 studies that included 4,381 obese people and 5,652 healthy control people. Five genetic polymorphisms showed significant association with increased obesity risk in the allelic model: rs9939609 (OR=1.38, 95% CI:1.27−1.50: I²=0%, p<0.001), rs8050136 (OR=1.23, 95% CI:1.03−1.46: I²=54%, p=0.02), rs3751812 (OR=1.31, 95% CI:1.18−1.45: I²=4%, p<0.001), rs1421085 (OR=1.29, 95% CI: 1.14−1.46: I²=4%, p<0.001) and rs1121980 (OR=1.45, 95% CI: 1.10−1.93: I²=66%, p=0.009). For the allelic model, no significant correlation was identified in the rs15588902 SNP FTO gene (OR=1.24, 95% CI:0.96−1.62: I²=62%, p=0.11). Therefore, this meta-analysis found six FTO SNPs linked to human FTO polymorphisms and the risk of obesity. Further research on the FTO gene and obesity should consider gene-gene and gene-environment interactions.

Abstract

At the end of data collection, a health survey supervisor aged 26 years old with normal body temperature conducted a PCR swab test as a research protocol. He already felt unwell three days before the PCR swab test. The test showed positive for COVID-19. He self-isolated and consumed nutritional supplements, including B vitamins, vitamin C, vitamin D, zinc, propolis, and probiotics. This case found persistent tonsillitis with odynophagia as the primary symptom. Other symptoms like sore throat, hot sensation in feet and hands, headache and mucus production behind the nose appeared only for one or two days. No fever, dry cough, and diarrhea were found. On day 8 of confirmation, the PCR swab test showed a negative result. Even several days after the negative result, an enlarged tonsil was still observed, but without odynophagia. We propose that persistent tonsillitis should be regarded as a new symptom of COVID-19, and immuno-nutrition supports the healing process in a patient with a history of tonsillitis.

Abstract

Obesity is a global pandemic that is closely related to the emergence of non-communicable diseases and premature death. Although obesity results from excess energy intake, deficiency of other nutrients are commonly found. Therefore, the use of the term overnutrition to describe obesity creates misperceptions. The objective of this review was to provide the evidence of a nutritional deficiency condition in obesity that can be used as a basis for considering the use of the term in Indonesia. The review was performed with unstructured literature search on several databases, such as PubMed, Scopus, ScienceDirect, and Google Scholar. Although obesity is caused by excess intake of macronutrients, the results of this review showed that many studies reported micronutrient deficiencies were present in people with obesity. The deficiencies were commonly reported, including deficiency of vitamin D, vitamin B12, folate, iron, calcium, and phosphor. The main causes were the habit of consuming food with a high content of macronutrients, but low in micronutrients, and metabolic consequences of body fat accumulation. Moreover, the deficiencies of micronutrients contributed to an increased risk of diseases, such as diabetes, cardiovascular disease and cancer. Meanwhile, the Global Nutrition Report has not used the term overnutrition, instead to use overweight and obesity. Therefore, the use of the term in Indonesia to describe obesity needs to be further reviewed. However as this is a narrative review, more comprehensive and systematic review is still needed.
